The membrane electrical characteristics of stomatal guard cells in epidermal strips from Vicia faba L. and Commelina communis L. were explored using conventional electrophysiological methods, but with double-barrelled microelectrodes containing dilute electrolyte solutions. When electrodes were filled with the customary 1–3 M KCl solutions, membrane potentials and resistances were low, typically decaying over 2–5 min to near-30 mV and <0.2 k·cm2 in cells bathed in 0.1 mM KCl and 1 mM Ca2+, pH 7.4. By contrast, cells impaled with electrodes containing 50 or 200 mM K+-acetate gave values of-182±7 mV and 16±2 k·cm2 (input resistances 0.8–3.1 G, n=54). Potentials as high as (-) 282 mV (inside negative) were recorded, and impalement were held for up to 2 h without appreciable decline in either membrane parameter. Comparison of results obtained with several electrolytes indicated that Cl- leakage from the microelectrode was primarily responsible for the decline in potential and resistance recorded with the molar KCl electrolytes. Guard cells loaded with salt from the electrodes also acquired marked potential and conductance responses to external Ca2+, which are tentatively ascribed to a K+ conductance (channel) at the guard cell plasma membrane.Measurements using dilute K+-acetate-filled electrodes revealed, in the guard cells, electrical properties common to plant and fungal cell membranes. The cells showed a high selectivity for K+ over Na+ (permeability ratio PNa/PK=0.006) and a near-Nernstian potential response to external pH over the range 4.5–7.4 (apparent PH/PK=500–600). Little response to external Ca2+ was observed, and the cells were virtually insensitive to CO2. These results are discussed in the context of primary, charge-carrying transport at the guard cell plasma membrane, and with reference to possible mechanisms for K+ transport during stomatal movements. They discount previous notions of Ca2+-and CO2-mediated transport control. 采用经蔗糖密度梯度法纯化的大豆 (GlycinemaxL .)下胚轴质膜微囊为材料 ,分析了胰蛋白酶处理对质膜H ATPase钒酸钠抑制效应的影响。实验结果显示 ,温和胰蛋白酶处理显著提高H ATPase的ATP水解活力。并且发现酶切处理降低了钒酸钠对ATPase的抑制效应 ,当钒酸钠浓度为 2mmol/L时 ,ATPase活力仅被抑制 5 3.49% ,而未经酶切的对照组则被抑制 6 4.13%。ATP水解动力学分析表明 ,胰蛋白酶酶切处理既不影响ATP水解的Km 值也不影响钒酸钠的抑制类型 ,酶切前后的Km 值都等于 0 .34mmol/L ,并且都属于反竞争抑制。以上结果显示胰蛋白酶酶切处理可能改变了磷酸酶结构域的结构而影响了钒酸钠的抑制效应 ,暗示C_末端调节着磷酸酶结构域的结构和功能  相似文献

【目的】本文采用低温氧等离子体,在自行设计的远程等离子体反应装置中,对位于不同放电区域(放电区,余辉区,远程区)的模拟染菌载体聚对苯二甲酸乙二醇酯(PET)表面的铜绿假单胞菌(Pseudomonas aeruginosa)的杀灭效果和机理进行了研究。【方法】采用扫描电子显微镜观察了等离子体处理前后细菌细胞的形貌变化,采用考马斯亮蓝法测定了等离子体处理后细菌蛋白质泄漏量,采用双悬浮探针对氧等子体的电子温度和离子浓度以及电子自旋共振波谱对自由基浓度进行了测定。【结果】强放电区、余辉区和远程区处理30s后的灭菌效果分别为4.2 ,3.8和2.6;扫描电镜观察结果和蛋白泄漏量测定结果证明细菌细胞被损毁,在强放电区是电子、离子、自由基和紫外光子的协同作用,而在余辉区和远程区的灭杀作用主要因自由基所为。【结论】证明该反应装置可有效实现活性粒子的分离,在远程等离子体场中揭示了等离子体灭菌的规律和机理。  相似文献

Matrix metalloproteinases (MMPs) and their tissue inhibitors (TIMPs) play a major role in extracellular matrix component degradation in several normal and abnormal tissue situations; they are also found in human seminal plasma. MMPs have been found in rat prostate secretions and are nearly lobe specific in expression pattern. The aim of this study was to evaluate whether TIMP-2, like other semen components, is expressed differently from different rat prostatic lobes. Immunohistochemical staining was performed in both young and adult rat ventral (VP), lateral (LP), dorsal (DP), and anterior (AP) prostatic lobes and confirmed by western blotting. TIMP-2 expression was found in the epithelial cells in the following sequence: LP>AP>DP>VP, in both young and adult rats. In this study, 100% of adult LP presented histological signs of prostatitis, where TIMP-2 immunostaining was positive in normal epithelium even with intraluminal neutrophils, but was reduced or absent in the epithelium with intraepithelial leukocytes or with periductal stroma disorganization associated with mononuclear cell infiltration. However, TIMP-2 expression in LP was not induced by prostatitis, since younger rat LPs were also strongly TIMP-2 positive. The distal and intermediate VP regions were TIMP-2 negative, but the proximal regions were strongly stained. Western blotting results confirmed the high TIMP-2 expression in the LP lobe. Thus, TIMP-2 is expressed differently between the prostatic lobes and is another nearly lobe-specific protein, which plays a role in the regulation of MMP activity in seminal plasma and glandular homeostasis. TIMP-2 is also another regional ductal variation of VP. Further studies should address whether TIMP-2 expression is related to the highest incidence of rat LP prostatitis and adenocarcinoma.  相似文献

Telemetry studies of feral pigs (Sus scrofa, L.) in different habitats were used to predict home range size, from (i) body mass and (ii) population density. Geometric mean regressions of the loge transformed data indicated that body mass of male and female feral pigs and mean population density were good predictors of the home range size of feral pigs (r2 = 0.81, 0.77 and 0.85 respectively). There were no sexual differences in home range size of feral pigs once the effect of body mass was taken into account. Use of these variables in models that incorporate spatial aspects along with the practical application of the results to the management of feral pig populations are briefly discussed.  相似文献

The vocal behavior of captive animals is increasingly exploited as an index of well‐being. Here we show that the terrestrial predator alarm (TPA) vocalization, a robust and acoustically distinctive anti‐predation vocal response present in many mammal and bird species, offers useful information on the relative well‐being and stress levels of captive animals. In a 16‐week experiment evaluating the effects of varying levels of physical environmental enrichment (control < toys < foraging box < foraging box and toys) in the cages of eight singly housed adult male brown capuchins, we quantified the 1) emission rate of TPAs, 2) proportions of normal and abnormal behavior sample intervals, and 3) fecal and plasma cortisol levels. Variation in TPA emission across the experimental conditions was significant. We found significant reductions in the mean TPA production rate by the group in the enriched (toys, foraging box, and foraging box and toys) compared to the control condition; pre‐ and post‐experimental conditions, however, did not differ from the control condition. Mean TPA production by the group was also significantly positively correlated to mean group levels of fecal cortisol and proportion of abnormal behavior sample intervals, and significantly negatively correlated to the average proportion of normal behavior sample intervals in the group. Based on group means, plasma cortisol levels were positively, but not significantly, related to increasing TPA rate. At the level of the responses of an individual subject, however, the covariation between the vocal and non‐vocal behavioral measures and the cortisol assays seldom attained significance. Nevertheless, the direction of the relationships among these parameters within individual subjects typically mirrored those correlations based on group means. At both the group mean and individual levels, our results are consistent with the interpretation that in conditions of low environmental enrichment the study subjects were more stressed, and therefore more reactive to the presence of a threatening terrestrial stimulus (human observer), than when in more enriched conditions. We suggest that protocols to evaluate the effectiveness of enrichment for captive species other than brown capuchins could also profitably exploit TPAs as a first‐line monitor or as corroboratory evidence of current well‐being. p53 levels were assessed in lymphocytes and in blood plasma of workers from two Czech nuclear power plants (NPP): 114 subjects working in Temelin and 108 subjects working in Dukovany. Ionizing radiation (IR) exposure data were available for 64 and 59 subjects working in the monitored zones from the NPP in Temelin and Dukovany, respectively. The short-term doses of IR for these subjects were 0.01 and 0.12 mSv, and the long-term doses were 0.46 and 5.68 mSv, in the Temelin and Dukovany NPP, respectively. As a control group, 46 subjects living in České Budějovice, a city nearby the Temelin NPP, were analyzed. The concentration of p53 in lymphocytes was significantly higher in workers from the monitored zone in the Dukovany NPP (median value 6.4 pg/μg protein, P < 0.001) than in workers from the Temelin NPP (3.2 pg/μg) as well as in the control group (3.5 pg/μg). In contrast, plasma levels of p53 were comparable in the control group (median value 116 pg/ml plasma) and workers from the monitored zone of Dukovany NPP (102 pg/ml), but lower in workers from Temelin NPP (5 pg/ml). Other factors affecting p53 levels were studied. Smoking resulted in increased p53 lymphocyte levels. The effect of polymorphisms in metabolic and DNA repair genes on p53 levels was analyzed. The correlation was found between p53 levels in lymphocytes and p53 codon 72 polymorphism in subjects working in NPPs, but not in the control group.

The results of measurement p53 levels in lymphocytes suggest that this biomarker could reflect the short-term as well as long-term effects of low doses IR. Its impact on human health should be further explored.  相似文献

土壤盐碱化是造成农作物减产的主要原因之一。质膜Na^+/H^+逆向转运蛋白能够介导植物根部Na^+的外排和体内Na^+的长距离运输, 并能够调控细胞K+的稳态平衡及细胞内pH值和Ca^2+的转运, 因此其在植物耐盐性方面具有重要作用。该文概述了植物质膜Na^+/H^+逆向转运蛋白的分子结构、功能、表达调控及其与植物耐盐性关系等方面的研究进展, 并对今后有关该蛋白的主要研究方向作了分析和展望。  相似文献

α-L-Rhamnosidase (EC 3.2.1.40) catalyzes the hydrolytic release of rhamnose from polysaccharides and glycosides. Bacillus sp. GL1 α-L-rhamnosidase (RhaB), a member of glycoside hydrolase (GH) family 78, is responsible for degrading the bacterial biofilm gellan, and also functions as a debittering agent for citrus fruit in the food and beverage industries through the release of rhamnose from plant glycoside, naringin. The X-ray crystal structure of RhaB was determined by single-wavelength anomalous diffraction using a selenomethionine derivative and refined at 1.9 Å resolution with a final R-factor of 18.2%. As is seen in the homodimeric form of the active enzyme, the structure of RhaB in crystal packing is a homodimer containing 1908 amino acids (residues 3-956), 43 glycerol molecules, four calcium ions, and 1755 water molecules. The overall structure consists of five domains, four of which are β-sandwich structures designated as domains N, D1, D2, and C, and an (α/α)6-barrel structure designated as domain A. Structural comparison by DALI showed that RhaB shares its highest level of structural similarity with chitobiose phosphorylase (Z score of 25.3). The structure of RhaB in complex with the reaction product rhamnose (inhibitor constant, Ki = 1.8 mM) was also determined and refined at 2.1 Å with a final R-factor of 19.5%. Rhamnose is bound to the deep cleft of the (α/α)6-barrel domain, as is seen in the clan-L GHs. Several negatively charged residues, such as Asp567, Glu572, Asp579, and Glu841, conserved in GH family 78 enzymes, interact with rhamnose, and RhaB mutants of these residues have drastically reduced enzyme activity, indicating that the residues are crucial for enzyme catalysis and/or substrate binding. To our knowledge, this is the first report on the determination of the crystal structure of α-L-rhamnosidase and identification of its clan-L (α/α)6-barrel as a catalytic domain.  相似文献

Isolated hepatocytes release 2–3 nmol Mg2+/mg protein or ~10% of the total cellular Mg2+ content within 2 minutes from the addition of agonists that increase cellular cAMP, for example, isoproterenol (ISO). During Mg2+ release, a quantitatively similar amount of Ca2+ enters the hepatocyte, thus suggesting a stoichiometric exchange ratio of 1 Mg2+:1Ca2+. Calcium induced Mg2+ extrusion is also observed in apical liver plasma membranes (aLPM), in which the process presents the same 1 Mg2+:1Ca2+ exchange ratio. The uptake of Ca2+ for the release of Mg2+ occurs in the absence of significant changes in Δψ as evidenced by electroneutral exchange measurements with a tetraphenylphosphonium (TPP+) electrode or 3H-TPP+. Collapsing the Δψ by high concentrations of TPP+ or protonophore carbonyl cyanide p-trifluoromethoxyphenylhydrazone (FCCP) does not inhibit the Ca2+-induced Mg2+ extrusion in cells or aLPM. Further, the process is strictly unidirectional, serving only in Ca2+ uptake and Mg2+ release. These data demonstrate the operation of an electroneutral Ca2+/Mg2+ exchanger which represents a novel pathway for Ca2+ accumulation in liver cells following adrenergic receptor stimulation.
