This essay explores connections between bacteriology and the disciplinary evolution of biochemistry in this country during
the 1930s. Many features of intermediary metabolism, a central component of biochemistry, originated as attempts to answer
fundamental bacteriological questions. Thus, many bacteriologists altered their research programs to answer these questions.
In so doing they changed their disciplinary focus from bacteriology to biochemistry. Chester Hamlin Werkman's (1893–1962)
Iowa State career illustrates the research perspective that many bacteriologists adopted. As a junior faculty member in the
Bacteriology Department in the late 1920s, Werkman faced a powerful professional dilemma: establishing a research identity
that distinguished him from his colleagues with flourishing national and international reputations. His solution was to radically
alter his research program from traditional bacteriology to a biochemistry program, which reflected the influence of the Dutch
microbiologist/biochemist, Albert Jan Kluyver (1888–1956). Werkman was extremely successful in this career change. His laboratory
made significant contributions to biochemistry, and Werkman achieved a notable degree of personal success. His career began
in the shadow of his departmental bacteriological colleagues; within a decade he became the department's dominant research figure, as a biochemist. Werkman's personal success, however, had profound consequences for the disciplinary future of bacteriology at Iowa State.

Streptococcus mutans is a major cause of tooth decay due to its promotion of biofilm formation and acid production. Several plant extracts have been reported to have multiple biological activities such as anti-inflammation and antibacterial effects. This study investigated the antibacterial activity of three plant extracts, phellodendron bark (PB), yucca, and black ginger, and found that PB had a stronger effect than the other extracts. Then, the minimum inhibitory concentration (MIC) of PB against 100 S. mutans strains was investigated. The MIC range of PB was 9.8–312.5 µg/mL. PB suppressed the growth kinetics of S. mutans in a dose-dependent manner, even at sub-MICs of PB. Then, we investigated the effect of PB on S. mutans virulence. The PB suppressed biofilm formation at high concentrations, although PB did not affect the expression of glucosyltransferase genes. Additionally, PB suppressed the decrease in pH from adding an excess of glucose. The expression of genes responsible for acid production was increased by the addition of excess glucose without PB, whereas their expression levels were not increased in the presence of 1× and 2× MIC of PB. Although PB showed a bacteriostatic effect on planktonic S. mutans cells, it was found that more than 2× MIC of PB showed a partial bactericidal effect on biofilm cells. In conclusion, PB not only showed antibacterial activity against S. mutans but also decreased the cariogenic activity in S. mutans. 相似文献

The genus Enterococcus is composed of 38 species, the most important of which are Enterococcus faecalis and Enterococcus faecium—both human intestinal colonizers. Hospitals within the United States and around the world commonly isolate these bacteria
because they are a cause of bacteremia, urinary tract infections (UTIs), endocarditis, wound infections, meningitis, intraabdominal
and pelvic infections, and nosocomial and iatrogenic infections. Given the ubiquity of enterococci within the human population,
it is important for laboratories to be able to distinguish these agents within hospitalized patients from other bacterial
genera and also differentiate different species within the Enterococcus genus as well as different strains within each species. Unfortunately, the enterococci are emerging as serious pathogens
in both the developed world, where surveillance needs to be improved and speciation procedures are inadequate or cumbersome,
and in developing nations, which lack the trained hospital personnel or funding to sufficiently identify enterococci to the
genus or species level. This review explores the Enterococcus genus and highlights some of the concerns for national and international clinical microbiology laboratories. 相似文献

Between 1946 and 1960, a new phenomenon emerged in the field of bacteriology. “Bacterial sex,” as it was called, revolutionized the study of genetics, largely by making available a whole new class of cheap, fast-growing, and easily manipulated organisms. But what was “bacterial sex?” How could single-celled organisms have “sex” or even be sexually differentiated? The technical language used in the scientific press – the public and inalienable face of 20th century science – to describe this apparently neuter organism was explicit: the cells “copulated,” had “intimate contract,” “conjugal unions,” and engaged in “ménage ã trois” relationships. And yet, to describe bacteria as sexually reproducing organisms, the definition of sex itself had to change. Despite manifold contradictions and the availability of alternative language, the notion of sexually active (even promiscuous) single-celled organisms has persisted, even into contemporary textbooks on cell biology and genetics. In this paper I examine the ways in which bacteria were brought into the genetic fold, sexualized, and given gender; I also consider the issues underlying the durability of “bacterial sex.” 相似文献

AbstractThe present study aimed to evaluate the suitability for drinking purpose of shallow groundwater near the Béni-Mellal wastewater treatment lagoon based on various physicochemical, heavy metals, and bacteriological parameter analyses. The physicochemical results revealed that some of the samples do not comply with the Moroccan and/or WHO standards for drinking water. Parameters including turbidity, TH, Na+, Li+, Ba2+, Ca2+ (~47.1% of samples), Cd (~52.9% of samples), Fe (~82.4% of samples), Pb (~58.8% of samples), T. coliforms, and E. coli exceeded the drinking limits. The statistical analyses revealed that the shallow groundwater chemistry is mainly controlled by geogenic and anthropogenic sources. For quality assessment, using the Moroccan groundwater assessment grid, the values of EC and Cl–, NO3–, NH4+, oxidability, and E. coli, fixed as pollution indicators, showed that most of the wells showed medium-to-poor quality, 14% of them have a very poor water quality, and 20% of them belong to the bad water quality. According to geometric and arithmetic DWQI values, the groundwater quality was frequently fair to good, needing treatment or at least disinfection before public consumption. A sensitivity analysis results indicated that Fe, Cd, Cr, Pb, and E. coli have an important impact on the DWQI computing. 相似文献
