The kinetic resolution of racemates constitutes one major route to manufacture optically pure compounds. The enzymatic kinetic resolution of (R,S)-1-phenylethanol over Candida antarctica lipase B (CALB) by using vinyl acetate as the acyl donor in the acylation reaction was chosen as model reaction. A systematic screening and optimization of the reaction parameters, such as enzyme, ionic liquid and substrates concentrations with respect to the final product concentration, were performed. The enantioselectivity of immobilized CALB commercial preparation, Novozym 435, was assayed in several ionic liquids as reaction media. In particular, three different ionic liquids: (i) 1-butyl-3-methylimidazolium hexafluorophosphate [bmim][PF6], (ii) 1-butyl-3-methylimidazolium tetrafluoroborate [bmim][BF4] and (iii) 1-ethyl-3-methylimidazolium triflimide [emim][NTf2] were tested. At 6.6% (w/w) of Novozym 435, dispersed in 9.520 M of [bmim][PF6] at 313.15 K, using an equimolar ratio of vinyl acetate/(R,S)-1-phenylethanol after 3 h of bioconversion, the highest possible conversion (50%) was reached with enantiomeric excess for substrate higher than 99%.  相似文献

The chiral separation of (±)‐catechin was investigated by capillary electrophoresis using characterized succinyl‐β‐cyclodextrins (Suc‐β‐CDs) with one to three degree of substitution values. The effects of nature and concentration of Suc‐β‐CDs and running buffer pH on the migration time and resolution of (±)‐catechin are discussed. All three kinds of Suc‐β‐CDs show a clear baseline separation of (±)‐catechin in capillary electrophoresis. Mono‐Suc‐β‐CD effectively separated (±)‐catechin, and additional substituted CDs (di‐ and tri‐Suc‐β‐CD) were capable of chiral separation at a broad pH range. The optimum running conditions were found to be 100 mM borate buffer (pH 9.8) containing 5 mM mono‐Suc‐β‐CD with no methanol organic modifier. Chirality, 2009. © 2009 Wiley‐Liss, Inc.  相似文献

ZnO nanowires were successfully introduced into a macroporous SiO2 by in situ hydrothermal growth in 3D pores. The obtained composites were characterized by SEM and XRD, and used as supports to immobilize Candida antarctica lipase B (CALB) through adsorption. The high specific surface area (233 m2/g) and strong electrostatic interaction resulted that the average loading amount of the composite supports (196.8 mg/g) was 3–4 times of that of macroporous SiO2 and approximate to that of a silica-based mesoporous material. Both adsorption capacity and the activity of the CALB immobilized on the composite supports almost kept unchanged as the samples were soaked in buffer solution for 48 h. The chiral resolution of 2-octanol was catalyzed by immobilized CALB. A maximum molar conversion of 49.1% was achieved with 99% enantiomeric excess of (R)-2-octanol acetate under the optimal condition: a reaction using 1.0 mol/L (R,S)-2-octanol, 2.0 mol/L vinyl acetate and 4.0 wt.% water content at 60 °C for 8 h. After fifteen recycles the immobilized lipase could retain 96.9% of relative activity and 93.8% of relative enantioselectivity.  相似文献

This article presents a research study on the deposition process of Ca2+ induced by Synechocystis sp. PCC6803 in BG11 liquid medium with different Ca2+ concentrations and different pH. The changes of Ca2+ concentrations were measured by using atomic absorption method and the corresponding dynamical models were studied. Minerals and cells were analyzed by high resolution transmission electron microscope, selected area electron diffraction, scanning electron microscope, energy dispersive X-ray spectroscope, X-ray diffraction. The selected area electron diffraction patterns were analyzed by Digital Micrograph 3.7 software. The result showed that Ca2+ concentrations decreased faster in the experimental group. The changes of calcium carbonate precipitation were fitting to an exponential model. PH 7 and Ca2+ concentration of 1.5 g/L were most conducive to calcium carbonate precipitation in the corresponding gradient range. The result of high-resolution transmission electron microscopy showed that minerals in the experimental group differed obviously from that of the control group in the surface morphology, but both of them were calcites. It also showed that a certain number of minute calcites adhesion to the outer surfaces of S. PCC6803 cells. The result of scanning electron microscopy displayed that many sunken holes emerged on the surfaces of the prismatic calcium carbonate minerals. The results of X-ray diffraction proved that minerals induced by S. PCC6803 were calcites with preferential orientation. This article discusses the process of carbonate formation and the possible role played by S. PCC6803. It may be useful to further study the mechanism of microbial carbonates deposition in the field of geology.  相似文献

The synthesis of Spi(τ‐dec), derived from the selective alkylation of L‐spinacine (4,5,6,7‐tetrahydro‐1H‐imidazo[4,5‐c]pyridine‐6‐carboxylic acid) at the τ‐nitrogen of its heteroaromatic ring, with a linear hydrocarbon chain of 10 carbon atoms, is described here for the first time. Spi(τ‐dec) was successfully employed in the past to prepare home‐made chiral columns for chiral ligand‐exchange high‐performance liquid chromatography. In the present article a new method is described, using Spi(τ‐dec) as a chiral selector in high‐performance thin‐layer chromatography (HPTLC): commercial hydrophobic plates were first coated with Spi(τ‐dec) and then treated with copper sulfate. The performance of this new chiral stationary phase was tested against racemic mixtures of aromatic amino acids, after appropriate optimization of both the conditions of preparation of the plates and the mobile phase composition. The enantioselectivity values obtained for the studied compounds were higher than those reported in the literature for similar systems. Five primer sets were designed in order to identify single nucleotide polymorphisms (SNPs) in middle and terminal exons (2 to 6) and in some flanking intronic regions of the bovine insulin-like growth factor 1 (IGF-1) gene. Sequencing results of PCR products for 10% of animals showed no variant in exons but a SNP at intron 4 was occurred. Both polymerase chain reaction–restriction fragment length polymorphism (PCR–RFLP) and high resolution melting (HRM) methods were developed to genotype samples. The PCR–RFLP results showed the presence of three fragments on agarose gel for the C allele due to two cleavage sites while two fragments for the T allele were observed. Melting curves of 123 bp fragments in HRM analysis showed a difference between temperature melting (Tm) of two homozygous genotypes as the CC genotypes had higher Tm than the TT genotypes. Melting curve of the CT genotype was different and crossed two parallel patterns of homozygous genotypes. The frequencies of the CC, CT and TT genotypes were 0.6, 0.37 and 0.03, respectively. Also, the estimated allele frequencies were 0.785 and 0.215 for the C and T alleles, respectively. Results showed higher accuracy of the HRM analysis compared to the PCR–RFLP method. Least square means (LSMs) comparison of the different genotypes in the SNP showed significant association with milk fat yield trait in the first lactation and open days after the second calving. The polymorphism did not have a significant effect on other milk production or reproduction traits. It seems that other variants or QTLs known in this region underlie genetic variation in the production and reproduction of dairy cattle.  相似文献
