Treatment of Dutch iris (Iris × hollandica Hoog. cv. Sapphire Beauty) bulbs with ethylene prior to precooling stimulated flowering in bulbs of various sizes. In large sized bulbs exposure to ethylene followed by precooling resulted in 100% flowering over a five months period after planting. Flowering in control bulbs which were not treated with ethylene prior to precooling was limited to 67% during the same five months period. In medium sized bulbs flowering in the ethylene treatment was 90% compared to 75% in the control. However, the biggest stimulation of flowering by ethylene was found in small sized bulbs (from 16 to 56%). Application of octanoic acid for a short time period prior to exposure to ethylene stimulated flowering in all bulb sizes. After five months the final percentage flowering in large and medium sized bulbs of the octanoic acid plus ethylene treatment did not differ from that of the ethylene only treatment. However, the initial rate of flowering was higher in the former treatment. In small bulbs the percentage flowering was much higher in the octanoic acid plus ethylene treatment than in the ethylene only treatment. The results of this study indicate that, just as in certain flowers, fruit and seeds, treatment with octanoic acid stimulates ethylene sensitivity in Dutch iris bulbs. The sensitivity of untreated bulbs to ethylene was highest in large bulbs and lowest in small bulbs. This correlated well with the endogenous octanoic acid content of the bulbs. Octanoic acid levels were highest in large bulbs and lowest in small Bulbs. It appears that the endogenous levels of octanoic acid in the bulbs is determined prior to the onset of dormancy.  相似文献

This immunohistochemical study of luteinizing hormone-releasing hormone (LHRH) in the olfactory bulbs in primates was undertaken in order to see whether there was an LHRH innervation in these species similar to that found in rodents. One old world (Macaca fascicularis) and two new world (Saimiri sciureus and Aotus trivirgatus) monkeys were studied. Aotus trivirgatus was of particular interest as it is noctural and so presumably more dependent upon olfactory cues. Animals were perfused with fixative, olfactory bulbs removed and sectioned, and tissues reacted immunocytochemically using LR1 (Benoit) antiserum to LHRH. Some LHRH innervation was found in the olfactory bulbs of all three species, comprising a few LHRH neurons and many fibers that ramified within the bulbs. The accessory bulb (not present as a distinct entity in old world primates) had more LHRH innervation than did the main olfactory bulb. Aotus trivirgatus had the greatest representation of LHRH of the three species. The layer of the olfactory bulb with the greatest number of LHRH fibers was the external plexiform layer. This is also true in rodents. There is evidence that LHRH has a role in the mediation of olfactory cues in reproductive behavior in rodents. It is not known how LHRH functions within the olfactory system in primates. However, the fact that it is distributed similarly in the two groups suggests that it may serve a similar function.  相似文献

Three protease inhibitors (OTI-1-3) have been purified from onion (Allium cepa L.) bulbs. Molecular masses of these inhibitors were found to be 7,370.2, 7,472.2, and 7,642.6 Da by matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF-MS), respectively. Based on amino acid composition and N-terminal sequence, OTI-1 and -2 are the N-terminal truncated proteins of OTI-3. All the inhibitors are stable to heat and extreme pH. OTI-3 inhibited trypsin, chymotrypsin, and plasmin with dissociation constants of 1.3×10-9 M, 2.3×10-7 M, and 3.1×10-7 M, respectively. The complete amino acid sequence of OTI-3 showed a significant homology to Bowman-Birk family inhibitors, and the first reactive site (P1) was found to be Arg17 by limited proteolysis by trypsin. The second reactive site (P1) was estimated to be Leu46, that may inhibit chymotrypsin. OTI-3 lacks an S-S bond near the second reactive site, resulting in a low affinity for the enzyme. The sequence of OTI-3 was also ascertained by the nucleotide sequence of a cDNA clone encoding a 101-residue precursor of the onion inhibitor.  相似文献

Plant bulbs are modified shoot systems comprised of short internodes with apical bud(s) surrounded by layers of leaf bases. Bulb diameters can vary greatly, with overall bulb size playing a role in flower formation and resource allocation. Despite the importance of bulb size to the overall fitness of an individual, evolutionary and ecological aspects of this trait have been almost completely neglected. Examining over 2,500 herbarium vouchers for 115 selected species, we analyzed monocot tunicate bulb size within a phylogenetic context in order to investigate its evolutionary significance. We recorded two bulb diameter optima and observed that as bulb size increases taxa inhabit warmer areas with less temperature seasonality. Furthermore, we found that hysteranthous taxa, a habit where leaves emerge separately from flowers, exhibit overall larger bulbs potentially due to reliance upon belowground stored resources to flower rather than on current environmental inputs. This work highlights the importance of including the belowground portion of plants into ecological and evolutionary studies in order to gain a more complete understanding of the evolution of plant forms and functions.  相似文献

The plant vacuole fulfills a variety of functions, and is essential for plant growth and development. We previously identified complex and mobile structures on the continuous vacuolar membrane, which we refer to as 'bulbs'. To ascertain their biological significance and function, we searched for markers associated with bulbs, and mutants that show abnormalities with respect to bulbs. We observed bulb-like structures after expression of non-membranous proteins as well as the functional soluble N-ethylmaleimide-sensitive factor attachment protein receptor (SNARE) molecules VAM3 and VTI11. Bulbs are formed in more tissues than previously reported, including flowering organs, suspension culture cells, endodermal cells in the flowering stem, and at very early stages of seed germination. Using existing and newly developed marker lines, we found that the frequency of bulb occurrence is significantly decreased in multiple shoot gravitropism (sgr) mutants, which are known to have a defect in vacuolar membrane properties in endodermal cells. Based on results with new marker lines, which enabled us to observe the process of bulb biogenesis, and analysis of the phenotypes of these mutants, we propose multiple mechanisms for bulb formation, one of which may be that used for formation of transvacuolar strands.  相似文献

The ultrastructure of the flame bulbs and capillaries of the protonephridia of Dactylogyrus (probably anchoratus) from Carassius auratus in southeastern Australia, and of an unidentified ancyrocephaline from the marine teleost Priacanthus macracanthus in southern Queensland is described. The cilia of the flame are anchored in the terminal cell by means of basal bodies without distinct rootlets. The nucleus of the terminal cell is basal, and (in Dactylogyrus) partly lateral to the basal bodies. The weir consists of a row of internal and a row of external ribs (rods) connected by a ‘membrane’. The external ribs are continuations of the cytoplasm of a thick-walled ‘cytoplasmic cylinder’ (proximal canal cell) which tightly surrounds most of the flame and contains a septate junction; the internal ribs are continuous with the terminal cell. Internal leptotriches arise from the perikaryon of the terminal cell, and, in the ancyrocephaline, also from the internal ribs. The wall of the protonephridial capillaries contains a septate junction, a reticulum of interconnected spaces and, in the ancyrocephaline, also lamellae. Lateral flames are common in the capillaries of Dactylogyrus.  相似文献

To find suitable candidates for biological control of the bulb mite, Rhizoglyphus robini Claparède (Acari: Astigmata) on lilies, exploration was undertaken in areas where the bulb mite is an established pest (The Netherlands, Taiwan and Japan). Among the predators, found in association with R. robini in the field and under storage conditions, mesostigmatic mites predominate. The most abundant species were Hypoaspis aculeifer (Canestrini), Lasioseius bispinosus Evans and Parasitus fimetorum (Berlese). These predators appeared to feed and reproduce on a diet of exclusively R. robini and they were able to control the bulb mite in small-scale population experiments initiated with a 1:20 predator-prey ratio. Under laboratory conditions corresponding to lily bulb propagation (lily scales mixed with vermiculite and stored at 23°C and >90% RH) the laelapid mite, H. aculeifer, was the most effective predator; the ascid predator, L. bispinosus, was much less effective, but being relatively small and being successful in attacking the juvenile stages of the bulb mite it may be better able to search for bulb mites hidden inside the lily bulb. The parasitid predator, P. fimetorum, failed to control the bulb mite when vermiculite was used as a medium, but turned out to suppress this prey when peat was used instead. Various strains of H. aculeifer or closely related species were compared with respect to their impact and performance on bulb mites as prey: two Dutch strains, one obtained from Breezand and the other from 'tZand, a Taiwanese strain, a German strain that in contrast to the previously mentioned strains was not collected from lily bulbs, but from agricultural areas near Bremen and, in addition, a Canadian strain of a related species (Hypoaspis miles Berlese), known to control sciarid fly larvae. These comparative experiments showed that H. miles died out without noticeable impact on the bulb mite population whereas all strains of H. aculeifer were able to suppress the bulb mites to very low numbers. However, the numerical responses of the H. aculeifer strains differed in that those collected in association with the pest (Breezand > Taiwan > 'tZand) were superior to the strain from Bremen. These results do not provide support to the Hokkanen and Pimentel hypothesis, which states that predators forming an evolutionary new association with the pest are often more effective in biological control.  相似文献

Aims:  Dickeya zeae is a pectinolytic bacterium responsible for soft rot disease in flower bulb crops. In this study, the possibility of controlling soft rot disease in hyacinth by using antagonistic bacteria isolated from hyacinth bulbs was explored.
Methods and Results:  Bacterial isolates with potential for biocontrol were selected on the basis of antibiosis against D. zeae , siderophore production, and the N -acyl homoserine lactones (AHLs)-inactivation. In in vitro assays, 35 out of 565 hyacinth-associated bacterial isolates produced antimicrobial substances against D. zeae, whereas 20 degraded AHLs, and 35 produced siderophores. Isolates of interest were identified by 16S rDNA sequence analysis and reaction in BIOLOG™ tests. Twenty-six isolates that differed in characteristics were selected for pathogenicity testing on hyacinth cultivars, Pink Pearl and Carnegie. Two strains identified as Rahnella aquatilis and one as Erwinia persicinus significantly reduced tissue maceration caused by D. zeae 2019 on hyacinth bulbs, but not on leaves.
Conclusions:  Hyacinth bulbs harbour bacteria belonging to different taxonomic groups that are antagonistic to D. zeae , and some can attenuate decay of bulb tissue.
Significance and Impact of the Study:  Selected hyacinth-associated bacterial isolates have potential for control of soft rot disease caused by D. zeae in hyacinth bulb production.  相似文献
