Three new eriophyid species (Phyllocoptinae), Shevtchenkella denticulata sp. n., Notallus pestehae sp. n. and Echinacrus ruthenicus sp. n., were described from Eryngium thyrsoideum Boiss. (Apiaceae), Pistacia vera L. (Anacardiaceae) and Lycium ruthenicum Murray (Solanaceae), respectively. All the three new species were collected from southwest of the East Azerbaijan province, Iran in 2011. It is the first record of an eriophyoid mite collected from E. thyrsoideum and L. ruthenicum and the first record of Notallus from Anacardiaceae plant family.  相似文献

Soil CO2 efflux (Fsoil) is the largest source of carbon from forests and reflects primary productivity as well as how carbon is allocated within forest ecosystems. Through early stages of stand development, both elevated [CO2] and availability of soil nitrogen (N; sum of mineralization, deposition, and fixation) have been shown to increase gross primary productivity, but the long‐term effects of these factors on Fsoil are less clear. Expanding on previous studies at the Duke Free‐Air CO2 Enrichment (FACE) site, we quantified the effects of elevated [CO2] and N fertilization on Fsoil using daily measurements from automated chambers over 10 years. Consistent with previous results, compared to ambient unfertilized plots, annual Fsoil increased under elevated [CO2] (ca. 17%) and decreased with N (ca. 21%). N fertilization under elevated [CO2] reduced Fsoil to values similar to untreated plots. Over the study period, base respiration rates increased with leaf productivity, but declined after productivity saturated. Despite treatment‐induced differences in aboveground biomass, soil temperature and water content were similar among treatments. Interannually, low soil water content decreased annual Fsoil from potential values – estimated based on temperature alone assuming nonlimiting soil water content – by ca. 0.7% per 1.0% reduction in relative extractable water. This effect was only slightly ameliorated by elevated [CO2]. Variability in soil N availability among plots accounted for the spatial variability in Fsoil, showing a decrease of ca. 114 g C m?2 yr?1 per 1 g m?2 increase in soil N availability, with consistently higher Fsoil in elevated [CO2] plots ca. 127 g C per 100 ppm [CO2] over the +200 ppm enrichment. Altogether, reflecting increased belowground carbon partitioning in response to greater plant nutritional needs, the effects of elevated [CO2] and N fertilization on Fsoil in this stand are sustained beyond the early stages of stand development and through stabilization of annual foliage production.  相似文献

Despite the availability of therapy and vaccine, tuberculosis (TB) remains one of the most deadly and widespread bacterial infections in the world. Since several decades, the sudden burst of multi- and extensively-drug resistant strains is a serious threat for the control of tuberculosis. Therefore, it is essential to identify new targets and pathways critical for the causative agent of the tuberculosis, Mycobacterium tuberculosis (Mtb) and to search for novel chemicals that could become TB drugs. One approach is to set up methods suitable for the genetic and chemical screens of large scale libraries enabling the search of a needle in a haystack. To this end, we developed a phenotypic assay relying on the detection of fluorescently labeled Mtb within fluorescently labeled host cells using automated confocal microscopy. This in vitro assay allows an image based quantification of the colonization process of Mtb into the host and was optimized for the 384-well microplate format, which is proper for screens of siRNA-, chemical compound- or Mtb mutant-libraries. The images are then processed for multiparametric analysis, which provides read out inferring on the pathogenesis of Mtb within host cells.  相似文献

Acidic exocellular class III chitinase (EC 3.2.1.14) was previously identified in healthy white lupin (Lupinus albus L.) plants and suspension-cultured cells by N-terminal microse-quencing. In this study, the detection of chitinase activity with Remazol Brilliant Violet 5R (RBV)-labelled chitin derivatives is described. Chitinase activity was observed in protein fractions of cytoplasmic or exocellular origin from roots, hypocotyls, cotyledons, and leaves of healthy white lupin plants. Using isoelectrofocusing followed by a new overlay technique with carboxymethyl chitin-RBV conjugate-containing gel, up to six different chitinase isoforms were visualised. Their activity was distributed fairly evenly within a plant with acidic isoforms predominating in cell walls and basic (or neutral) ones found intracellularly. Exocellular location of some chitinase isoforms were also confirmed by detection of their activities in intercellular washing fluids from white lupin tissues. Chitinase activity was demonstrated in culture filtrates and cell walls of suspension-cultured white lupin cells.  相似文献

This article describes the removal of heavy metals from contaminated clayey soils by soil washing using various extractants. Two clayey soils, kaolin, a low buffering soil with pH of 5, and glacial till, a high buffering soil with pH of 8, were used to represent various soil conditions. These soils were spiked with chromium (Cr), nickel (Ni), and cadmium (Cd) to simulate improper disposal of typical electroplating waste constituents. The following extracting solutions were investigated for the removal of heavy metals from the soils: deionized water, distilled water, and tap water; acetic acid and phosphoric acid; chelating agents ethylenediaminetetraacetic acid (EDTA) and citric acid; and the oxidizing agents potassium permanganate and hydrogen peroxide. The effect of extractant concentration on removal of heavy metals was also investigated. Complete removal of Cr was achieved using 0.1?M potassium permanganate for kaolin, while a maximum of 54% was removed from glacial till. A maximum Ni removal of 80% was achieved using tapwater for kaolin, while a maximum removal of 48 to 52% was achieved using either 1?M acetic acid or 0.1?M citric acid for glacial till. A maximum Cd removal of 50% was achieved using any of the extractants for kaolin, while a maximum removal of 45 to 48% was obtained using either acids or chelating agents for glacial till. Overall, this study showed that complete removal of Cr, Ni, and Cd from clayey soils is difficult to achieve using the soil-washing process, and also the use of one extractant may not be effective in removing all metals. A sequential extraction using different extractants may be needed for the removal of multiple metal contaminants from clayey soils.  相似文献

Digital point‐occurrence records from the Global Biodiversity Information Facility (GBIF) and other data providers enable a wide range of research in macroecology and biogeography. However, data errors may hamper immediate use. Manual data cleaning is time‐consuming and often unfeasible, given that the databases may contain thousands or millions of records. Automated data cleaning pipelines are therefore of high importance. Taking North American Ephedra as a model, we examined how different data cleaning pipelines (using, e.g., the GBIF web application, and four different R packages) affect downstream species distribution models (SDMs). We also assessed how data differed from expert data. From 13,889 North American Ephedra observations in GBIF, the pipelines removed 31.7% to 62.7% false positives, invalid coordinates, and duplicates, leading to datasets between 9484 (GBIF application) and 5196 records (manual‐guided filtering). The expert data consisted of 704 records, comparable to data from field studies. Although differences in the absolute numbers of records were relatively large, species richness models based on stacked SDMs (S‐SDM) from pipeline and expert data were strongly correlated (mean Pearson''s r across the pipelines: .9986, vs. the expert data: .9173). Our results suggest that all R package‐based pipelines reliably identified invalid coordinates. In contrast, the GBIF‐filtered data still contained both spatial and taxonomic errors. Major drawbacks emerge from the fact that no pipeline fully discovered misidentified specimens without the assistance of taxonomic expert knowledge. We conclude that application‐filtered GBIF data will still need additional review to achieve higher spatial data quality. Achieving high‐quality taxonomic data will require extra effort, probably by thoroughly analyzing the data for misidentified taxa, supported by experts.  相似文献

Understanding how wheat (Triticum aestivum L.) plants under high temperature (HT) regulate lipid composition is critical to developing climate‐resilient varieties. We measured 165 glycerolipids and sterol derivatives under optimum and high day and night temperatures in wheat leaves using electrospray ionization‐tandem mass spectrometry. Levels of polar lipid fatty acyl chain unsaturation were lower in both heat‐tolerant genotype Ventnor and susceptible genotype Karl 92 under HT, compared with optimum temperature. The lower unsaturation was predominantly because of lower levels of 18:3 acyl chains and higher levels of 18:1 and 16:0 acyl chains. Levels of 18:3‐containing triacylglycerols increased threefold/more under HT, consistent with their possible role in sequestering fatty acids during membrane lipid remodelling. Phospholipids containing odd‐numbered or oxidized acyl chains accumulated in leaves under HT. Sterol glycosides (SG) and 16:0‐acylated sterol glycosides (ASG) were higher under HT than optimum temperatures. Ventnor had lower amounts of phospholipids with oxidized acyl chains under HT and higher amounts of SG and 16:0‐ASG than Karl 92. Taken together, the data demonstrate that wheat leaf lipid composition is altered by HT, in which some lipids are particularly responsive to HT, and that two wheat genotypes, chosen for their differing physiological responses to HT, differ in lipid profile under HT.  相似文献

土壤淋洗修复技术是一种行之有效的污染土壤治理技术,适合于快速修复受高浓度重金属和有机物污染土壤与沉积物。本文综述了土壤淋洗修复技术的特点、技术流程、土壤淋洗剂的研究与应用进展,指出异位土壤淋洗修复技术因修复效果稳定,易于实现系统控制和废弃物减量化等优点而具有更广阔的应用前景,天然螯合剂和生物表面活性剂等环境友好型淋洗剂正逐渐取代人工螯合剂和化学表面活性剂成为土壤淋洗剂研究的主流方向,而现代超分子化学的引入和发展有可能对复合污染土壤的高效淋洗修复研究产生新的影响。  相似文献

摘要 目的:评价放散式体外冲击波(rESW)穴位治疗联合中药熏洗在老年膝骨关节炎(KOA)中的应用效果。方法:选入2021年2月~2022年8月我院收治的老年KOA患者76例,根据治疗方法不同分为对照组和观察组,各38例,两组均予以中药熏洗,观察组加用rESW穴位治疗。评价两组的治疗效果、膝关节功能、疼痛程度等指标,并进行统计比较。结果:与治疗前相比,两组治疗2周、治疗4周时的VAS评分明显下降(P<0.05),而观察组下降幅度更大,与对照组差异显著(P<0.05);两组治疗前Lysholm评分无明显差异(P>0.05),而观察组治疗2周、治疗4周时的Lysholm评分显著高于对照组(P<0.05);观察组治疗显效率和有效率均显著高于对照组(P<0.05);观察组治疗2周、4周时的血清IL-1β、TNF-α和MMP-13水平较对照组低(P<0.05)。结论:rESW穴位联合中药熏洗治疗老年KOA患者疗效显著,可缓解疼痛症状,改善膝关节功能,控制炎症反应,且操作简单,值得推荐。  相似文献
