Multicopper oxidases, such as laccase or bilirubin oxidase, are known to reduce molecular oxygen at very high redox potentials, which makes them attractive biocatalysts for enzymatic cathodes in biological fuel cells. By designing an enzymatic gas‐diffusion electrode, molecular oxygen can be supplied through the gaseous phase, avoiding solubility and diffusion limitations typically associated with liquid electrolytes. In doing so, the current density of enzymatic cathodes can theoretically be enhanced. This publication presents a material study of carbon/Teflon composites that aim to optimize the functionality of the gas‐diffusion and catalytic layers for application in enzymatic systems. The modification of the catalytic layer with multiwalled carbon nanotubes, for example, creates the basis for stronger π–π stacking interactions through tethered enzymatic linkers, such as pyrenes or perylene derivates. Cyclic voltammograms show the effective direct electron contact of laccase with carbon nanotube‐modified electrodes via tethered crosslinking molecules as a model system. The polarization behavior of laccase‐modified gas‐diffusion electrodes reveals open‐circuit potentials of +550 mV (versus Ag/AgCl) and current densities approaching 0.5 mA cm2 (at zero potential) in air‐breathing mode.  相似文献

Lignin, a major component of the cell wall of vascular plants, has long been recognized for its negative impact on forage quality, paper manufacturing, and, more recently, cellulosic biofuel production. Over the last two decades, genetic and biochemical analyses of brown midrib mutants of maize, sorghum and related grasses have advanced our understanding of the relationship between lignification and forage digestibility. This work has also inspired genetic engineering efforts aimed at generating crops with altered lignin, with the expectation that these strategies would enhance forage digestibility and/or pulping efficiency. The knowledge gained from these bioengineering efforts has greatly improved our understanding of the optimal lignin characteristics required for various applications of lignocellulosic materials while also contributing to our understanding of the lignin biosynthetic pathway. The recent upswing of interest in cellulosic biofuel production has become the new focus of lignin engineering. Populus trichocarpa and Brachypodium distachyon are emerging as model systems for energy crops. Lignin research on these systems, as well as on a variety of proposed energy crop species, is expected to shed new light on lignin biosynthesis and its regulation in energy crops, and lead to rational genetic engineering approaches to modify lignin for improved biofuel production.  相似文献

Arundo donax L., commonly known as giant reed, is promising biomass feedstock that is also a notorious invasive plant in freshwater ecosystems around the world. Heretofore, the salt tolerance of A. donax had not been quantified even though anecdotal evidence suggests halophytic qualities. To test whole-plant and leaf level responses, we established a pot experiment on 80 scions propagated from an A. donax population that has naturalized on the shore of the San Francisco Bay Estuary. To quantify growth and physiological responses to salinity (NaCl), A. donax scions were divided into eight treatments and grown for 60 days across a range of salinities (0–42 dS m−1). Classic growth analysis showed >80% reduction in overall growth at the highest salinities. Yet, there was zero mortality indicating that A. donax is able to tolerate high levels of salt. Declining photosynthesis rates were strongly correlated (R2 > 0.97) with decreasing stomatal conductance, which was in turn closely related to increasing salinity. Leaf gas exchange revealed that stomata and leaf limitations of carbon dioxide were three times greater at high salinities. Nonetheless, even when salinities were 38–42 dS m−1 A. donax was able to maintain assimilation rates 7–12 μmol m−2 s−1. Further, by maintaining 50% relative growth at salinities ~12 dS m−1 A. donax can now be classified as ‘moderately salt tolerant’. A. donax leaf gas exchange and whole-plant salt tolerance are greater than many important food crops (i.e. maize, rice), the bioenergy feedstock Miscanthus × giganteus, as well as some uncultivated plant species (i.e. Populus and Salix) that are indigenous in regions A. donax currently invades. The results of this study have implications for both agronomists wishing to expand A. donax to fields dominated by saline soils, and for others who are concerned about the spread of A. donax with altered stream hydrology or sea-level rise.  相似文献

C4 perennial grasses are being considered for bioenergy because of their high productivity and low inputs. In side-by-side replicated trials, Miscanthus ( Miscanthus x giganteus ) has previously been found more than twice as productive as switchgrass ( Panicum virgatum ). The hypothesis that this difference is attributable to higher leaf photosynthetic rates was tested on established plots of switchgrass and Miscanthus in central Illinois with >3300 individual measurements on 20 dates across the 2005 and 2006 growing seasons. Seasonally integrated leaf-level photosynthesis was 33% higher in Miscanthus than switchgrass ( P  < 0.0001). This increase in carbon assimilation comes at the expense of additional transpiration since stomatal conductance was on average 25% higher in Miscanthus ( P  < 0.0001). Whole-chain electron transport rate, measured simultaneously by modulated chlorophyll fluorescence, was similarly 23% higher in Miscanthus ( P  < 0.0001). Efficiencies of light energy transduction into whole chain photosynthetic electron transport, leaf nitrogen use and leaf water use were all significantly higher in Miscanthus. These may all contribute to its higher photosynthetic rates, and in turn, productivity. Systematic measurement of photosynthesis over two complete growing seasons in the field provides a unique dataset explaining why the productivity of these two species differs and for validating mechanistic production models for these emerging bioenergy crops.  相似文献

Switchgrass is being evaluated as a potential feedstock source for cellulosic biofuels and is being cultivated in several regions of the United States. The recent availability of switchgrass land cover maps derived from the National Agricultural Statistics Service cropland data layer for the conterminous United States provides an opportunity to assess the environmental conditions of switchgrass over large areas and across different geographic locations. The main goal of this study is to develop a data-driven multiple regression switchgrass productivity model and identify the optimal climate and environment conditions for the highly productive switchgrass in the Great Plains (GP). Environmental and climate variables used in the study include elevation, soil organic carbon, available water capacity, climate, and seasonal weather. Satellite-derived growing season averaged Normalized Difference Vegetation Index (GSN) was used as a proxy for switchgrass productivity. Multiple regression analyses indicate that there are strong correlations between site environmental variables and switchgrass productivity (r = 0.95). Sufficient precipitation and suitable temperature during the growing season (i.e., not too hot or too cold) are favorable for switchgrass growth. Elevation and soil characteristics (e.g., soil available water capacity) are also an important factor impacting switchgrass productivity. An anticipated switchgrass biomass productivity map for the entire GP based on site environmental and climate conditions and switchgrass productivity model was generated. Highly productive switchgrass areas are mainly located in the eastern part of the GP. Results from this study can help land managers and biofuel plant investors better understand the general environmental and climate conditions influencing switchgrass growth and make optimal land use decisions regarding switchgrass development in the GP.  相似文献

Switchgrass (Panicum virgatum) has been planted extensively for habitat restoration across the United States, such as with the Conservation Reserve Program (CRP). However, genetic profiles of these populations have never been studied nor compared with those of remnant prairies or cultivars. In this study, we sampled 16 CRP and 17 prairie populations across eastern Kansas. We assessed ploidy levels of all populations and compared genetic diversity and structure of 10 prairies, 10 CRP areas, and 5 standard cultivars, using nine simple sequence repeat (SSR) DNA markers. All CRP and prairie populations were octaploid (8x), except two prairies with both hexaploid (6x) and octaploid (8x) individuals. Based on the results of SSR analyses, there were no significant differences between CRP and prairie populations in genetic diversity, and 94% of total variation was partitioned within populations. Similarities among prairie and CRP populations were also observed in Bayesian clustering algorithms and principal coordinate analysis, suggesting that they had similar genetic compositions. In addition, positive spatial autocorrelations were detected up to 42 and 46 km among prairie and among CRP populations, respectively. To conclude, the CRP and prairie populations shared similar genetic profiles. However, remnant prairies still harbored unique genotypes and a high level of genetic diversity, highlighting the importance of seed sources for restoration efforts, that is using local wild seeds or cultivars from the same geographical region. A popular tetraploid (4x) cultivar known as “Kanlow” was genetically distinct from the prairie populations and therefore is not recommended for habitat restoration in this region.  相似文献

Many countries see biofuels as a replacement to fossil fuels to mitigate climate change. Nevertheless, some concerns remain about the overall benefits of biofuels policies. More comprehensive tools seem required to evaluate indirect effects of biofuel policies. This article proposes a method to evaluate large‐scale biofuel policies that is based on life cycle assessment (LCA), environmental extensions of input‐output (I‐O) tables, and a general equilibrium model. The method enables the assessment of indirect environmental effects of biofuels policies, including land‐use changes (LUCs), in the context of economic and demographic growth. The method is illustrated with a case study involving two scenarios. The first one describes the evolution of the world economy from 2006 to 2020 under business as usual (BAU) conditions (including demographic and dietary preferences changes), and the second integrates biofuel policies in the United States and the European Union (EU). Results show that the biofuel scenario, originally designed to mitigate climate change, results in more greenhouse gas emissions when compared to the BAU scenario. This is mainly due to emissions associated with global LUCs. The case study shows that the method enables a broader consideration for environmental effects of biofuel policies than usual LCA: Global economic variations calculated by a general equilibrium economic model and LUC emissions can be evaluated. More work is needed, however, to include new biofuel production technologies and reduce the uncertainty of the method.  相似文献

Microalgae are considered a very promising alternative for biofuel production. Several strategies were developed to modulate and improve algae metabolites production to meet the requirements for biodiesel production. Most previous research evidenced that the increase of the lipid content is accompanied by a decrease of the biomass production, which increases the cost of the downstream processing. Hence, the challenge is to find special culture conditions that increase the lipid and the biomass productivities simultaneously. In the present work, we developed a strategy for the improvement of biomass and lipid productivities in a novel local microalga isolate, Chlorocystis sp. QUCCCM14, which was not previously known as a promising strain. Indeed, culturing QUCCCM14 using f/2 medium with 10× NaH2PO4 (0.15 g L?1 NaNO3 and 5.6 mg L?1 NaH2PO4) resulted in an improvement of 3.178 folds the lipid productivity reaching 56.121 mg L?1 day?1 and enhanced the biomass productivity reaching 141.363 mg L?1 day?1, simultaneously. Comparative analyses of the FAME profiles demonstrated that fed‐batch culture with phosphate or nitrate separately leads to a high production of the omega 3 fatty acids (Linolenic acid), whereas fed‐batch culture with phosphate and nitrate simultaneously increased the production of fatty acids suitable for biodiesel production.  相似文献
