The investigation of cell-mediated events in man has been largely limited to the study of the cells in the peripheral circulation. The study of T cells from localized anatomic compartments has been difficult due to the small numbers of cells usually obtainable from these sites. Investigation of such compartmentalized responses theoretically may yield information relating to both normal immunoregulation and autoimmune diseases--information that may not be obtainable through the investigation of the circulating cellular immune system. Utilizing cerebrospinal fluid (CSF) lymphocytes from patients with multiple sclerosis as a model of compartmentalized immunologically relevant cells, the technology for the generation of long-term T-cell lines from compartments both in continuous culture and after cryopreservation and that consist of both helper/inducer and suppressor/cytotoxic phenotypes have been generated. The 10(4) to 10(5) CSF cells obtained initially from individual patients have often been expanded into greater than 10(8) total cells within 4 months. The ability to generate large, stable, cryopreservable helper and suppressor/cytotoxic T-cell lines from limited access compartments will allow for new investigative approaches into both normal immunoregulation and autoimmune diseases in man.  相似文献

A rapid radioreceptor assay for measuring ß-endorphin (ß-EP) in unextracted serum has been developed. The method is based upon the inhibition by ß-EP of 3H-naloxone binding to the specific receptors on rat brain membranes, prepared in a stable form of pellets. Effect of serum on the assay was minimized by adding pooled serum to the equal dilution of total serum in the assay mixture. Pharmacokinetic analysis of pharmacologically active ß-EP equivalents (ß-EP eq.) in rats was performed using this method. The serum disappearance of ß-EP eq. after iv administration followed a biexponential decline and pharmacokinetic parameters were calculated by a two-compartment open model. The half-lives of α-phase and ß-phase were 2.6 ± 0.5 min and 6.2 ± 1.6 hr (mean ± SE; n=6), respectively. The volume of the central compartment (V1) and that of steady-state (Vdss) were 67 ± 16 and 480 ± 75 ml/kg (mean ± SE; n=6), respectively. The total body serum clearance (CLtot) was 2.1 ± 0.9 ml/min/kg (mean ± SE; n=6). The serum disappearance curve of ß-EP eq. obtained in the present study was similar to that previously reported by Houghten et al. (Proc. Natl. Acad. Sci. U.S.A. 77, 4588–4591 (1980)), in which the disapperance of total radioactivity of tritiated ß-EP in rats was examined.  相似文献

Summary The previous simple model for treating concerted evolution of multigene families has been revised to be compatible with various new observations on the immunoglobulin variable region family and other families. In the previous model, gene conversion and unequal crossing-over were considered, and it was assumed that genes are randomly arranged on the chromosome; neither subdivision nor correlation of gene identity and chromosomal distance were considered. Although this model satisfactorily explains the observed amino acid diversity within and between species, it fails to predict the very ancient branching of the mouse immunoglobulin heavy chain V-gene family. By incorporating subdivided structure and genetic correlation with chromosomal distance into the simple model, the data of divergence may be satisfactorily explained, as well as the rate of nucleotide substitution and the amino acid diversity. The rate at which a V-gene is duplicated or deleted by conversion or by unequal crossing-over is estimated by the new model to be on the order of 10–6 per year. An area of artificial intelligence known as experts systems (or knowledge-based systems) is being applied in many areas of science, technology and commerce. It is likely that the techniques will have an impact on vegetation science and ecology in general. This paper discusses some of those impacts and concludes that the main effects will be in areas of applied ecology especially where ecological expertise is needed either quickly (e.g. disaster management) or across a wide range of ecological disciplines (e.g. land management decisions). Expert systems will provide ecologists with valuable tools for managing data and interacting with other fields of expertise. The impact of expert systems on ecological theory will depend on the degree to which deep knowledge (i.e. knowledge based on first principles rather than on more empirical rules) is used in formulating knowledge bases.  相似文献

Summary There are many contradictory observations on the mechanohydraulic relation of growing higher plant cells and tissues. Graphical analysis of the simultaneous equations which govern irreversible wall yielding and water absorption has made more comprehensive the understanding of this relation when relative growth rate is plotted against turgor pressure. It suggests that some respiration-dependent and auxin sensitive process might regulate the difference of osmotic potential between cells and water source. Based on anatomical and electrophysiological knowledge of the pea stem xylem, we propose the wall canal system as the mechanism of respiration-dependent water uptake which is sensitive to auxin. This system consists of the xylem apoplastic walls, the xylem proton pumps, active solute uptake system and cell membranes. In the simplest case, third-order simultaneous differential equations are involved. Numerical analysis showed that net uptake of solutes enables water to be taken up against an opposing gradient of water potential. The behaviour of this wall canal system describes well the mechano-hydraulic relation of enlarging plant cells and tissues. Summary Dynamic change of plastid nucleoids (pt nucleoids) was followed by fluorescence microscopy after staining with 46-diamidino-2-phenyl indole (DAPI). The fluorescence image was quantified with a supersensitive photonic microscope system based on photon counting and image analysis. The results showed that small pt nucleoids located in the center of proplastids in the dry seed increased in size after imbibition and formed highly organized ring structures in the dark, which divided into ca. 10 pieces within 3 days. Corresponding to this morphological change, DNA content of a plastid multiplied 7.5 fold. Total increase in DNA content of pt nucleoids per cell was 34 times as that of dry seed, as plastid multiplied 4.6 times in the average during this period. Upon light illumination small pt nucleoids having basic genome size were separated from divided pt nucleoids, suggesting a relationship with the formation of thylakoid system. Cardiac copper,magnesium, and zinc in recent and old myocardial infarction   总被引:1,自引:0,他引:1  
X-ray fluorescence spectrometry and atomic absorption spectrometry were used in a quantitative study of zinc, copper, and magnesium in 71 postmortal human hearts. Samples were obtained from individuals who had demonstrated no previous clinical or subsequent pathological findings of myocardial infarction and from victims of a recent or an old infarction. A significant difference (p<0.001) in the elemental levels was observed between the noninfarct and the recent infarct groups. The noninfarct group had higher cardiac levels of all three elements. However, the difference in elemental concentrations between the noninfarct and the old-infarct groups was not significant. Cardiac levels of zinc (p<0.001) and copper (p<0.01) were significantly greater in the old-infarct group than in the recent-infarct group. Magnesium levels were higher in the recent-and-old-infarct group than in the recent infarct group (p<0.01). It is possible that the elements are redistributed during myocardial infarction, and that uptake of these elements (from the serum pool) by the heart may be important in maintaining myocardial integrity and function.  相似文献

A comparative study on the activity profile of catalase and superoxide dismutase, the two scavenging enzymes, as well as the developmental profile of lipid peroxidation in the human fetal brain, liver and kidney have been done for gestation periods ranging from 12 weeks to 28 weeks and beyond. The activity of the scavenging enzymes increase gradually inall the tissues with the advancement of pregnancy. Brain is an exception in case of catalase where the activity remains more or less same throughout the developmental period except in the case of fetuses, 28 weeks and above where significant decrease in the catalase activity is observed. A high level of lipid peroxidation is observed during early stages of development which declines thereafter.  相似文献
