Video microscopy and digital time-lapse recording were used to monitor locomotion and proliferation of bovine pulmonary artery endothelial (BPAE) cells cultured with varying concentrations of basic fibroblast growth factor (bFGF). Cell trajectories were reconstructed using a generalized nearest-neighbor algorithm and analyzed to determine how cell motility is affected by cell-cell collisions, cell divisions, and increasing cell density. The temporal evolution patterns of the average speed of locomotion for all cells in a culture were computed and the effects of varying bFGF concentrations were analyzed. Intermediate concentrations of bFGF (30 and 50 ng/mL) significantly increased the speed of locomotion above the levels we observed with 0 and 100 ng/mL concentrations of bFGF. Increases in cell density due to proliferation were immediately accompanied by a decrease in the average speed of locomotion of the cell population. Finally, the effect of bFGF concentration on the overall cell proliferation rates was assessed. With the addition of 30 or 50 ng/mL of bFGF to the culture media, the observed cell proliferation rates increased significantly. The proliferation rates decreased when the bFGF concentration increased to 100 ng/mL. These results show that bFGF concentrations that increase the motility of BPAE cells also increase the observed cell proliferation rates. (c) 1994 John Wiley & Sons, Inc.  相似文献

The maintenance of protein function and structure constrains the evolution of amino acid sequences. This fact can be exploited to interpret correlated mutations observed in a sequence family as an indication of probable physical contact in three dimensions. Here we present a simple and general method to analyze correlations in mutational behavior between different positions in a multiple sequence alignment. We then use these correlations to predict contact maps for each of 11 protein families and compare the result with the contacts determined by crystallography. For the most strongly correlated residue pairs predicted to be in contact, the prediction accuracy ranges from 37 to 68% and the improvement ratio relative to a random prediction from 1.4 to 5.1. Predicted contact maps can be used as input for the calculation of protein tertiary structure, either from sequence information alone or in combination with experimental information. © 1994 John Wiley & Sons, Inc.  相似文献

For many cell types, initial receptor-mediated attachment to a ligand-coated surface is followed by the formation of focal contacts - strong, specialized, discrete adhesive connections between cell and substrate in which receptors are clustered and simultaneously linked to extracellular ligand and cytoskeletal proteins. Since adhesion affects many aspects of cellular physiology including growth, differentiation, and motility, understanding the biochemical factors which regulate focal contact assembly should enhance our understanding of these phenomena. In this paper, we present a mathematical model to examine how receptor-ligand, receptor-cytoskeleton, and cytoskeleton-cytoskeleton interactions affect the formation of receptor clusters which serve as precursors to mature focal contacts. Receptor clustering is presumed to occur through self-recognition of cytoskeletal elements which induce the polymerization of ligand-receptor-cytoskeleton complexes. Polymerization only occurs when the ligand density is above a critical value and a decrease in the receptor-ligand affinity shifts the critical ligand density to higher values. While cytoskeletal protein expression and receptor-cytoskeleton affinity influence the concentration of monomeric complexes, the formation of polymeric ligand-receptor-cytoskeleton aggregates is most sensitive to changes in the self-association affinity between cytoskeletal proteins. We find that a 100-fold enhancement in the affinity between cytoskeletal elements can produce a substantial increase in the total fraction of adhesion receptors associated with focal contact precursors (from 5% to over 90%). Our results suggest that under physiological conditions, cellular control of focal contact assembly most likely occurs through modulation of specific cytoskeletal proteins to solidify cytoskeleton-cytoskeleton connections within precursor focal contact structures.  相似文献

On any given day, about 35% of 80- to 85-day-old socially na?ve male (SNM) lobster cockroaches (Nauphoeta cinerea) spontaneously adopted an aggressive posture (AP) without encountering another male [spontaneous AP (SAP)]. Although SAP SNMs showed significantly higher release of the pheromone 3-hydroxy-2-butanone (3H-2B) than non-SAP SNMs, there was no significant difference in hemolymph juvenile hormone (JH) III titer. When different body parts were tested for induction of the attack behavior, the antenna was found to be the most effective. After 1 min of contact with an antenna from another SAP SNM, attack behavior was induced in 100% of SAP and 76.2% of non-SAP SNMs, and the JH III titer was significantly increased in all responders. Among the non-SAP SNMs, the JH III titer before antenna contact was significantly lower in the non-responders than in the responders, and, although the JH III increase induced by 1 min antenna contact was similar between responders and non-responders, the final JH III titer of the non-responders was significantly lower. A similar attack response, JH III titer change, and 3H-2B release were seen when the individual's own antenna was used. After 5 min of contact with an antenna from another SAP SNM, attack behavior was induced in 100% of SAP and 82% of non-SAP SNMs; in the former, 3H-2B release was similar before and after antenna contact, but the JH III titer was significantly increased after antenna contact, while, in the latter, both 3H-2B release and JH III titer were significantly increased after antenna contact. Among the non-SAP SNMs, JH III titer in the non-responders was not elevated after 5 min antenna contact, and was significantly lower than that in the responders. A pentane-washed antenna did not induce attack behavior or increase the hemolymph JH III titer, and a pentane-washed antenna coated with 3H-2B also failed to induce attack behavior. These results indicate that N. cinerea male-male agonistic interactions, to which the vertebrate challenge hypothesis can be applied, are due to contact pheromone on the antenna, resulting in the concomitant expression of attack behavior and an increase in 3H-2B release and JH III titer.  相似文献

Genistein is a phytoestrogen, particularly abundant in soybeans that can bind estrogen receptors and sex hormone binding proteins, exerting both estrogenic and antiestrogenic activity. In this study we used the Japanese quail embryo as a test end-point to investigate the effects of early embryonic exposure to genistein on male copulatory behavior and on vasotocin parvocellular system. Both differentiate by the organizational effects of estradiol during development and may therefore represent an optimal model to study the effects of xenoestrogens. We injected two doses of genistein (100 and 1000 microg) into the yolk of 3-day-old Japanese quail eggs. Other eggs were treated with either 25 microg of estradiol benzoate or sesame oil as positive and negative controls. At the age of 6 weeks, behavioral tests revealed a significant decrease of all aspects of copulatory behavior (in comparison to the control group) in estradiol-treated birds. In contrast, genistein-treated animals demonstrated various degrees of decrease in the mean frequencies of some aspects of the sexual behavior. The computerized analysis of vasotocin innervation in medial preoptic, stria terminalis and lateral septum nuclei revealed a statistically significant decreased immunoreactivity in treated animals compared to control ones. These results demonstrate that genistein, similarly to estradiol, has an organizational effect on quail parvocellular vasotocin system and on copulatory behavior. In conclusion, present results confirm, in this avian model, that embryonic exposure to phytoestrogens may have life-long effects on sexual differentiation of brain structures and behaviors.  相似文献

Protein folding experiments demonstrate that the folding behaviors of many proteins can be roughly classified into two types: two-state kinetics and multi-state kinetics. Although the two types of protein folding kinetics have been observed for a long time, what determines the folding type of a protein is still largely unclear. The present work performed a comparative study based on a dataset of 43 two-state and 42 multi-state folders at different levels of proteins' intrinsic properties from the simplest sequence length to native structure topology. The results show that protein's amino acids composition and the long-range interaction-based topological complexity rather than secondary structure contents are the major determinants of protein folding type. Furthermore, a sequence-based folding type prediction achieved an accuracy of more than 80%. These findings implicate that there is no clear boundary between secondary and tertiary structure formation during the protein folding process and support the existence of a continuum of folding mechanism between the two ends of hierarchic and nucleation folding scenarios.  相似文献

Biologists have repeatedly rediscovered classical models from physics predicting collision rates in an ideal gas. These models, and their two-dimensional analogues, have been used to predict rates and durations of encounters among animals or social groups that move randomly and independently, given population density, velocity, and distance at which an encounter occurs. They have helped to separate cases of mixed-species association based on behavioural attraction from those that simply reflect high population densities, and to detect cases of attraction or avoidance among conspecifics. They have been used to estimate the impact of population density, speeds of movement and size on rates of encounter between members of the opposite sex, between gametes, between predators and prey, and between observers and the individuals that they are counting. One limitation of published models has been that they predict rates of encounter, but give no means of determining whether observations differ significantly from predictions. Another uncertainty is the robustness of the predictions when animal movements deviate from the model's assumptions in specific, biologically relevant ways. Here, we review applications of the ideal gas model, derive extensions of the model to cover some more realistic movement patterns, correct several errors that have arisen in the literature, and show how to generate confidence limits for expected rates of encounter among independently moving individuals. We illustrate these results using data from mangabey monkeys originally used along with the ideal gas model to argue that groups avoid each other. Although agent-based simulations provide a more flexible alternative approach, the ideal gas model remains both a valuable null model and a useful, less onerous, approximation to biological reality.  相似文献

ABSTRACT

The article is divided into two parts. In the first, I explore what is known about the precontact history of Collingwood Bay based on archaeological, linguistic, and oral evidence. While much must be left to speculation, the evidence strongly suggests that the Bay has long been a meeting point between Austronesian and Papuan peoples. The second and longer part of the paper attends to the political system found in most Collingwood Bay communities. I focus here primarily on the Maisin-speaking people residing in the southern part of the Bay with whom I’ve worked since 1981. More specifically, I describe how this system incorporates both hierarchical and egalitarian aspects and has proven remarkably adaptable to social changes from the time of European first contact in the 1890s.  相似文献

Syncytia are formed when individual cells fuse. SARS-CoV-2 induces syncytia when the viral spike (S) protein on the surface of an infected cell interacts with receptors on neighboring cells. Syncytia may potentially contribute to pathology by facilitating viral dissemination, cytopathicity, immune evasion, and inflammatory response. SARS-CoV-2 variants of concern possess several mutations within the S protein that enhance receptor interaction, fusogenicity and antibody binding. In this review, we discuss the molecular determinants of S mediated fusion and the antiviral innate immunity components that counteract syncytia formation. Several interferon-stimulated genes, including IFITMs and LY6E act as barriers to S protein-mediated fusion by altering the composition or biophysical properties of the target membrane. We also summarize the effect that the mutations associated with the variants of concern have on S protein fusogenicity. Altogether, this review contextualizes the current understanding of Spike fusogenicity and the role of syncytia during SARS-CoV-2 infection and pathology.  相似文献
