Oryzalin (3,5-dinitro-N4,N4-dipropyl-sulfanilamide) and BAS 083 (l,l-dimethylpiperdinium chloride) reduced root-knot infection in tomato roots when respectively applied as a soil drench at 20 ppm and 10,000 ppm. Oryzalin reduced knot counts with various intervals between treatment and inoculation. BAS 083 reduced knot counts only when applied before inoculation. Oryzalin was shown not to be a contact nematicide, and BAS 083 was only a weak one. Neither compound reduced penetration by infective larvae. Postinfection reduction in knot counts by Oryzalin and BAS 083 resulted, in part, from activation of natural defense mechanisms of the host. Giant-cell development in cotton roots inoculated with nematodes was inhibited by Oryzalin. Lateral root development was inhibited by BAS 083.  相似文献

Previous research showed that addition of nutrient nitrogen to ligninolytic (stationary, nitrogen-starved) cultures of the wood-decomposing basidiomycete Phanerochaete chrysosporium causes a suppression of lignin degradation. The present study examined early effects on nitrogen metabolism that followed addition of NH 4 + and l-glutamate at concentrations that yield similar patterns of suppression. Both nitrogenous compounds were rapidly assimilated (>80% in 6 h). Both caused an initial 80% or greater increase in the intracellular glutamate pool and had similar effects in increasing the specific activities of NADP- and NAD-glutamate dehydrogenases and glutamine synthetase. Differences between the effects of added NH 4 + and glutamate showed that suppression was not correlated with intracellular pools of arginine or glutamine, nor was the maintenance of an elevated glutamate pool required to maintain the suppressed state. While a portion of the initial glutamate suppression could be attributed to an effect on central carbon metabolism through glutamate catabolism by NAD-glutamate dehydrogenase, the long term suppression by glutamate and the suppression by NH 4 + were more specific. Suppression by NH 4 + or glutamate in the presence or absence of protein synthesis (cycloheximide) followed essentially identical kinetics during 12 h. These results indicate that nitrogen additions cause a biochemical repression of enzymes associated with lignin degradation. Initial changes in the fine morphology of apparently normal mouse zygotes and embryos were studied in serial sections of mouse oviducts that had been fixed in situ. These changes included extra sperm in the perivitelline space, nuclear budding, the presence of large nucleoli, perinuclear vesicles, the concentration of cytoplasmic organelles, and the extrusion of cytoplasmic ground substance. Initial changes in the nucleus and cytoplasm were undetectable when the zygotes and embryos were examined with the light microscope. It was concluded that serious abnormalities in zygotes and embryos, which may not be identified at the light microscope level, may be detected if they are examined in the electron microscope. Therefore, zygotes and embryos should be critically evaluated before being rated as normal.  相似文献

Intracellular electrical recordings in onion (Allium cepa L.) guard cells show that they maintain a membrane potential difference (MPD), inside negative. The MPD of intact cells averaged -72±29 mV (n=45); MPD of cells partially digested with a cellulolytic enzyme, -39±7 mV (n=65). Evidence indicates that the guard cells have two electrically distinct compartments, presumably delimited by the plasmalemma and tonoplast. Epidermal cells in partially digested preparations also showed MPD that could be either positive (+15±7 mV; n=23) or negative (-15 ±8 mV; n=13). Guard cells exposed to light-dark cycles hyperpolarized in the light and depolarized in the dark. The largest observed voltage changes reached 52 mV during hyperpolarizations and 60 mV during depolarizations. The light responses saturated with roughly exponential kinetics, with the depolarizations exhibiting a slower second phase that might be related to the contracting movements of the guard cells. Initial rates of the responses averaged about 14 mV min-1 in the dark and about 8 mV min-1 in the light. The results can be interpreted as electrical correlates of fluctuations in intracellular potassium concentration, as light-induced changes in membrane permeability, or as the photoactivation of an electrogenic proton pump. The last possibility seems to be the simplest interpretation of the data that also provides us with a mechanism driving the ion fluxes associated with stomatal function.  相似文献

The third petiolar bud ofHypolepis punctata appears on the basiscopic lateral side of the petiole above the fairly developed first petiolar bud. This investigation clarified the fact that the third bud is formed neither by the activity of the meristem of the first bud nor by the meristem directly detached from the shoot apical meristem, but is initiated in the cells involved in the abaxial basal part of the elevated portion of the leaf primordium. Thus the third bud is of phyllogenous origin. This investigation further revealed that the cells to initiate the third bud are originally located in the abaxial side of the leaf apical cell complex like the cells to initiate the first bud, but are not incorporated into the meristem of the first. After the first, second and third petiolar buds have been initiated, they are carried up into fairly high regions on the petiolar base by the intercalary growth which occurs in the leaf base below the insertion level of the first and the second buds.  相似文献

Shoot tip, bud, leaf, stem and root explants from bearing trees, offshoots, seedlings, and asexual plantlets ofPhoenix dactylifera L. were cultured on modified Murashige and Skoog nutrient media containing 3 g/l activated charcoal, 100 mg/l 2,4-dichlorophenoxyacetic acid, 3 mg/l N 6-(Δ2-isopentyl)adenine to obtain callus. Differential morphogenetic responses were obtained from calli dependent on the explant type and parent source. Subcultured shoot tips and leafy lateral buds callus on nutrient media devoid of charcoal and supplemented with 0.1 mg/l α-naphthaleneacetic acid (NAA) produced adventitious plantlets. Subcultured leaf calli produced roots only. Root callus failed to exhibit any morphogenetic response upon subculturing. Undifferentiated non-leafy buds and stem tissues did not give rise to callus, regardless of the parent source. Generally, the best callus and embryogenetic responses from explants were obtained from seedling and plantlet parent sources. Similarly, organogenetic responses such as root formation and shoot development from shoot tips cultured on media containing 10 mg/l NAA were also related to the parent explant source. 以走马胎(Ardisia gigantifolia)幼嫩茎段为外植体, 通过腋芽增殖的方式进行组织培养和快速繁殖研究。结果表明, 培养基MS+1.0 mg·L -1 6-BA+0.2 mg·L -1NAA和MS+0.5 mg·L -1 ZT均可用于腋芽的诱导和前期继代培养, 诱导率分别为89.3%和85.7%; 芽增殖最佳培养基为MS+0.5 mg·L -16-BA+0.1 mg·L -1ZT+0.1 mg·L -1NAA, 增殖系数为4.3倍; 根诱导最佳培养基为1/2MS+1.5 mg·L -1 IAA+1.0 mg·L -1 NAA, 生根率达92.3%, 且根系发达, 植株健壮; 生根苗在混合基质园土:泥炭:珍珠岩=3:1:1 (v/v/v )中移栽成活率为82%。该研究建立了走马胎种苗的组织培养快速繁殖技术体系, 且可应用于规模化生产。  相似文献

Glycogen, trehalose, glucose, and total lipid contents of six nematode species were studied. Anhydrobiotic Anguina tritici and Ditylencbus dipsaci stored trehalose in preference to glycogen and only small amounts of glucose were detected. Glycogen content was also reduced in anhydrobiotic Aphelenchus avenae. Conversely, Panagrellus redivivus and Turbatrix aceti contained large amounts of glycogen, appreciable amounts of glucose, and minimal amounts of trehalose. Ditylenchus myceliophagous "curds" contained low amounts of glycogen and very little trehalose; total lipid was 60% of that in fresh samples. The lipid contents of fresh samples of P. redivivus, T. aceti, and A. avenae were high (23.1, 21.9, and 36.7% dry weight, respectively), but in anhydrobiotic A. avenae larvae the level was reduced by over 60%. In contrast, lipid levels remained high in anhydrobiotic A. tritici and D. dipsaci larvae (40.6 and 38.3%, respectively). Analysis of lipid composition in anhydrobiotic A. tritici and A. avenae did not indicate any specific metabolic adaptations to desiccation survival.  相似文献

The influence of various c oncentrations of K⁺, nitrogen sources, and inoculation with root-knot nematode Meloidogyne javanica were evaluated in tomato plants. Increased potassium concentration increased top and root fresh weights of intact plants and fresh weights of excised roots. Nitrate-fertilized plants weighed more than plants receiving ammonium independent of the K level in the medium. Nematode counts on roots were not affected by nutritional differences in intact or excised roots. In intact roots a high percentage of males was recorded at low K⁺ levels, whereas in excised roots the proportion of males in the population rose as the K⁺ levels increased. Inoculated intact roots accumulated K⁺ when the level of potassium supply was low; infected excised roots contained less K⁺ than did nematode-free roots.  相似文献

Continuous cultures of Merismopedia tenuissima Lemmerman, limited by phosphorus, nitrogen, sulfur, or carbon, were compared to non limited batch cultures by two methods. The cellular content of photosynthetic pigments (chlorophyll and phycocyanin) was found to decrease in all nutrient limited cultures, except for the carbon limited culture. The ratio of carbohydrate to protein was 4- to 7-fold higher in P, N or S limited cultures than in non-limited or C limited cultures. The macromolecular products of photosynthesis were determined in samples to which NaH14CO3 was added. Relative incorporation into protein decreased in P or N limited cultures, increased accumulation of low molecular weight compounds was found in S and P limited cultures, and little change was noted in C limited cultures as compared to non-limited cultures. Although relative incorporation into protein was significantly greater at 20μEin·m?2·s?1 light intensity than at 180 μEin·m?2.s?1 in non-limited cultures, this effect was abolished in all nutrient limited cultures. These results suggest that measurement of the cellular carbohydrate to protein ratio and the products of photosynthesis would be useful in the analysis of algal population dynamics in nature.  相似文献
