Alfalfa is sensitive to waterlogging, and its yields are significantly reduced under this condition. We investigated the effects of soil flooding on free abscisic acid (ABA) accumulation in shoots and roots of alfalfa in relation to plant growth and stomatal conductance responses. The production of dry matter in alfalfa was significantly affected by flooding mainly as a result of a rapid reduction in root growth. Shoot dry matter accumulation was maintained during the first 10 d of treatment and started to decline thereafter. Foliar concentration of the major mineral elements (N, P, K) was reduced by flooding, whereas only K concentration decreased in roots of flooded plants. Regrowth declined with duration of flooding and was less than 50% of controls after 2 weeks. While no changes in ABA concentration could be detected in flooded roots, an increase was noted within a few days in leaves when compared to unflooded controls. This increase in free ABA coincided with the accumulation of large quantities of starch in leaves and a rapid decline in leaf stomatal conductance. Our results support the suggestion that leaf ABA originates from the leaf itself and may be accumulating along with starch as a result of reduced translocation to the roots. Our observation of large accumulations of sucrose in flooded roots agrees with previous reports that supply of carbohydrates is not a limiting factor to root anaerobic metabolism in flooded alfalfa. 相似文献

An infection and treatment protocol involving infection with a mixture of three parasite isolates and simultaneous treatment with oxytetracycline is currently used to vaccinate cattle against Theileria parva. While vaccination results in high levels of protection in some regions, little or no protection is observed in areas where animals are challenged predominantly by parasites of buffalo origin. A previous study involving sequencing of two antigen-encoding genes from a series of parasite isolates indicated that this is associated with greater antigenic diversity in buffalo-derived T. parva. The current study set out to extend these analyses by applying high-throughput sequencing to ex vivo samples from naturally infected buffalo to determine the extent of diversity in a set of antigen-encoding genes. Samples from two populations of buffalo, one in Kenya and the other in South Africa, were examined to investigate the effect of geographical distance on the nature of sequence diversity. The results revealed a number of significant findings. First, there was a variable degree of nucleotide sequence diversity in all gene segments examined, with the percentage of polymorphic nucleotides ranging from 10% to 69%. Second, large numbers of allelic variants of each gene were found in individual animals, indicating multiple infection events. Third, despite the observed diversity in nucleotide sequences, several of the gene products had highly conserved amino acid sequences, and thus represent potential candidates for vaccine development. Fourth, although compelling evidence for population differentiation between the Kenyan and South African T. parva parasites was identified, analysis of molecular variance for each gene revealed that the majority of the underlying nucleotide sequence polymorphism was common to both areas, indicating that much of this aspect of genetic variation in the parasite population arose prior to geographic separation. 相似文献

Glycopeptides can be valuable tools in determining the influence of carbohydrate moieties on the intrinsic properties of glycoproteins. However, glycopeptides of sufficient quantity and purity are as yet not readily available from biological sources. The chemical coupling of a -glycosylamino group of an unprotected carbohydrate with an activated aspartic acid residue of an unprotected peptide is a simple method for synthesizing asparagine-linked glycopeptides. In this report we demonstrate that the use of this method is not restricted to -glycosylamines of simple monosaccharides or short aspartic acid-containing pentapeptides. Carbohydrate food sources have emerged as a critical factor limiting occupation of rainforests by hunter—gatherer peoples globally. In the wet tropics bioregion of northeastern Australia, Kuku–Yalanji aboriginal people occupied the rainforests through a hunter–gatherer subsistence economy prior to European occupation. Collaborative environmental research between a researcher at the James Cook University and Kuku–Yalanji people has established that their fire management protected carbohydrate resources in the fire-sensitive rainforests and their margins, and ensured ongoing access to the critical dry season carbohydrate resource, Cycas media, growing in patches of fire-prone open forest on each clan estate. Carbohydrate resources in the wet season were obtained predominantly from seeds of rainforest tree nuts, a high proportion of which are wet tropics endemic species. Several of the genera utilized by aboriginal people in the wet tropics bioregion also occur in the rainforests of eastern Cape York Peninsula, where they were not utilized as foods. It is hypothesized that use of rainforest seeds for carbohydrate is a cultural adaptation that occurred in the wet tropics bioregion, stimulated by the unique availability of the substantial number of large-seeded rainforest trees that are wet tropics endemics. The implications of these data for concepts about the impact of aboriginal fires on Australian rainforests are considered. Aboriginal fires imposed a fine patterning on the vegetation at the local scale, with little effect on the vegetation at the regional scale, which is determined by environmental factors. 相似文献

The monoclonal L5 antibody reacts with an N-glycosidically linked carbohydrate structure which is present on the neural cell adhesion molecule L1, neural chondroitin sulfate proteoglycans, and other not yet identified glycosylated proteins. Using this antibody, we isolated and characterized proteoglycans from adult mouse brain and cultured astrocytes biosynthetically labeled with Na2 35SO4 and a 3H-amino acid mixture. Our data suggest that the L5 proteoglycans of both sources are identical in their biochemical properties. The apparent molecular mass of the L5 proteoglycan is approximately 500 kDa. Digestion of the iodinated L5 proteoglycan from mouse brain and of the [35S]methionine-labeled L5 proteoglycan from cultured astrocytes with proteinase-free chondroitinases ABC and AC revealed three major core proteins with apparent molecular masses of approximately 380, 360, and 260 kDa. Summary Adult mice were found to show regional variation in the epithelial expression of some molecules of the blood-group antigen series. To investigate connective tissue influences on such differences, heterotypic recombinants of epithelia and connective tissues from various regions were prepared and examined using monoclonal antibodies directed against bloodgroup antigens H and Ley. Callus cultures from 300 genotypes of alfalfa (Medicago sativa L.) were initiated from leaf, petiole, and internode explants placed on Blaydes medium containing 10.74 M -naphthaleneacetic acid, 11.42 M indole-3-acetic acid, and 9.29 M kinetin. Five genotypes produced somatic embryos. Upon transfer of these embryos to growth regulator-free Murashige and Skoog medium with B5 vitamins, new somatic embryos repeatedly formed directly on older somatic embryos without an intervening callus phase in a cycle lasting about 30 days. These cultures have been maintained for two years, during which time their embryogenic capacity has remained stable. New embryogenic cultures could be started repeatedly from these genotypes. The elimination of sugars from the medium could stop recurrent embryogenesis. Glucose, maltose, and fructose stimulated recurrent embryogenesis more effectively than sucrose. Sucrose was superior to lactose, while sorbitol and mannitol did not stimulate recurrent somatic embryogenesis. The absence of nicotinic acid in the medium, as long as sucrose was present, was lethal to embryos of three of the five tested genotypes. The ability of this system to propagate embryos exponentially offers potential for development of new gene transfer systems and application to artificial seed technology.Abbreviations NAA
