In vitro clonal propagation of Lagerstroemia flos-reginae Retz   总被引:1,自引:0,他引:1  
Multiple shoots were obtained from nodal segments of young and mature trees of Lagerstroemia flos-reginae Retz on MS medium with 7.50–20 mg/l of benzyl amino purine. Rooting was achieved on transfer of the excised shoots to MS medium with 1 mg/l of indole butyric acid. The plantlets have been successfully transferred to soil.  相似文献

Summary I examined the germination characteristics of weed and outcrop populations of Crepis tectorum to test the hypothesis that the presumably more ephemeral weed habitat favors the highest levels of seed dormancy. The winter annual habit characterizing most plants of this species was reflected in a rapid germination of seeds sown in late summer. A slightly higher fraction of surface-sown seeds of weed plants delayed germination. Buried seeds of weed plants also survived better than seeds produced by plants in most outcrop populations, supporting the idea that weediness favors seed dormancy and a persistent seed bank. However, the differences in seed dormancy between the two ecotypes were small and not entirely consistent. Furthermore, high levels of seed dormancy were induced during burial in the outcrop group, suggesting that there is a potential for a dormant seed population in this habitat as well. Demographic data from one of the outcrop populations verified the presence of a large between-year seed bank. Possible environmental factors favoring seed dormancy in outcrop populations are discussed. The unusually large seeds of weedy Crepis contrasts with the relatively small difference in seed dormancy between the two ecotypes.  相似文献

Short-term applications of very high concentrations of 1-naphthaleneacetic acid (NAA) to expiants from flower stalks of tobacco (Nicotiana tabacum L. cv. Samsun) induced flower-bud regeneration to the same extent as longer or continuous incubation on lower concentrations. The maximum number of flower buds per explant after 15 d of culture was obtained not only by continuous culturing at 1 mol·l–1 NAA but also by 12 h of culturing at 22 mol·l–1 or 0.5 h at 220 mol· l–1, followed by incubation on medium without auxin for the remaining period. Continuous application of such high concentrations resulted in callus formation or caused the death of the explanted tissue. In all experiments in which auxin concentration and time of application were independently varied, the product of concentration and time determined the number of buds formed. Most, but not all, of the NAA taken up by the tissues was converted into conjugates. In expiants which had received a dose which was optimal for regeneration, the internal concentration of free NAA remaining beyond the pulse period was between 1.7 and 6.2 mol·l–1. Suboptimal applications led to lower values, supraoptimal treatments to much higher internal concentrations. Summary Keratin filaments of epithelial- and taste-bud cells in the circumvallate papillae of adult and developing mice were studied by immunocytochemistry using monoclonal antikeratin antibodies (PKK2 and PKK3) and by conventional electron microscopy. Elongated cells (type-I,-II, and-III cells) of the taste buds were stained by PKK3 antibody, which reacts with 45-kdalton keratin, whereas basal cells of the taste buds and surrounding epithelial cells showed negative staining with PKK3. Such PKK3-reactive cells occurred at 0 day after birth, when taste-buds first appeared in the dorsal surface epithelium of the papillae. Thus 45-kdalton keratin seems to be an excellent immunocytochemical marker for identifying taste-bud cells. Epithelial cells in all layers of the trench wall and basal layer cells of the dorsal surface contained densely aggregated bundles of keratin filaments that reacted with PKK2 antibody, but not with PKK3. In contrast, taste-bud cells and spinous and granular layer cells of the dorsal surface possessed loose aggregated bundles of filaments that reacted with PKK3, but not with PKK2. These results suggest that the aggregation and distribution pattern of keratin filaments may reflect differences in the keratin subtypes that comprise these filaments.  相似文献

野黄桂为常绿乔木,既是重要的药用和香料植物,也有观赏价值。原为野生,现已在中国科学院武汉植物研究所引种驯化成功,可以推广栽培。它的种子休眠的原因在胚的外部。这种休眠能被流水浸泡,GA和KN处理及冷层积打破。种子在3—5℃层积2—3个月,发芽率达85—89%。种子在15—30℃的温度中都可发芽,但最适发芽温度为20℃。种子应于采收后即播或及时层积至春播。当年生幼苗夏天应遮荫,冬季应防冻。它对气候和土壤的适应性较强,可塑性较大,在武汉地区生长发育良好。  相似文献

Summary A highly specific recognition system, capable of foreseeing and distinguishing between two critical points in time, exists in Mamestra brassicae (Lepidoptera: Noctuidae). Both points in time, the onset of a drought period and the end of the growth season, require different growth patterns of the pupae. In order to minimize the likelihood of weather-induced mortality and to maximize fitness, individuals of M. brassicae must enter aestival dormancy or hibernal diapause, respectively, before the onset of drought or frost. This study is primarily concerned with aestival dormancy. Normally, the pupal period of dormancy-free developing individuals amounts to approximately 20 to 30 days. A modified pupal period of approximately 35 to 80 days is defined as aestival dormancy. The onset of aestival dormancy is triggered by day lengths exceeding an innate individaul-specific threshold. The results reported in this paper indicate that the photoperiodic response curve represents largely the genetic variability within a population with respect to the thresholds triggering aestival dormancy. This variability in thresholds is considered to reflect the frequency of correlation of a distinct day length with a certain onset of drought period in the past. Furthermore, the innate thresholds are characterized by a temperature dependent norm of reaction. Our results also indicate, that a strong genetical component is involved in variability of duration of the pupal period. This variability in duration of aestivation reflects the frequency of drought periods of a certain length in the past. The adaptive significance of both the variation in day length thresholds and duration of aestival dormancy is discussed with respect to the number of generations per season, and the synchronization of the individual life cycles with the seasonal changing environmental conditions.This study is dedicated to Prof. H.J. Abstract Endogenous elemental sulfur (S0) has been studied in dormant spores and in spores in the early stages of germination, of Phomopsis viticola . S0 was measured by high-pressure liquid chromatography (HPLC). The rapid and almost total disappearance of endogenous S0 during the early stages of spore germination was directly related to a sharp increase of the respiratory activity and the ATP concentration. This was followed by the synthesis of DNA, RNA, proteins and lipids. Respiratory activity, S0 reduction and germination were inhibited in high concentrations of spores. Endogenous S0 disappearance, due to its reduction at the level of the mitochondrial respiratory chain with hydrogen sulfide production, may play a key role in the breaking of dormancy and the induction of germination in spores of P. viticola .  相似文献

Changes in activity of endogenous cytokinin-like substances were examined in intact plants and excised leaves of Begonia x chemantha Everett cv. Prinsesse Astrid (Christimas Begonia) by means of the tobacco callus bioassay. Cytokinin activity in the leaves of intact plants was higher in plants grown at 18°C than in those grown at 21° or 24°C. In excised leaves, an increase in cytokinin activity was observed during the first 4 days following leaf detachment. However, after the seventh day cylokinin activity decreased again. This decrease was more profound in leaves exposed to 24°C than in those exposed to 18°C.
Treatment of detached leaves with gibberellic acid (2.8 m M ) caused an increase in measurable cytokinin activity. This increase was more profound in the zones of activity which correspond with zeatin glucosides on paper and Sephadex LH-20 chromatography. Additional zones of activity appeared after Sephadex chromatography. These were of a more slow moving nature with elution volumes corresponding to Nb-(Δ2-isopentenyl)adenine and its derivaties. Water-treated control leaves had higher activity in the regions corresponding to zeatin and zeatin riboside.  相似文献
