Axillary buds from adult field-grown plants of Lavandula dentata L. were used to evaluate the effect of growth regulators and culture media on the in vitro shoot proliferation and growth. The highest multiplication rate was obtained using Murashige and Skoog (MS) medium supplemented with a combination of 2.2 μM of benzyladenine and 2.5 μM indole-3-butyric acid. The best condition for rooting was MS medium plus 2.5 μM naphthaleneacetic acid. Rooted plantlets were successfully transferred to soil. Short-term culture derived plants (6 month) exhibited a normal development, but a low frequency of not heritable morphological changes were detected in long term culture derived plants (more than 1 year).This work was supported by a grant from the University of Caxias do Sul and CNPq, Brazil.  相似文献

While it is clear that the normal branching morphogenesis of the ureteric bud (UB) is critical for development of the metanephric kidney, the specific patterns of branching and growth have heretofore only been inferred from static images. Here, we present a systematic time-lapse analysis of UB branching morphogenesis during the early development of the mouse kidney in organ culture. Metanephric primordia from Hoxb7/GFP transgenic embryos were cultured for 3-4 days, and GFP images of the UB taken every 30 min were assembled into movies. Analysis of these movies (available as )revealed that the UB is a highly plastic structure, which can branch in a variety of complex patterns, including terminal bifid, terminal trifid, and lateral branching. To examine kinetic parameters of branching and elongation, skeletal representations of the UB were used to measure the number of segments and branch points and the length of each segment as a function of time and of branch generation. These measurements provide a baseline for future studies on mutant kidneys with defects in renal development. To illustrate how these quantitative methods can be applied to the analysis of abnormal kidney development, we examined the effects of the MEK1 inhibitor PD98059 on renal organ cultures and confirmed a previous report that the drug has a specific inhibitory effect on UB branching as opposed to elongation.  相似文献

Feeding of Helicoverpa zea larvae on cotton (Gossypium hirsutum L.) flower buds (squares) for 24 or 48 h induced the release of a number of terpenes [(E)--ocimene, linalool, (E)--farnesene, (E,E)--farnesene, (E)-4,8-dimethyl-1,3,7-nonatriene, (E,E)-4,8,12-trimethyl-1,3,7,11-tridecatetraene], isomeric hexenyl butyrates, 2-methylbutyrates, indole and (Z)-3-hexenyl acetate. These compounds are not released in significant amounts from undamaged squares and freshly damaged squares. The release of inducible compounds was not limited to the damaged squares themselves. The compounds were also released systemically from the upper undamaged leaves of the same plant after 72 h. However, the composition of the blend of systemically released volatiles differed from the blend released by damaged squares. The compounds that were systemically released from undamaged leaves in response to feeding on the squares were (Z)-3-hexenyl acetate, (E)--ocimene, linalool, (E)-4,8-dimethyl-1,3,7-nonatriene, (E)--farnesene, (E,E)--farnesene, and indole. This study shows that insect damage inflicted to the reproductive parts of a plant causes a systemic emission of volatiles from its vegetative parts.  相似文献

BACKGROUND AND AIMS: The formation of zonal communities may be attributed to differences in germination across the community and to timing of germination of seeds present in the seed bank. Our goals were two-fold: (1) to assess the annual germination pattern of Spergularia marina; and (2) to determine whether germination of S. marina differed across zonal communities. METHODS: Fresh seeds were buried in an experimental garden in polyester bags. Bags were harvested monthly for 1 year and exposed to differing 12 h/12 h temperature regimes (5/15 degrees C, 5/25 degrees C, 15/25 degrees C and 20/35 degrees C) with a 12 h dark/12 h light photoperiod. Replicate seeds were exposed to 24 h dark. Seeds were also placed in different zonal communities to assess germinability in the field. KEY RESULTS: Spergularia marina has a primary physiological dormancy. Conditional dormancy occurs from December to May and non-dormancy from June to November. Field germination initiates in the spring when temperatures are cool and salinity is low due to flooding, and ceases in the summer when temperatures exceed germination requirements. Spergularia marina has a light requirement for germination. CONCLUSIONS: If seeds become buried in the field or are light inhibited by Phragmites australis, they will remain dormant until they receive an adequate amount of light for germination. Since S. marina can germinate across all zones in a salt-marsh community, the formation of zonal communities is not determined at the germination stage, but at some later stage of development.  相似文献

Direct regeneration from explants without an intervening callus phase has several advantages, including production of true type progenies. Axillary bud explants from 6-month-old sugarcane cultivars Co92061 and Co671 were co-cultivated with Agrobacterium strains LBA4404 and EHA105 that harboured a binary vector pGA492 carrying neomycin phosphotransferase II, phosphinothricin acetyltransferase (bar) and an intron containing -glucuronidase (gus-intron) genes in the T-DNA region. A comparison of kanamycin, geneticin and phosphinothricin (PPT) selection showed that PPT (5.0 mg l–1) was the most effective selection agent for axillary bud transformation. Repeated proliferation of shoots in the selection medium eliminated chimeric transformants. Transgenic plants were generated in three different steps: (1) production of putative primary transgenic shoots in Murashige-Skoog (MS) liquid medium with 3.0 mg l–1 6-benzyladenine (BA) and 5.0 mg l–1 PPT, (2) production of secondary transgenic shoots from the primary transgenic shoots by growing them in MS liquid medium with 2.0 mg l–1 BA, 1.0 mg l–1 kinetin (Kin), 0.5 mg l–1 -napthaleneacetic acid (NAA) and 5.0 mg l–1 PPT for 3 weeks, followed by five more cycles of shoot proliferation and selection under same conditions, and (3) rooting of transgenic shoots on half-strength MS liquid medium with 0.5 mg l–1 NAA and 5.0 mg l–1 PPT. About 90% of the regenerated shoots rooted and 80% of them survived during acclimatisation in greenhouse. Transformation was confirmed by a histochemical -glucuronidase (GUS) assay and PCR amplification of the bar gene. Southern blot analysis indicated integration of the bar gene in two genomic locations in the majority of transformants. Transformation efficiency was influenced by the co-cultivation period, addition of the phenolic compound acetosyringone and the Agrobacterium strain. A 3-day co-cultivation with 50 M acetosyringone considerably increased the transformation efficiency. Agrobacterium strain EHA105 was more effective, producing twice the number of transgenic shoots than strain LBA4404 in both Co92061 and Co671 cultivars. Depending on the variety, 50–60% of the transgenic plants sprayed with BASTA (60 g l–1 glufosinate) grew without any herbicide damage under greenhouse conditions. Differential display analysis using dormant and non-dormant Arabidopsis thaliana (L.) Heynh seeds resulted in a set of genes that were associated with either dormancy or germination. Expression of the germination-associated genes AtRPL36B and AtRPL27B, encoding two ribosomal proteins, was undetectable in the dry seed, low in dormant seed, and high under conditions that allowed completion of germination. Expression of these genes was also found to be light-regulated and to correlate with germination speed. Expression of the dormancy-associated genes ATS2 and ATS4, encoding a caleosin-like protein and a protein similar to a low-temperature-induced protein respectively, was high in the dry seed and decreased during germination. Expression of ATS2 and ATS4 was high in primary and secondary dormant seed but low in after-ripened or chilled seed. The expression of both genes was also light-regulated, but no relationship with temperature-dependent germination speed was found.  相似文献

BACKGROUND AND AIMS: Plants have complex mechanisms of aerial biomass exposition, which depend on bud composition, the period of the year in which shoot extension occurs, branching pattern, foliage persistence, herbivory and environmental conditions. METHODS: The influence of water availability and temperature on shoot growth, the bud composition, the leaf phenology, and the relationship between partial leaf fall and branching were evaluated over 3 years in Cerrado woody species Bauhinia rufa (BR), Leandra lacunosa (LL) and Miconia albicans (MA). KEY RESULTS: Deciduous BR preformed organs in buds and leaves flush synchronously at the transition from the dry to the wet season. The expansion time of leaves is <1 month. Main shoots (first-order axis, A1 shoots) extended over 30 d and they did not branch. BR budding and foliage unfolds were brought about independently of inter-annual rainfall variations. By contrast, in LL and MA evergreen species, the shoot extension rate and the neoformation of aerial organs depended on rainfall. Leaf emergence was continuous for 2-6 months and lamina expansion took place over 1-4 months. The leaf life span was 5-20 months and the main A1 shoot extension happened over 122-177 d. Both evergreen species allocated biomass to shoots, leaves or flowers continuously during the year, branching in the middle of the wet season to form second-order (A2 shoots) and third-order (A3 shoots) axis in LL and A2 shoots in MA. Partial shed of A1 shoot leaves would facilitate a higher branching intensity A2 shoot production in LL than in MA. MA presented a longer leaf life span, produced a lower percentage of A2 shoots but had a higher meristem persistence on A1 and A2 shoots than LL. CONCLUSIONS: It was possible to identify different patterns of aerial growth in Cerrado woody species defined by shoot-linked traits such as branching pattern, bud composition, meristem persistence and leaf phenology. These related traits must be considered over and above leaf deciduousness for searching functional guilds in a Cerrado woody community. For the first time a relationship between bud composition, shoot growth and leaf production pattern is found in savanna woody plants.  相似文献

BACKGROUND AND AIMS: Seeds of east Australian Grevillea species generally recruit post-fire; previous work showed that the seed coat was the controller of dormancy in Grevillea linearifolia. Former studies on seed development in Grevillea have concentrated on embryology, with little information that would allow testing of hypotheses about the breaking of dormancy by fire-related cues. Our aim was to investigate structural and chemical characteristics of the seed coat that may be related to dormancy for three Grevillea species. METHODS: Seeds of Grevillea linearifolia, Grevillea buxifolia and Grevillea sericea were investigated using gross dissection, thin sectioning and histochemical staining. Water movement across the seed coat was tested for by determining the water content of embryos from imbibed and dry seeds of G. sericea. Penetration of intact seeds by Lucifer Yellow was used to test for internal barriers to diffusion of high-molecular-weight compounds. KEY RESULTS: Two integuments were present in the seed coat: an outer testa, with exo-, meso- and endotestal (palisade) layers, and an inner tegmen of unlignified sclerenchyma. A hypostase at the chalazal end was a region of structural difference in the seed coat, and differed slightly among the three species. An internal cuticle was found on each side of the sclerenchyma layer. The embryos of imbibed seeds had a water content six times that of dry seeds. Barriers to diffusion of Lucifer Yellow existed at the exotestal and the endotestal/hypostase layers. CONCLUSIONS: Several potential mechanisms of seed coat dormancy were identified. The embryo appeared to be completely surrounded by outer and inner barriers to diffusion of high-molecular-weight compounds. Phenolic compounds present in the exotesta could interfere with gas exchange. The sclerenchyma layer, together with strengthening in the endotestal and exotestal cells, could act as a mechanical constraint.  相似文献

  • Cycling of sensitivity to physical dormancy (PY) break has been documented in herbaceous species. However, it has not been reported in tree seeds, nor has the effect of seed size on sensitivity to PY‐breaking been evaluated in any species. Thus, the aims of this study were to investigate how PY is broken in seeds of the tropical legume tree Senna multijuga, if seeds exhibit sensitivity cycling and if seed size affects induction into sensitivity.
  • Dormancy and germination were evaluated in intact and scarified seeds from two collections of S. multijuga. The effects of temperature, moisture and seed size on induction of sensitivity to dormancy‐breaking were assessed, and seasonal changes in germination and persistence of buried seeds were determined. Reversal of sensitivity was also investigated.
  • Fresh seeds were insensitive to dormancy break at wet–high temperatures, and an increase in sensitivity occurred in buried seeds after they experienced low temperatures during winter (dry season). Temperatures ≤20 °C increased sensitivity, whereas temperatures ≥30 °C decreased it regardless of moisture conditions. Dormancy was broken in sensitive seeds by incubating them at 35 °C. Sensitivity could be reversed, and large seeds were more sensitive than small seeds to sensitivity induction.
  • Seeds of S. multijuga exhibit sensitivity cycling to PY‐breaking. Seeds become sensitive during winter and can germinate with the onset of the spring–summer rainy season in Brazil. Small seeds are slower to become sensitive than large ones, and this may be a mechanism by which germination is spread over time. Sensitive seeds that fail to germinate become insensitive during exposure to drought during summer. This is the first report of sensitivity cycling in a tree species.
