该研究以从匈牙利引进的良种芦竹带腋芽茎段为外植体,以MS为基本培养基,通过设计不同的生长调节剂配比,对芦竹腋芽诱导、继代增殖及生根培养基进行了研究。结果表明:良种芦竹初代诱导最佳培养基为MS+6-BA 4.0 mg·L~(-1)+蔗糖30.0 g·L~(-1)+琼脂3.5 g·L~(-1),萌发率为90.0%;继代增殖最佳培养基为MS+6-BA 8.0 mg·L~(-1)+IBA 0.8 mg·L~(-1)+蔗糖30.0 g·L~(-1)+琼脂3.5 g·L~(-1),芽健壮均匀,增殖系数为4.3/30 d,基部保留3~5个芽作为继代培养物增殖效果最佳;选择高度5.0 cm以上的植株进行生根,最佳生根培养基为1/2MS+NAA 0.2 mg·L~(-1)+蔗糖20.0 g·L~(-1)+活性碳1.0 g·L~(-1),不添加琼脂,培养1周生根率为100.0%,平均生根4条,根长2.0~4.0 cm;将所得生根苗炼苗1周,以河沙、黄泥或腐质土作为栽培基质,移栽于阴蔽度70的大棚中,1个月后移栽成活率≥98.0%;移栽于实验田中,按常规方式进行管理,当年亩产量约为5 500.0 kg。该研究结果为良种芦竹的大规模产业化应用提供了技术支撑。  相似文献

Summary Plants of the inbreeding perennial herb Epilobium montanum L. were defoliated at two stages in development and the formation and abortion of flower buds, flowers, copsules and ovules was compared with control plants. A comparison was also made of the effects of self- and cross-pollination on fecundily. Defoliation reduced the number of flower buds formed but its greatest effect was to increase the abortion of flower buds. Defoliation also caused earlier abortions which were generally lower on the racemes of the treated than on the control plants. The abortion of flowers and capsules was much less significant. The number of ovules formed per capsule decreased with height on the raceme and the abortion rate of ovules was increased by defoliation. The pattern of ovule abortion within the capsules differed between cross- and self-pollinated plants but the number of seeds ripened was not affected. The overall survivorship of ovules to mature seeds was 89.3% which compares with the average of 85% found by Wiens for inbreeding annuals. The findings suggest that such high values of ovule survivorship may be characteristics of normally inbreeding species irrespective of whether they are annuals or perennials.  相似文献

The seeds of Fraxinus excelsior L. are dormant after harvest, since they need a period of chilling for germination. Moist treatment at 20°C for 2–3 months followed by stratification at 4°C for 7 months breaks dormancy. We observed that germination occurred during stratification and was spread over a period of 3 months. Germination at low temperature was temporarily inhibited by a moderate reduction of the seed water content initiated after the third month of stratification. This allowed the afterripening process to continue.
The following procedure was developed to suppress dormancy and to induce uniform germination:
  • 1. 

    Imbibition of the seeds and moist treatment at 20°C for 2–3 months;

  • 2. 

    stratification for 3 months;

  • 3. 

    treatment at low temperature and low water potential for at least 4 months, this treatment should not exceed 6 months;

  • 4. 

    complete rehydration of the seeds at 16°C.

Beffa, T., Pezet, R. and Turian, G. 1987. Multiple-site inhibition by colloidal elemental sulfur (S°) of respiration by mitochondria from young dormant α spores of Phomopsis viticola. Mitochondria from young dormant α spores of Phomopsis viticola Sacc. (ATCC 44940) were isolated by grinding and differential centrifugation. They presented a good integrity of their inner and outer membranes as measured by biochemical assays. Electron microscopic analysis revealed an homogenous population. The highest respiratory activities were observed with NADH and ascorbate + tetra-methyl-p-phenylenediamine (TMPD). Malate stimulated the oxidation of pyruvate, citrate or α-ketoglutarate. The coupling of respiration to oxidative phosphorylation appeared at the time of spore germination. The respiratory activities of mitochondria isolated from young dormant α spores of P. viticola were strongly inhibited by S°. The sensitivity of mitochondrial oxidation of different substrates (NADH, pyruvate + malate, succinate and ascorbate + TMPD) to S° was heterogenous and indicated multiple-site action. Thus preincubation of mitochondria with 30 μM S° before addition of substrates fully prevented NADH oxidation (>98%), and strongly inhibited oxidation of pyruvate + malate (85%), succinate (60%) and ascorbate + TMPD (74%). S° inhibited more rapidly the oxidation of succinate than that of other substrates. In the presence of dithiothreitol (DTT), S°-inhibited oxidation of all substrates (except ascorbate + TMPD) could only be transiently and weakly reestablished. The inhibitory action of S° on the oxidation of NADH, pyruvate + malate and succinate was higher than that observed with sulfhydryl group reagents such as mersalyl, Hg-acetate or p - chloromercuribenzoate. In contrast to S° these SH-group reagents could not inhibit oxidation of ascorbate + TMPD. S°, by its dual capacity to oxidize the SH-groups and to self-reduce, probably at the level of cytochrome c oxidase, could produce a modification of the oxidation state of the respiratory complexes thereby disturbing the electron flux.  相似文献

Indole-3-acetic acid (IAA) levels in the cambial region were measured by radioimmunoassay during the annual cambial activity - rest - quiescence transition at the crown top (current-year shoots) and at the middle and base of the stem of two 21-year-old Abies balsamea (L.) Mill. trees about 7.5 m in height. The IAA level declined throughout the experimental period (July 24 - December 3) at all positions in both trees. The earlywood - latewood transition, which occurred throughout each tree about the end of July, was associated in time with the largest decrease in the absolute amount of IAA. Tracheid production ceased towards the end of September, the cessation occurring earlier at the top of the tree than at the base. The cessation of tracheid production was only poorly correlated in time and space with the decline in IAA level, and it was not prevented by exogenous IAA. The results suggest that IAA level is involved in the control of tracheid radial enlargement, but not in the regulation of the cessation of tracheid production.  相似文献

Lateral buds may be preferred to shoot tips for in vitro propagation of papaya because of its unbranched nature. Proliferating shoot cultures from lateral buds appeared extremely compact with shortened internodes and leaf lamina of the cytokinin level (BAP 2 M) reported for multiple shoot production from shoot tips. ZEA (4 M) and 2iP (8 M) although reduced the proliferation rate, resulted in better growth of the shoot from lateral bud. Rooting was observed with IBA 20 M but plantlets so produced remained stunted.  相似文献

Multiple shoots were obtained from nodal segments of young and mature trees of Lagerstroemia flos-reginae Retz on MS medium with 7.50–20 mg/l of benzyl amino purine. Rooting was achieved on transfer of the excised shoots to MS medium with 1 mg/l of indole butyric acid. The plantlets have been successfully transferred to soil.  相似文献

Summary I examined the germination characteristics of weed and outcrop populations of Crepis tectorum to test the hypothesis that the presumably more ephemeral weed habitat favors the highest levels of seed dormancy. The winter annual habit characterizing most plants of this species was reflected in a rapid germination of seeds sown in late summer. A slightly higher fraction of surface-sown seeds of weed plants delayed germination. Buried seeds of weed plants also survived better than seeds produced by plants in most outcrop populations, supporting the idea that weediness favors seed dormancy and a persistent seed bank. However, the differences in seed dormancy between the two ecotypes were small and not entirely consistent. Furthermore, high levels of seed dormancy were induced during burial in the outcrop group, suggesting that there is a potential for a dormant seed population in this habitat as well. Demographic data from one of the outcrop populations verified the presence of a large between-year seed bank. Possible environmental factors favoring seed dormancy in outcrop populations are discussed. The unusually large seeds of weedy Crepis contrasts with the relatively small difference in seed dormancy between the two ecotypes.  相似文献

Short-term applications of very high concentrations of 1-naphthaleneacetic acid (NAA) to expiants from flower stalks of tobacco (Nicotiana tabacum L. cv. Samsun) induced flower-bud regeneration to the same extent as longer or continuous incubation on lower concentrations. The maximum number of flower buds per explant after 15 d of culture was obtained not only by continuous culturing at 1 mol·l–1 NAA but also by 12 h of culturing at 22 mol·l–1 or 0.5 h at 220 mol· l–1, followed by incubation on medium without auxin for the remaining period. Continuous application of such high concentrations resulted in callus formation or caused the death of the explanted tissue. In all experiments in which auxin concentration and time of application were independently varied, the product of concentration and time determined the number of buds formed. Most, but not all, of the NAA taken up by the tissues was converted into conjugates. In expiants which had received a dose which was optimal for regeneration, the internal concentration of free NAA remaining beyond the pulse period was between 1.7 and 6.2 mol·l–1. Suboptimal applications led to lower values, supraoptimal treatments to much higher internal concentrations.
