Mitochondria are essential organelles because of their function in energy conservation. Here, we show an involvement of mitochondria in phytochrome‐dependent light sensing in fungi. Phytochrome photoreceptors are found in plants, bacteria, and fungi and contain a linear, heme‐derived tetrapyrrole as chromophore. Linearization of heme requires heme oxygenases (HOs) which reside inside chloroplasts in planta. Despite the poor degree of conservation of HOs, we identified two candidates in the fungus Alternaria alternata. Deletion of either one phenocopied phytochrome deletion. The two enzymes had a cooperative effect and physically interacted with phytochrome, suggesting metabolon formation. The metabolon was attached to the surface of mitochondria with a C‐terminal anchor (CTA) sequence in HoxA. The CTA was necessary and sufficient for mitochondrial targeting. The affinity of phytochrome apoprotein to HoxA was 57,000‐fold higher than the affinity of the holoprotein, suggesting a “kiss‐and‐go” mechanism for chromophore loading and a function of mitochondria as assembly platforms for functional phytochrome. Hence, two alternative approaches for chromophore biosynthesis and insertion into phytochrome evolved in plants and fungi.  相似文献

来源于水母Aequorea victoria的绿色荧光蛋白(green fluorescent protein, GFP)现已成为在生物化学和细胞生物学中研究和开发应用得最广泛的蛋白质之一. 其内源荧光基团在受到紫外光或蓝光激发时(λmax=395 nm, 小峰在479 nm)可高效发射清晰可见的绿光. GFP的高分辨率晶体结构为了解和研究蛋白质结构和光谱学功能关系提供了一个极好的机会. GFP已成为一个监测在完整细胞和组织内基因表达和蛋白质定位的理想标记. 通过突变和蛋白质工程构建的GFP嵌合蛋白在生理指示剂、生物传感器、光化学领域以及生产发光纤维等方面展示了广阔前景.  相似文献

The red fluorescent protein KillerRed, engineered from the hydrozoan chromoprotein anm2CP, has been reported to induce strong cytotoxicity through the chromophore assisted light inactivation (CALI) effect. Here, we present the X-ray structures of KillerRed in its native and bleached states. A long water-filled channel is revealed, connecting the methylene bridge of the chromophore to the solvent. This channel facilitates the transit of oxygen and of reactive oxygen species (ROS) formed by reaction with the excited chromophore. The functional roles of key mutations used to produce KillerRed are discussed, strong chromophore distortions in the bleached state are revealed, and mechanisms for ROS production and self protection are proposed. The presence of a partially mature, photo-resistant, green-emitting state is characterized, which accounts for enhanced CALI by “pre-bleached” KillerRed.  相似文献

Summary Copper-deficient cells ofPseudomonas stutzeri strain ZoBell synthesize catalytically inactive nitrous oxide (N2O) reductase which is activated by added Cu(II) in the absence of de novo protein synthesis. The apparentK m for the activation process is 0.13 M. Activation is temperature-dependent and is inhibited by Cd(II)(K i 1.27 M) and less strongly by Zn(II), Ni(II), and Co(II). The same metal ions at 20 M have little or no effect on N2O reduction of intact cells. Apo-N2O reductase of transposon Tn5-inducednos mutants with defective Cu-chromophore biosynthesis is not reactivated by Cu(II). N2O reductase of Cu-sufficient and Cu-deficient wild type, and ofnos mutants is localized in the periplasm, the latter providing the likely site of metal incorporation into the apoenzyme.  相似文献

During the visual process, light absorption in the 11-cis retinylidene chromophore leads to a rapid cis-trans-isomerization which initiates the phototransduction step. Important spectroscopic properties of this chromophore can be derived from polarized UV-absorption spectra of crystalline 11-cis-retinal if a parallel X-ray structure analysis is performed. Several questions about the relation between molecular geometry and spectroscopic behavior could not be answered from these spectra. All crystal forms of 11-cis-retinal contain this molecule in its 6-s-cis-ring conformation. For the retinal homologue, -8-apocarotenal (APC), however, two crystal forms with different ring conformation can be grown. The spectrum of -APC (6-s-cis) shows a vibronic structure whereas that of -APC (6-s-trans) is diffuse but has a distinct shoulder on the low energy side of the main band. This S-band is typical for retinal spectra and has been ascribed to a transition into a 1A g -* -state. The appearance of the S-band is not correlated with a 6-s-cis-conformation as suggested by the retinal spectra but is due to intermolecular interactions: -APC has a dense dimer packing and a strong electrostatic interaction between the -electron systems. This might cause the forbidden 1A g -* -transition. On the other hand, this interaction is missing in the loose and polar packing of -APC which favors vibration in the polyene chain. Following UV mutagenesis of protonemal tissue of the moss Ceratodon purpureus we have isolated different aphototropic mutant lines that can be divided into two distinct classes. One class, represented by the line ptr1, shows characteristic features of phytochrome chromophore deficiency. ptrl shows negligible photoreversibility (<5% of wild type), whereas immunoblots show normal apoprotein levels. The aphototropic phenotype could be partially restored with biliverdin, a precursor of the phytochrome chromophore. It was found that, whereas in wild type formation of Pfr leads to suppression of gravitropism, there is no such suppression ptrl. In addition, ptr1 shows lower chlorophyll levels than the wild type. These findings indicate that, as expected for a chromophore-deficient mutant, multiple phytochrome effects are lost. The other class of mutants, represented by the line ptr103, shows more specific effects. In this mutant, only phototropism is affected. Suppression of gravitropism, the content of chlorophyll and photoreversibility of phytochrome were similar to those of the wild type.  相似文献

Photoreceptor proteins serve as efficient nano-machines for the photoenergy conversion and the photosignal transduction of living organisms. For instance, the photoactive yellow protein derived from a halophilic bacterium has the p-coumaric acid chromophore, which undergoes an ultrafast photoisomerization reaction after light illumination. To understand the structure-function relationship at the atomic level, we used a computational method to find functionally important atoms for the photoisomerization reaction of the photoactive yellow protein. In the present study, a "direct" measure of the functional significance was quantitatively evaluated for each atom by calculating the partial atomic driving force for the photoisomerization reaction. As a result, we revealed the reaction mechanism in which the specific role of each functionally important atom has been well characterized in a systematic manner. In addition, we observed that this mechanism is strongly conserved during the thermal fluctuation of the photoactive yellow protein. We compared the experimental data of fluorescence decay constant of several different mutants and the present analysis. As a result, we found that the reaction rate constant is decreased when a large positive driving force is missing.  相似文献

The synthesis of ortho-substituted (S)-1-phenethylamines via ortho-lithiation of its N,N-dimethyl derivative followed by reactions with different reagents is described. Circular dichroism spectra of synthesized compounds were measured. It is shown that the observed short-wave Cotton effects greatly depend on the existence of cyclic structure due to possible interactions such as hydrogen bond or electrostatic interaction.  相似文献

Bryophytes, which include mosses, liverworts and hornworts, indeed play a significant role in the natural dynamics of native forests. They contribute to fundamental ecological processes such as tree regeneration. In this context, the objective was to analyse the association of bryophytes with Nothofagus seedlings, as well as whether the substrates where bryophytes grew influenced bryophyte-tree seedling association, in sub-Antarctic forests at two contrasting landscapes (coastal and mountain) of Tierra del Fuego (Argentina), to better understand their impacts on the natural regeneration processes. We hypothesized that bryophytes act as a substrate for seed germination and initial regeneration growth in pure and mixed forests of N. pumilio and N. betuloides, resulting in widespread tree seedlings–bryophyte associations in all landscapes. In each forest and landscape location, 60 transects were established (10 m in length) to evaluate bryophyte cover (by point intercept method) and substrate type (bare soil, decaying wood and litter cover) where they were growing. An adaptation of the relative interaction index (RII) based on tree seedling cover associated with bryophyte species (%) and tree seedling cover in the absence of bryophytes (%) was calculated and analysed to assess competition or facilitation between bryophytes and tree seedlings. Nothofagus pumilio seedlings were less abundant in bryophytes compared to other substrates, suggesting an inhibitory effect on the germination and/or survival of N. pumilio. In contrast, the seedlings of N. betuloides, in both pure and mixed forests, exhibited higher abundance on bryophyte substrates, particularly in mosses at mountain landscape (RII = 0.83 ± 0.08 in litter and 0.62 ± 0.11 in decaying wood). These findings suggest that bryophytes play a facilitating role for N. betuloides seedlings during germination and initial phases of growth, mainly in the mountain. Therefore, their conservation promotes N. betuloides forest continuity, both in pure and mixed structures.  相似文献
