Summary A five and a half weeks old female Kestrel exhibiting osteopathy of the pectoral and pelvic limbs, including symmetrical hyperdactyly, was investigated in order to clarify the pattern of the involved anatomical alterations and the possible causes of this developmental malformation. In the pectoral limb it consisted of a triplication of the alular digit, in the pelvic limb of a duplication of digit I. The live young Kestrel was observed for a period of two weeks to ascertain that it was unable to fly or procure prey on its own. After its death radiographs were taken and, apart from an eidonomic inspection including the wing claws, a detailed macroscopic dissection of the musculature of the pectoral and pelvic limbs was carried out using the in-water-method. Consecutive dissection steps were documented by a series of photographic slides. The relevant musculature, particularly that of the supernumerary digits, was recorded in proportional drawings. Subsequent to maceration of the limbs the isolated bones were reassembled according to the radiographs and also documented by means of photographs and drawings. This anatomical approach produced a reliable reconstruction of the skeletomuscular apparatus of the hyperdactylous limb parts. The eidonomic inspection revealed that at least young Kestrels may have two (alular and major digit) or even three wing claws per side. The proximal skeletal elements of both pectoral and pelvic limb were more sturdily built than in a typical Kestrel of comparable age. The proximal elements of the pelvic limb, the tarsometatarsus in particular, were shorter than in a typical Kestrel. In addition, the long axis of the tarsometatarsus was laterally bent in the transverse plane so that its proximal articular surfaces were medially inclined. Duplication of the cutaneous and osseous elements in the foot was accompanied by a duplication of some of the muscular and/or tendinous elements supplying digit I proper and the accessory digit I'. There were left-to-right asymmetries of the pedal musculature concerned. In contrast, the two accessory alular digits of each wing were almost completely devoid of musculature. Apart from atypical points of origin or insertion of the remaining distal musculture, left-to-right asymmetries and the two accessory alulae per wing, presumably, affected aerodynamic properties and resulted in flightlessness.A juvenile Kestrel of similar age and without hyperdactyly was dissected for comparison. In addition, the external appearance of the carpometacarpal region of two female Silkies, an obligatory pentadactylous breed of domestic fowl, was inspected and the skeletal parts of their pectoral and pelvic limbs compared with those of the hyperdactylous Kestrel. Our results and a literature review suggest that the symmetrical hyperdactyly in the Kestrel bears striking similarities to the hereditary hyperdactyly observed in certain breeds of domestic fowl. In addition, there is a striking resemblance between the hyperdactyly of the young Kestrel and certain forms of hyperdactyly induced by molecular genetical experiments of other authors on chicks. Comparison with these results taken from the literature suggest that the symmetrical hyperdactyly in the young Kestrel, including the alterations of the proximal skeletal elements, is caused by an unusually early expression of the Hoxd-11 gene group during embryological development. Most likely, this gene group is situated on the 2nd chromosome in birds just as it is in mammals.
Osteopathie der Vorder - und Hinterextremitäten, verbunden mit einer symmetrischen Hyperdactylie bei einem jungen Turmfalken (Falco tinnunculus)
Zusammenfassung Ein fünfeinhalb Wochen alter weiblicher Turmfalke mit einer Osteopathie der Vorder- und Hinterextremitäten, verbunden mit einer symmetrischen Hyperdactylie, wurde untersucht, um das Muster der beteiligten anatomischen Veränderungen und die möglichen Ursachen dieser Mißbildung zu erkennen. An der Vorderextremität bestand sie aus einer Verdreifachung des Alula-Fingers, an der Hinterextremität aus einer Verdoppelung der Zehe I. Die Beobachtung des lebenden jungen Turmfalken während eines Zeitraumes von zwei Wochen ergab, dass er flugunfähig war und keine Beute schlagen konnte.Nach seinem Tod und einer Inspektion der Eidonomie, einschließlich der Flügelkrallen, wurden Röntgenaufnahmen angefertigt. Danach folgte eine detaillierte makroskopische Präparation der Flügel- und Beinmuskulatur unter Verwendung der In-Wasser-Methode. Die einzelnen Präparationsschritte wurden anhand von Dia-Serien dokumentiert. Die relevante Muskulatur, insbesondere die der überzähligen Digiti, wurde in proportionsgetreuen Zeichnungen festgehalten. Nach Mazeration der Extremitäten wurden die Einzelknochen, entsprechend den Röntgenbildern, wieder zusammengesetzt und ebenfalls mit Fotografien und Zeichnungen dokumentiert. Dieser anatomische Ansatz lieferte eine zuverlässige Rekonstruktion des Skelett-Muskel-Apparates der hyperdactylen Extremitätenanteile.Die eidonomische Inspektion ergab, dass zumindest junge Turmfalken zwei (Digitus alularis und majoris) oder sogar drei Flügelkrallen haben können. Die proximalen Skelettelemente der Vorder- und Hinterextremität waren deutlich robuster gebaut als bei einem typischen Turmfalken vergleichbaren Alters. Die proximalen Elemente der Hinterextremität, insbesondere der Tarsometatarsus, waren kürzer als bei einem typischen Turmfalken. Darüberhinaus war die Längsachse des Tarsometatarsus in der Transversalebene laterad gekrümmt, so dass sich seine proximalen Gelenkflächen schräg mediad richteten. Entsprechend der kutanen und knöchernen Doppelbildungen des Fußes waren auch einige der Muskeln und Sehnen doppelt vorhanden, welche die eigentliche erste Zehe und die akzessorische erste Zehe versorgten. Es traten Rechts-/Links-Asymmetrien der betreffenden Muskulatur auf. Im Gegensatz dazu waren die beiden akzessorischen Alula-Finger jedes Flügels fast vollständig ohne Muskulatur. Abgesehen von atypischen Ursprungs- und Insertionspunkten der verbleibenden distalen Muskulatur, beeinträchtigten Rechts-/Links-Asymmetrien und die beiden akzessorischen Alulae pro Flügel vermutlich die aerodynamischen Eigenschaften und führten zur Flugunfähigkeit.Ein junger Turmfalke ähnlichen Alters ohne Hyperdactylie wurde zum Vergleich präpariert. Zusätzlich wurde die äußere Erscheinung der Carpometacarpal-Region zweier Seidenhühner, einer obligatorisch pentadactylen Hühnerrasse, inspiziert und die Skelettelemente ihrer Vorder- und Hinterextremitäten mit denen des hyperdactylen Turmfalken verglichen. Unsere Ergebnisse und ein Überblick der Literatur lassen auffallende Übereinstimmungen zwischen der symmetrischen Hyperdactylie des jungen Turmfalken und der erblichen Hyperdactylie bestimmter Hühnerrassen erkennen. Darüberhinaus besteht eine auffallende übereinstimmung zwischen der Hyperdactylie des jungen Turmfalken und bestimmten Formen der Hyperdactylie, welche von anderen Autoren durch molekulargenetische Experimente an Hühnerküken induziert wurden. Ein Vergleich mit diesen Ergebnissen aus der Literatur legt nahe, dass die symmetrische Hyperdactylie des jungen Turmfalken, einschließlich der Veränderungen der proximalen Skelettelemente, durch eine ungewöhnlich frühe Expression der Hoxd-11 Gengruppe im Laufe der Embryonalentwicklung verursacht wurde. Sehr wahrscheinlich ist diese Gengruppe bei Vögeln auf dem zweiten Chromosom lokalisiert — ebenso wie bei Säugetieren.

We examined the effects of habitat fragmentation of the white-starred robin Pogonocichla stellata metapopulation in the Taita Hills archipelago, a hotspot for biodiversity which was fragmented approximately 40 years ago. Using seven microsatellite markers, we analysed the robin's genetic structure and tested for equilibrium between migration and drift (testing the probability of decreased dispersal) as well as between mutation and drift (test for recent reduction in effective population size, i.e. bottlenecks). This metapopulation was found to retain relatively high levels of genetic variability (H(E) between 0.63 and 0.71) and to be in migration-drift equilibrium, suggesting that increased isolation between fragments did not have much effect on the dispersal between them. Furthermore, this equilibrium test greatly enhanced the interpretation of parameters (e.g. F(ST)) assumed to have reached an equilibrium value. In contrast to previous findings on the related and sympatric Taita thrush Turdus helleri (which is critically endangered), there were no indications for recent bottlenecks in any of the robin subpopulations. This difference can be attributed to the higher dispersal capacity of the robin compared with the thrush (deduced from both the genetic and capture-recapture data). Our results stress the importance of sustained dispersal for species conservation.  相似文献

Oxidative glutamate toxicity in the neuronal cell line HT22 is a model for cell death by oxidative stress, where an excess of extracellular glutamate inhibits import of cystine, a building block of the antioxidant glutathione. The subsequent decrease in glutathione then leads to the accumulation of reactive oxygen species (ROS) and programmed cell death. We used pharmacological compounds known to interact with heterotrimeric G-protein signalling and studied their effects on cell survival, morphology, and intracellular events that ultimately lead to cell death. Cholera toxin and phorbol esters were most effective and prevented cell death through independent pathways. Treating HT22 cells with cholera toxin attenuated the glutamate-induced accumulation of ROS and calcium influx. This was, at least in part, caused by an increase in glutathione due to improved uptake of cystine mediated by the induction of the glutamate/cystine-antiporter subunit xCT or, additionally, by the up-regulation of the antiapoptotic protein Bcl-2. Gs activation also protected HT22 cells from hydrogen peroxide or inhibition of glutathione synthesis by buthionine sulfoximine, and immature cortical neurones from oxidative glutamate toxicity. Thus, this pathway might be more generally implicated in protection from neuronal death by oxidative stress.  相似文献

Lake Bogoria, in the Rift Valley of Kenya is an extreme saline lake (conductivity 40–80 mS cm–1, alkalinity 1500 m equ l–1). It is hydrologically more stable than the other, endorheic lakes in Kenya, because it is deep – maximum depth at present just over 10 m in an area of 3000 ha – and so does not have periods when it is dry. It is ecologically simple, with only one species dominating the phytoplankton – the cyanobacterium `spirulina', Arthrospira fusiformis. Its biomass and productivity were very high – biomass between 38 and 365 g l–1 chlorophyll `a' and 3.4–21 × 103 coils ml–1 and net production between 0.24 and 1 gm C m3 h, the latter in a narrow zone of less than a metre. There were no macro-zooplankton in the plankton and the only grazer of A. fusiformis was the lesser flamingo, Phoeniconaias minor,which occurred irregularly in very high concentrations (in excess of 1 × 106). Detritivory in the benthos was effected by a single chironomid species, Paratendipes sp., at a maximum density of 4 × 104 m–2. The mean daily emergence of adult chironomids was estimated to be 1 × 103 m–2, the maximum 3. There was no littoral plant community within the lake but 44 dicotyledonous and 31 monocotyledonous plant species in the drawn-down zone and adjacent to it. A diverse draw-down terrestrial invertebrate fauna, only superficially described here, processed the flamingo feathers and carcasses, with other detritus such as chironomid pupal exuviae and decaying A. fusiformis scum. About 50 bird species depended upon the chironomids, either as they emerged through the water column as flying adults or later on the shoreline as floating pupal exuvia and dead adults. The lake has high conservation value because of three bird species in particular – lesser flamingo, Cape teal and black-necked grebe. The former provides real economic value in a region otherwise impoverished, because of the spectacle of tens of thousands of flamingos set against the landscape of hot springs and fumaroles at the lake edge, which draws 15000 visitors per annum. P. minor has experienced three periods during the past ten years when major mortalities have occurred, the last of which killed 700 birds day–1. This could have involved as many as 200000 birds (about 1/5th of the maximum population at this lake) if mortality was at a constant rate for the nine months it was observed. Causes of mortality have been suggested as avian tuberculosis, poisoning from cyanobacterial toxins or from heavy metal contamination at Lake Nakuru, but it is still not yet clear what contribution each makes to the problem.  相似文献

Using the quail-chick chimera technique, we followed the fate of Rauber's sickle cells in older whole blastoderms (cultured for approximately 2 days): after removal of the autochthonous Rauber's sickle from an unincubated chicken blastoderm, a quail Rauber's sickle was grafted isotopically and isochronically in its place. In transverse sections through these chimeras, the grafted quail Rauber's sickle cells were seen to have transformed into a broad row or ridge of quail junctional endoblast cells extending at the inner border of the area containing blood islands. After unilateral removal of the junctional endoblast from an intermediate streak chicken blastoderm (Stage 3; Hamburger and Hamilton [1951] J Morphol 88:49-92), we observed during further in vitro culture that at the operated side, in the area previously occupied by this junctional endoblast, blood islands no longer developed. If after such a unilateral removal of the chicken junctional endoblast quail junctional endoblast was apposed in its place, then blood islands reappeared in the operated area. The intimate contact between the apposed quail junctional endoblast and the recently formed blood islands, derived from peripherally migrating mesoderm, was very obvious on sections through such chimeras. We further demonstrate that Rauber's sickle vs. junctional endoblast is indispensable for the anlage of blood islands in avian blastoderms. Indeed, in the absence of Rauber's sickle material no blood islands develop (even when mesoderm is present after ingression of the upper layer via a primitive streak) in the isolated central region of the area centralis of unincubated chicken blastoderms after culture in vitro. Also, no junctional endoblast and no sickle canal appear in these explants. By contrast, if a Rauber's sickle fragment is placed on such an isolated central blastoderm region, then blood islands develop. These blood islands start to develop from peripherally migrating mesoderm in the neighborhood of the Rauber's sickle-derived junctional endoblast.  相似文献

In this article, we develop a new reconstruction of the pelvic and hindlimb muscles of the large theropod dinosaur Tyrannosaurus rex. Our new reconstruction relies primarily on direct examination of both extant and fossil turtles, lepidosaurs, and archosaurs. These observations are placed into a phylogenetic context and data from extant taxa are used to constrain inferences concerning the soft-tissue structures in T. rex. Using this extant phylogenetic bracket, we are able to offer well-supported inferences concerning most of the hindlimb musculature in this taxon. We also refrain from making any inferences for certain muscles where the resulting optimizations are ambiguous. This reconstruction differs from several previous attempts and we evaluate these discrepancies. In addition to providing a new and more detailed understanding of the hindlimb morphology of T. rex--the largest known terrestrial biped--this reconstruction also helps to clarify the sequence of character-state change along the line to extant birds.  相似文献

Recombinant cholera toxin B subunit (rCTB) is a safe and potent mucosal adjuvant. As a clue to the mechanism of the adjuvant effect of rCTB, the profile of cytokines secreted in vitro by the mouse peritoneal macrophage (Mphi) treated with rCTB was examined. IL-1beta secretion, intracellular production, and expression of its mRNA of LPS-stimulated Mphi was greatly enhanced by treatment with rCTB. IL-1beta production in response to other microbial stimulators, such as Pansorbin, Sansorbin, insoluble peptidoglycan, and Taxol, was also potentiated by rCTB. Mphi pretreated with rCTB before 24 hr could maintain the ability to produce a high level of IL-1beta, suggesting that this ability may be involved in the adjuvant activity of rCTB on Mphi stimulation. The possibility of close association between rCTB and signal transduction of a Toll-like receptor family in Mphi is discussed.  相似文献

The application of molecular genetic techniques has revolutionized our view of avian mating systems. Contrary to prior expectations, birds are only very rarely sexually monogamous, with 'extra-pair offspring' found in approximately 90% of species. Even among socially monogamous species, over 11% of offspring are, on average, the result of extra-pair paternity (EPP). Based on over 150 molecular genetic studies of EPP in birds, we review two topical areas: (i) ecological explanations for interspecific variation in the rate of EPP; and (ii) evidence bearing on the adaptive function of EPP. We highlight the remaining challenges of understanding the relative roles of genes and ecology in determining variation between taxa in the rate of extra paternity, and testing for differences between extra-pair offspring and those sired within-pair.  相似文献

Avian reovirus (ARV) structural protein, C, the prime candidate for vaccine against ARV, was expressed using a baculovirus/insect cell system. The expressed protein remained intracellular and reached 96 g/106 cells. Total product yield from a 200 ml suspension culture was 19 mg. When the protein was fused with a histidine tag and an enterokinase (EK) cleavage site, purification of 94% was achieved in a single step. The histidine tag was removed by EK.  相似文献
