Summary This study reports the selection and characterization of osteogenic precursors from human bone marrow which were isolated by two “clonings” and successive subculturing. These cell lines express alkaline phosphatase activity. Gel electrophoresis of [3H]-proline labeled cultures showed that the cloned cells produce only type I collagen. They synthetize osteocalcin and osteonectin. They respond to 1,25 dihydroxy vitamin D3 by increasing osteocalcin synthesis and secretion, and to parathyroid hormone by increasing cyclic AMP synthesis. After the third subculture in the absence of β-glycerophosphate, these cell lines formed lots of clusters which exhibit high alkaline phosphatase activity and positive von Kossa staining. X-ray energy spectrum shows that these cells are surrounded by “budding” structures containing calcium and phosphorus with a ratio Ca:P identical to those of pure hydroxyapatite. This process was associated with45Ca uptake into the cells. All these data support the selection of osteogenic cells which may be of considerable clinical importance.  相似文献

Niobium is used as an alloy in the industrial and biomedical fields. The concentration of the toxic element in organs of a number of animal species has been defined by using radioactive niobium (95Nb). However, tissue lesions induced by niobium have only been studied at the light microscopy level. In this study, we used an electron probe X-ray analyzer equipped with a transmission electron microscope to define the localization of this element in kidney and bone marrow cells. Results demonstrated that niobium is located in the lysosome and that this element coprecipitates with phosphate. In kidney, lysosomes and precipitates are eliminated in the tubular lumen. In contrast, precipitates appear to be eliminated more slowly from the lysosomes of bone marrow macrophages. These processes therefore correspond to one of the mechanisms by which lysosomes eliminate certain toxic mineral elements and thus play a role in the more general process of the body's defenses.  相似文献

Amphibians and reptiles are sensitive to changes in the thermal environment, which varies considerably in human-modified landscapes. Although it is known that thermal traits of species influence their distribution in modified landscapes, how herpetofauna respond specifically to shifts in ambient temperature along forest edges remains unclear. This may be because most studies focus on local-scale metrics of edge exposure, which only account for a single edge or habitat patch. We predicted that accounting for the combined effect of multiple habitat edges in a landscape would best explain herpetofaunal response to thermally mediated edge effects. We (1) surveyed herpetofauna at two lowland, fragmented forest sites in central Colombia, (2) measured the critical thermal maximum (CTmax) of the species sampled, (3) measured their edge exposure at both local and landscape scales, and (4) created a thermal profile of the landscape itself. We found that species with low CTmax occurred both further from forest edges and in areas of denser vegetation, but were unaffected by the landscape-scale configuration of habitat edges. Variation in the thermal landscape was driven primarily by changes in vegetation density. Our results suggest that amphibians and reptiles with low CTmax are limited by both canopy gaps and proximity to edge, making them especially vulnerable to human modification of tropical forest. Nuclear protein 1 (NUPR1) is a stress-induced protein activated by various stresses, such as inflammation and oxidative stress. We previously reported that Nupr1 deficiency increased bone volume by enhancing bone formation in 11-week-old mice. Analysis of differentially expressed genes between wild-type (WT) and Nupr1-knockout (Nupr1-KO) osteocytes revealed that high temperature requirement A 1 (HTRA1), a serine protease implicated in osteogenesis and transforming growth factor-β signaling was markedly downregulated in Nupr1-KO osteocytes. Nupr1 deficiency also markedly reduced HtrA1 expression, but enhanced SMAD1 signaling in in vitro-cultured primary osteoblasts. In contrast, Nupr1 overexpression enhanced HtrA1 expression in osteoblasts, suggesting that Nupr1 regulates HtrA1 expression, thereby suppressing osteoblastogenesis. Since HtrA1 is also involved in cellular senescence and age-related diseases, we analyzed aging-related bone loss in Nupr1-KO mice. Significant spine trabecular bone loss was noted in WT male and female mice during 6−19 months of age, whereas aging-related trabecular bone loss was attenuated, especially in Nupr1-KO male mice. Moreover, cellular senescence-related markers were upregulated in the osteocytes of 6−19-month-old WT male mice but markedly downregulated in the osteocytes of 19-month-old Nupr1-KO male mice. Oxidative stress-induced cellular senescence stimulated Nupr1 and HtrA1 expression in in vitro-cultured primary osteoblasts, and Nupr1 overexpression enhanced p16ink4a expression in osteoblasts. Finally, NUPR1 expression in osteocytes isolated from the bones of patients with osteoarthritis was correlated with age. Collectively, these results indicate that Nupr1 regulates HtrA1-mediated osteoblast differentiation and senescence. Our findings unveil a novel Nupr1/HtrA1 axis, which may play pivotal roles in bone formation and age-related bone loss.  相似文献

The recovery process of aSasa tsuboiana population after a mass flowering and death in 1977 was investigated by 15 years of observation in the Hira Mountains, Kinki district, western Japan. Seed production was high (6600–13 800 seeds m−2 inSasa plots and 3900 seeds m−2 in a forest plot) but emergent seedling density was low (14–21 seedlings m−2), probably because of seed predation byMicrotus montebelli occurring between seed shedding and the next spring. The seedling density had decreased further by the next year and theS. tsuboiana population recovered from only a limited number of seedlings. In spite of such a low initial density, theS. tsuboiana population was able to regenerate successfully and attained the previous full stand height in 7–16 years.Miscantbus sinensis invaded and delayed the recovery ofS. tsuboiana in one plot, butS. tsuboiana became dominant as it caught up with the height ofM. sinensis. Seedling growth patterns, such as frequent tillering, the onset of rhizome extension in the early stage of seedling growth and frequent culm production from rhizomes, played important roles in the successful regeneration ofS. tsuboiana.  相似文献

The power conversion efficiencies (PCEs) of all-polymer solar cells (all-PSCs) have already exceeded 17%. However, the limited absorption range of an all-polymer system results in significantly reduced short-circuit current density (Jsc), which eventually influences the PCE improvement. To broaden the light absorption of polymer acceptors, herein, benzotriazole is introduced in the core unit of small molecule acceptors and thus two narrow-bandgap polymer acceptors named PTz-BO and PTz-C11 featuring the same molecular backbone and different side-chain length are synthesized. Compared with PTz-C11, the PTz-BO based-all PSCs deliver a slightly reduced Jsc, a large open-circuit voltage (Voc) and a low voltage loss below 0.50 V. Moreover, ternary all-PSCs are constructed by introducing PTz-C11 as a guest component. Benefiting from the reduced recombination, improved exciton generation and dissociation, and balanced charge transport, a high efficiency of 16.58% is obtained for the ternary all-PSCs, with a high Jsc over 25 mA cm−2 without sacrificing the Voc. Such result represents the highest efficiency reported for benzotriazole-based all-PSCs in the literature thus far. This study examined the linkage between xylem vulnerability, stomatal response to leaf water potential (ΨL), and loss of leaf turgor in eight species of seasonally dry tropical forest trees. In order to maximize the potential variation in these traits species that exhibit a range of leaf habits and phenologies were selected. It was found that in all species stomatal conductance was responsive to ΨL over a narrow range of water potentials, and that ΨL inducing 50% stomatal closure was correlated with both the ΨL inducing a 20% loss of xylem hydraulic conductivity and leaf water potential at turgor loss in all species. In contrast, there was no correlation between the water potential causing a 50% loss of conductivity in the stem xylem, and the water potential at stomatal closure (ΨSC) amongst species. It was concluded that although both leaf and xylem characters are correlated with the response of stomata to ΨL, there is considerable flexibility in this linkage. The range of responses is discussed in terms of the differing leaf‐loss strategies exhibited by these species.  相似文献

Land use induced changes of organic carbon storage in soils of China   总被引:29,自引:0,他引:29  
Using the data compiled from China's second national soil survey and an improved method of soil carbon bulk density, we have estimated the changes of soil organic carbon due to land use, and compared the spatial distribution and storage of soil organic carbon (SOC) in cultivated soils and noncultivated soils in China. The results reveal that ~ 57% of the cultivated soil subgroups ( ~ 31% of the total soil surface) have experienced a significant carbon loss, ranging from 40% to 10% relative to their noncultivated counterparts. The most significant carbon loss is observed for the non‐irrigated soils (dry farmland) within a semiarid/semihumid belt from northeastern to southwestern China, with the maximum loss occurring in northeast China. On the contrary, SOC has increased in the paddy and irrigated soils in northwest China. No significant change is observed for forest soils in southern China, grassland and desert soils in northwest China, as well as irrigated soils in eastern China. The SOC storage and density under noncultivated conditions in China are estimated to ~ 77.4 Pg (1015 g) and ~ 8.8 kg C m?2, respectively, compared to a SOC storage of ~ 70.3 Pg and an average SOC density of ~ 8.0 kg C m?2 under the present‐day conditions. This suggests a loss of ~ 7.1 Pg SOC and a decrease of ~ 0.8 kg C m?2 SOC density due to increasing human activities, in which the loss in organic horizons has contributed to ~ 77%. This total loss of SOC in China induced by land use represents ~ 9.5% of the world's SOC decrease. This amount is equivalent to ~ 3.5 ppmv of the atmospheric CO2 increase. Since ~ 78% of the currently cultivated soils in China have been degraded to a low/medium productivities and are responsible for most of the SOC loss, an improved land management, such as the development of irrigated and paddy land uses, would have a considerable potential in restoring the SOC storage. Assuming a restoration of ~ 50% of the lost SOC during the next 20–50 years, the soils in China would absorb ~ 3.5 Pg of carbon from the atmosphere.  相似文献
