Earlier studies from the authors' laboratory showed that malnourishment induces alterations in the cardiovascular homeostasis increasing the basal mean arterial pressure and heart rate. In this study, the authors evaluated whether the sympathetic and parasympathetic efferent activities contribute to changes in the cardiovascular homeostasis through altered modulation of the arterial baroreflex of malnourished rats. After weaning, male Fischer rats were given 15% (Normal Protein--NP) or 6% (Low Protein--LP) protein diet for 35 d. The baroreflex gain and latency were evaluated before and after selective autonomic blockades in control and malnourished rats. It was observed that malnourishment affected the baroreflex gain in response to activation and deactivation of the arterial baroreflex. Moreover, malnourished rats showed increased baroreflex latency as compared to that of control rats. Regarding the autonomic efferent activity directed to the heart, the data showed increased sympathetic and decreased parasympathetic efferent activities in malnourished rats, and such alterations could be related to the observed changes in the arterial baroreflex gain as well as in the basal mean arterial pressure and heart rate.  相似文献

Thiazolidinediones (TZDs) are currently the most efficacious class of oral antidiabetics. However, they carry the burden of weight gain and haemodilution, which may lead to cardiovascular complications. The present study was designed to ascertain whether a combination of dipeptidyl peptidase IV (DPP IV) inhibitor with low dose of a thiazolidinedione absolves TZD associated weight gain and oedema without compromising its efficacy. In this study, we examined the efficacy and safety of lower dose (1 mg/kg/day) of rosiglitazone, a thiazolidinedione, in combination with 5 mg/kg/day dose of LAF-237 (vildagliptin), a known DPP IV inhibitor, in aged db/db mice after 14 days of treatment and compared the combination with therapeutic dose (10 mg/kg) of rosiglitazone. The combination therapy showed similar efficacy as that of 10 mg/kg/day rosiglitazone in lowering random blood glucose (53.8%, p<0.001 and 54.3%, p<0.001 respectively), AUC ((0-120) min) during oral glucose tolerance test (OGTT) (38.6 %, p<0.01; 38.3%, p<0.01 respectively) and triglyceride levels (63.9% and 61% respectively; p<0.01). Plasma active glucagon like peptide-1 (GLP-1) and insulin levels were found to be elevated significantly (p<0.01 and p<0.05 respectively) in both LAF-237 and combination treated groups following oral glucose load. LAF-237 alone had no effect on random glucose and glucose excursion during OGTT in severely diabetic db/db mice. Interestingly, the combination treatment showed no significant increase in body weight as compared to the robust weight gain by therapeutic dose of rosiglitazone. Rosiglitazone at 10 mg/kg/day showed significant reduction (p<0.05) in haematocrit, RBC count, haemoglobin pointing towards haemodilution associated with increased mRNA expression of Na(+), K(+)-ATPase-alpha and epithelial sodium channel gamma (ENaCgamma) in kidney. The combination therapy escaped these adverse effects. The results suggest that combination of DPP IV inhibitor with low dose of thiazolidinedione can interact synergistically to represent a therapeutic advantage for the clinical treatment of type 2 diabetes without the adverse effects of haemodilution and weight gain associated with thiazolidinediones.  相似文献

We studied the influence of daily temperature gradients on organogenesis in apical and axil shoot meristems at different developmental stages in Cucumis sativus L. The level of organogenic activity of meristems was determined according to the number of leaf primordia on the main and lateral shoots, number of 2nd order shoots, and rudiments of flowers of different levels of development. At the studied ontogenetic stages (mesotrophic seedling or juvenile state), plants were grown under the controlled conditions: photoperiod 12 h, light intensity 100 Wt/m2, range of mean daily temperatures 20…30°C, and daily temperature gradients ?20…+20°C. After the temperature treatment, some plants were returned to the optimal, for growth and development, conditions for two weeks (aftereffect). Three types of organogenic activity of meristems in response to the influence of variable daily temperatures were described: stimulation, inhibition, or absence of effect. The phenomenon of stimulation includes two subtypes: optimization, when a maximum effect, observed at other constant temperatures, was attained under the influence of variable temperatures and maximization, when maximum values markedly exceeded those at constant temperatures. The patterns described are preserved on the whole in the aftereffect of daily temperatures.  相似文献

Although spliceosomal introns are present in all characterized eukaryotes, intron numbers vary dramatically, from only a handful in the entire genomes of some species to nearly 10 introns per gene on average in vertebrates. For all previously studied intron-rich species, significant fractions of intron positions are shared with other widely diverged eukaryotes, indicating that 1) large numbers of the introns date to much earlier stages of eukaryotic evolution and 2) these lineages have not passed through a very intron-poor stage since early eukaryotic evolution. By the same token, among species that have lost nearly all of their ancestral introns, no species is known to harbor large numbers of more recently gained introns. These observations are consistent with the notion that intron-dense genomes have arisen only once over the course of eukaryotic evolution. Here, we report an exception to this pattern, in the intron-rich diatom Thalassiosira pseudonana. Only 8.1% of studied T. pseudonana intron positions are conserved with any of a variety of divergent eukaryotic species. This implies that T. pseudonana has both 1) lost nearly all of the numerous introns present in the diatom-apicomplexan ancestor and 2) gained a large number of new introns since that time. In addition, that so few apparently inserted T. pseudonana introns match the positions of introns in other species implies that insertion of multiple introns into homologous genic sites in eukaryotic evolution is less common than previously estimated. These results suggest the possibility that intron-rich genomes may have arisen multiple times in evolution. These results also provide evidence that multiple intron insertion into the same site is rare, further supporting the notion that early eukaryotic ancestors were very intron rich.  相似文献

Several facets of spliceosomal intron in apicomplexans remain mysterious. First, intron numbers vary across species by 2 orders of magnitude, indicating massive intron loss and/or gain. Second, previous studies have shown very different evolutionary patterns over different timescales, with 100-fold higher rates of intron loss/gain between genera than within genera. Third, the timing and dynamics of nearly complete intron loss in Cryptosporidium species, as well as reasons for retention of the few remaining introns, remain unknown. We compared intron positions in 785 orthologous genes between 3 moderate to intron-rich apicomplexan species. We estimate that the Theileria-Plasmodium ancestor had 4.5 times as many introns as modern Plasmodium species and 38% more than modern Theileria species, and that subsequent intron losses have outnumbered intron gains by 5.8 to 1 in Theileria and by some 56 to 1 in Plasmodium. Several patterns suggest that these intron losses occurred by recombination with reverse-transcribed mRNAs. Intriguingly, this finding suggests significant retrotransposon activity in the lineages leading to both Theileria and Plasmodium, in contrast to the dearth of known retrotransposons and intron loss within modern species from both genera. We also compared genomes from Cryptosporidium parvum and C. hominis and found no evidence of ongoing intron loss, nor of intron gain. By contrast, Cryptosporidium introns are less evolutionary conserved with Toxoplasma than are introns from other apicomplexans; thus the few remaining introns are not simply indispensable ancestral introns.  相似文献

Goal, Scope and Background The life cycles of many products including textiles contain chemicals for which process flow data are not known or are too time consuming to collect. Although each chemical may not contribute significantly to the LCA results of the product, which might justify excluding them, but together their contribution could be significant. Similarly, rough estimates of the process flows for the production of a single chemical may be very uncertain and considered meaningless, while the estimates of the cumulative data of process flows for several chemicals may be less uncertain and be a meaningful contribution to the quality of the LCA results. There are methods for estimation of process flows for different types of products, with varying demands regarding input data and time and with varying accuracy of the results. This work contributes to the available methods, focusing on simple estimations for production of chemical substances. The goal was to create a fast method for estimation of emissions, resource and energy flows (process flows) for the production of chemicals, based on easily available data on the properties of the chemicals. The process flows investigated were limited to those normally associated with process industries and contributing most to depletion of resources, to global warming, acidification, eutrophication and photochemical ozone production, i.e. use of energy, crude oil, coal, natural gas, uranium in ore and emissions of CO2, SOx, NOx, NMVOC, methane, BOD, COD and total N. Toxic substances were excluded, since toxic emissions are substance specific and cannot be included in a generalization. Method Available data for the process flows for the production of chemicals of mainly fossil origin were correlated to properties of chemicals such as amount of carbon in the molecule, heat of formation and average number of chemical reaction steps in the production. The production procedures were found in readily available literature. Up to about six reaction steps were evaluated in the correlation study. The variations in the process flows among the chemicals studied were calculated. Results and Discussion There were weak correlations between average number of chemical reaction steps in the production and energy use, COD measured in water emissions, and SOx and NOx emissions to air. For the remaining properties of chemicals and process flows, there were only weak correlations for share of double bonding in the molecule if only molecules containing double bondings were included. Conclusions The precision in estimation of the process flows increases non-significantly when adding information on the number of reaction steps or share of double bonding for chemicals containing double bonding is added. Recommendations and Outlook It seems reasonable to start with a simple grouping method to estimate the process flows for the production of a chemical of fossil origin. Further investigations might investigate whether there is a correlation between process flows and the costs of chemicals, and further study the correlations between process flows and share of double bonding for chemicals containing double bondings.  相似文献

The Mutation‐Minimization Method (MuMi) to study the local response of proteins to point mutations has been introduced here. The heat shock protein Hsp70 as the test system since it displays features that have been studied in great detail has been used here. It has many conserved residues, serves several different functions on each of its domains, and displays interdomain allostery. For the analysis of spatial arrangement of residues within the protein, the network properties of the wild type (WT) protein as well as its all single alanine residue mutants using MuMi has been investigated. The measures to express the amount of change from the WT structure upon mutation and compare these deviations to find potential critical sites have been proposed. The functional significance of the potential sites to the parameter that uncovers them has been mapped. It was found that sites directly involved in binding were sensitive to mutations and were characterized by large displacements. On the other hand, sites that steer large conformational changes typically had increased reachability upon alanine mutations occurring elsewhere in the protein. Finally, residues that control communication within and between domains reside on the largest number of paths connecting pairs of residues in the protein. Concentrations of 16 priority polycyclic aromatic hydrocarbons (PAHs) were determined in 40 brands of biscuits in the Nigerian market. The analyses were performed by gas chromatography-flame ionisation detection (GC-FID) after Soxhlet extraction of the sample with hexane/dichloromethane and clean-up of the extract. The concentrations of Σ16 PAHs in these biscuit samples were in the range of 35.7–645.3 μg kg?1, 75.9–490.7 μg kg?1, 91.5–537 μg kg?1, 18.4–522.2 μg kg?1, 123.5–393.8 μg kg?1, 167.2–880 μg kg?1, 136.5–316 μg kg?1, and 135.5–241.6 μg kg?1 for shortcake, digestives, cookies, shortbread, wafers, crackers, Pringles, and cabin, respectively. The concentrations of Σ8 carcinogenic PAHs in the samples ranged from not detected (nd)–323.3 μg kg?1, 15.7–138 μg kg?1, 9.7–312.9 μg kg?1, nd–331.7 μg kg?1, nd–220.9 μg kg?1, nd–53.3 μg kg?1, 18.4–56.6 μg kg?1, and 6.6–170.8 μg kg?1 for shortcake, digestives, cookies, shortbread, wafers, crackers, Pringles, and cabin, respectively. The margin of exposure (MOE), based on PAH8 as an indicator for the occurrence and effects of PAHs in food, was less than 10,000, the serious health effects value, in 30% and 8% of the brands for the child and adult scenarios, respectively.  相似文献

Minerals can enter the food chain through industrial and mining activities. Soil-to-vegetable transfer is higher than soil-to-cereal, but human consumption of metals is attributable to balanced diets containing both vegetables and cereals and drinking water. However, the impact of location on intakes of metals from predominantly cereal-based Indian diets is not clear. Hence, the present study was undertaken in selected Agricultural, Industrial, and Coal Mining Areas (AA, IA, CMA) around the Allahabad District in Northern India to compare transfer of toxic metals, Pb, Cd, Cr and essential metals, Fe, Zn, Cu, Co in soil and water to common crops: cereals (rice, wheat, maize) and vegetables (spinach, potato), and to assess Daily Intake of Metal (DIM) and consequent Health Risk Index (HRI). The overall content of all metals, except Cu, in water, soils, and crops followed the pattern CMA > IA > AA. Transfer factors (TFs) followed the sequence spinach > potato > cereals. Quantitatively, however, cereals contribute maximally to a balanced diet, so DIM and HRI were higher from cereals than vegetables. Even though spinach had the highest TFs, cereals contributed maximally to HRI. CMA had the highest metal content so locally grown cereals contributed significantly to intake of both toxic and essential metals.  相似文献
