The fine structure of the green stem of Dianthus caryophyllus, the leaf petiole and the flower pedicel of Zantedeschia aethiopica were studied using light and scanning electron microscopy. It was revealed that these non-foliar plant parts of both species possess epidermis with numerous stomata. Stomatal density of D. caryophyllus stem was found to be relatively high (79 vs 100 per mm2 found on leaf surface). Z. aethiopica petioles and pedicels also possess numerous stomata (17 per mm2), yet stomatal density was found to be about half of that of leaves. Anatomical differences observed between petioles and pedicels were only minor. Stems of D. caryophyllus as well as petioles and pedicels of Z. aethiopica have a chlorenchyma-type tissue whose fine structure is quite similar to the leaf palisade chlorenchyma. Yet, the palisade of Z. aethiopica petioles and pedicels shows a peculiar arrangement: palisade cells are arranged with their long axis parallel to the longitudinal organ axis. Palisade tissue found in the aforementioned non-foliar plant organs in both species shows strong red chlorophyll auto-fluorescence under epi-fluorescence optics, consists of cells with abundant chloroplasts, possesses high percentage of intercellular spaces (13 and 20%, respectively) and its cells expose considerable part of their surface to the intercellular air. The fine structure of this stem palisade tissue along with the abundance of functional stomata found on the epidermis may support efficient photosynthesis.  相似文献

For almost 150 years, the two monotypic genera Trochodendron and Tetracentron (Trochodendraceae) have been considered to share an unusual and primitive feature in angiosperms - the lack of vessels in their wood. Therefore, they have been classified in a basal position in the angiosperms. Our observations by light microscopy, low-vacuum environmental scanning electron microscopy (ESEM) and high-vacuum scanning electron microscopy (SEM) both in fresh and FAA-fixed materials consistently showed the presence of tracheary elements differentiated into two types in both genera. In Trochodendron, the tracheary elements can be divided into perforate vessel elements and imperforate fiber-tracheids and tracheids. The vessel elements show end and lateral walls. The pits on the end walls are elongate- broadened and do not have membranes or only a few remnants of them forming the perforation plates. The fiber-tracheids show crossfield pit pairs and sharp ends, and the tracheids show bordered pits. In Tetracentron, the tracheary elements comprise vessel elements and fibers. The vessel elements are similar to those of Trochodendron, whereas the fibers have no crossfield pit pairs but, rather, elliptical pits and sharp ends. Thus, both Trochodendron and Tetracentron are vessel bearing rather than vesselless, although their vessel elements are primitive.  相似文献

Background and Aims

Cleomaceae is one of 19 angiosperm families in which C4 photosynthesis has been reported. The aim of the study was to determine the type, and diversity, of structural and functional forms of C4 in genus Cleome.

Methods

Plants of Cleome species were grown from seeds, and leaves were subjected to carbon isotope analysis, light and scanning electron microscopy, western blot analysis of proteins, and in situ immunolocalization for ribulose bisphosphate carboxylase oxygenase (Rubisco) and phosphoenolpyruvate carboxylase (PEPC).

Key Results

Three species with C4-type carbon isotope values occurring in separate lineages in the genus (Cleome angustifolia, C. gynandra and C. oxalidea) were shown to have features of C4 photosynthesis in leaves and cotyledons. Immunolocalization studies show that PEPC is localized in mesophyll (M) cells and Rubisco is selectively localized in bundle sheath (BS) cells in leaves and cotyledons, characteristic of species with Kranz anatomy. Analyses of leaves for key photosynthetic enzymes show they have high expression of markers for the C4 cycle (compared with the C3–C4 intermediate C. paradoxa and the C3 species C. africana). All three are biochemically NAD-malic enzyme sub-type, with higher granal development in BS than in M chloroplasts, characteristic of this biochemical sub-type. Cleome gynandra and C. oxalidea have atriplicoid-type Kranz anatomy with multiple simple Kranz units around individual veins. However, C. angustifolia anatomy is represented by a double layer of concentric chlorenchyma forming a single compound Kranz unit by surrounding all the vascular bundles and water storage cells.

Conclusions

NAD-malic enzyme-type C4 photosynthesis evolved multiple times in the family Cleomaceae, twice with atriplicoid-type anatomy in compound leaves having flat, broad leaflets in the pantropical species C. gynandra and the Australian species C. oxalidea, and once by forming a single Kranz unit in compound leaves with semi-terete leaflets in the African species C. angustifolia. The leaf morphology of C. angustifolia, which is similar to that of the sister, C3–C4 intermediate African species C. paradoxa, suggests adaptation of this lineage to arid environments, which is supported by biogeographical information.  相似文献

Background and Aims

Our knowledge about the influences of environmental factors on tree growth is principally based on the study of dominant trees. However, tree social status may influence intra-annual dynamics of growth, leading to differential responses to environmental conditions. The aim was to determine whether within-stand differences in stem diameters of trees belonging to different crown classes resulted from variations in the length of the growing period or in the rate of cell production.

Methods

Cambial activity was monitored weekly in 2006 for three crown classes in a 40-year-old silver-fir (Abies alba) plantation near Nancy (France). Timings, duration and rate of tracheid production were assessed from anatomical observations of the developing xylem.

Key Results

Cambial activity started earlier, stopped later and lasted longer in dominant trees than in intermediate and suppressed ones. The onset of cambial activity was estimated to have taken 3 weeks to spread to 90 % of the trees in the stand, while the cessation needed 6 weeks. Cambial activity was more intense in dominant trees than in intermediate and suppressed ones. It was estimated that about 75 % of tree-ring width variability was attributable to the rate of cell production and only 25 % to its duration. Moreover, growth duration was correlated to tree height, while growth rate was better correlated to crown area.

Conclusions

These results show that, in a closed conifer forest, stem diameter variations resulted principally from differences in the rate of xylem cell production rather than in its duration. Tree size interacts with environmental factors to control the timings, duration and rate of cambial activity through functional processes involving source–sink relationships principally, but also hormonal controls.  相似文献

Most birds use at least two modes of locomotion: flying and walking (terrestrial locomotion). Whereas the wings and tail are used for flying, the legs are mainly used for walking. The role of other body segments remains, however, poorly understood. In this study, we examine the kinematics of the head, the trunk, and the legs during terrestrial locomotion in the quail (Coturnix coturnix). Despite the trunk representing about 70% of the total body mass, its function in locomotion has received little scientific interest to date. This prompted us to focus on its role in terrestrial locomotion. We used high-speed video fluoroscopic recordings of quails walking at voluntary speeds on a trackway. Dorso-ventral and lateral views of the motion of the skeletal elements were recorded successively and reconstructed in three dimensions using a novel method based on the temporal synchronisation of both views. An analysis of the trajectories of the body parts and their coordination showed that the trunk plays an important role during walking. Moreover, two sub-systems participate in the gait kinematics: (i) the integrated 3D motion of the trunk and thighs allows for the adjustment of the path of the centre of mass; (ii) the motion of distal limbs transforms the alternating forward motion of the feet into a continuous forward motion at the knee and thus assures propulsion. Finally, head bobbing appears qualitatively synchronised to the movements of the trunk. An important role for the thigh muscles in generating the 3D motion of the trunk is suggested by an analysis of the pelvic anatomy.  相似文献

Myocardial infarction is one of the leading causes of death in the Western world. The similarity of the mouse heart to the human heart has made it an ideal model for testing novel therapeutic strategies.In vivo magnetic resonance imaging (MRI) gives excellent views of the heart noninvasively with clear anatomical detail, which can be used for accurate functional assessment. Contrast agents can provide basic measures of tissue viability but these are nonspecific. Positron emission tomography (PET) is a complementary technique that is highly specific for molecular imaging, but lacks the anatomical detail of MRI. Used together, these techniques offer a sensitive, specific and quantitative tool for the assessment of the heart in disease and recovery following treatment.In this paper we explain how these methods are carried out in mouse models of acute myocardial infarction. The procedures described here were designed for the assessment of putative protective drug treatments. We used MRI to measure systolic function and infarct size with late gadolinium enhancement, and PET with fluorodeoxyglucose (FDG) to assess metabolic function in the infarcted region. The paper focuses on practical aspects such as slice planning, accurate gating, drug delivery, segmentation of images, and multimodal coregistration. The methods presented here achieve good repeatability and accuracy maintaining a high throughput.  相似文献

The impressive roaring of adult male muskoxen most often occurs during rutting contests. Roaring in adult females is primarily relevant to mother–infant communication. Loud roars are produced by taking up a specific roaring posture. Acoustic recordings were made in a small herd of zoo muskoxen during three successive rutting seasons. Earlier recordings of a different herd were used for comparison. Head-and-neck specimens were subjected to vascular injection, macroscopic anatomical dissection, computer tomographic analysis and skeletonization. Isolated preserved larynges of young animals were dissected for ontogenetic comparison. Despite a pronounced sexual dimorphism of head mass, larynx size is almost identical in adult male and female muskoxen, as is the fundamental frequency of their roars. Remarkably, the larynges of both sexes of muskoxen are provided with an unpaired ventrorostral ventricle. Probably, this ventricle is inflated during the initial phase of a roar. The ventricle may have two functions: to increase the amplitude of roaring and to darken the timbre of the roars by acting as an additional resonance space. The vocal fold of adult female and young individuals has a sharp rostral edge and a vocal ligament is still present. During male ontogeny the vocal ligament becomes transformed into a large fat pad extending into the wall of the laryngeal vestibulum. Accordingly, the glottic region in the adult male lacks any sharp edges of the mucosa. In both sexes the thyroarytenoid muscle is divided into three portions. A single roar may comprise phases of different sound volume. The roars of both muskox sexes are characterized by a pulsed structure. We suggest that two oscillating systems are involved in the production of roars: one comprising only the medial portion of the vocal fold and one including its lateral portion.  相似文献
