The crystal structure of the seed lectin from the tropical legume Bowringia milbraedii was determined in complex with the disaccharide ligand Man(alpha1-2)Man. In solution, the protein exhibits a dynamic dimer-tetramer equilibrium, consistent with the concanavalin A-type tetramer observed in the crystal. Contacts between the tetramers are mediated almost exclusively through the carbohydrate ligand, resulting in a crystal lattice virtually identical to that of the concanavalin-A:Man(alpha1-2)Man complex, even though both proteins have less than 50% sequence identity. The disaccharide binds exclusively in a "downstream" binding mode, with the non-reducing mannose occupying the monosaccharide-binding site. 非结构性碳水化合物(NSC)在植物体内的含量及分配对植物生长和存活至关重要。开展氮沉降和降雨变化对幼苗NSC影响的研究,为揭示干旱导致幼苗死亡机理及预测气候变化背景下幼苗自然更新及培育提供依据。本研究以1年生红砂幼苗为对象,测定了不同降雨(降雨减少(W-)、自然降水(W)和降雨增多(W+))和氮添加(N0(0 g N·m-2·a-1)、N1(4.6 g N·m-2·a-1)、N2(9.2 g N·m-2·a-1)、N3(13.8 g N·m-2·a-1))条件下红砂幼苗各器官NSC及其组分含量。结果表明:红砂幼苗各器官NSC含量为28.8~71.8mg·g-1,叶片含量最高,茎含量最低。氮添加和降雨变化对红砂幼苗叶片和根系淀粉及总NSC含量有显著影响,而对茎无显著影响。各降雨条件下,氮添加均促进了红砂幼苗叶片淀粉和总NSC累积,在降雨增加30%的条件下氮的促进效应更显著,中高氮(N2和N3)叶片淀粉与总NSC含量显著高于低氮水平(N1和N0);在低氮降雨减少30%(N1W-)处理下,红砂叶片淀粉和NSC含量最小,而根系淀粉和NSC含量最大,即低氮干旱胁迫下红砂可通过NSC在不同器官的重... 相似文献

ABSTRACT. The detailed behavioural mechanisms underlying an instance of compensation for changes in dietary nutrients are described for the first time in an insect. Nymphs of Locusta migratoria L. were given one of four artificial diets on the third day of the fifth instar, and their feeding patterns recorded in detail for 12 h. The diets represented combinations of two protein and two digestible carbohydrate levels (28% and 14% dry weight) presented in an otherwise complete nutrient mix. At the nutrient levels used, locusts regulated their intake of food with respect to protein but not digestible carbohydrate in the diet. They ate more of the lower protein diets by eating the same sized meals more frequently than insects fed on the higher protein diets. Compensation was not complete over the 12 h observation period: insects on the lower-protein diets ingested and absorbed 72% as much nitrogen as those insects fed on the higher-protein diets. An abundant supply of cavity-bearing trees is important for maintaining wildlife communities in harvested forests. During harvesting, suitable trees and cavities are directly removed, and the longevity of cavities in retained trees may be reduced by increased exposure to wind and other disturbance factors. We examined patterns of cavity survival in retained trembling aspen (Populus tremuloides) trees in harvested stands compared with those in unharvested mature stands by monitoring the persistence of individual cavities. We followed 930 cavities in 3 harvest treatments for up to 17 years in pre-cut and uncut forest, and up to 13 years post-harvest (reserve patches and dispersed retention), in temperate-mixed forests of interior British Columbia, Canada. Average annual cavity loss rates were 5.6% in pre-cut and uncut forest, 7.2% for cavities in trees retained in reserves, and 8.1% for cavities in retained trees dispersed throughout cuts. Correspondingly, median cavity longevity was 15 years for cavities in pre-cut and uncut forest, 10 years for cavities retained in reserves, and 9 years for those in dispersed retention. Risk of loss increased most for cavities in living trees (factor of 2.17), but we found no detectable difference for cavities in recently dead trees and trees with advanced decay. We suggest retention of a range of aspen size and decay classes to allow for future cavity-tree recruitment in harvested stands. Inclusion of wildlife reserves as part of an overall forest management plan will also help to mitigate the effects of windthrow and maintain long-lived cavity resources required by a large portion of forest wildlife. © 2013 The Wildlife Society 相似文献

Survival of the green alga Scenedesmus acuminatus Lagerh. in complete darkness was studied in axenic batch cultures at 7°C and 22°C for three months. The decrease in cell numbers was insensitive to temperature and slower than the loss of dry weight. However, the lag phase before cells began to lyse was more than twice as long at 7° C than at 22°C. The decline in cellular carbohydrates and proteins occurred in two phases. During the first 3-4 days, the decrease in cellular carbohydrate levels was significantly accelerated and temperature-sensitive. Pyrenoids disappeared within 5 days of darkness. Proteins showed 20-fold higher degradation rates at 22°C than at 7°C during the first 4 days. Thereafter, the rates of carbohydrate and protein decomposition were slow and temperature-independent. By contrast, lipids degraded only little at virtually constant and temperature-insensitive rates over the entire experimental period. After three months of dark incubation, about 40% of the remaining cells had retained their growth potential. However, the lag phase, after which cell division was resumed when exposed to light, increased with the duration of the previous dark period. The decrease in photo synthetic potential, which was more pronounced at 22° C than at 7° C, was apparent both in declining maximum assimilation numbers and maximum quantum yields. Cellular chlorophyll a concentrations in surviving cells decreased only slightly. We conclude that the primary means by which S. acuminatus survives extended dark periods is by reduction of catabolic reactions. This was suggested by the slow loss of cell weight. No evidence of significant heterotrophic acetate uptake was found.
