Aromatic compounds derived from lignin are of great interest for renewable biotechnical applications. They can serve in many industries e.g. as biochemical building blocks for bioplastics or biofuels, or as antioxidants, flavor agents or food preservatives. In nature, lignin is degraded by microorganisms, which results in the release of homocyclic aromatic compounds. Homocyclic aromatic compounds can also be linked to polysaccharides, tannins and even found freely in plant biomass. As these compounds are often toxic to microbes already at low concentrations, they need to be degraded or converted to less toxic forms. Prior to ring cleavage, the plant- and lignin-derived aromatic compounds are converted to seven central ring-fission intermediates, i.e. catechol, protocatechuic acid, hydroxyquinol, hydroquinone, gentisic acid, gallic acid and pyrogallol through complex aromatic metabolic pathways and used as energy source in the tricarboxylic acid cycle. Over the decades, bacterial aromatic metabolism has been described in great detail. However, the studies on fungal aromatic pathways are scattered over different pathways and species, complicating a comprehensive view of fungal aromatic metabolism. In this review, we depicted the similarities and differences of the reported aromatic metabolic pathways in fungi and bacteria. Although both microorganisms share the main conversion routes, many alternative pathways are observed in fungi. Understanding the microbial aromatic metabolic pathways could lead to metabolic engineering for strain improvement and promote valorization of lignin and related aromatic compounds. 相似文献

Diesel engine exhaust (DEE) was found to induce lipid peroxidation (LPO) in animal exposure studies. LPO is a class of oxidative stress and can be reflected by detecting the levels of its production, such as malondialdehyde (MDA) and 4-hydroxynonenal (4-HNE), and etheno-DNA adducts including 1,N6-etheno-2′-deoxyadenosine (?dA) and 3,N4-etheno-2′-deoxycytidine (?dC). However, the impact of DEE exposure on LPO has not been explored in humans. In this study, we evaluated urinary MDA, 4-HNE, ?dA, and ?dC levels as biomarkers of LPO among 108 workers with exclusive exposure to DEE and 109 non-DEE-exposed workers. Results showed that increased levels of urinary MDA and ?dA were observed in subjects occupationally exposed to DEE before and after age, body mass index (BMI), smoking status, and alcohol use were adjusted (all p?0.001). There was a statistically significant relationship between the internal exposure dose (urinary ΣOH-PAHs) and MDA, 4-HNE, and ?dA (all p?0.001). Furthermore, significant increased relations between urinary etheno-DNA adduct and MDA, 4-HNE were observed (all p?0.05). The findings of this study suggested that the level of LPO products (MDA and ?dA) was increased in DEE-exposed workers, and urinary MDA and ?dA might be feasible biomarkers for DEE exposure. LPO induced DNA damage might be involved and further motivated the genomic instability could be one of the pathogeneses of cancer induced by DEE-exposure. Ellagic acid, a common plant phenol, was shown to be a potent inhibitor of epidermal microsomal aryl hydrocarbon hydroxylase (AHH) activity in vitro, and of benzo[a]pyrene (BP)-binding to both calf thymus DNA in vitro and to epidermal DNA in vivo. The in vitro addition of ellagic acid (0.25-2.0 microM) resulted in a dose-dependent inhibition of AHH activity in epidermal microsomes prepared from control or carcinogen-treated animals. The I50 of ellagic acid for epidermal AHH was 1.0 microM making it the most potent inhibitor of epidermal AHH yet identified. In vitro addition of ellagic acid to microsomal suspensions prepared from control or coal tar-treated animals resulted in 90% inhibition of BP-binding to calf thymus DNA. Application of ellagic acid to the skin (0.5-10.0 mumol/10 gm body wt) caused a dose-dependent inhibition of BP-binding to epidermal DNA. Our results suggest that phenolic compounds such as ellagic acid may prove useful in modulating the risk of cutaneous cancer from environmental chemicals. 相似文献

Several biological assays were used to evaluate the toxic effects of contaminants in soil after phytoremediation. During the treatment process, significant decreases in overall toxicity were observed. Specifically, earthworm survivability and lettuce germination increased over the study period. Microbial respiration improved, but only in planted treatments. Toxicity and total polycyclic aromatic hydrocarbon concentrations showed some correlation, but the relationships generally were not significant. Soil moisture was less of a predictor for biological responses. The presence of plants did not provide a clear advantage for improving toxicity compared to unplanted treatments. 相似文献

Acetone extracts of filter-collected urban airborne particulate matter contain compounds which can competitively inhibit 2,3,7,8-[1,6-3H]tetrachlorodibenzo-p-dioxin (TCDD) binding to the rat liver TCDD-receptor protein. The concentration of conventional polycyclic aromatic hydrocarbons (PAHs) or chlorinated dioxins and dibenzofurans cannot account for more than 1-30% of the observed competition for [3H]TCDD binding to the receptor protein. The difference in potency between samples collected in urban areas during different periods of the year and a background sample is 25-400-fold. Collecting samples in the presence of increased concentrations of nitrogen dioxide, nitrous acid, nitric acid or ozone did not increase the amount of compounds with receptor affinity. However, with nitrogen dioxide and especially with nitric acid, a substantial increase of the mutagenic effects in the Ames Salmonella assay in the absence of mammalian activation as well as a degradation of several PAHs were noted. Affinity for the TCDD-receptor protein, mutagenicity in the absence of mammalian metabolic activation in the Ames Salmonella assay and PAH-content are characteristics of urban particulate matter showing the presence of compounds, that represent potential health risks. The compounds with affinity for the receptor may constitute a group of substances different from both conventional PAHs and direct-acting mutagens. 相似文献

该文依托于小兴安岭典型阔叶红松(Pinus koraiensis)林9 hm2森林动态监测样地,对样地内林窗边缘主要树种红松和臭冷杉(Abies nephrolepis)进行生长释放判定分析,重建了冠层树木的干扰历史。结果表明:整体上林窗木与非林窗木的生长变化百分率变化规律基本一致,而不同林窗间生长变化百分率存在明显的差异,林窗干扰及其产生的影响存在较大的变异性。在1733–1738、1748–1752、1769–1771、1798–1801、1827–1833、1841–1844、1935–1939及1968–1973年间红松生长释放较强,其中1752、1770、1800、1830、1842、1937及1970年出现了明显的干扰峰;在1889–1904、1932–1938、1947–1973和1986–2005年间臭冷杉生长释放较强,其中1894、1934、1951、1968和1990年出现了明显的干扰峰。红松干扰存在2.0 a、3.5 a、3.8 a、7.3–7.9 a和9.1–18.2 a的显著周期,臭冷杉干扰存在3.5–3.6 a、7.5–48.8 a和65–85 a的显著周期。风干扰是典型阔叶红松林干扰释放的主要因子,异常温度也影响该地区树木生长释放事件。太阳活动通过影响局地风速、温度、降水等气候因子以及其他大尺度气候模态影响林窗动态,可能是小兴安岭典型阔叶红松林的干扰机制之一。 相似文献

Arctic shrubs have a strong potential for climate and environmental reconstructions in the chronically understudied regions of the high northern latitudes. The climate dynamics of these regions are important to understand because of large-scale feedbacks to the global climate system. However, little is known about other factors influencing shrub ring growth, possibly obscuring their climate signal. For example, as of yet we are not able to differentiate between herbivory or climatically induced growth depressions. Here, we use one of the most common Arctic shrubs, Alnus viridis as a test case to address this question. We sampled Alnus in Kobbefjord, Greenland, measured shrub-ring width and cell wall thickness and built site chronologies of each parameter. We analysed climate-growth relationships, tested their stability over time and employed a pointer-year analysis to detect growth depressions. We employed bootstrapped transfer function stability tests (BTFS) to assess the suitability of our shrub chronologies for climate reconstruction. Correlations with climate data showed strong significantly positive and stable correlations between summer temperature and ring-width with the exception of the recent decade. A climate reconstruction model failed stability tests, when the complete period of record was used for calibration and verification. Wood anatomy analysis uncovered the occurrence of unusual cell structure (very thin cell walls) in the exceptionally narrow ring of 2004, a recorded insect outbreak year in other parts of Greenland. When excluding the affected ring and a recovery period, the reconstruction model passed all tests, suggesting that the unusual 2004 ring was not climate driven, but rather the result of an insect attack. When combining anatomical analysis with traditional ring-width measurements, we move a step further in potentially distinguishing small rings caused by insect attacks from small rings formed in climatically challenging years. While this study does not provide unambiguous evidence, it does provide potential useful methodological combinations to enable more robust climate reconstructions in areas where climatic records are extremely sparse. 相似文献

Creosote was evaluated as an inexpensive carbon source for growing inocula of a polycyclic aromatic hydrocarbon (PAH)-degrading
bacterial community (community five). Creosote was a poor growth substrate when provided as sole carbon source in a basal
salts solution (BSM). Alternatively, peptone, yeast extract or glucose in BSM supported high growth rates, but community five
could not subsequently degrade pyrene. A combination of creosote and yeast extract in BSM (CYEM) supported growth and maintained
the pyrene-degrading capacity of community five. Optimum pyrene-degrading activity occurred when the inocula were grown in
creosote and yeast extract concentrations of 2 ml L−1 and 1 g L−1 respectively: concentrations outside these values resulted in either low biomass yields or loss of PAH-degrading activity.
CYEM-grown community five inocula degraded 250 mg L−1 of pyrene in BSM at a rate comparable to cultures inoculated with community five grown in BSM-pyrene. However, the CYEM-grown
community showed a 40% lower rate of PAH degradation in a synthetic PAH mixture compared with pyrene-grown cells and there
was an increase in the lag period before the onset of PAH degradation. This appears to reflect a weaker induction of PAH catabolism
