Aims

Microbiological, biotechnological and chemical characterization of a previously undescribed aryl‐crowned polyketide from Bacillus subtilis MTCC 10403 isolated from brown seaweed Anthophycus longifolius with activity against opportunistic Gram‐negative food‐borne pathogenic bacterial strains.

Methods and Results

A culture‐dependent method was used to isolate heterotrophic B. subtilis associated with A. longifolius and assessed for its antimicrobial properties. Minimum inhibitory concentration (MIC) of the title compound against the test pathogens was analysed by microtube dilution coupled with the 3‐(4,5‐dimethylthiazol‐2‐yl)‐2,5‐diphenyltetrazolium bromide‐based colorimetric endpoint detection. Bacillus subtilis MTCC 10403 was found to be antagonistic against Gram‐negative food‐borne pathogenic Pseudomonas aeruginosa, Escherichia coli, Klebsiella pneumoniae, Salmonella enterica serotype Typhi, Aeromonas hydrophila and Vibrio sp. (diameter of zone of growth inhibition 13–22 mm). Bacillus subtilis was assessed for the presence of secondary metabolite coding polyketide synthase (pks) gene ( KC589397 , 700‐bp gene product) and carboxylate siderophore framework in the aryl‐crowned polyketide designated as 7‐O‐6′‐(2″‐acetylphenyl)‐5′‐hydroxyhexanoate‐macrolactin by exhaustive spectroscopic techniques. The MIC assay showed that the reference antibiotics tetracycline and ampicillin were active at 25 μg ml?1 against the test pathogens, whereas the newly isolated polyketide displayed anti‐infective properties against E. coli, A. hydrophilla, P. aeruginosa and Vibrio sp. at a lower concentration (MIC <13 μg ml?1). The MIC of the aryl macrolactin against K. pneumoniae was comparable with that of the referral antibiotics (~25 μg ml?1). The mode of antimicrobial action of acryl‐crowned macrolactin was found to be iron chelating similar to siderophores. Putative biosynthetic pathway of the pks gene product further validated its molecular attributions.

Conclusions

This study recognized new variant of antimicrobial aryl‐crowned polyketide bearing methyl 6′‐(2″‐acetylphenyl)‐5′‐hydroxyhexanoate moiety at the C‐7 position of the macrolactin system from A. longifolius‐associated bacterium B. subtilis.

Significance and Impact of the Study

This study revealed seaweed‐associated micro‐organisms as promising biological strata to produce new‐generation anti‐infective agents.  相似文献

The energy expending and glucose sink properties of brown adipose tissue (BAT) make it an attractive target for new obesity and diabetes treatments. Despite decades of research, only recently have mechanistic studies started to provide a more complete and consistent picture of how activated brown adipocytes handle glucose. Here, we discuss the importance of intracellular glycolysis, lactate production, lipogenesis, lipolysis, and beta‐oxidation for BAT thermogenesis in response to natural (temperature) and artificial (pharmacological and optogenetic) forms of sympathetic nervous system stimulation. It is now clear that together, these metabolic processes in series and in parallel flexibly power ATP‐dependent and independent futile cycles in brown adipocytes to impact on whole‐body thermal, energy, and glucose balance.  相似文献

The aim of this study was to determine and compare the phenotypic variation in Çoruh trout (Salmo coruhensis), Rize trout (Salmo rizeensis), brown trout (Salmo trutta fario) and their native hybrids (S. rizeensis × S. t. fario; S. t. fario × S. coruhensis; S. rizeensis × S. coruhensis). The numbers and diameters of red and black spots were determined on operculum, fins and above and below the lateral line. The results from this study indicate that there were differences in spotting pattern, colour pattern or fin pigmentation traits between S. rizeensis, S. t. fario, S. coruhensis and their hybrids. Consequently, native S. rizeensis, S. t. fario and S. coruhensis were affected by interaction and phenotypic plastic aspects of traits in S. rizeensis were dominant in their hybrids (S. rizeensis × S. t. fario; S. rizeensis × S. coruhensis).  相似文献

The intramembrane particles and cellulose synthesis of the brown alga Syringoderma phinneyi Henry et Müller were examined using replicas of freeze‐fractured apical cells. Like in other brown algae, linear terminal complexes (TCs) were found in the plasmatic fracture face (PF) of the plasmalemma, which are the putative cellulose synthases. Terminal complexes consist of a single row of particles, each particle composed of two sub‐units, and are found in close relationship with cellulose microfibril imprints. Examination of the distribution of TCs revealed a clear apico‐basal gradient, with a higher density of TCs in the apical part. This seems to reflect the tip growth of the apical cells. The rate of cellulose synthesis per TC subunit was calculated based on the dimensions of the TCs and cellulose microfibrils.  相似文献

The number of genes controlling slow rusting resistance to leaf rust (Puccinia triticina) was estimated in five spring wheat (Triticum aestivum) cultivars using quantitative formulae. Parents and F6 families were evaluated in replicated field trials under epidemics initiated by artificial inoculation. The F6 families resulted from a diallel cross involving the fast-rusting cultivar Yecora 70 and five slow-rusting wheat cultivars: Sonoita 81, Tanager ‘S’, Galvez 87, Ures 81, and Moncho ‘S’. The area under the disease progress curve (AUDPC) was used to measure leaf rust severity over time. Results indicate that cultivar Sonoita 81 has three or four genes, Tanager ‘S’ has two or three genes, Galvez 87 has three genes, and both Ures 81 and Moncho ‘S’ have two genes for slow rusting resistance to leaf rust. Based on this result and previously reported moderate to high narrow-sense heritability estimates for slow rusting resistance in these materials, early-generation selection for slow leaf rusting would be effective.  相似文献
