The blue light dependent utilization of nitrate by green algae under common air and high irradiances, besides its assimilatory nature, is associated with the release of NO2 and NH4 + to the culture medium. If the CO2 content of the sparging air was increased up to 2%, previously excreted NO2 and NH4 + were rapidly assimilated. When under air and high irradiances the cell density in the culture reached values corresponding to 25 g Ch 1.ml-1, no further growth was observed and the highest values of NO3 consumption and NO2 and NH4 + release were attained. Besides low CO2 tensions, increasing NO3 concentrations in the medium stimulated the release of NO3 and NH4 +. Under CO2-free air the consumption of NO3 and the release of NO2 and NH4 + on a total N bases were almost stoichiometric and their rates saturated at much lower irradiances than under air. Under CO2-free air high rates of NO2 release were only observed under the blue radiations that were effectively absorbed by photosynthetically active pigments, i.e. 460 nm, but not under 404 and 630 nm radiations. However, the simultaneous illumination of the cells with 404 and 630 nm monochromatic light showed a remarkable synergistic effect on NO2 release.The results are discussed in terms of the close relationship between C and N metabolism, the photosynthetic reducing power required to convert NO inf3 sup± -N into R – NH2-N and the blue light activation of nitrate reductase.  相似文献

Unilateral irradiation with red light (R) or blue light (BL) elicits positive curvature of the mesocotyl of maize (Zea mays L.) seedlings raised under R for 2 d from sowing and kept in the dark for 1 d prior to curvature induction. The fluenceresponse curve for R-induced mesocotyl curvature, obtained by measuring curvature 100 min after phototropic induction, shows peaks in two fluence ranges, designated first positive range (from the threshold to the trough), and second positive range (above the trough). The fluence-response curve for BL is similar to that for R but shifted two orders of magnitude to higher fluences. Blue light elicits the classical first positive curvature of the coleoptile, whereas this response is not found with R. Positive mesocotyl curvature induced by either R or BL is eliminated by R given from above just before the unilateral irradiation, whereas BL-induced coleoptile curvature is not eliminated. The above results collectively offer evidence that phototropic curvature of the mesocotyl is induced by R-sensitive photosystem(s). Mesocotyl curvature in the second positive range is reduced by vertical far-red light (FR) applied after phototropic induction with R, but is not affected by FR applied before R. Unilateral irradiation with FR following vertical irradiation with a high R fluence leads to negative curvature of the mesocotyl. It is concluded that mesocotyl curvature in the second positive range results from a gradient in the amount of the FR-absorbing form of phytochrome (Pfr) established across the plant axis. Mesocotyl curvature in the first positive range is inhibited by vertical FR given either before or after phototropic induction with R. Since the FR used here is likely to produce more Pfr than the very low fluences of R eliciting the mesocotyl curvature in the first positive range, it is assumed that FR reduces the response in this case by adding Pfr at both sides of the plant axis. By rotating seedlings on a clinostat with its axis horizontal, the kinetics of mesocotyl curvature can be studied in the absence of a counteracting gravitropic response. On the clinostat, the R-induced mesocotyl curvature develops after a lag, through two successive phases having different curvature rates, the late phase is slower than the early phase. Negative curvature of the coleoptile can be induced by either R or BL; the BL-induced negative curvature is found at fluences higher than those giving positive curvature. Abstract Washed cell suspensions of Crithidia oncopelti oxidizing a variety of substrates gave complex plots for the inhibition of respiration by potassium cyanide or azide. The data indicated the presence of at least two and possibly three terminal oxidases on the basis of their differential sensitivity to these inhibitors. The oxidase most sensitive to cyanide, azide and CO accounted for approx. 65–70% of whole cell respiration and is probably cytochrome oxidase a/a3. A second oxidase exhibiting low affinity for CO required high concentrations of KCN or azide for inhibition. This haemoprotein had the spectral characteristics of cytochrome o and accounted for 15–20% of cell respiration. Incomplete inhibition of respiration by high concentrations of KCN or azide suggested the presence of a third oxidase which was CO-unreactive.  相似文献

The fluence response of the blue light induced carotenoid synthesis inNeurospora is biphasic. Using fluence rates between 0.3 and 40 Wm-2, increasing illumination times beyond 16 min (at 20°C) result in a second rise of the amount of carotenoids synthesized in the subsequent dark period. On altering the temperature, the transition point to the second phase of the response is shifted to shorter/longer illumination periods with increasing/decreasing temperature, respectively. The transition point can also be shifted by administering high fluence rates of near UV light: The start of the second phase is already triggered after an irradiation time of 2 min. The findings suggest that elements of the transduction sequence become depleted and senstivity recovers in a temperature-dependent process. The effect of light on carotene accumulation was studied by analyzing the -carotene content of 4--old mycelia continuously exposed to illumination of different intensities. The wild-type, mutants defective in phototropism, mutants defective in carotene regulation, and newpic mutants specifically defective for photocarotenogenesis were examined. The results indicate that photocarotenogenesis depends on a single sensory pathway which shares its earlier steps (governed by genesmadA andmadB) with the sensory pathway for phototropism. It shares its later steps (probably governed by genescarA andpicB) with one of the pathways for carotene regulation, and includes at least one specific step (governed by genepicA) not known to be involved in other responses.  相似文献

Mild sonication was used to obtain single cell suspensions of Paracoccidioides brasiliensis. These cells were intact by microscopic criteria. Direct cell counts in a given inoculum and colony formation on various media were used to determine plating efficiency. Sonicated and nonsonicated cell suspensions were used to study plating efficiency and to estimate viability by means of vital dyes. Methylene blue, Erythrosin B, and Janus green were unreliable when used with P. brasiliensis, but vital dyes were accurate when tested with Candida albicans.Acridine orange gave more meaningful results of viability. Estimates of viability, however, changed significantly as a result of relatively minor alterations in the composition of the suspending medium.In initial experiments, the plating efficiency of P. brasiliensis was dismally low. It descended abruptly with increasing dilution of inoculum. Efficiency was much improved if horse serum was added to brain heart infusion plates or if glucose glycine yeast extract (GGY) plates were incubated at room temperature and mycelial colonies were counted. With the technique we report, current plating efficiency of sonicated suspensions is of the order of 25 %. Our results and procedures have an important bearing upon those studies concerned with in vitro killing of P. brasiliensis in suspensions or with isolating this fungus from clinical or environmental specimens.  相似文献

Sandoz 6706 pretreatment of white light grown barley seedlings causes a 60% increase in saponarin (6-C-glucosyl-7-O-glucosylapigenin) but a 300% increase in lutonarin (3′-hydroxysaponarin). Norflurazon has little effect on saponarin levels but is almost as effective as Sandoz 6706 in enhancing lutonarin net synthesis. Barley roots contain saponarin and lutonarin only after herbicide treatment. Mung bean seedlings respond to Sandoz 6706 by accumulating higher levels of rutin and delphinidin 3-glucoside. The results are discussed in relation to the site of action of the herbicides, the High Energy photoresponse, and control of flavonoid 3′-hydroxylation.  相似文献

Oxidation of ferrocytochrome c by molecular oxygen catalysed by cytochrome c oxidase (cytochrome aa3) is coupled to translocation of H+ ions across the mitochondrial membrane. The proton pump is an intrinsic property of the cytochrome c oxidase complex as revealed by studies with phospholipid vesicles inlayed with the purified enzyme. As the conformation of cytochrome aa3 is specifically sensitive to the electrochemical proton gradient across the mitochondrial membrane, it is likely that redox energy is primarily conserved as a conformational “strain” in the cytochrome aa3 complex, followed by relaxation linked to proton translocation. Similar principles of energy conservation and transduction may apply on other respiratory chain complexes and on mitochondrial ATP synthase.  相似文献

SYNOPSIS. Four new species of Henneguya (myxosporidan) are described from Ictalurus punctatus Rafinesque (channel catfish). They are as follows: Henneguya postexilis sp. n. and Henneguya longicauda sp. n. from the gills; Henneguya adiposa sp. n. from the adipose fin; and Henneguya diversis sp. n. from the liver, kidney, connective tissue of muscles and fins, and tumor-like external growths. Henneguya pellis sp. n. is described from the dermis of Ictalurus furcatus (LeSueur) (blue catfish). The development stages of all described species are discussed. Henneguya exilis Kudo was found on the gills of one I. punctatus; notes on its spore characteristics are presented.  相似文献

Light-induced chloroplast movements were found to cause changes in chlorophyll fluorescence emission, closely matching those in leaf absorptance, both in terms of the kinetics and the maximum extent of the changes observed in different species. The results demonstrate that chloroplast movements can have a significant effect on the efficiency of light utilization in photosynthesis. They further show that chloroplast movements need to be taken into account in measurements of fluorescence quenching and especially in measurements of light-induced optical changes used to monitor zeaxanthin formation and pH associated light scattering in leaves.
