The coal fields of Pakistan and their microbiology have not been fully explored. Therefore, a study was conducted on the coal mines of Dara Adam Khel located in the Federally Administered Tribal Areas of Pakistan. For this purpose, sampling was done from nine different mines with varying depths. A total of 32 bacterial strains were isolated and their colony size, form, texture, color, margin, elevation and opacity were noted. The majority of the strains (75%) were found Gram negative. The bacterial strains were then characterized in detail by different biochemical tests including catalase, citrate, oxidase, indole, triple sugar iron, motility, methyl red-Vogues Proskeur, nitrate reduction and phenylalanine deaminase, and an enormous physiological diversity was observed. The Gram positive strains were further characterized on molecular level using 16S rRNA gene amplification and sequence analysis. Based on molecular analysis, seven strains were identified as Bacillus tequilensis, B. cereus, Janibacter melonis, Kocuria atrinae, B. anthracis, K. rosea and B. simplex. The other two strains (strains 6 and 41) had molecular similarity of only 98% and 97% with Brachybacterium spp. and Arthrobacter spp. respectively. The phylogenetic analysis further suggested that the strains 6 and 41 may be potential candidates for novel species; however, further work is needed for confirmation.  相似文献

Endothelial dysfunction is associated with increase in oxidative stress and low NO bioavailability. The endothelial NO synthase (eNOS) uncoupling is considered an important factor in endothelial cell oxidative stress. Under increased oxidative stress, the eNOS cofactor tetrahydrobiopterin (BH4) is oxidized to dihydrobiopterin, which competes with BH4 for binding to eNOS, resulting in eNOS uncoupling and reduction in NO production. The importance of the ratio of BH4 to oxidized biopterins versus absolute levels of total biopterin in determining the extent of eNOS uncoupling remains to be determined. We have developed a computational model to simulate the kinetics of the biochemical pathways of eNOS for both NO and O2•− production to understand the roles of BH4 availability and total biopterin (TBP) concentration in eNOS uncoupling. The downstream reactions of NO, O2•−, ONOO, O2, CO2, and BH4 were also modeled. The model predicted that a lower [BH4]/[TBP] ratio decreased NO production but increased O2•− production from eNOS. The NO and O2•− production rates were independent above 1.5 μM [TBP]. The results indicate that eNOS uncoupling is a result of a decrease in [BH4]/[TBP] ratio, and a supplementation of BH4 might be effective only when the [BH4]/[TBP] ratio increases. The results from this study will help us understand the mechanism of endothelial dysfunction.  相似文献

土地同时具有生态功能、生产功能和生活功能,相应地具有生态-生产-生活综合承载力,土地生态承载力、生产承载力和生活承载力是一个相互支持、相互依存的有机整体。土地生态-生产-生活系统承载力是指土地资源与生态环境的供容能力、经济活动能力和满足一定生活水平人口数量的社会发展能力的有机综合体,由处于支持层的生态承载力和处于表现层的生产承载力和生活承载力三部分组成,简称土地"三生"承载力。以党的"十八大"报告提出的"优化国土空间开发格局,促进生产空间集约高效,生活空间宜居适度,生态空间山清水秀"为目标,分析了土地生态-生产-生活综合承载力的基本构成及作用机制,在此基础上,以县级尺度作为测度对象,通过多方案比选,构建了由1个一级指标、3个二级指标、21个三级指标构成的土地生态-生产-生活综合承载力测度指标体系,给出了不同层级具体指标的量化辨识方法;进一步优选出土地生态-生产-生活综合承载力核算的SD情景模型。该研究从理论上旨在为推进国土生态、生产、生活空间集约高效利用提供科学判据,为开展全国市县尺度的土地综合承载力监测预警提供定量的科学依据。  相似文献

Aldabra Atoll has the largest population of giant tortoises (Aldabrachelys gigantea) in the world. As such an important biological resource, it is necessary to understand how the effects of climate change will impact this keystone species; in particular the frequency of drought, which is likely to affect tortoise habitat. To assess whether drought frequency has changed over the last 50 years on Aldabra, we calculated the standardized precipitation index (SPI) to identify drought periods using monthly rainfall data collected during 1969–2013. We found that drought frequency has increased to more than six drought months per year today compared with about two months per year in the 1970s (t = 2.884, p = 0.006). We used MODIS normalized difference vegetation index (NDVI) as a proxy for vegetation activity, to determine how vegetation has responded to the changing drought frequency between 2000 and 2013. We found that Aldabra’s vegetation is highly responsive to changes in rainfall: anomalies in long-term mean monthly NDVI across Aldabra were found to decrease below the mean during most drought periods and increase above the mean during most non-drought periods. To investigate the response of tortoise habitat to rainfall, we extracted mean NDVI anomalies for three key habitat types. Open mixed scrub and grasslands, the preferred habitat of tortoises, showed the greatest decrease in vegetation activity during drought periods, and the greatest increase in average greenness during non-drought periods. Recent analysis has shown vegetation changes on Aldabra in recent decades. If these changes are caused by decreased precipitation, then the increased frequency of drought could impact the tortoise population, in both the short and long term, by limiting the quality and quantity of forage and/or shade availability within favoured habitats, and by changing the habitat composition across the atoll.  相似文献

We have isolated cell wall peptidoglycan associated proteins (CW-Pr) of Mycobacterium tuberculosis H37Ra by chemical treatment with trifluoromethanesulfonic acid:anisole (2:1), which further resolved into 71, 60 and 45 kDa proteins on SDS-PAGE. A study was carried out to investigate the immunoreactivity of these proteins with blood samples from 4 categories, including 15 tuberculous patients (TB), 5 tuberculous patients on ATT (TBT), 10 PPD non-reactive healthy controls (HPPD?) and 11 PPD reactive healthy controls (HPPD+). Comparing the proliferative responses to cell wall protein antigens, it was observed that the 71 kDa protein gave maximum stimulation with PBMCs from the TB and HPPD+ groups. The adherent PBMCs from the TB group also demonstrated enhanced phagocytosis, particularly in the presence of 71 and 45 kDa proteins, and the phagocytic index was significantly higher (P < 0.05) than the TBT group. However, PBMCs from of the groups recognized the 60 kDa cell wall antigen. Our results suggest that the 71 kDa protein from the cell wall of M. tuberculosis is highly immunogenic.  相似文献

Flagellar and basal body development during cell division was studied in the biflagellate green alga Spermatozopsis similis Preisig et Melkonian by light microscopy of immobilized living cells, statistical analysis of flagellar lengths during the cell cycle, and electron microscopy of cells and isolated cytoskeletons. Interphase cells display two flagella of unequal/subequal length. An eyespot located in an anterior lobe of the chloroplast is connected to the basal body bearing the shorter flagellum by means of a five-stranded microtubular root. Until cell division, the two parental flagella attain the same length. During cell division, each cell forms two new flagella that grow to a length of 1.5 μm before they are distributed in a semiconservative fashion together with the parental flagella to the two progeny cells at cytokinesis. During the following interphase, the flagella newly formed during the preceding cell division grow to attain the same length as the parental flagella until the subsequent cell division. The shorter of the two flagella of a cell thus represents the developmentally younger flagellum, which transforms to the mature state during two consecutive cell cycles. Interphase cells display only two flagella-bearing basal bodies; two nascent basal bodies are formed during cell division and are connected to the microtubular d-roots of respective parental basal bodies with which the newly formed basal bodies are later distributed to the progeny cells. During segregation, basal body pairs shaft into the 11/5 o'clock direction, thus conserving the 1/7 o'clock configuration of basal body pairs of interphase cells. Prior to chloroplast and cell division, an eyespot is newly formed near the cell posterior in close association with a 1s microtubular root, while the parental eyespot is retained. During basal body segregation, eyespot-root connections for both the old and newly formed eyespots are presumably lost, and new associations of the eyespots with the 2s roots of the newly formed basal bodies are established during cytokinesis. The significance of this “eyespot-flagellar root developmental cycle” for the absolute orientation of the progeny cells is discussed.  相似文献

In this study, the genotoxic and cytotoxic potential of extremely low frequency magnetic fields (ELF-MF) was investigated in Wistar rat tibial bone marrow cells, using the chromosomal aberration (CA) and micronucleus (MN) test systems. In addition to these test systems, we also investigated the mitotic index (MI), and the ratio of polychromatic erythrocytes (PCEs) to normochromatic erythrocytes (NCEs). Wistar rats were exposed to acute (1 day for 4 h) and long-term (4 h/day for 45 days) to a horizontal 50 Hz, 1 mT uniform magnetic field generated by a Helmholtz coil system. Mitomycin C (MMC, 2 mg/kg BW) was used as positive control. Results obtained by chromosome analysis do not show any statistically significant differences between the negative control and both acute and long-term ELF-MF exposed samples. When comparing the group mean CA of long-term exposure with the negative control and acute exposure, the group mean of the long-term exposed group was higher, but this was not statistically significant. However, the mean micronucleus frequency of the longer-term exposed group was considerably higher than the negative control and acutely exposed groups. This difference was statistically significant (p < 0.01). The results of the MI in bone marrow showed that the averages of both A-MF and L-MF groups significantly decreased when compared to those in the negative control (p < 0.001 and p < 0.01, respectively). No significant differences were found between the group mean MI of A-MF exposure with L-MF. We found that the average of PCEs/NCEs ratios of A-MF exposed group was significantly lower than the negative control and L-MF exposed groups (p < 0.001 and p < 0.01, respectively). In addition, the group mean of the PCEs/NCEs ratios of L-MF was significantly lower than negative control (p < 0.01). We also found that the MMC treated group showed higher the number of CA and the frequency of MN formation when compared to those in all other each groups (p-values of all each groups <0.01) and also MMC treated group showed lower MI and the PCEs/NCEs ratios when compared to those in all other each groups (p-values of all groups <0.01). These observations indicate the in vivo suspectibility of mammals to the genotoxicity potential of ELF-MF.  相似文献

The present study is aimed at determining some haematological and biochemical parameters in the wild Indian bonnet monkeys as also the microscopic and ultrastructural characteristics of their pancreatic islets. Adult wild Indian bonnet monkeys (Macaca radiata radiata) of both sexes weighing between 2.5 and 4 kg were used in these experiments. Their platelet, reticulocyte and total leukocyte counts and the blood concentrations of hemoglobin and plasma proteins and the serum concentrations of aspartate amino transferase, alanine amino transferase and calcium are similar to the values reported for M. mulatta. Plasma glucose is lower when compared with reported values of M. mulatta and M. fascicularis. Insulin levels are comparable with those of M. mulatta and M. nigra. Histology of islets is similar to that of humans. Ovoid cell collections of islet cells are scattered throughout the pancreas. Ultrastructure of A, B and D cells is similar to humans. These findings suggest that this relatively underutilized macaques may be a suitable model for biomedical research.  相似文献

目的测定早期断奶仔猪血常规和血清生化参数、氨基酸浓度以及脏器指数常值,以供相关研究参考。方法分别采用全血细胞计数分析仪、全自动血液生化分析仪和全自动氨基酸分析仪等。结果断奶后第0、71、4、28天仔猪的血常规和血清生化参数、氨基酸浓度以及脏器指数均取得平均值和标准差。随着日龄的增加,WBC总数和分类计数不断增加,然后趋于稳定;MCV和PDW先降低后升高,第7天时最低;MCH和MCHC不断升高,第14天时最高。TP、LAC和Mg2+的浓度有升高趋势;P、BUN、TCHO和GLU的浓度以及LPS、CK和GPT的活性先降低后升高;AMM的活性和Zn2+、Na+、Cl-的浓度先升高后降低,第7天时最高,然后趋于稳定;TG的浓度在第14天时开始显著下降,然后趋于稳定;ALP的活性先升高后降低,第14天时最高。Asp的浓度不断增加,Cys的浓度不断降低,Ser、Ala、Leu、Phe、Gly、Val、Met、Lys、His、Pro、BCAA、AAA和总氨基酸的浓度先升高后降低,第7天时的浓度最高。肾脏指数不断下降,至第28天时趋于稳定;胃、颌下淋巴结和肝脏指数第7天时最大,脾脏指数第14天时最大。结论随着日龄的增加,早期断奶仔猪多数血常规和血清生化参数、氨基酸浓度及脏器指数表现不同的变化规律。  相似文献

Carbon-based secondary compounds (CBSCs), such as phenols or tannins, have been considered as one of the most important and general chemical barriers of woody plants against a diverse array of herbivores. Herbivory has been described as a critical factor affecting the growth and survival of newly established tree seedlings or juveniles then, the presence of secondary metabolites as defences against herbivores should be a primary strategy to reduce foliar damage. We examined whether light-induced changes in leaf phenolic chemistry affected insect herbivory on seedlings of two rainforest tree species, Drimys winteri (Winteraceae) and Gevuina avellana (Proteaceae). Seedlings of both species were planted under closed canopy and in a canopy gap within a large remnant forest patch. Half of the seedlings in each habitat were disinfected with a wide-spectrum systemic insecticide and the other half were used as controls. Seedling growth, survival, and foliar damage (estimated by an herbivory index) due to insect herbivores were monitored over a period of 16 months (December 2001–April 2003). The total leaf content of phenols and condensed tannins were assessed in seedlings from both habitats. As expected, access to light induced a greater production of CBSCs in seedlings of both tree species, but these compounds did not seem to play a significant defensive role, as seedlings grown in gaps suffered greater leaf damage than those planted in forest interior. In addition, in both habitats, seedlings without insecticide treatment suffered a greater foliar damage than those with insecticide, especially 16 months after the beginning of the experiment. Canopy openness and herbivory had positive and negative effects, respectively, on seedling growth and survival in both tree species. In conclusion, despite the higher levels of defence in tree-fall gap, the higher densities of herbivore override this and lead to higher damage levels.  相似文献
