Turnover in phosphate and potassium limited chemostat cultures of Bacillus subtilis W23 results in the release of over 80% of the wall material present at the time of chasing equilibrium-labelled cultures. The rate at which turnover proceeds is faster in potassium limited cultures than in phosphate limited cultures but in both cases a fraction of the wall material appears to be conserved, or to undergo turnover at a lower rate. Previously we have shown that the polar wall is less active metabolically than the cylindrical wall and it is possible that the apparently conserved wall is that present in the pole.  相似文献

Summary Exchange isotherms of Cu2+ vs Zn2+-ions were performed on the cell wall of a fresh water alga,Nitella flexilis. The relevant thermodynamic parameters are calculated. The cell wall absorbs copper selectively. The selectivity is explained by a stronger chelation of the cupric ion, due to the Jahn-Teller effect. The wall acts like a two-site model, based on the nature of the ligands: a first group of aminesites reacts exothermically and a second of hydroxylic-carboxylic sites of lower affinity, reacts endothermically.  相似文献

Incubation in vitro of adult Brugia pahangi in an apparatus which permitted the separate exposure of the anterior, middle, or posterior region of the worms to medium-containing radioactively labeled d-glucose, l-leucine, and adenosine has provided evidence that these materials are taken up in physiologically significant amounts by a transcuticular route. No evidence for an oral ingestion of materials has been obtained from worms in vitro, but in vivo an oral uptake of Trypan blue has been demonstrated. The ultrastructure and cytochemical staining reactions for nonspecific esterase, acid phosphatase (EC-3.1.3.2), and leucine naphthylamidase of the gut and body wall are described.  相似文献

During the life cycle of the mermithid nematode Gastromermis boophthorae the main components of the body wall, namely the cuticle, hypodermis and somatic musculature, undergo radical changes in structure and relative proportions. In the non-feeding, free-living stages the cuticle is a well-defined, multilayered structure which lies upon a generally thin hypodermis; the somatic musculature has the extensive and highly organised appearance typical of motile nematodes. In parasitic juveniles the body wall has a completely different structure which is adapted for the suggested purpose of nutrient uptake from the host insect's haemocoelomic fluid; the cuticle is characteristically thin and rather diffuse, the underlying hypodermis becomes very extensive and metabolically active, and there is a corresponding reduction in the proportion of somatic musculature. The sub-cuticular surface of the enlarged hypodermis has a microvillous configuration and has associated alkaline phosphatase activity.  相似文献

SYNOPSIS. The sporogonic stages of Leucocytozoon dubreuili in the midgut and salivary glands of the simuliid vectors was studied by electron microscopy. Young uninucleate oocysts have a pellicle that initially resembles that of the ookinetc. Numerous electron-dense bodies and microtubules in the peripheral cytoplasm may be involved in the formation of the cyst wall. The dense bodies appear to give rise to the amorphous material of the wall. The tubules which run circumferentially beneath the oocyst's boundary probably serve as a skeletal support for the cell surface during deposition of the wall material. A subcapsular “space” which provides area for expansion of the developing sporozoites is formed in early multinucleate oocysts. The subcapsular “space” appears to be formed through a condensation of the peripheral cytoplasm, resulting in an osmotic gradient across the oocyst's limiting membrane. Consequently water diffuses out, creating a fluid-filled space. Sporozoite formation begins with localized thickenings on the oocyst's limiting membrane. Subsequent extension of the thickened regions into the subcapsular “space” marks the onset of sporozoite budding. The process is highly synchronized, and culminates with the production of up to 150 sporozoites about the sporoblastoid body. The structure of sporozoites from mature oocysts and of the salivary glands of the vector is basically similar, although salivary gland sporozoites are more elongate and have numerous electron-dense micronemes. The paired rhoptries in the latter sporozoites are more elongate and uniformly electron-dense than in oocyst sporozoites.  相似文献

Phages are highly abundant in the environment and pose a major threat for bacteria. Therefore, bacteria have evolved sophisticated defence systems to withstand phage attacks. Here, we describe a previously unknown mechanism by which mono- and diderm bacteria survive infection with diverse lytic phages. Phage exposure leads to a rapid and near-complete conversion of walled cells to a cell-wall-deficient state, which remains viable in osmoprotective conditions and can revert to the walled state. While shedding the cell wall dramatically reduces the number of progeny phages produced by the host, it does not always preclude phage infection. Altogether, these results show that the formation of cell-wall-deficient cells prevents complete eradication of the bacterial population and suggest that cell wall deficiency may potentially limit the efficacy of phage therapy, especially in highly osmotic environments or when used together with antibiotics that target the cell wall.  相似文献

Genetic improvement of forage digestibility, especially utilizing marker assisted selection and recombinant DNA techniques, requires identification of specific biochemical traits and associated genes that impact digestibility. We undertook a study to identify cell wall (CW) traits of lucerne (Medicago sativa L.) stems that were consistently and strongly correlated with in vitro neutral detergent fibre (NDF) digestibility, a measurement that has been shown to correlate with animal performance. Spring and summer harvested lucerne stem material, for 2 years, from 24 individual plants in each of two germplasm sources were analyzed for 16 and 96 h in vitro NDF digestibility, and cell wall concentration and composition (monosaccharide constituents of cellulose, hemicellulose, and pectin; and Klason lignin (KL)) by the Uppsala dietary fibre method using near-infrared reflectance spectroscopy (NIRS). Pearson correlation coefficients were calculated for the relationships among these cell wall traits and with in vitro NDF digestibility. Concentrations of the pectin monosaccharide components were all negatively correlated (r=−0.73 to −0.94) with total cell wall concentration. In contrast, the three most abundant cell wall components glucose (Glc), xylose (Xyl) and Klason lignin were not correlated, or only weakly positively correlated (r<0.35), with cell wall concentration. Cell wall concentration was consistently negatively correlated (r=−0.60 to −0.94) with both 16 and 96 h in vitro NDF digestibility. In contrast, Klason lignin concentration was only marginally correlated (r<0.30) with 16 h in vitro NDF digestibility, but strongly negatively correlated (r=−0.71 to −0.74) with 96 h in vitro NDF digestibility. This is consistent with previous reports which show that lignin affects potential extent of digestion, but not rate. Cell wall glucose and xylose concentrations were inconsistently correlated with fibre digestibility. The monosaccharide components of pectin were consistently positively correlated (r=0.54–0.90) with in vitro NDF digestibility, except for 96 h in vitro NDF digestibility of spring harvested stems. Growth environment (year) and germplasm source had only minor impacts on the preceding correlation patterns, whereas spring versus summer harvests accounted for the inconsistencies observed among correlations for cell wall traits. The results of this study indicate that genetic improvement of fibre digestibility of lucerne stems should target genes that reduce total cell wall concentration, perhaps by reducing the rate of xylem tissue deposition during maturation, and reduce Klason lignin and increase pectin concentrations in the cell wall to improve potential extent and rate of fibre digestibility, respectively.  相似文献

Carbohydrate-hybridization probes (Vreeland and Laetsch, 1989, Planta (177, 423–434) were used to localize the homogalacturonan (pectate) component of pectins in the cell walls of leaves and soybean root nodules. Leaves of two species of the dicotyledon Dubautia were compared; these species contain much pectin but differ in their tissue water relations with respect to their cell-wall properties. Maturation of the primary cell walls in nodules was studied in the Bradyrhizobium japonicum-Glycine max symbiosis. Probe labelling was based on the divalent-cation-mediated association between pectate in tissue sections and fluorescein-conjugated pectate fragments. Pectate was also labelled by mixed-dimer formation with fluorescent polyguluronate derived from alginate. The specificity of the probe for unesterified polygalacturonate was indicated by increased cell-wall labelling after chemical or enzymatic deesterification of tissue sections, in contrast to elimination of labelling by chemical esterification. Postfixation of tissue sections improved retention of soluble pectate. Pectate differences were found in the leaves among cell types, in degree of esterification, and between plant species. The cell walls of soybean nodules were strongly labelled by the pectate probe in nodules one week and three weeks after infection. Pectate was more highly esterified in the central infected zone than in the surrouding cortex. Within the infected zone, walls of uninfected cells and infected cells were similarly labelled by the pectate probe. Abstract Bacteria grow by enlarging their envelope in such a way that osmotic pressure does not normally cause physical rupture. The strategy of Bacillus subtilis for both cylindrical elongation and pole formation is now substantially defined. Side-wall growth takes place by laying down new peptidoglycan, which is then displaced outwards, stretched and discarded; cross walls are laid down in the absence of stress, and then stretched and bulged outward as the septum is split and the pole is formed.  相似文献
