Of the three defining properties of circadian rhythmicity--persisting free-running rhythm, temperature compensation, and entrainment--the last is often poorly understood by many chronobiologists. This paper gives an overview of entrainment. Where have we been? Where are we now? Whence should we be going? Particular emphasis is given to a discussion of the Discrete vs. Continuous models for entrainment. We provide an integrated mechanism for entrainment from a limit-cycle perspective.  相似文献

Chlorophyll fluorescence measurements have a wide range of applications from basic understanding of photosynthesis functioning to plant environmental stress responses and direct assessments of plant health. The measured signal is the fluorescence intensity (expressed in relative units) and the most meaningful data are derived from the time dependent increase in fluorescence intensity achieved upon application of continuous bright light to a previously dark adapted sample. The fluorescence response changes over time and is termed the Kautsky curve or chlorophyll fluorescence transient. Recently, Strasser and Strasser (1995) formulated a group of fluorescence parameters, called the JIP-test, that quantify the stepwise flow of energy through Photosystem II, using input data from the fluorescence transient. The purpose of this study was to establish relationships between the biochemical reactions occurring in PS II and specific JIP-test parameters. This was approached using isolated systems that facilitated the addition of modifying agents, a PS II electron transport inhibitor, an electron acceptor and an uncoupler, whose effects on PS II activity are well documented in the literature. The alteration to PS II activity caused by each of these compounds could then be monitored through the JIP-test parameters and compared and contrasted with the literature. The known alteration in PS II activity of Chenopodium album atrazine resistant and sensitive biotypes was also used to gauge the effectiveness and sensitivity of the JIP-test. The information gained from the in vitro study was successfully applied to an in situ study. This is the first in a series of four papers. It shows that the trapping parameters of the JIP-test were most affected by illumination and that the reduction in trapping had a run-on effect to inhibit electron transport. When irradiance exposure proceeded to photoinhibition, the electron transport probability parameter was greatly reduced and dissipation significantly increased. These results illustrate the advantage of monitoring a number of fluorescence parameters over the use of just one, which is often the case when the FV/FM ratio is used. Analysis of thermal melting curves represents one important approach for evaluating protein stability and the consequences of amino acid substitution on protein structure. By use of the van't Hoff relationship, the differential melting curve can be robustly fit to only three parameters, two of which are the underlying physical constants of melting temperature (Tm) and van't Hoff enthalpy (deltaHvH). Calculated Tm and deltaHvH values are insensitive to the choice of pre- and post-transition baselines. Consequently, the method accurately computes Tm and deltaHvH for extremely truncated data sets, in the complete absence of baseline information, and for proteins with low melting temperatures, where the traditional direct approach routinely fails. Moreover, agreement between deltaHvH values obtained using points derived from pre- vs. post-transition data provide an independent method for detecting some classes of non-two-state transitions. Finally, fitting of the differential denaturation curve should prove useful for analysis of abbreviated data sets obtained from high throughput array analysis of protein stability.  相似文献

The contribution of indigenous arbuscular mycorrhizal fungi (AMF) to growth and phosphorus (P) uptake by oilseed flax (Linum usitatissimum L.) was examined in two field experiments covering soil P levels from 20–86 mg kg-1 NaHCO3-extractable P. The fumigant dazomet was applied to the soil in half of the plots to obtain control plants with reduced mycorrhiza formation. An extensive AMF colonization of up to 48% of the root length was established in untreated soil of both experiments, although P fertilization reduced colonization to 28–39% at the latest harvests. Fumigation markedly decreased or totally prevented AMF colonization throughout the experiments. Root growth responded to fumigation by increased total and specific root length. Shoot P uptake was decreased by fumigation at soil P levels lower than ca. 50 mg kg-1 whereas shoot growth was reduced by fumigation at soil P levels lower than ca. 40 mg kg-1. The effects of fumigation were ascribed to the suppression of mycorrhiza formation. The effect of the AMF increased with decreasing soil P levels. Phosphorus inflow through roots (based on shoot P uptake) was reduced more strongly by fumigation than total P uptake. The P inflow through fungal tissue in roots was estimated to 4 × 10-14 mol P cm-1 s-1. We conclude that AMF are essential to flax growth at soil P levels below ca. 40 mg P kg-1, which is representative of the conditions under which most flax is grown.  相似文献

This paper describes development of a multi-pathway arsenic exposure model. The model uses information on arsenic concentrations in food, water, soil, and dust, combined with estimates of intake and medium-specific absorption. Urinary arsenic is predicted assuming that 60% of absorbed arsenic is excreted in urine under steady state conditions. Fecal arsenic is predicted assuming all unabsorbed arsenic is excreted in feces. We applied this model at a former copper smelter site. Site specific distributions were available for the following parameters: soil and dust arsenic concentration (geometric mean approximately 100 to 200?ppm and 50 to 100?ppm, respectively); the combined childhood soil and dust ingestion rate (geometric mean of 20?mg/d); soil and dust arsenic relative bioavailability (geometric mean 0.20 and 0.28, respectively); exposure duration; water arsenic concentration; air arsenic concentration; and total arsenic in food. Monte Carlo simulation was used to predict daily arsenic uptake and excretion in urine and feces for children. Predicted urine arsenic levels were less than measured levels (73% to 88% of measured values, depending on region of site). On the other hand, predicted fecal arsenic levels exceeded measured levels by a factor of 1.7 to 4.6. We were able to improve the correspondence between predicted and measured arsenic excretion rates by decreasing the assumed value of the combined soil and dust ingestion rate, and increasing the assumed bioavailability of arsenic in soil and dust.  相似文献

The effects of grazing by livestock on plant species composition and spatial distribution have been studied at Sadoré, Niger. Herbaceous species were recorded in plots of increasing size from 1/64 to 1024 m2 in ten fallow plots subjected to five different grazing treatments over the previous three years. Treatments consisted of three intensities of grazing, and of protection from grazing for either 3 or 14 years. For all treatments, the number of species fitted a normal distribution with the log (ln) of the area inventoried. However, the fit improved slightly when the model included two successive log-normal distributions respectively considered as species distribution within a patch and at the patch mosaic scale. Across treatments, the optimal sampling areas averaged 3.8 ± 1.1 m2 for the within-patch and 725 ± 113 m2 for the mosaic scale distributions. It is argued that similarity between treatments in the overall log-normal distribution resulted from compensations between the divergent trends that affected species distribution within and across patches depending on the grazing status. Long-term protection resulted in a regular spatial arrangement of highly contrasted, but internally homogeneous patches. Heavy grazing ensued the dominance of a few species in contagious patches but also left niches for scattered individuals of other species. Tests of the relative frequency of each plant species, together with the average area needed to record that species, were used to characterize specific response to grazing. A majority of species encountered in old Sahelian fallows were either fostered by grazing, indifferent or tolerant to grazing. However, more than a third of the species appeared sensitive to heavy grazing, and no relationships were found between species response to grazing and palatability.  相似文献

Aim The sequential break‐up of Gondwana is thought to be a dominant process in the establishment of shared biota across landmasses of the Southern Hemisphere. Yet similar distributions are shared by taxa whose radiations clearly post‐date the Gondwanan break‐up. Thus, determining the contribution of vicariance versus dispersal to seemingly Gondwanan biota is complex. The southern freshwater crayfishes (family Parastacidae) are distributed on Australia and New Guinea, South America, Madagascar and New Zealand and are unlikely to have dispersed via oceans, owing to strict freshwater limitations. We test the hypotheses that the break‐up of Gondwana has led to (1) a predominately east–west (((Australia, New Zealand: 80 Ma) Madagascar: 160–121 Ma) South America: 165–140 Ma), or (2) a southern (((Australia, South America: 52–35 Ma) New Zealand: 80 Ma) Madagascar: 160–121 Ma) pattern for parastacid crayfish. Further, we examine the evidence for a complete drowning of New Zealand and subsequent colonization by freshwater crayfish. Location Southern Hemisphere. Methods The evolutionary relationships among the 15 genera of Parastacidae were reconstructed using mitochondrial [16S, cytochrome c oxidase subunit I (COI)] and nuclear (18S, 28S) sequence data and maximum likelihood and Bayesian methods of phylogenetic reconstruction. A Bayesian (multidivtime ) molecular dating method using six fossil calibrations and phylogenetic inference was used to estimate divergence time among crayfish clades on Gondwanan landmasses. Results The South American crayfish are monophyletic and a sister group to all other southern crayfish. Australian crayfish are not monophyletic, with two Tasmanian genera, Spinastacoides and Ombrastacoides, forming a clade with New Zealand and Malagasy crayfish (both monophyletic). Divergence of crayfish among southern landmasses is estimated to have occurred around the Late Jurassic to Early Cretaceous (109–178 Ma). Main conclusions The estimated phylogenetic relationships and time of divergence among the Southern Hemisphere crayfishes were consistent with an east–west pattern of Gondwanan divergence. The divergence between Australia and New Zealand (109–160 Ma) pre‐dated the rifting at around 80 Ma, suggesting that these lineages were established prior to the break‐up. Owing to the age of the New Zealand crayfish, we reject the hypothesis that there was a complete drowning of New Zealand crayfish habitat.  相似文献

Previously, multivariate calibration techniques have been successfully applied to model and predict the antioxidant activity of green tea from its chromatographic fingerprint. Since the selectivity differences between dissimilar chromatographic systems have already been valuably used in several applications, in this paper it is studied whether combining the complementary information contained in two dissimilar fingerprints can improve the predictive capacity of the multivariate calibration model. The simplest way of combining the data is concatenating both fingerprints for each sample. The resulting matrix can then be subjected to Orthogonal Projections to Latent Structures (O-PLS). Unfortunately, this approach resulted in a more complex model with a prediction error of about the average of the errors obtained with the individual fingerprints. Secondly, only the peaks with high loading and low orthogonal loading from both chromatograms were included in the O-PLS model. This resulted in a reduced complexity, but not in better predictions, probably due to a lack of complementarity of the information concerning the antioxidant capacity. Finally, the concatenated fingerprints were subjected to stepwise multiple linear regression (MLR) in order to build a model based on the variables most correlated with the antioxidant capacity. The obtained prediction error was lower than those of both previous approaches, but still higher than the error of the model based on a single analysis. This is probably again caused by a lack of complementarity in the variables. Nevertheless, it was advantageous to develop fingerprints on dissimilar system, because it enables to choose the most suited chromatographic profile to build a multivariate calibration model for the considered purpose. In contrast to what was expected, the study showed that the most simple (so the worst separated) fingerprints resulted in the best predictions. On the other hand, a more complex fingerprint in which more compounds are separated is still important to improve the interpretability of the model.  相似文献

Yield reduction of pea (Pisum sativum) due to various types of infections by Mycosphaerella pinodes on pods was assessed. A range of disease severities was created on pods of pea plants grown in the glasshouse, by painting the pods with different concentrations of spore suspensions, at three different pod development stages: lag phase, the beginning of seed filling (BSF) and mid-filling of the seeds. Seed number at harvest was reduced only if the pods were infected before BSF, as shown previously for whole plant infections. Pod infections led to individual seed weight (ISW) losses from zero (for late infections, at mid-filling) to 20% (for earlier infections and severe disease). Infection during the lag phase affected ISW by reducing seed growth rate, whereas infection at BSF tended to reduce the duration of seed filling. There was a linear relationship between the area under the disease progress curve and the percentage decrease in ISW. This model should be complemented by the effect of leaves and stem infections, in order to predict ISW losses in diseased crop conditions, in which epidemics occur on all aerial parts of the pea plant.  相似文献

随着世界人口老龄化步伐的加快,衰老机制及抗衰老药物的研究日益成为生物医学领域的热点前沿之一.国内外已有大量研究报道抗衰老药物能够延长多种模式生物包括线虫、果蝇、小鼠、大鼠及灵长类等的寿命,然而,这些药物延缓衰老方式的差异仍缺乏系统研究.因此,本研究以衰老研究的热点模式生物线虫为对象,搜集1990年以来国内外刊物上正式发表的有关抗衰老药物寿命试验的研究文献及其涉及的生存曲线,利用荟萃分析比较了不同抗衰老药物与生存曲线变化类型间的关系,并结合药物的药理作用探讨其延寿机制.生存曲线特征聚类结果与药物生物学分类交叉分析结果表明,药理作用类型与增益类型具有很强相关性,提示这2种分类方法的结果是匹配的.抗氧化剂类药物和控制血糖类药物对生存曲线的改善总体增益虽然不是最高,但相对于正常组生存曲线其增益部分呈平移形,表明该类药物可以通过改善年龄结构对整个群体产生显著增益,且其衰老曲线的特征与自然衰老相似.抗癫痫药物及胃肠道菌群相关药物总体增益较大,其曲线增益形状呈梯形,提示该类药物(尤其是胃肠道菌群相关药物)尽管显著延长了少数个体的最大寿命,但是从整个群体来看,大多数个体寿命延长程度并不明显,受益的少数个体可能需要较长的外界资源支持方能保持较长时间的存活状态,这种模式既不大众化也不经济.综上所述,清除自由基和控制血糖类药物对健康老年有着更为积极合理的作用与效应.  相似文献
