应用与小麦第六同源群有关的分子和生化标记,包括DNA探针pSc5·3H3和pSR167以及同工酶Est-5和a-Amy-1,对来自六倍体小黑麦Beagle与普通小麦科冬58杂交后代F1花粉植株的抗白粉病株系M24.M09及M17进行了分析。结果表明,M24、M09及M17不同程度地含有黑麦染色体成分,而且电泳谱带差别较大,据此推断,M09为6RL的易位系。因此,生化和分子标记不仅可以用于确定外源片段的存在,而且可以帮助确定染色体组型和外源片段的位置  相似文献

食用菌因其显著的营养、保健、药用及综合利用价值受到世人青睐。原生质体融合技术则为食用菌的育种和遗传学研究提供了一条很好的途径。食用菌的种内、种间乃至属间、目间的原生质体融合均已有过报道,但这些工作大多利用营养缺陷型标记的亲株,采用化学融合法完成。这种融合的效率较低,对亲株的标记繁琐费时,甚至会对菌株性状产生不良影响。最近,一种具有较高融合效率的物理融合法——电融合法以及非遗传性标记法分别被一些研究者所采用。本研究将原生质体电融合法与灭活标记法相结合来获得食用菌的属间融合产物,并采用包括同工酶分析和RAPD(或AP-PCR)方法在内的分子生物学方法,对食用菌属间杂交后代的遗传学特性进行初步探讨。1 材料和方法  相似文献

The effects of different seed sizes, planting densities and planting patterns on the transmission of silver scurf (Helminthosporium solani) and black scurf (Rhizoctonia solani) diseases of potatoes were examined in five field experiments with cv. Estima in 1991–93.
In all experiments, silver scurf was more severe with increase in seed size and planting density. At high planting density, silver scurf was less severe from a square planting pattern than from planting in wide rows. The incidence of black scurf also tended to increase with increase in seed size and was increased markedly high planting density.  相似文献

Cell Tr2 is a neuron in the subesophageal ganglion of the leech that can trigger swim episodes. In this report, we describe the ability of Tr2 to terminate ongoing swim episodes as well as to trigger swimming. Stimulation of Tr2 terminated ongoing swim episodes in nearly every preparation tested, while Tr2 stimulation triggered swim episodes in only a minority of the preparations. We suggest that the primary role of Tr2 is in the termination rather than the initiation of swimming activity.The swim trigger neuron Tr3 and a swim-gating neuron, cell 21, hyperpolarized during Tr2-induced swim termination. Another swim-gating neuron, cell 204 was sometimes slightly excited, but more often, hyperpolarized during Tr2-induced swim termination. In contrast to these cells, Tr2 stimulation excited another swim-gating neuron, cell 61. The responses of the swimgating cells were variable in amplitude and sometimes not evident during Tr2-induced swim termination. Hence, the effects of Tr2 stimulation on swim-gating neurons seem unlikely to be the direct cause of swim termination.Oscillator cells examined during Tr2-induced swim termination include: 27, 28, 33, 60, 115, and 208. The largest effect seen in an oscillator neuron was in cell 208, which was repolarized by up to 10 mV during Tr2 stimulation. Tr2 stimulation did not produce any obvious synaptic effects in motor neurons DI-1, VI-1, and DE-3. Our findings indicate that other, yet undiscovered, connections are likely to be important in Tr2-induced swim termination. Neuronal peptides exert neurohormonal and neurotransmitter (neuromodulator) functions in the central nervous system (CNS). Besides these functions, a group of neuropeptides may have a capacity to create cell proliferation, growth, and survival. Axotomy induces transient (1–21 d) upregulation of synthesis and gene expression of neuropeptides, such as galanin, corticotropin releasing factor, dynorphin, calcitonin gene-related peptide, vasoactive intestinal polypeptide, cholecystokinin, angiotensin II, and neuropeptide Y. These neuropeptides are colocalized with “classic” neurotransmitters (acetylcholine, aspartate, glutamate) or neurohormones (vasopressin, oxytocin) that are downregulated by axotomy in the same neuronal cells. It is more likely that neuronal cells, in response to axotomy, increase expression of neuropeptides that promote their survival and regeneration, and may downregulate substances related to their transmitter or secretory activities.  相似文献

We have developed a biophysical model of a pair of reciprocally inhibitory interneurons comprising an elemental heartbeat oscillator of the leech. We incorporate various intrinsic and synaptic ionic currents based on voltage-clamp data. Synaptic transmission between the interneurons consists of both a graded and a spike-mediated component. By using maximal conductances as parameters, we have constructed a canonical model whose activity appears close to the real neurons. Oscillations in the model arise from interactions between synaptic and intrinsic currents. The inhibitory synaptic currents hyperpolarize the cell, resulting in activation of a hyperpolarization-activated inward currentI h and the removal of inactivation from regenerative inward currents. These inward currents depolarize the cell to produce spiking and inhibit the opposite cell. Spike-mediated IPSPs in the inhibited neuron cause inactivation of low-threshold Ca++ currents that are responsible for generating the graded synaptic inhibition in the opposite cell. Thus, although the model cells can potentially generate large graded IPSPs, synaptic inhibition during canonical oscillations is dominated by the spike-mediated component.  相似文献

In the mid-western Himalaya (altitude 1350 m, rainfall 1100 mm), multipurpose trees found as escapees in agricultural fields or naturally growing in the forests, play an important role in providing fuel, fooder and small timber to the farmers. Shoot elogation, and tree architecture of 4 year old trees of Grewia optiva, Robinia pseudoacacia and Celtis australis (early successionals), and Quercus leucotrichophora, Q. glauca and Ilex odorata (late successionals), were analyzed. All the late successional species showed a proleptic type of bud and branch production, while the early successional trees made growth through syllepsis. The shoot elongation differed significantly (P <0.05) with the crown position, and ranged from 11 to 30 cm in different species. Early successional species tended to maintain a comparatively narrow crown and showed a significantly (P <0.05) higher ramification ratio, and multilayered canopy. The late successionals, in contrast, showed a wide crown with monolayered canopy, adapted to the weak light intensity. There was only one flush of leaves in Q. leucotrichophora and Q. glauca while in the rest of the species there were two distinct flush periods. The results are important for the management of agroforestry trees.  相似文献

The Drosophila neuromusculin (nrm) gene encodes an immunoglobulin-like (Ig-like) cell adhesion molecule expressed in the precursors of the embryonic peripheral nervous system (PNS), in the midline precursors of the central nervous system (CNS), and in muscles. During the initial phases of CNS axonogenesis, nrm is expressed in cells involved in the development of commissures and longitudinal tracts. Mutations which alter expression of nrm mRNAs cause aberrant development of commissures and longitudinal axon pathways. Defects in the PNS and muscles of nrm mutants are also observed. In most nrm embryos, abnormal development can be detected in a subset of abdominal segments; however, in approximately 1 of 10 nrm embryos, the defects extend to all segments. Herein, we present evidence that nrm plays an important role in early morphogenesis, possibly by mediating or facilitating inductive cell contacts and movements.  相似文献

Type II antifreeze proteins (AFP), which inhibit the growth of seed ice crystals in the blood of certain fishes (sea raven, herring, and smelt), are the largest known fish AFPs and the only class for which detailed structural information is not yet available. However, a sequence homology has been recognized between these proteins and the carbohydrate recognition domain of C-type lectins. The structure of this domain from rat mannose-binding protein (MBP-A) has been solved by X-ray crystallography (Weis WI, Drickamer K, Hendrickson WA, 1992, Nature 360:127-134) and provided the coordinates for constructing the three-dimensional model of the 129-amino acid Type II AFP from sea raven, to which it shows 19% sequence identity. Multiple sequence alignments between Type II AFPs, pancreatic stone protein, MBP-A, and as many as 50 carbohydrate-recognition domain sequences from various lectins were performed to determine reliably aligned sequence regions. Successive molecular dynamics and energy minimization calculations were used to relax bond lengths and angles and to identify flexible regions. The derived structure contains two alpha-helices, two beta-sheets, and a high proportion of amino acids in loops and turns. The model is in good agreement with preliminary NMR spectroscopic analyses. It explains the observed differences in calcium binding between sea raven Type II AFP and MBP-A. Furthermore, the model proposes the formation of five disulfide bridges between Cys 7 and Cys 18, Cys 35 and Cys 125, Cys 69 and Cys 100, Cys 89 and Cys 111, and Cys 101 and Cys 117.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献
