迁移的鼻咽癌细胞容积激活性氯电流 总被引:6,自引:1,他引:5
用膜片钳技术研究了Transwell小室趋化迁移后的鼻咽癌CNE-2Z细胞容积激活性CT电流。47%低渗刺激迁移后的CNE-2Z细胞诱发容积激活性氯电流,与未迁移细胞相比,其特性以及其对氯通道阻断剂的敏感性发生明显的变化,此电流的密度明显高于未迁移细胞,而且该电流几乎完全被氯通道阻断剂adenosine-5'-triphosphate(ATP,10 mmol/L)、5-nitro-2-3-phenylpropylamino benzoic acid(NPPB,100μmol/L)和他莫昔芬(30μmol/L)抑制,其中NPPB和他莫昔芬对迁移细胞的抑制作用明显强于未迁移细胞。迁移后的CNE-2Z细胞容积激活性氯通道对阴离子的通透性为:Br>Cl>I>葡萄糖酸,与未迁移细胞(I>Br>Cl>葡萄糖酸)不同。结果提示,容积激活性氯通道可能参与CNE-2Z细胞的迁移过程。 相似文献

采用全细胞膜片钳技术和细胞容积测量技术,在低分化鼻咽癌细胞株CNE-2Z上观察ATP 诱导的Cl- 电流的特性及其对细胞容积的影响。细胞外微摩尔水平的ATP 以剂量依赖性的方式激活一个具有弱外向整流特性,没有时间依赖性失活的电流,此电流的反转电位 [(-0.05 ± 0.03) mV]接近Cl- 的平衡电位(-0.9 mV)。用葡萄糖酸置换细胞外液Cl- 后, ATP 激活的电流明显减小并且反转电位发生改变。氯通道抑制剂NPPB (200 μmol/L)可以抑制这一电流 [(81.03 ± 9.3)%] 。此电流亦可被嘌呤受体(P2Y) 拮抗剂反应蓝 2 抑制 [(67.39 ± 5.06)%]。50 μmol/L 的 ATP 使在等渗状态下的细胞容积缩小, 替代和耗竭细胞外、内的Cl- 后, ATP 的这一作用消失。这些结果提示细胞外微摩尔水平的 ATP 可通过兴奋 P2Y 受体激活氯通道而产生与细胞容积调节相关的Cl- 电流。 相似文献

Nitryl chloride, formed by reaction of hypochlorous acid with nitrite, might contribute to nitrative damage of biomolecules in addition to peroxynitrite. Damage of DNA by these reactive nitrogen oxide species is implicated in carcinogenesis associated with chronic infections and inflammation. Nitrated DNA adducts, such as 8-nitroguanine and 8-nitroxanthine, are not stable in DNA since they undergo spontaneous depurination, leading to apurinic site formation. In this report, we investigate the protective effect of biological and dietary antioxidants in inhibiting DNA nitration induced by nitryl chloride. The effect of inhibition was evaluated by decrease of 8-nitroxanthine and 8-nitroguanine formation. Among the 21 compounds examined, dihydrolipoic acid is the most effective in preventing DNA nitration, followed by N-acetyl-L-cysteine and folic acid. For sulfur-containing compounds, the more highly reduced compounds are stronger inhibitors of DNA nitration. The major product of N-acetyl-L-cysteine reaction with nitryl chloride is characterized as the (R)-2-acetylamino-3-sulfopropionic acid, a physiologically irreversible product, suggesting that nitryl chloride is a strong oxidizing agent. 相似文献

One year old, individually tagged Lake Inari Arctic charr, Salvelinus alpinus, were reared at three constant temperatures, 10.3°C, 14.1°C and 18.1°C, over four weeks. Blood samples were collected from a group of unstressed fish after the cultivation period at the same time as another group of fish were subjected to acute handling stress treatment (2min netting in air and 40min (± 20min) recovery period in water). Plasma cortisol, calcium, sodium, potassium and chloride concentrations were measured on both groups. To study the effect of minor daily temperature fluctuations on the stress response of Arctic charr, two additional daily fluctuating temperature (14 ± 1°C, 18 ± 1°C) treatments were established. The samples were taken in the same manner as those in the constant temperature treatments. Growth was fastest at 10.3–14.1°C and clearly lower at 18.1°C. Pre-stress plasma cortisol levels were low but increased slightly with increasing temperature. After stressor treatment, the cortisol concentrations of Arctic charr were clearly higher in all temperature treatments but there were no significant differences in plasma cortisol concentrations among temperatures. Plasma calcium levels increased during the stress treatment but temperature did not modulate this effect. The plasma potassium concentrations declined at 14.1–18.1°C after acute stress but the response was not affected by temperature within this range. The concentrations of sodium and chloride were unaffected by acute stress. Temperatures of 10.3–18.1°C and fluctuating temperature treatments had no influence on any plasma ion concentrations. Arctic charr were able to maintain the plasma ion concentrations in fresh water at 10.3–18.1°C and after acute stress treatment. Results indicate that the optimum temperature for growth of Arctic charr has little to do with the plasma ion concentrations or the ability to maintain those concentrations after short-term stress. The plasma cortisol responses further indicate that the optimum temperature for growth of Arctic charr is not related to the suppressed ability to react to an acute handling stressor. This study (1) analyzed fetal development of mouse embryos after oocyte cryopreservation in CJ2, a choline-based medium, (2) examined the effect of culture duration in vitro on subsequent fetal development, and (3) compared survival and fetal development of zygotes frozen in embryo transfer freeze medium (ETFM; sodium-based medium) or CJ2. Unfertilized oocytes and zygotes were cryopreserved using a slow-cooling protocol. After thawing, oocytes were inseminated after drilling a hole in their zona, cultured in vitro either to the two-cell or blastocyst stage, and transferred to the oviducts or uterine horns of recipient mice. In parallel experiments, frozen-thawed zygotes were similarly cultured and transferred. Implantation rates for transferred embryos were high (range 66-88%), regardless of whether they had been frozen as oocytes or zygotes and whether they had been transferred to the oviduct or uterus. However, fetal development was significantly higher when two-cell embryos were transferred. With blastocyst transfer, control embryos implanted and produced a greater proportion of fetuses than did oocytes frozen in CJ2, whereas transfer at the two-cell stage resulted in similar proportions of implantation sites and fetuses. Blastocyst transfer of zygotes cryopreserved in ETFM or CJ2 produced similar fetal development rates (23.6% vs 20.0%), but when frozen-thawed zygotes were transferred at the two-cell stage the fetal development rates were higher in the ETFM group (53.3%) than in the CJ2 group (32.0%). A high proportion (46.7%) of oocytes frozen in CJ2 in a nonprogrammable freezer and plunged at -20 degrees C developed into live offspring. This study shows that in the mouse (1) oocytes frozen in CJ2 can develop into viable fetuses, (2) prolonging culture in vitro has a detrimental effect on embryo transfer outcome, and (3) CJ2 offers no advantage for zygote cryopreservation. 相似文献

Recently we have reported that bilirubin UDP-glucuronosyltransferase (UGT1A1) is induced in rat liver by chronic ethanol treatment. Several studies have shown that Kupffer cells play a central role in the mediation of various hepatic effects of chronic alcohol consumption. In the present work, the participation of Kupffer cells in the ethanol dependent induction of UGT1A1 was investigated. A group of rats was pretreated with gadolinium chloride, a known Kupffer-cell-depleting agent. We compared the effect of chronic ethanol ingestion on UGT1A1 expression in the liver of normal and gadolinium chloride treated rats. The effect of ethanol on bilirubin glucuronidation was completely prevented in Kupffer cell deficient rats. The western and northern blot analyses showed that the increase of both the protein and mRNA of UGT1A1 was prevented in these animals. These results suggest that Kupffer cells play a major role in the mediation of ethanol-stimulated induction of UGT1A1 in liver parenchymal cells. 相似文献

Smooth muscle cells undergo substantial increases in length, passively stretching during increases in intraluminal pressure in vessels and hollow organs. Active contractile responses to counteract increased transmural pressure were first described almost a century ago (Bayliss, 1902) and several mechanisms have been advanced to explain this phenomenon. We report here that elongation of smooth muscle cells results in ryanodine receptor-mediated Ca(2+) release in individual myocytes. Mechanical elongation of isolated, single urinary bladder myocytes to approximately 120% of slack length (DeltaL = 20) evoked Ca(2+) release from intracellular stores in the form of single Ca(2+) sparks and propagated Ca(2+) waves. Ca(2+) release was not due to calcium-induced calcium release, as release was observed in Ca(2+)-free extracellular solution and when free Ca(2+) ions in the cytosol were strongly buffered to prevent increases in [Ca(2+)](i). Stretch-induced calcium release (SICR) was not affected by inhibition of InsP(3)R-mediated Ca(2+) release, but was completely blocked by ryanodine. Release occurred in the absence of previously reported stretch-activated currents; however, SICR evoked calcium-activated chloride currents in the form of transient inward currents, suggesting a regulatory mechanism for the generation of spontaneous currents in smooth muscle. SICR was also observed in individual myocytes during stretch of intact urinary bladder smooth muscle segments. Thus, longitudinal stretch of smooth muscle cells induces Ca(2+) release through gating of RYR. SICR may be an important component of the physiological response to increases in luminal pressure in smooth muscle tissues. 相似文献

The cystic fibrosis transmembrane conductance regulator (CFTR) is an ion channel that conducts Cl- current. We explored the CFTR pore by studying voltage-dependent blockade of the channel by two organic anions: glibenclamide and isethionate. To simplify the kinetic analysis, a CFTR mutant, K1250A-CFTR, was used because this mutant channel, once opened, can remain open for minutes. Dose-response relationships of both blockers follow a simple Michaelis-Menten function with K(d) values that differ by three orders of magnitude. Glibenclamide blocks CFTR from the intracellular side of the membrane with slow kinetics. Both the on and off rates of glibenclamide block are voltage dependent. Removing external Cl- increases affinity of glibenclamide due to a decrease of the off rate and an increase of the on rate, suggesting the presence of a Cl- binding site external to the glibenclamide binding site. Isethionate blocks the channel from the cytoplasmic side with fast kinetics, but has no measurable effect when applied extracellularly. Increasing the internal Cl- concentration reduces isethionate block without affecting its voltage dependence, suggesting that Cl- and isethionate compete for a binding site in the pore. The voltage dependence and external Cl- concentration dependence of isethionate block are nearly identical to those of glibenclamide block, suggesting that these two blockers may bind to a common binding site, an idea further supported by kinetic studies of blocking with glibenclamide/isethionate mixtures. By comparing the physical and chemical natures of these two blockers, we propose that CFTR channel has an asymmetric pore with a wide internal entrance and a deeply embedded blocker binding site where local charges as well as hydrophobic components determine the affinity of the blockers. 相似文献

The goal of this study was to calculate the average CO2 emissions for manufacturing three commodity plastics, polyethylene (PE), polypropylene (PP), and polyvinyl chloride (PVC)
in Japan. The CO2 emissions were calculated from cradle to gate, excluding the calcination processes after use. As the results, the followings
were observed:
1) The gross CO2 emissions for the manufacture of plastics in Japan were 1.3, 1.4, and 1.7 kg-CO2/kg-PE, PP, and PVC, respectively. These mainly reflected the difference of CO2 emissions for the in-house electricity generation. 2) The CO2 emissions for the electricity used for manufacturing PVC were higher than that used for PE and PP, because additional electricity
was required for the electrolysis to produce chlorine. The gross electricity consumption for manufacturing PVC was 1.3 kWh/kg-PVC,
and the other plastics consumed 0.5 kWh/kg-Products. In addition, the effects of energy saving were studied using a projected
gas-diffusion electrode for the electrolysis of salt on the reduction of CO2 emissions. It was estimated that the reduction in CO2 emissions was 7% compared with the present PVC manufacturing processes. 相似文献

Effect of Modulators of Protein Kinase C Activity on Ca2+ Transport in Retinal Rod Microsomes 总被引:1,自引:0,他引:1
The effect of modulators of protein kinase C (PKC) activity on Ca2+ translocation in retinal rod microsomes was studied. It is shown that PKC activators (phorbol 12-myristate-13-acetate (PMA) and diacylglycerol (DAG)) and inhibitors (chelerythrine chloride, polymyxin B, and phloretin) stimulate and inhibit ATP-dependent Ca2+ uptake in retinal rod microsomes, respectively. This effect is apparently due to an influence of PKC on Ca-ATPase contained in these vesicular structures. It was found that PKC inhibitors (chelerythrine chloride, polymyxin B, and phloretin) and activators (PMA and DAG) potentiate Ca2+ release from Ca2+ -loaded retinal rod microsomes. Specific and nonspecific mechanisms of Ca-release stimulation by the modulators of PKC activity are discussed. 相似文献
