Abstract Stony downs consist of grassy areas that alternate with areas that have a substantial stone cover. The stone-covered areas are impermeable, and most rain falling on them runs off, substantially increasing the effective rainfall in adjacent grassy areas. As a result, 20–25 mm of rain on stony downs wetted the soil around the grass to a depth of 140–170 mm and allowed sustained grass response. This is much less than the 35–40 mm of rain required for the same response on red clay or grey clay plains. Grasses respond very rapidly after rain. Some have green shoots the day after rain, and all have responded by the second day. Ephemerals dry off in 4–6 weeks, but most tussock grasses still have some green foliage 8–10 weeks after rain. Deeper rooted tussock grasses remain green for so long because most of the moisture that reaches deeper roots after rain remains there. Most moisture loss is through the soil surface and is recognizable as a drying front that descends through the soil profile. Soil above the drying front is nearly air dry (<5% moisture) while soil below the front has substantial moisture (14–16%). By about a month after rain in summer, the drying front is at a depth of about 80–120 mm. This is near the tips of the roots of ephemeral grasses and the ephemerals then dry off rapidly. Only the tips of the leaves of deep rooted grasses like Mitchell grass (Astrebla spp.) dry off. Their leaves continue to remain mostly green during most of the second month after rain and they do not dry off completely until the third month when the drying front reaches the bottom of the main root system.  相似文献

A tree-ring densitometric transect from Alaska to Labrador   总被引:1,自引:0,他引:1  
We describe a recently completed network of densitometric tree-ring time series representing various aspects of tree-growth for up to 200 years at 69 sites spread across the northern North American conifer zone from Yukon to Labrador. Duplicate cores, from 12 to 15 trees per site, provide time series for a suite of growth parameters including earlywood (spring), latewood (summer) and total (annual) ring widths and mean earlywood, mean latewood, minimum and maximum ring density. These data form the basis for extensive analyses of intra- and inter-site parameter comparisons and regional climate/tree-growth comparisons. Five large-scale regional chronologies do not suggest that any anomalous growth increases have occurred in recent decades, at least on these regional scales, despite the observed changes in atmospheric composition and climate.  相似文献

A comprehensive study of the application of continuous zone electrophoresis to preparative separation of proteins in free solution is presented. First, the influence of electric field strength, buffer residence time in the chamber, sample flow rate, and sample concentration on separation resolution and throughput were studied. Using multiple injections of sample into the electrophoresis chamber, a throughput of 500 mg protein/h was achieved for partially purified model proteins. Experiments on Escherichia coli crude extracts yielded a fivefold purification of beta-galactosidase along with a simultaneous separation of proteins from cell debris in a single step. Experiments correlating the electrophoretic mobility in continuous electrophoresis with the elution behavior in ion-exchange chromatography were performed on more than a dozen proteins which conclusively showed that separation of proteins in continuous zone electrophoresis is governed by net surface charge. Based on these results, the fraction numbers in which the proteins eluted could be correctly predicted. Proteins and enzymes with differences >0.5 M elution molarities in ion-exchange chromatography were separated by continuous zone electrophoresis on a preparative scale (mg/h or g/h) with >90% recovery. This corresponds to a preparative scale separation of proteins and enzymes which differ in apparent electrophoretic mobility by only 0.70 x 10(-5) cm(2)/V . s. The extent to which alleles can disperse across a hybrid zone depends on the selection they are subjected to in the hybrid genetic background or, for those that are selectively neutral, on their ability to escape from the unfavourable environment by recombination. Three markers spanning a 45 cM segment in the center of the X chromosome were used to investigate the degree to which selection against X chromosome linked genes helps to maintain the barrier to gene flow in the hybrid zone between Mus musculus domesticus and M. m. musculus in Denmark. The introgression of all the sex chromosome specific markers was more limited than that of the autosomal enzymes (Idh1, Amy, Gpd1, Pgm1, Es1, Es2, Mpi, Np1, Es10, Sod1) and the mitochondrial DNA. The cline for DXPas2, which is in the center of the X chromosome, is extremely steep and shows that certain genes located in this region are strongly selected against in the hybrid background. The clines of the other two X-linked markers, Hprt and DXPas1, and of the Y chromosome are not as abrupt and all three have similar asymmetric introgression patterns. Although the musculus variants appear to behave in much the same way as those of the autosomal genes, the domesticus variants do not introgress. The results show that X-linked and to a lesser extent Y-linked genes are more strongly selected against in the hybrid genome than the mitochondrial genome or the different autosomal loci. This suggests that co-adapted gene systems involving the sex chromosomes may play an important role in the hybrid breakdown between the two subspecies.  相似文献

Microtubular basal bodies and epiplasm (membrane skeleton) are the main components of the cortical skeleton of Tetrahymena. The aim of this report was to study functional interactions of basal bodies and epiplasm during the cell cycle. The cortex of Tetrahymena cells was stained with anti-epiplasm antibody. This staining produced a bright epiplasmic layer with a dark pattern of unstained microtubular structures. The fluorescence of the anti-epiplasm antibody disappeared at sites of newly formed microtubular structures, so the new basal body domains and epiplasmic layer could be followed throughout the cell cycle. Different patterns of deployment of new basal bodies were observed in early and advanced dividers. In advanced dividers the fluorescence of the epiplasmic layer diminished locally within the forming fission line where the polymerization of new basal bodies largely extincted. In wild type Tetrahymena, the completion of the micronuclear metaphase/anaphase transition was associated with a transition from the pattern of new basal body deployment and epiplasm staining of the early divider to the pattern of the advanced dividers. The signal for the fission line formation in Tetrahymena (absent in cdaA1 Tetrahymena mutationally arrested in cytokinesis) brings about 1) transition of patterns of deployment of basal bodies and epiplasmic layer on both sides of the fission line; and 2) coordination of cortical divisional morphogenesis with the micronuclear mitotic cycle.  相似文献

Interaction of a 70-amino acid zinc-binding polypeptide from the regulatory chain of aspartate transcarbamoylase (ATCase) with the catalytic (C) subunit leads to dramatic changes in enzyme activity and affinity for ligand binding at the active sites. The complex between the polypeptide (zinc domain) and wild-type C trimer exhibits hyperbolic kinetics in contrast to the sigmoidal kinetics observed with the intact holoenzyme. Moreover, the Scatchard plot for binding N-(phosphonacetyl)-L-aspartate (PALA) to the complex is linear with a Kd corresponding to that evaluated for the holoenzyme converted to the relaxed (R) state. Additional evidence that the binding of the zinc domain to the C trimer converts it to the R state was attained with a mutant form of ATCase in which Lys 164 in the catalytic chain is replaced by Glu. As shown previously (Newell, J.O. & Schachman, H.K., 1990, Biophys. Chem. 37, 183-196), this mutant holoenzyme, which exists in the R conformation even in the absence of active site ligands, has a 50-fold greater affinity for PALA than the free C subunit. Adding the zinc domain to the C trimer containing the Lys 164-->Glu substitution leads to a 50-fold enhancement in the affinity for the bisubstrate analog yielding a value of Kd equal to that for the holoenzyme. A different mutant ATCase containing the Gln 231 to Ile replacement was shown (Peterson, C.B., Burman, D.L., & Schachman, H.K., 1992, Biochemistry 31, 8508-8515) to be much less active as a holoenzyme than as the free C trimer. For this mutant holoenzyme, the addition of substrates does not cause its conversion to the R state. However, the addition of the zinc domain to the Gln 231-->Ile C trimer leads to a marked increase in enzyme activity, and PALA binding data indicate that the complex resembles the R state of the holoenzyme. This interaction leading to a more active conformation serves as a model of intergenic complementation in which peptide binding to a protein causes a conformational correction at a site remote from the interacting surfaces resulting in activation of the protein. This linkage was also demonstrated by difference spectroscopy using a chromophore covalently bound at the active site, which served as a spectral probe for a local conformational change. The binding of ligands at the active sites was shown also to lead to a strengthening of the interaction between the zinc domain and the C trimer.  相似文献

The coevolutionary relationship between fungus-growing ants (Formicidae: Attini: Attina) and their symbionts has been well studied in the Panamanian rain forests. To further understand the ecological context of these evolutionary relationships, we have examined the population-genetic structure of the fungus-growing ant species Mycetomoellerius mikromelanos Cardenas, Schultz, Adams 2021 in the Panama Canal Zone. We specifically investigated the presence of population structure, the significance of geographic features (i.e., creeks) limiting gene flow, and relatedness between ant colonies. To accomplish this, we genotyped 85 ant colonies from nine creeks across an approximately 30 km transect in Parque National Soberanía, Panama, using double-digest restriction-site-associated DNA sequencing. We did not find distinct population structure using two genetic clustering methods; however, we did detect an effect of isolation by distance. Furthermore, related colonies were frequently detected on the same creek or neighboring creeks, and some at further geographic distances. Collectively, these findings demonstrate that new colonies tend to establish on natal creeks and occasionally on distant creeks following long-distance dispersal events. We discuss how population-genetic patterns reveal the natural history of M. mikromelanos in Parque National Soberanía and how these results fit into the context of fungus-growing ant mutualisms. Nitrogen-fixing (acetylene-reducing) activity of intact saplings ofAlnus maximowiczii was measured under natural conditions in the subalpine zone of Mt Fuji. The nitrogen-fixing activity was detected from the middle of June when expansion of leaves had just begun to the end of October when the shedding of leaves was almost completed. Diurnal changes in the activity were almost parallel with those of ground temperature. The measured nitrogen-fixing activity was related to ground temperature and total leaf area. Using this relation, annual nitrogen fixation was estimated from the data of ground temperature and leaf area measured in the field. The amount of annual nitrogen fixation was almost the same as that of nitrogen used for annual growth. It was concluded that nitrogen fixation by nodules made a considerable contribution to the nitrogen economy in the saplings ofA. maximowiczii.  相似文献

动物能量代谢相关的生理生态特征与其地理分布密切相关。为探讨温州地区迁徙鸟类小杓鹬(Numenius minutus)的代谢产热特征及体温调节,本文在环境温度(Ta)5.0~42.5℃范围内,测定了小杓鹬的代谢率(Rm,以单位时间耗氧量表示,ml/h)和体温,并计算不同环境温度的热传导。结果显示:在环境温度为5~35℃的范围内,小杓鹬的体温维持相对恒定,平均体温为(42.8±0.10)℃;热中性区为27.5~40.0℃;在热中性区温度范围内,代谢率即基础代谢率为(221.31±6.01)ml/h,是体重预期值的141%;环境温度在5.0~27.5℃范围内,代谢率与环境温度(Ta,℃)呈负相关,回归方程为Rm=587.10﹣11.78 Ta;在5.0~27.5℃的环境温度范围内,小杓鹬的热传导最低,平均为(0.11±0.00)ml/(g·h·℃),是体重预期值的212%;代谢预期比和热传导预期比的比值(F值)为1.21,表明该物种有较好的体温调节能力。小杓鹬具有较高的体温和基础代谢...  相似文献
