Polymer chains of (13)--d-glucan were dissolved with 1 M NaOH at 4° C from native microfibrillar protoplast nets. The chains associated into microfibrils during NaOH neutralization or dialysis. In contrast to the native microfibrils which are of uniform width individually (10 to 20 nm) and arranged in flat bundles, the microfibrils formed in vitro showed no band formation and consisted of fibrous spindle-shaped subunits of variable width or loose elementary fibrils about 1.7 nm wide. X-ray diagrams of native nets indicated a fairly high crystallinity and were different for wet and dry specimens. They corresponded to those of paramylon. Precipitated glucans produced diagrams different from the former and revealing a lower crystallinity especially with the dry samples.The X-ray pattern, combined with other data, allowed the precipitated microfibrils to be identified as aggregates of molecular strands composed each of three intertwined helical glucan chains. Since these triple helical chains are about 1.7 nm wide the elementary fibrils of this width can represent only single triple-helical strands. These helices have 7 glucose residues per turn and therefore a low symmetry which explains the poor crystallizing properties. The 7 membered helix represents a basic difference with the well crystallized native glucan which is built of highly symmetrical triple helices with 6 glucose residues per turn. Since 61 helical conformation is not formed in vitro at normal temperatures its generation in vivo must be due to the action of synthesizing enzymes at the protoplast membrane. The intertwining of these helices and crystallization of the strands are determined by their symmetry and physical properties of the chains. This characterizes the native microfibrils as products of self-assembly of enzymegenerated 61 helices.  相似文献

In order to demonstrate in detail the relationship between the longevity and productivity of leaves within a canopy, a new life table approach, the ‘bioeconomic life table’, was applied to the leaves of kidney bean plants (Phaseolus vulgaris L.) in relation to planting density and their position within the canopy. The net photosynthetic rate for upper leaves under full daylight tended to decline gradually due to leaf senescence from about 20 days after leaf emergence, and for the lower leaves the decrease was very rapid due to both shading and senescence about 10 days after emergence. Analysis of the survivorship curves and daily surplus production of leaves suggested that the lower and middle leaves, especially the latter, survived without surplus production of dry matter after they had reached mean longevity, and while the upper leaves at high density had a much shorter mean longevity, they had very large values of daily surplus production throughout the survival period. For the total foliage, the summed value of accumulated surplus production during the survival period was about five times as large as the summed value of the dry weight of the dead leaves, regardless of planting density. The daily rate of canopy leaf respiration was almost proportional to that of canopy gross photosynthesis for the various leaf area indices of the canopy, so that there was no optimum leaf area index that maximized canopy daily surplus production.  相似文献

Skeletonema costatum (Grev.) Cl. exhibits an asexual means, of increasing cell size that is more common than auxosporulation. Summary We describe the in vitro influence of 3,5,3′-triiodo-l-thyronine (T3),l-thyroxine (T4), a thyroid-stimulating hormone (TSH), and/or estradiol (E2: chosen as the control of the methodology) on the cell kinetics (cell distribution in the S+G2+M phases) of mouse MXT and human MCF-7 mammary cancer cells. Experiments were performed by means of a cell image processor, analyzing MCF-7 or MXT cells that had been grown on glass cover slips and whose nuclei had been stained by the Feulgen reaction, which is selective and quantitative (stoichiometric) with respect to DNA. We show that T3, T4, and TSH at 0.01 μM dramatically stimulate the cell kinetics of the MXT mouse and the MCF-7 human mammary cancer cell lines. Indeed, the three hormones bring about a significant transient increase in the S+G2+M fraction as does E2. Furthermore, our data indicate that E2 and TSH are antagonistic with regards to MXT or MCF-7 cell kinetics. This investigation assessed the efficacy of a biobehavioral intervention in the adjunctive treatment of mild pregnancy-induced hypertension (PIH), a potentially serious complication of pregnancy in which normotensive women develop hypertension, proteinuria, and edema of unknown etiology late in gestation. Forty-five women with symptoms of PIH were randomly assigned to one of three treatment conditions: bed rest alone (the most common obstetrical treatment), bed rest with individualized compliance enhancement training, or a four-session biobehavioral treatment consisting of bed rest, compliance enhancement training, and individualized thermal biofeedback-assisted relaxation training. Results indicated that while blood pressure for the bed rest and compliance enhancement groups continued to rise and pose an increasing health risk to maternal and fetal well-being, subjects in the biobehavioral group maintained their blood pressure at a significantly lower, and presumably safer, level. The biobehavioral treatment is hypothesized to affect blood pressure levels in subjects with mild PIH through the mediation of the sympathetic nervous system, decreasing peripheral vascular resistance and cardiac output. The results of this investigation suggest that the biobehavioral intervention may be an effective adjunct to bed rest in the treatment of mild PIH remote from term.  相似文献

Morphology and life history of 3 cephaline gregarines found in the gut of the pest of stored fruit, Oryzaephilus mercator. are described. Of these 3, 2 are new species. The 3 species are (1) Hirmocystis minuta (Ishii, 1914) (LP TL = 1:7 – 1:22, WP/WD = 1:1, 6–1:7); (2) Amsotobus indicus n. sp. (LP/TL = 1:3–1:6, WP/WD = 1:1 – 1:1.3); (3) Leidyana oryzaephili n. sp.
