The effect of hydroxyurea on DNA repair replication has been studied in Chinese hamster ovary cells. Mitotic cells were treated with UV irradiation, methyl methanesulfonate or nitrogen mustard and incuated in the presence of each of the 4 [3H]deoxyribonucleosides plus BrdUrd and FdUrd for 2 h. The amount of repair replication was quantitated on CsCl gradients and similar values were obtained for each nucleoside. In all cases addition of HU during the incubation period increased these values approximately 2-fold. Following MMS treatment, pool sizes for each of the nucleosides were estimated by varying the amount of exogenously supplied nucleoside. They were found to be insensitive to the addition of HU and it is concluded that the increased incorporation of [3H]deoxyribonucleosides in the presence of HU reflects an increased amount of repair replication.  相似文献

The vomeronasal sensory epithelium contains two distinct populations of vomeronasal sensory neurons. Apical neurons express Gi2α‐linked V1R vomeronasal receptors and project to the anterior portion of the accessory olfactory bulb, while basal neurons express Goα‐linked V2R receptors and project to the posterior portion. Sensory neurons expressing V1R and V2R vomeronasal receptors are sensitive to different stimuli. Neurons in the vomeronasal system undergo continuous cell turnover during adulthood. To analyze over time neurogenesis of the different sensory cell populations, adult mice were injected with bromodeoxyuridine (BrdU) and sacrificed at postinjection days 1, 3, 5, 7, and 11. Newborn vomeronasal neurons were revealed by antibodies against BrdU while subclasses of vomeronasal neurons were identified using antibodies against Goα or Gi2α proteins. To ascertain whether G proteins are early expressed during neurogenesis, multiple labeling experiments using PSA‐NCAM and doublecortin were performed. Distribution of BrdU‐labeled cells was analyzed in angular segments from the margin of the sensory epithelium. No sexual differences were found. Within survival groups, BrdU‐Goα labeled cells were found more marginally when compared with BrdU‐Gi2α labeled cells. The number of BrdU‐positive cells decreased from day 1 to day 3 to remain constant afterwards. The relative proportions of BrdU‐Gi2α and BrdU‐Goα labeled cells remained similar and constant from postinjection day 1 onwards. This rate was also comparable with BrdU‐positive cells starting day 3. These results indicate an early, constant, and similar rate of neurogenesis in the two major subclasses of vomeronasal neurons, which suggests that both cell populations maturate independently. © 2010 Wiley Periodicals, Inc. Exposure of mouse epidermal cell cultures to β-retinyl acetate (RA) affects a number of parameters presumed to be important in chemical carcinogenesis. (1) RA alters the course of differentiation of the epidermal cells in culture resulting in a reduced rate of cell death which normally follows cellular maturation during the first two weeks in culture. The extended life span of the cultures appeared due to prolonged survival of cells and not to increased growth rate since RA inhibited the rate of cellular proliferation. This inhibition took place only after completion of a full cell cycle in the presence of RA. (2) DNA repair in response to physical and chemical agents was quantitatively unaffected in the presence of RA. (3) The activity of constitutive aryl hydrocarbon hydroxylase (AHH) was slightly decreased after exposure to RA but the level of enzyme induced by benz[a]anthracene was strongly reduced to 20% of the controls. (4) In the presence of RA, binding of 7,12-dimethylbenz[a]anthracene to epidermal cell DNA was markedly decreased. In contrast, binding to cellular protein was significantly increased by the retinoid.  相似文献

To compare the size of micronuclei induced by clastogens and by spindle poisons, bone-marrow smears were prepared 24 h after a single intraperitoneal injection of triethylenemelamine (TEM) (0.3 mg/kg) or vincristine (VCR) (0.125 mg/kg) into male mice. 100 micronucleated erythrocytes (MNEs) were randomly selected from each group and photomicrographed. Diameters of the cytoplasm (D) and the micronucleus (d) of each MNE were measured on the photographs. Relatively large micronuclei (d?D/4) were frequent (74%) in the VCR-treated group, and infrequent (2%) in the TEM-treated group. The frequencies of MNEs resulting in d?D/4 were determined for several other mutagens. All clastogens tested could be classified as TEM type, and all spindle poisons as VCR type.These findings indicate that it is possible to analyze the action site of micronucleus-inducing agents on the basis of the relative sizes of micronuclei.  相似文献
