This study investigated the potential effect of poultry dung (biostimulation) and stubborn grass (Sporobolus pyramidalis) (phytoremediation) on microbial biodegradation of gasoline and nickel uptake in gasoline-nickel-impacted soil. In addition, the potential stimulatory effects of nickel on hydrocarbon utilization were investigated over a small range of nickel concentrations (2.5–12.5 mg/kg). The results showed that an increase in nickel concentration increased hydrocarbon degraders in soil by a range of 8.4–17.2% and resulted in a relative increase in gasoline biodegradation (57.5–62.4%). Also, under aerobic conditions, total petroleum hydrocarbons’ (TPH) removal was 62.4% in the natural gasoline-nickel microcosm (natural attenuation), and a maximum of 78.5%, 85.7%, and 95.8% TPH removal was obtained in phytoremediation, biostimulation, and a combination of biostimulation- and phytoremediation-treated microcosms, respectively. First-order kinetics described the biodegradation of gasoline and nickel uptake very well. Half-life times obtained were 28.88, 18.24, 14.44, and 8.56 days for gasoline degradation under natural attenuation, phytoremediation, biostimulation, and combined biostimulation and phytoremediation treatment methods, respectively. The results indicate that these remediation methods have promising potential for effective remediation of soils co-contaminated with petroleum hydrocarbons and heavy metals.  相似文献

The intent of this study was to determine whether aging affects signaling pathways involved in mitochondrial biogenesis in response to a single bout of contractile activity. Acute stimulation (1 Hz, 5 min) of the tibialis anterior (TA) resulted in a greater rate of fatigue in old (36 month), compared to young (6 month) F344XBN rats, which was associated with reduced ATP synthesis and a lower mitochondrial volume. To investigate fiber type-specific signaling, the TA was sectioned into red (RTA) and white (WTA) portions, possessing two- to 2.5-fold differences in mitochondrial content. The expression and contraction-mediated phosphorylation of p38, MKK3/6, CaMKII and AMPKα were assessed. Kinase protein expression tended to be higher in fiber sections with lower mitochondrial content, such as the WTA, relative to the RTA muscle, and this was exaggerated in tissues from senescent, compared to young animals. At rest, kinase activation was generally similar between young and old animals, despite the age-related variations in mitochondrial volume. In response to contractile activity, age did not influence the signaling of these kinases in the high-oxidative RTA muscle. However, in the low-oxidative WTA muscle, contraction-induced kinase activation was attenuated in old animals, despite the greater metabolic stress imposed by contractile activity in this muscle. Thus, the reduction of contraction-evoked kinase phosphorylation in muscle from old animals is fiber type-specific, and depends on factors which are, in part, independent of the metabolic milieu within the contracting fibers. These findings imply that the downstream consequences of kinase signaling are reduced in aging muscle.  相似文献

Arsenite (As(III)) is the predominant arsenic (As) species in reducing environments. As(III) is less strongly adsorbed than As(V) at circumneutral pH conditions by common non‐iron metal oxides in sediments such as those of aluminum. Therefore, oxidation of As(III) to As(V) could contribute to an improved immobilization of As and thus help mitigate As contamination in groundwater. Microbial oxidation of As(III) is known to readily under aerobic conditions, however, the dissolved oxygen (O2) concentration in groundwater may be limited due to the poor solubility of O2 and its high chemical reactivity with reduced compounds. Nitrate (${\rm NO}_{3}^{{-} } $ ), can be considered as an alternative electron acceptor, which can support oxidation of As(III) to As(V) by denitrifying bacteria. In this study, two up‐flow sediment columns packed with activated alumina (AA) were utilized to demonstrate the role of denitrification on the oxidation of As(III) to As(V) and its contribution to improved As adsorption onto AA. One column was supplied with ${\rm NO}_{3}^{{-} } $ (C1) and its performance was compared with a control column lacking ${\rm NO}_{3}^{{-} } $ (C2). During most of the operation when the pH was in the circumneutral range (days 50–250), the release of arsenic was greater from C2 compared to C1. The effluent As concentrations started increasing on days 60 and 100 in C2 and C1, respectively. Complete breakthrough started on day 200 in C2; whereas in C1, complete breakthrough was never achieved. The effluent and solid phase As speciation was dominated by As(V) in C1, indicating the occurrence of As(III) oxidation due to ${\rm NO}_{3}^{{-} } $ ; whereas in C2, only As(III) was dominant. This study illustrates a bioremediation or natural attenuation process based on anoxic microbial ${\rm NO}_{3}^{{-} } $ ‐dependent oxidation of As(III) to more readily adsorbed As(V) as a means to enhance the immobilization of As on alumina oxide particles in subsurface environments. A fuzzy-logic light attenuation model was developed and validated for a stirred-tank photobioreactor. Based on this model, local light intensities were used to calculate local specific growth rates of the cyanobacteria Synechococcus PCC 7942. The light regime for maximization of biomass space-time yield in a batch process was estimated using a genetic algorithm taking into account the integral average of the individual growth rates.  相似文献

Abstract: Plant species vary widely in their average leaf lifespan (LL) and specific leaf area (SLA, leaf area per dry mass). The negative LL–SLA relationship commonly seen among species represents an important evolutionary trade‐off, with higher SLA indicating greater potential for fast growth (higher rate of return on a given investment), but longer LL indicating a longer duration of the revenue stream from that investment. We investigated how these leaf‐economic traits related to aggregate properties of the plant crown. Across 14 Australian sclerophyll shrub species, those with long LL accumulated more leaf mass and leaf area per unit ground area. Light attenuation through their canopies was more severe. Leaf accumulation and light attenuation were more weakly related to SLA than to LL. The greater accumulation of foliage in species with longer LL and lower SLA may counterbalance their generally lower photosynthetic rates and light‐capture areas per gram of leaf.  相似文献

Live attenuated Shigella vaccines have shown promise in inducing protective immune responses in human clinical trials and as carriers of heterologous antigens from other mucosal pathogens. In the past, construction of Shigella vaccine strains relied on classical allelic exchange systems to genetically engineer the bacterial genome. These systems require extensive in vitro engineering of long homologous sequences to create recombinant replication-defective plasmids or phage. Alternatively, the lambda red recombination system from bacteriophage facilitates recombination with as little as 40 bp of homologous DNA. The process, referred to as recombineering, typically uses an inducible lambda red operon on a temperature-sensitive plasmid and optimal transformation conditions to integrate linear antibiotic resistance cassettes flanked by homologous sequences into a bacterial genome. Recent advances in recombineering have enabled modification of genomic DNA from bacterial pathogens including Salmonella, Yersinia, enteropathogenic Escherichia coli, or enterohemorrhagic E. coli and Shigella. These advances in recombineering have been used to systematically delete virulence-associated genes from Shigella, creating a number of isogenic strains from multiple Shigella serotypes. These strains have been characterized for attenuation using both in vivo and in vitro assays. Based on this data, prototypic Shigella vaccine strains containing multiple deletions in virulence-associated genes have been generated.  相似文献

In order to develop an optical model to map the extent of coastal waters, the authors analyzed variations in bio-optical constituents and submarine optical properties along a transect from the nutrient-enriched coastal bay, Himmerfjärden, out into the open Baltic Sea. The model is a simple implementation of the “ecosystem approach,” because the optical constituents are proxies for important components of ecosystem state. Yellow substance or colored dissolved organic matter (CDOM) is often a marker for terrestrial freshwater or decay processes in the littoral zone. Phytoplankton pigments, especially chlorophyll a, are used as a proxy for phytoplankton biomass that may be stimulated by fluvial or coastal inputs of anthropogenic nutrients. Suspended particulate matter (SPM) is placed in suspension by tidal or wind-wave stirring of shallow seabeds, and is therefore an indicator for physical forcing. It is the thesis of this article that such constituents, and the optical properties that they control, can be used to provide an ecological definition of the extent of the coastal zone. The spatial distribution of the observations was analyzed using a steady-state model that assumes diffusional transport of bio-optical variables along an axis perpendicular to the coast. According to the model, the resulting distribution along this axis can be described as a low-order polynomial (of order 1–3) when moving from a “source” associated with land to the open-sea “sink.” Order 1 implies conservative mixing, and the higher orders imply significant biological or chemical processes within the gradient. The analysis of the transect data confirmed that the trend of each optical component could be described well using a low-order polynomial. Multiple regression analysis was then used to weigh the contribution of each optical component to the spectral attenuation coefficient K d(490) along the transect. The results showed that in this Swedish Baltic case study, the inorganic fraction of the SPM may be used to distinguish between coastal and open-sea waters, as it showed a clear break between coastal and open-sea waters. Alternative models may be needed for coastal waters in which fronts interrupt the continuity of mixing.  相似文献

As a result of mixing and light attenuation, algae in a photobioreactor (PBR) alternate between light and dark zones and, therefore, experience variations in photon flux density (PFD). These variations in PFD are called light/dark (L/D) cycles. The objective of this study was to determine how these L/D cycles affect biomass yield on light energy in microalgae cultivation. For our work, we used controlled, short light path, laboratory, turbidostat‐operated PBRs equipped with a LED light source for square‐wave L/D cycles with frequencies from 1 to 100 Hz. Biomass density was adjusted that the PFD leaving the PBR was equal to the compensation point of photosynthesis. Algae were acclimated to a sub‐saturating incident PFD of 220 µmol m?2 s?1 for continuous light. Using a duty cycle of 0.5, we observed that L/D cycles of 1 and 10 Hz resulted on average in a 10% lower biomass yield, but L/D cycles of 100 Hz resulted on average in a 35% higher biomass yield than the yield obtained in continuous light. Our results show that interaction of L/D cycle frequency, culture density and incident PFD play a role in overall PBR productivity. Hence, appropriate L/D cycle setting by mixing strategy appears as a possible way to reduce the effect that dark zone exposure impinges on biomass yield in microalgae cultivation. A 3-compartment mechanistic model is proposed to explain the attenuation of the electrocardiographic QRS complexes (↓QRSV) observed in patients with heart failure (HF). This includes the effects of increased intracardiac blood volume and decreased hematocrit due to blood dilution (1st compartment), the heart's alteration in electrogenesis due to possible ischemia or inflammation, leading to myocardial edema, (2nd compartment), and the passive volume conductor of the tissue and organ constituents of the thorax and the entire body, with their resistivity changes due to increased fluid content (pulmonary and peripheral edema) (3rd compartment).
