The sugar kelp Saccharina latissima experiences a wide range of environmental conditions along its geographical and vertical distribution range. Temperature and salinity are two critical drivers influencing growth, photosynthesis and biochemical composition. Moreover, interactive effects might modify the results described for single effects. In shallow water coastal systems, exposure to rising temperatures and low salinity are expected as consequence of global warming, increased precipitation and coastal run‐off. To understand the acclimation mechanisms of S. latissima to changes in temperature and salinity and their interactions, we performed a mechanistic laboratory experiment in which juvenile sporophytes from Brittany, France were exposed to a combination of three temperatures (0, 8 and 15°C) and two salinity levels (20 and 30 psu (practical salinity units)). After a temperature acclimation of 7 days, sporophytes were exposed to low salinity (20 psu) for a period of 11 days. Growth, and maximal quantum yield of photosystem II (Fv/Fm), pigments, mannitol content and C:N ratio were measured over time. We report for the first time in S. latissima a fivefold increase in the osmolyte mannitol in response to low temperature (0°C) compared to 8 and 15°C that may have ecological and economic implications. Low temperatures significantly affected all parameters, mostly in a negative way. Chlorophyll a, the accessory pigment pool, growth and Fv/Fm were significantly lower at 0°C, while the de‐epoxidation state of the xanthophyll cycle was increased at both 0 and 8°C compared to 15°C. Mannitol content and growth decreased with decreased salinity; in contrast, pigment content and Fv/Fm were to a large extent irresponsive to salinity. In comparison to S. latissima originating from an Arctic population, despite some reported differences, this study reveals a remarkably similar impact of temperature and salinity variation, reflecting the large degree of adaptability in this species. 相似文献

The structure of microbial communities of brown algae, red algae, and of the red alga Gracilaria verrucosa, healthy and affected with thallus rot, were comparatively investigated; 61 strains of heterotrophic bacteria were isolated and characterized. Most of them were identified to the genus level, some Vibrio spp., to the species level according to their phenotypic properties and the fatty acid composition of cellular lipids. The composition of the microflora of two species of brown algae was different. In Chordaria flagelliphormis, Pseudomonas spp. prevailed, and in Desmarestia viridis, Bacillus spp. The composition of the microflora of two red algae, G. verrucosa and Camphylaephora hyphaeoides, differed mainly in the ratio of prevailing groups of bacteria. The most abundant were bacteria of the CFB cluster and pseudoalteromonads. In addition, the following bacteria were found on the surface of the algae: Sulfitobacter spp., Halomonas spp., Acinetobacter sp., Planococcus sp., Arthrobacter sp., and Agromyces sp. From tissues of the affected G. verrucosa, only vibrios were isolated, both agarolytic and nonagarolytic. The existence of specific bacterial communities characteristic of different species of algae is suggested and the relation of Vibrio sp. to the pathological process in the tissues of G. verrucosa is supposed. 相似文献

A growth factor (TJF) for a malo-lactic fermentation bacterium has been isolated from tomato juice, and found to be a β-glucoside. The NMR spectra of TJF and its acetate revealed that the glucosyl residue linked to the hydroxyl group at C-2′ or C-4′ of d- or l-pantothenic acid moiety. Then, 2′-O-(β-d-glucopyranosyl)-dl-pantothenic acid (I), 4′-O-(β-d-glucopyranosyl)-dl-pantothenic acid (II) and 4′-O-(β-d-glucopyranosyl)-d(R)-pantothenic acid (II-a) were synthesized, and Il-a and 4′-O-(β-d-glucopyranosyl)-l-pantothenic acid (II-b) were obtained by the optical resolution of the acetate of II. Among the above compounds, II-a was identical with natural TJF regarding to the biological activity, NMR and ORD spectra, and thin-layer chromatography. 相似文献

Techniques are described for measuring gas exchange in seaweeds held in moist air (air suspension). In the species we have examined, oxygen has little or no effect on photosynthesis except at very low (50 μ1·1?1) CO2 concentration. Photorespiration could not be detected unless the seaweeds were treated simultaneously with high O2 and low CO2 or with the carbonic anhydrase inhibitor, diamox. However, sporulating and meristematic tissues exhibit oxygen-insensitive light respiration (CO2production in light not associated with photorespiratory metabolism). Elevated pH in the surface water of seaweeds also caused light respiration. Oxygen-sensitive wound respiration was observed that could easily be mistaken for photorespiration. C4 photosynthesis could not be detected. On the basis of several experimental approaches it was concluded that these seaweeds normally absorb bicarbonate rather than CO2 from sea water. High CO2 concentrations are required in gas streams aerating seaweed cultures in air or water suspension to maintain the bicarbonate concentration at levels normally found in sea water and to support normal levels of photosynthesis. 相似文献

Extraction with hydrochloric acid (pH 2.5) of the brown alga Padina pavonia afforded water-soluble and water-insoluble polysaccharides comprising D-glucuronic acid, L-fucose, D-xylose, D-mannose, D-glucose and D- galactose residues. The water-soluble polysaccharide was fractionated by using ethanol, and cetylpyridinium chloride and by chromatography on DEAE-cellulose. A neutral laminaran-like glucan, a sulphated heteropolysaccharide composed of the aforementioned sugars and a protein moiety were obtained. RESUME. La Microsporidie Nosema bombycis, Protozoaire parasite agent de la pébrine du ver à soie, a étéétudiée cytochimiquement à la fois en microscopie photonique et électronique. Les examens ont porté sur la détection et la localisation des acides nucléiques (ADN et ARN), des polysaccharides, de la phosphatase acide, au cours des différents stades du développement dans les cellules de I'hôte (du schizonte à la spore). Les principaux résultats concernent les observations relatives aux polysaccharides et à la phosphatase qui ne sont détectés qu'au stade de la spore et ne sont pas observés au stade du schizonte. Les polysaccharides sont présents au niveau du sac polaire, du filament polaire et sur la membrane cytoplasmique; la phosphatase acide est localisée au niveau du sac polaire, du filament polaire et dans la vacuole postérieure. SYNOPSIS. Nosema bombycis, agent of pebrine disease of silkworm, was studied cytochemically, using both light and electron microscopy. Presence of nucleic acids (DNA and RNA), polysaccharides, and acid phosphatases was demonstrated and localization of these substances was determined in various stages of the parasite (from the schizont to the spore). DNA and RNA were detected in all these stages. Polysaccharides and acid phosphatase were found in the spore but not in the schizogonic stages. Polysaccharides were detected in the polar cap, the polar filament, and the limiting membrane of the cytoplasm of the spore. Nilaparvata lugens, the brown planthopper (BPH) feeds on rice phloem sap, containing high amounts of sucrose as a carbon source. Nutrients such as sugars in the digestive tract are incorporated into the body cavity via transporters with substrate selectivity. Eighteen sugar transporter genes of BPH (Nlst) were reported and three transporters have been functionally characterized. However, individual characteristics of NlST members associated with sugar transport remain poorly understood. Comparative gene expression analyses using oligo-microarray and quantitative RT-PCR revealed that the sugar transporter gene Nlst16 was markedly up-regulated during BPH feeding. Expression of Nlst16 was induced 2 h after BPH feeding on rice plants. Nlst16, mainly expressed in the midgut, appears to be involved in carbohydrate incorporation from the gut cavity into the hemolymph. Nlst1 (NlHT1), the most highly expressed sugar transporter gene in the midgut was not up-regulated during BPH feeding. The biochemical function of NlST16 was shown as facilitative glucose transport along gradients. Glucose uptake activity by NlST16 was higher than that of NlST1 in the Xenopus oocyte expression system. At least two NlST members are responsible for glucose uptake in the BPH midgut, suggesting that the midgut of BPH is equipped with various types of transporters having diversified manner for sugar uptake. 相似文献

The competitive ability for N uptake by four intertidal seaweeds, Stictosiphonia arbuscula (Harvey) King et Puttock, Apophlaea lyallii Hook. f. et Harvey, Scytothamnus australis Hook. f. et Harvey, and Xiphophora gladiata (Labillardière) Montagne ex Harvey, from New Zealand is described by the uptake kinetics for NO3?, NH4+, and urea. This is the first study to report uptake kinetics for N uptake by a range of southern hemisphere intertidal seaweeds in relation to season and zonation. Species growing at the highest shore positions had higher NO3? and urea uptake at both high and low concentrations and had unsaturable NH4+ uptake in both summer and winter. Although there was evidence of some feedback inhibition of Vmax for NO3? uptake by Stictosiphonia arbuscula growing at the lower vertical limits of its range, rates were high compared with species growing lower on the shore. Our results highlight the superior competitive ability for N uptake of certain high intertidal seaweeds, and consistent with our previous findings we can conclude that intertidal seaweeds in southeast New Zealand are adapted to maximizing N acquisition in a potentially N‐limiting environment. 相似文献
