Birefringence can be induced in liposome suspensions using electric fields. The fields interact predominantly with anisotropic electrical polarisabilities which give rise to induced dipole moments. Using pulsed electric fields, the optical and electrical polarisabilities and the geometrical size of the liposomes can be measured simultaneously. These parameters have been found to be very sensitive to the presence of small amounts of fluidising additives of polar and ionic nature. Illustrative data are presented for the influence of the amines ammonium chloride, methyl ammonium chloride and lignocaine and of benzyl alcohol on phosphatidylcholine/serine liposomes. Structural changes in the vesicle membranes were detected, which appeared to correlate with the biological functions, thus indicating that electric birefringence is a rapid and useful method for studying interactive phenomena in lipid membrane systems.  相似文献

Complexes of DNA with benzocrown derivatives of actinocin were studied by viscometry and dynamic birefringence. Changes in the macromolecular structure of DNA caused by complex formation were determined. Models of DNA binding to the studied compounds were suggested on the basis of data obtained. The intercalation of actinocin chromophore of benzocrown derivatives of actinocin was shown to occur only when benzocrown groups were bound to the chromophore via the glycine spacer. A change of the distance between the crown group and the chromophore prevented ligand intercalation. Increase of the ionic strength resulted in appearance of a new, nonintercalative binding mode for crown-containing compounds determined by interaction of the crown groups with DNA.  相似文献

A polarization‐sensitive spectral domain optical coherence tomography (PS‐SD‐OCT) system is used to measure phase retardation and birefringence of the human retinal nerve fiber layer (RNFL) in vivo. The instrument records three parameters simultaneously: intensity, phase retardation and optic‐axis orientation. 3D data sets are recorded in the optic nerve‐head area of a healthy and a glaucomatous eye, and the results are presented in various ways: En‐face phase‐retardation maps of the RNFL are generated from the recorded 3D data and results are compared with scanning laser polarimetry (SLP). The depth information provided by OCT is used to segment the RNFL in the intensity image and measure the RNFL thickness. From the retardation and thickness data, 2D birefringence maps of the RNFL are derived. 我们研制了一种基于光纤的位相分辨偏振灵敏光学相干层析成像系统。该系统中的偏振状态控制设量在参考臂而非光源臂上,因而使得光抵达样品的传输效率大大提高。鉴于光源的部分偏振性,入射于样品上的光含有任意偏振状态的分量,通过对参考光偏振状态的调制,就可相干地提取对应于入射光四种正交偏振状态并经样品后向散射的光信号。基于斯托克斯矢量夹角在无损光纤系统传输的变换不变性,我们能利用测量臂中光信号的斯托克斯参数来确定双折射样品深度分辨的位相延迟信息。利用所研制的偏振灵敏光学相干层析成像系统,不仅确认了韧带和软骨的双折射性质,而且定量分析了不同条件下韧带的双折射变化.研究结果表明:韧带松弛可使其双折射特性明显减弱,而韧带经拉伸后,其双折射特性的变化却不明显。  相似文献

We have used a combination of techniques to study the structure and properties of C-type starch from pea seeds. It was found that all C-type starch granules contain both types of polymorph; the B polymorphs are in the center of the granule and are surrounded by the A polymorphs. During heating in excess salt solution the A and B polymorphs within C-type granules melt independently, giving a double transition in heat capacity and a two-step swelling, compared with single transitions for A- and B-type starches. It was shown that B polymorphs gave a transition with a lower peak temperature than A. The disruption of crystallinity during gelatinization began from the hilum area and was propagated along the granule, accompanied by swelling of disrupted areas. It is proposed that the swelling of disrupted parts of the granule decreases the melting temperature of the neighboring crystallites resulting in the progressive disruption of crystalline areas. Pathological crystal identification is routinely practiced in rheumatology for diagnosing arthritis disease such as gout, and relies on polarized light microscopy as the gold standard method used by medical professionals. Here, we present a single‐shot computational polarized light microscopy method that reconstructs the transmittance, retardance and slow‐axis orientation of a birefringent sample using a single image captured with a pixelated‐polarizer camera. This method is fast, simple‐to‐operate and compatible with all the existing standard microscopes without extensive or costly modifications. We demonstrated the success of our method by imaging three different types of crystals found in synovial fluid and reconstructed the birefringence information of these samples using a single image, without being affected by the orientation of individual crystals within the sample field‐of‐view. We believe this technique will provide improved sensitivity, specificity and speed, all at low cost, for clinical diagnosis of crystals found in synovial fluid and other bodily fluids.  相似文献

The leaf hairs (trichomes) on the aerial surface of many plant species play important roles in phytochemical production and herbivore protection, and have significant applications in the chemical and agricultural industries. Trichome formation in the model plant Arabidopsis thaliana also presents a tractable experimental system to study cell differentiation and pattern formation in plants and animals. Studies of this developmental process suggest that trichome positioning may be the result of a self‐forming pattern, emerging from a lateral inhibition mechanism determined by a network of regulatory factors. Critical to the continued success of these studies is the ability to quantitatively characterize trichome pattern phenotypes in response to mutations in the genes that regulate this process. Advanced protocols for the observation of changes in trichome patterns can be expensive and/or time consuming, and lack user‐friendly analysis tools. In order to address some of these challenges, we describe here a strategy based on polarized light microscopy for the quick and accurate measurement of trichome positions, and provide an online tool designed for the quantitative analyses of trichome number, density and patterning.  相似文献

The complex refractive indices of two benchmark nematic liquid crystal,4-4 -n-pentyl-cyanobiphenyl (5CB) and 4-(trans-4pentylcyclohexyl)-benzonitrile (PCH5) have been determinedin the frequency range from 0.2 to 0.8 THz. The technique of coherent THztime-domain spectroscopy (THz-TDS) was used. We show that the birefringenceof 5CB is in the range of 0.15 to 0.21, while that of PCH is from 0.01 to0.08. Both liquid crystals exhibit relatively small absorption in thisfrequency range. The large birefringence of 5CB indicates possible applications of liquid-crystal-based devices for modulation and polarizationcontrol of electromagnetic radiation in the THz frequency range.  相似文献

The interactions of DNA with histone H4 and with its fragments N-H4 (1-84) and C-H4 (85-102) have been studied by using electrooptical techniques, viscosity and electron microscopy. Electron microscopy reveals that histone H4 induces a large folding of DNA molecules : this is in agreement with electrooptical measurements which indicate that, with the increase of their ratio, H4/DNA complexes undergo a gradual process of condensation. Viscosity measurements show that complexes at ratios up to 0.20-0.25 become more rigid as compared to DNA. It appears that C-H4, and not the N-H4 fragment, causes a great distorsion to the structure of DNA, accompanied by an increase of rigidity at ratios up to 0.20-0.25, as occurs for H4/DNA complexes. Electrooptical studies of C-H4/DNA complexes show, along a range of histone/DNA ratios, an important permanent dipole component. These effects reveal a particular mode of interaction of C-H4 with DNA, indicating that some charged residues of the peptide are kept distant enough from the DNA backbone. As no dipole character, in addition to that shown for DNA, has been detected for H4/DNA complexes, it is concluded that the conformation of the H4 molecule modifies to some extent the interaction of the C-terminal region. Our results show that this histone, and particularly its C-terminal region, is important as a determinant factor in the folding of DNA within artificial complexes.  相似文献
