Many complex behaviors are genetically hardwired. Based on previous findings on genetic control of mating and other behaviors in invertebrate and mammalian systems, I suggest that genetic control of complex behaviors is modular: first, dedicated genes specify different behavioral patterns; secondly, separable genetic networks govern distinct behavioral components. I speculate that modular genetic encoding of complex behaviors may in part reflect modularity in brain development and function. The misfolding of human islet amyloid polypeptide (hIAPP) is regarded as one of the causative factors of type 2 diabetes mellitus (T2DM). Salvia miltiorrhiza (Danshen), one of the most commonly used of traditional Chinese medicines, is often used in Compound Recipes for treating diabetes, however with unclear mechanisms. Since salvianolic acid B (SalB) is the most abundant bioactive ingredient of salvia miltiorrhiza water‐extract. In this study, we tested whether SalB has any effect on the amyloidogenicity of hIAPP. Our results clearly suggest that SalB can significantly inhibit the formation of hIAPP amyloid and disaggregate hIAPP fibrils. Furthermore, photo‐crosslinking based oligomerization studies suggest SalB significantly suppresses the toxic oligomerization of hIAPP monomers. Cytotoxicity protection effects on pancreatic INS‐1 cells by SalB were also observed using MTT‐based assays, potentially due to the inhibition on the membrane disruption effects and attenuated mitochondria impairment induced by hIAPP. These results provide evidence that SalB may further be studied on the possible pharmacological treatment for T2DM. Proteins 2013. © 2012 Wiley Periodicals, Inc.  相似文献

Abstract

We have determined the extent to which individual responses of domestic cats on being handled by an unfamiliar person are stable between 2 and 33 months of age. Twenty-nine household cats from nine litters were tested at 2, 4, 12, 24 and 33 months of age, by being held for 1 minute by a standard, unfamiliar person. Between 4 and 33 months of age, individual differences in the number of attempts made by the cat to escape, and in whether or not it showed signs of distress, were stable, with the partial exception of the test at 12 months. There was no consistency between tests in whether or not a particular cat purred. At 2 months of age, the number of escape attempts was highest in cats which had been handled the least in the second month of life, but this trend was reversed in the number of escape attempts made at 4 months. The lack of distress exhibited by all cats in the test at 2 months indicated that all had received at least adequate socialization to people, and that none were therefore comparable with the unsocialized cats used in several previous studies. We conclude that under normal domestic conditions, the behavior of a cat when handled by an unfamiliar person reflects a stable character trait, and that extensive handling during the socialization period may be subsequently associated with a reduction in inhibited behavior when interacting with an unfamiliar person.  相似文献

Bacillus anthracis MoxXT is a Type II proteic Toxin–Antitoxin (TA) module wherein MoxT is a ribonuclease that cleaves RNA specifically while MoxX interacts with MoxT and inhibits its activity. Disruption of the TA interaction has been proposed as a novel antibacterial strategy. Peptides, either based on antitoxin sequence or rationally designed, have previously been reported to disrupt the MoxXT interaction but cause a decrease in MoxT ribonuclease activity. In the present study, we report the crystal structure of MoxT, and the effect of several peptides in disrupting the MoxXT interaction as well as augmentation of MoxT ribonuclease activity by binding to MoxT in vitro. Docking studies on the peptides were carried out in order to explain the observed structure activity relationships. The peptides with ribonuclease augmentation activity possess a distinct structure and are proposed to bind to a distinct site on MoxT. The docking of the active peptides with MoxT showed that they possess an aromatic group that occupies a conserved hydrophobic pocket. Additionally, the peptides inducing high ribonuclease activity were anchored by a negatively charged group near a cluster of positively charged residues present near the pocket. Our study provides a structural basis and rationale for the observed properties of the peptides and may aid the development of small molecules to disrupt the TA interaction.  相似文献

An enzyme that catalyzes the synthesis of S-carboxymethyl- l-cysteine from 3-chloro- l-alanine (3-Cl-Ala) and thioglycolic acid was found in Escherichia coli W3110 and was designated as S- carboxymethyl-l-cysteine synthase. It was purified from the cell-free extract to electrophoretic homogeneity and was crystallized. The enzyme has a molecular weight of 84,000 and gave one band corresponding to a molecular weight of 37,000 on SDS-polyacrylamide gel electrophoresis. The purified enzyme catalyzed the β-replacement reactions between 3-CI-AIa and various thiol compounds. The apparent Km values for 3-Cl-Ala and thioglycolic acid were 40 mM and 15.4 mM. The enzyme showed very low activity as to the α,β-elimination reaction with 3-Cl-Ala and l-serine. It was not inactivated on the incubation with 3-Cl-Ala. The absorption spectrum of the enzyme shows a maximum at 412 nm, indicating that it contains pyridoxal phosphate as a cofactor. The N-terminal amino acid sequence was determined and the corresponding sequence was detected in the protein sequence data bank, but no homogeneous sequence was found.  相似文献

The ammonia-oxidizing bacterium Nitrosomonas sp. strain ENI-11 contains three copies of the hao gene (hao 1, hao 2, and hao 3) coding for hydroxylamine oxidoreductase (HAO). Three single mutants (hao 1::kan, hao 2::kan, or hao 3::kan) had 68 to 75% of the wild-type growth rate and 58 to 89% of the wild-type HAO activity when grown under the same conditions. A double mutant (hao 1::kan and hao 3::amp) also had 68% of the wild-type growth and 37% of the wild-type HAO activity.  相似文献

【目的】研究杀粉蝶菌素A1产生菌中甲基转移酶基因pieB2的功能。【方法】利用接合转移和同源重组双交换的方法,构建pieB2基因缺失突变株,以及利用接合转移的方法,构建回补菌株。通过高保真PCR克隆pieB2基因到表达载体pET28a上,构建质粒pJTU5997,转化入大肠杆菌E.coliBL21(DE3)/pLysE中诱导表达。利用高效液相色谱检测PieB2的体外酶活。【结果】获得了pieB2基因缺失的双交换突变株。发酵结果显示,该突变株不再产生杀粉蝶菌素A1,而是积累了一种脱甲基产物。N-末端融合组氨酸标签的PieB2在大肠杆菌中获得可溶性表达,通过体外催化证明了PieB2甲基转移酶的功能。【结论】体内遗传实验和体外生化实验证明了PieB2作为甲基转移酶在杀粉蝶菌素A1合成中的作用。  相似文献

为了研究红缘天牛成虫对寄主植物杏树衰弱树及木段挥发物的生理及行为反应,本文用6种挥发物,分别以不同浓度的单一组份对红缘天牛进行了电生理和行为测试;又在这6种单组份化合物对天牛行为选择表现为引诱活性的浓度范围内选择EAG值最高的浓度,以4-6种化合物等体积制成7种组合配方进行EAG和行为测试。结果表明:单一组份R-柠檬烯、反-2-己烯醛、丁酸丁酯、S-柠檬烯、异戊醇和3-蒈烯等6种单一化合物在预设的8个浓度梯度范围内,都能引起红缘天牛产生一定的生理反应,但行为选择实验的结果没有统计学意义。7种组合配方中,R4对红缘天牛成虫引诱活性最强(P0.01),引诱率达到76.67%,R1次之(P0.05),R4与R1的区别在于增加了丁酸丁酯;即丁酸丁酯在R4配方中具有明显的增效作用。  相似文献
