Anabaena sp. grew with mono- and di-ester phosphate compounds as sources of phosphate, indicating the presence of phosphomonoesterase (PMEase) and phosphodiesterase (PDEase) activities. Cell-bound PMEase and PDEase activities were detected during growth in 0.5 and 10 mg PO4l–1 only when the cellular phosphate concentration fell to 0.46% of cell protein and the activities increased as cellular phosphate content decreased. The Km values for these enzymes were 0.3mm forp-nitrophenyl phosphate and 0.2mm for bis-p-nitrophenyl phosphate, respectively. Only PMEase activity was found extracellularly. The pH optima for PMEase and PDEase were 10.2 and 10.4, respectively, and the temperature optima at pH 10.2 were 37°C and 40°C, respectively. Ca2+ increased the enzyme activities while Zn2+ caused marked inhibition. The inorganic phosphate repressed the cellular PMEase activity after a lag of 4 h.  相似文献

We aimed at the investigation of the airborne fungiand their outdoor incidence in five vegetable growingareas in Edirne province (Turkey) by exposing a petridish with potato dextrose agar medium to air for 15minutes and then counting the number of growingcolonies. Sampling procedure for fungi was performed6 times in research stations at an interval of onemonth between April–September 1996. From the 90petri dishes obtained fungi were isolated and 1166colonies were counted. 12 genera (Absidia,Alternaria, Aspergillus, Botryotrichum, Chlamydomyces,Cladosporium, Endocochlus, Fusarium, Nematochtonus,Penicillium, Trichoderma and Torula) and 25species were identified. Among them, Aspergillusclavato-nanica and Penicillium estinogenum arevery likely to be new records for Turkey. Cladosporium carpophilum and Alternariaalternata were the most abundant species in the studyarea. Correlation analyses were applied to the data.  相似文献

Aims The introduction of potentially invasive species through ornamental cultivation or for rehabilitation purposes is a serious environmental problem. They cause damage to biodiversity through loss, increased mortality or ' in situ ' selection phenomena in natural flora. Spartium junceum is a Mediterranean shrub that is not native in most areas of the Iberian Peninsula, although it is extensively grown for the rehabilitation of roadsides. We have investigated the effect on the native vegetation of an old S. junceum (Fabaceae) plantation in a conservation area in the centre of the Iberian Peninsula: the Cuenca Alta del Manzanares nature reserve in Madrid.Methods We compared S. junceum stands with the native nanophanerophytic Cistus ladanifer community at different ecosystem properties: soil properties, temporal soil seed bank contents, standing vegetation and net primary production of annual grasslands growing in these shrublands.Important findings The results highlighted S. junceum 's ability to become established in the new environment (marginal areas of the nature reserve) and ensure its successful growth. This is more apparent in northern and eastern exposures where this formation contacts with the core of the best conserved native vegetation in the nature reserve. Soils under Spartium showed a higher nitrogen content, indicating its capacity—in common with other legumes—to fix N, and conferring an advantage over Cistus, which is N-limited. Other soil nutrients such as phosphorus, magnesium and calcium and water availability are also higher in Spartium soils than in Cistus. Phosphorus is usually a constraint for N-fixers, but in our study, it is the most significant soil variable in both shrub formations and is important to the success of Spartium. Water availability is a key factor for Mediterranean vegetation, and particularly in autumn when soils are recharged. The Spartium formation is able to retain water as its growth produces a closer canopy than Cistus, thereby preventing water evaporation and contributing to the success of this species. Perennials are more frequent in the standing vegetation than in the seed bank, whereas therophytes are similar. The standing vegetation has therophytes and chamaephytes as the predominant growth forms in Spartium sites, and hemicryptophytes and phanerophytes in Cistus. Therophytes are dominant in Spartium and Cistus seed banks, although the first formation has more species. Spartium has a higher number of hemicryptophytes and Cistus is higher in phanerophytes. Northern and eastern aspects show significant differences in richness—with a predominance of annual weed species in Spartium —and in above-ground net primary production, probably as a consequence of the nutrients present in the soils. Ruderal annual species under Spartium (Bromus tectorum, Chenopodium album) have higher germination rates in the greenhouse than in the standing vegetation, suggesting they are at potential risk if environmental conditions change.  相似文献

The visual fields of vultures contain a small binocular region and large blind areas above, below and behind the head. Head positions typically adopted by foraging vultures suggest that these visual fields provide comprehensive visual coverage of the ground below, prohibit the eyes from imaging the sun and provide extensive visual coverage laterally. However, vultures will often be blind in the direction of travel. We conclude that by erecting structures such as wind turbines, which extend into open airspace, humans have provided a perceptual challenge that the vision of foraging vultures cannot overcome.  相似文献

研究了植物源杀虫剂苦豆碱及顺式氯氰菊酯对菜田蚜虫群落结构的影响 ,并测定了其对第一优势种菜缢管蚜的杀伤毒力。苦豆碱在 1 %浓度下 ,以药膜法和喷雾法 ,测定其击倒作用 ,KT50 分别为2 9 74和 2 5 38min ;顺式氯氰菊酯在 0 1 %浓度下 ,KT50 分别为 1 73和 0 78min。以点滴法测定此两种杀虫剂对菜缢管蚜的触杀作用 ,LD50 分别为 7 7× 1 0 -4 及 9 2 0 1 μg 头 ,田间以 0 1 %喷雾 ,苦豆碱 1 0d校正防效均可达到 80 % ,而顺式氯氰菊酯仅能达到 30 %。结果显示 ,苦豆碱对菜缢管蚜有极强的毒杀作用 ,同时喷药前后菜田蚜虫群落结构也发生着明显的变化。  相似文献

Recent studies of visually guided reaching in monkeys support the hypothesis that the visuomotor transformations underlying arm movements to spatial targets involve a parallel mechanism that simultaneously engages functionally related frontal and parietal areas linked by reciprocal cortico-cortical connections. The neurons in these areas possess similar combinations of response properties. The multimodal combinatorial properties of these neurons and the gradient architecture of the parieto-frontal network emerge as a potential substrate to link the different sensory and motor signals that arise during reaching behavior into common hybrid reference frames. This convergent combinatorial process is evident at early stages of visual information processing in the occipito-parietal cortex, suggesting the existence of re-entrant motor influences on cortical areas once believed to have only visual functions.  相似文献

Fusarium species were isolated from plant debris in soil samples collected from cultivated maize fields and from undisturbed grasslands in two areas of the Transkei region. A total of 1205 Fusarium isolates were recovered from 27 soil samples. Fifteen Fusarium species were recovered from plant debris from Bizana soils and 13 Fusarium species from plant debris from Centane soils. The two dominant Fusarium species in both areas were F. oxysporum and F. equiseti. Very few isolates of F. moniliforme and F. subglutinans were recovered, but both of these species had significantly higher relative densities in cultivated soils than in undisturbed soils. 用细胞分析成像光盘记录系统测量了3T3细胞在某些ELF磁场和温度条件下生长周期的变化.实验结果表明,只有某些频率的ELF磁场才对细胞生长产生影响,磁场对细胞生长的影响还与培养细胞的生化环境有关.温度和ELF磁场都能影响细胞的生长.但二者的机理是不一样的,当撤除ELF磁场后,细胞在短时间内(2天以上)继续保持着ELF场对其生长的影响.而细胞生长周期能在短时间内(2天以内)随着温度的变化而变化.温度引起的细胞生长的变化可能与细胞内的各种生长因子、生物离子的活性有关.ELF磁场引起的细胞生长的变化可能与ELF磁场对细胞膜的影响有关,与细胞内细胞生长必不可少的生物离子(如Ca~(2+))的浓度有关.  相似文献

Magnetic fields (MFs) have been used as an external stimulus to increase cell proliferation in chondrocytes and extracellular matrix (ECM) synthesis of articular cartilage. However, previously published studies have not shown that MFs are homogeneous through cell culture systems. In addition, variables such as stimulation times and MF intensities have not been standardized to obtain the best cellular proliferative rate or an increase in molecular synthesis of ECM. In this work, a stimulation device, which produces homogeneous MFs to stimulate cell culture surfaces was designed and manufactured using a computational model. Furthermore, an in vitro culture of primary rat chondrocytes was established and stimulated with two MF schemes to measure both proliferation and ECM synthesis. The best proliferation rate was obtained with an MF of 2 mT applied for 3 h, every 6 h for 8 days. In addition, the increase in the synthesis of glycosaminoglycans was statistically significant when cells were stimulated with an MF of 2 mT applied for 5 h, every 6 h for 8 days. These findings suggest that a stimulation with MFs is a promising tool that could be used to improve in vitro treatments such as autologous chondrocyte implantation, either to increase cell proliferation or stimulate molecular synthesis. PurposeThe management of the occupational exposure to electromagnetic fields (EMF), an Occupational Health and Safety (OHS) issue of great scientific, social and economic significance, was under intense negotiations at European level over the last twenty years; the Directive 2013/35/EU is the new legislative tool. The presented study deals with the practical aspects of the Directive’s implementation.MethodsThe appropriate, extensive measurements and the overall EMF exposure assessments (i.e. exposure mapping, identification of hot spots, proposition of solutions) were conducted in specific workplaces, including power production, railway, broadcasting, clinical Magnetic Resonance Imaging (MRI) systems, industrial and research sites, as well as common office workplaces.ResultsThe vast majority of the performed EMF assessments did not reveal occupational overexposures; moreover in most of the cases, even the general public exposure limits (in the above occupational areas) were not exceeded. The very few localized overexposures detected, were manageable on the basis of the technical and organizational OHS principles.On the contrary, the maintenance procedures of the EMF emitting equipment, as recorded in this survey, presented overexposures revealing a challenging field.ConclusionsThis study lays a firm basis for the clarification of the occupational EMF environment, where potential exposures might be high. The proper risk assessment demands precise exposure identification and deep understanding of the EMF nature and hazards. Misconceptions range from the common exposure overestimation to the rarer case of the maintenance hazards underestimation, while attention is needed concerning the proper application of the complex limiting system of the Directive.  相似文献
