Pathogenicity of eight Bacillus strains to seedlings of four cotton cultivars was evaluated under greenhouse conditions. Each of the tested cultivars was individually treated with powdered inoculum of each bacterial strain. Untreated seeds were planted as control treatments in autoclaved soil. Effects of the tested strains on levels and activities of some biochemical components of the infected seedlings were also assayed. The biochemical components included total soluble sugars, total soluble proteins, total free amino acids, peroxidase, polyphenol oxidase, phenols, and lipid peroxidation. ANOVA showed that Bacillus strain (B) was a very highly significant source of variation in damping-off and dry weight. Cotton cultivar (V) was a nonsignificant source of variation in damping-off while it was a significant source of variation in dry weight. B × V interaction was a significant source of variation in damping- off and a nonsignificant source of variation in dry weight. Bacillus strain was the most important source of variation as it accounted for 59.36 and 64.99% of the explained (model) variation in damping-off and dry weight, respectively. The lack of significant correlation between levels and activities of the assayed biochemical components and incidence of damping-off clearly demonstrated that these biochemical components were not involved in the pathogenicity of the tested strains. Therefore, it was hypothesized that the pathogenicity of the tested strains could be due to the effect of cell wall degrading enzymes of pathogenic toxins. Based on the results of the present study, Bacillus strains should be considered in studying the etiology of cotton seedling damping-off. 相似文献

以低能氮离子束为诱变源,对甘草种进行注入处理后,以萌发的幼苗为材料对其在干旱胁迫条件下保护酶活性及MDA含量变化进行研究.结果表明注入后甘草种的存活率呈现马鞍形曲线变化,其中在4.1×1016N+/cm2剂量处达到鞍点,在剂量4.6×1016N+/cm2处存活率高于对照.在干旱胁迫下,随着胁迫时间的延长,离子束注入后萌发的幼苗MDA含量明显低于对照.酶活结果显示,在干旱胁迫后期,剂量4.6×1016N+/cm2处理后的甘草幼苗SOD和CAT酶活性都高于对照,说明该注入剂量处理甘草种有助于提高甘草幼苗的抗旱性能,SOD同工酶结果也证明该剂量的离子注入对干旱胁迫下同工酶谱带有所影响. 相似文献

目的了解临床分离的大肠埃希菌耐药性及氨基糖苷类修饰酶(AMEs)基因的存在状况。方法测定临床分离的60株大肠埃希菌对19种抗菌药物的敏感性,采用PCR技术检测氨基糖苷类修饰酶基因。结果60株大肠埃希菌呈现多重耐药,氨基糖苷类修饰酶基因aac(3)-Ⅱ、aac(6′)-Ⅰb、aac(6′)-Ⅱ、ant(3′′)-Ⅰ、ant(2′′)-Ⅰ的阳性率分别为36.7%、18.3%、0%、10%、1.6%。携带1种或1种以上基因的菌株有33株(55%)。结论临床分离的大肠埃希菌多重耐药严重,氨基糖苷类修饰酶基因携带率较高。 相似文献

咖啡因是从茶叶、咖啡果中提取出来的一种生物碱,常用于治疗神经衰弱和昏迷复苏。咖啡因体内代谢过程复杂,现已发现咖啡因的15种代谢产物,参与代谢过程的酶类也被逐一证实。尿液咖啡因代谢产物比率(UCMRs)常用于多种药物代谢酶活性的评估,主要包括CYP1A2、CYP2A6、N-乙酰转移酶和黄嘌呤氧化酶,咖啡因及其代谢产物检测方法也逐步得到改进与更新。总之,咖啡因与人类健康密切相关,以及其重要的科研应用价值,一直受到普遍关注。 相似文献

4-Hydroxynonenal (HNE) is one of the major end products of lipid peroxidation and may have either physiological or pathological significance regulating cell proliferation. We studied some biochemical effects of HNE, at various concentrations (0.1-100 μM), on Jurkat T cells incubated thereafter for 24, 48 and 72 h. HNE at low concentrations significantly enhanced the proliferation index, whereas at higher concentrations progressively blocked cell proliferation. Caspase 3 activity increased significantly at HNE concentrations between 1 and 10 μM and decreased at higher concentrations. Superoxide dismutase (SOD), glutathione peroxidase (GSH-Px) and glutathione reductase (GSH-Rd) increased progressively with HNE concentrations, particularly GSH-Px. Glucose-6-phosphate dehydrogenase (G6PDH) showed a different pattern, increasing at low HNE (1-5 μM) concentrations and rapidly declined thereafter. These results show that HNE may induce growth inhibition of Jurkat T cells and regulate the activity of typical antioxidant enzymes. Furthermore, the protective effect of doubling the foetal calf serum still points out the risk that cultured cells undergo oxidative stress during incubation. 相似文献

比较了内蒙古高原荒漠区4种锦鸡儿属(Caragana)优势植物——柠条锦鸡儿、狭叶锦鸡儿、垫状锦鸡儿和荒漠锦鸡儿的生理适应特性。研究发现:4种锦鸡儿属植物中,荒漠锦鸡儿的叶片含水量、束缚水/自由水比值、水势以及水分利用效率最低,气孔导度日变化表现为与气温变化相似的早晚低、中午高的单峰曲线。4种锦鸡儿蒸腾速率日变化状况相似,均在10:00达到最大,以后逐渐降低,但日蒸腾积累值垫状锦鸡儿<荒漠锦鸡儿<柠条锦鸡儿<狭叶锦鸡儿。相比较,柠条锦鸡儿、狭叶锦鸡儿和垫状锦鸡儿对当地的水分条件有良好的适应性,能够保持较好的水分状况,水分亏缺在15%以下,而荒漠锦鸡儿叶水分亏缺日变幅较大,保水能力不及其它3种。渗透调节比较研究发现:荒漠锦鸡儿的渗透势最低,细胞质离子浓度最高,无机渗透调节物产生的渗透势所占的比例也更大,说明其低渗透势的维持主要来自无机渗透调节物质的较多积累。保护酶和自由基比较研究发现:POD和SOD活性荒漠锦鸡儿明显高于其它3个种,但CAT活性在4种植物中无显著差别;叶片自由基含量狭叶锦鸡儿>荒漠锦鸡儿>柠条锦鸡儿>垫状锦鸡儿。这些结果表明:(1)荒漠锦鸡儿对干旱环境的适应方式与其它3种不同,柠条锦鸡儿、狭叶锦鸡儿和垫状锦鸡儿以强保水能力、维持稳定的水分而适应,而荒漠锦鸡儿以更负的渗透势、更高细胞质离子浓度弥补其更多的水分消耗和对水分变化的强耐性而适应,较高的保护酶活性可能是其强耐性的生理基础之一。(2)保水能力弱的锦鸡儿种主要通过无机离子的积累,调节细胞质渗透势,保持水分平衡,这是一种相对节省能量的适应对策。 相似文献

The effects of excess copper (Cu) on the accumulation of hydrogen peroxide (H2O2) and antioxidant enzyme activities in roots of the Cu accumulator Elsholtzia haichowensis Sun were investigated. Copper at 100 and 300 μM significantly increased the concentrations of malondialdehyde and H2O2, and the activities of catalase (E.C. 1.11.1.6), ascorbate peroxidase (E.C. 1.11.1.11), guaiacol peroxidase (GPOD, E.C. 1.11.1.7)
and superoxide dismutase (SOD, E.C. 1.15.1.1). Isoenzyme pattern and inhibitor studies showed that, among SOD isoforms, only
copper–zinc superoxide dismutase (CuZn–SOD) increased. Excess Cu greatly increased the accumulation of superoxide anion (O2
·−) and H2O2 in E. haichowensis roots. This study also provides the first cytochemical evidence of an accumulation of H2O2 in the root cell walls as a consequence of Cu treatments. Experiments with diphenyleneiodonium as an inhibitor of NADPH oxidase,
1,2-dihydroxybenzene-3,5-disulphonic acid as an O2
·− scavenger, and N-N-diethyldithiocarbamate as an inhibitor of SOD showed that the source of H2O2 in the cell walls could partially be NADPH oxidase. The enzyme can use cytosolic NADPH to produce O2
·−, which rapidly dismutates to H2O2 by SOD. Apoplastic GPOD and CuZn–SOD activities were induced in roots of E. haichowensis with 100 μM Cu suggesting that these two antioxidant enzymes may be responsible for H2O2 accumulation in the root apoplast. 相似文献

Antioxidant enzyme (AE) activities were studied in normal hamster kidney proximal tubules and in estrogen-induced hamster kidney cancer. In vivo, kidney tumor had lower activities of manganese superoxide dismutase (MnSOD), copper, zinc superoxide dismutase, catalase, and glutathione peroxidase than kidney proximal tubules. Differences in AE activities were, in general, maintained in tissue culture, with AE activities remaining low in tumor cells compared to normal cells. Normal proximal tubular cells showed significant induction of MnSOD activity as a function of time in culture of following exposure to diethylstilbestrol, a synthetic estrogen, while MnSOD activity remained low in tumor cells under these conditions. Our results suggest that antioxidant enzymes, particularly MnSOD, are regulated differently in estrogen-induced hamster kidney tumor cells than in normal kidney proximal tubular cells, demonstrating that cancers arising from hormonal influence have similar AE profiles to those previously described in cancers arising from viral or chemical etiologies. 相似文献

Efficient polysaccharide degradation depends on interaction between enzymes acting on the main chain and the side chains. Previous studies demonstrated cooperation between several enzymes, but not all enzyme combinations have been explored. A better understanding of enzyme cooperation would enable the design of better enzyme mixtures, optimally profiting from synergistic effects. In this study, we analyzed the cooperation of several enzymes involved in the degradation of xylan, glucan, xyloglucan and crude plant biomass from Aspergillus nidulans by single and combined incubations with their polymeric substrate. Positive effects were observed between most enzymes, although not always to the same extent. Moreover, the tailor made cocktails formulated in this study resulted in efficient release of glucose from plant biomass. This study also serves as an example for the complex cooperation that occurs between enzymes in plant biomass saccharification and how expression in easily‐accessible hosts, such as Pichia pastoris, can help in revealing these effects. 相似文献
