Catalases are the most important enzymatic systems used to degrade hydrogen peroxide (H2O2) into water and oxygen, thereby lowering intracellular hydrogen peroxide levels. Entomopathogenic fungi display increased catalase activity during germination and growth, which is necessary to counteract the hyperoxidant state produced by oxidative metabolism. We studied the influence of five different hydrocarbons on catalase production by Lecanicillium muscarium to determine the importance of catalase induction in fungal germination, stress tolerance and virulence. Conidia produced by colonies grown on different hydrocarbons showed higher rates of catalase activity compared to the control and the catalase activity of conidia produced on n-octacosane was three times higher than the activity of the control. This increase in catalase activity was accompanied by a higher level of resistance to exogenous hydrogen peroxide and a reduction in the germination time. Our study has helped to identify that increased catalase activity improves the germination and tolerance to different antioxidant stress response of L. muscarium.  相似文献

Protein–peptide interactions are a common occurrence and essential for numerous cellular processes, and frequently explored in broad applications within biology, medicine, and proteomics. Therefore, understanding the molecular mechanism(s) of protein–peptide recognition, specificity, and binding interactions will be essential. In this study, we report the first detailed analysis of antibody–peptide interaction characteristics, by combining large‐scale experimental peptide binding data with the structural analysis of eight human recombinant antibodies and numerous peptides, targeting tryptic mammalian and eukaryote proteomes. The results consistently revealed that promiscuous peptide‐binding interactions, that is, both specific and degenerate binding, were exhibited by all antibodies, and the discovery was corroborated by orthogonal data, indicating that this might be a general phenomenon for low‐affinity antibody–peptide interactions. The molecular mechanism for the degenerate peptide‐binding specificity appeared to be executed through the use of 2–3 semi‐conserved anchor residues in the C‐terminal part of the peptides, in analogue to the mechanism utilized by the major histocompatibility complex–peptide complexes. In the long‐term, this knowledge will be instrumental for advancing our fundamental understanding of protein–peptide interactions, as well as for designing, generating, and applying peptide specific antibodies, or peptide‐binding proteins in general, in various biotechnical and medical applications.  相似文献

Aims: To evaluate the diversity and antimicrobial activity of bacteria from the marine sponges Suberites carnosus and Leucosolenia sp. Methods and Results: Two hundred and thirty‐seven bacteria were isolated from the sponges S. carnosus (Demospongiae) and Leucosolenia sp. (Calcarea). Isolates from the phyla Actinobacteria, Bacteroidetes, Firmicutes and Proteobacteria were obtained. Isolates of the genus Pseudovibrio were dominant among the bacteria from S. carnosus, whereas Pseudoalteromonas and Vibrio were the dominant genera isolated from Leucosolenia sp. Approximately 50% of the isolates from S. carnosus displayed antibacterial activity, and c. 15% of the isolates from Leucosolenia sp. demonstrated activity against the test fungal strains. The antibacterial activity observed was mostly from Pseudovibrio and Spongiobacter isolates, while the majority of the antifungal activity was observed from the Pseudoalteromonas, Bacillus and Vibrio isolates. Conclusions: Both sponges possess a diverse range of bioactive and potentially novel bacteria. Differences observed from the sponge‐derived groups of isolates in terms of bioactivity suggest that S. carnosus isolates may be a better source of antibacterial compounds, while Leucosolenia sp. isolates appear to be a better source of antifungal compounds. Significance and Impact of the Study: This is the first study in which cultured bacterial isolates from the marine sponges S. carnosus and a Leucosolenia sp. have been evaluated for their antibacterial activity. The high percentage of antibacterial isolates from S. carnosus and of antifungal isolates from Leucosolenia sp. suggests that these two sponges may be good sources for potentially novel marine natural products.  相似文献

Arsenical compounds exhibit a differential toxicity to cancer cells. Microtubules are a primary target of a number of anticancer drugs, such as arsenical compounds. The interaction of 1-NAA (1-naphthylarsonic acid) has been investigated on microtubule polymerization under in vitro and cellular conditions. Microtubules were extracted from sheep brain. Transmission electron microscopy was used to show microtubule structure in the presence of 1-NAA. Computational docking method was applied for the discovery of ligand-binding sites on the microtubular proteins. Proliferation of HeLa cells and HF2 (human foreskin fibroblasts) was measured by the MTT [3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl-2H-tetrazolium bromide] assay method following their incubation with 1-NAA. Fluorescence microscopic labelling was done with the help of α-tubulin monoclonal antibody and Tunel kit was used to investigate the apoptotic effects of 1-NAA on the HeLa cells. 1-NAA inhibits the tubulin polymerization by the formation of abnormal polymers having high affinity to the inner cell wall.  相似文献

Bactenecin (Bac) is a 12‐residue disulfide‐linked antimicrobial peptide isolated from the granules of bovine neutrophils. In this study, to develop novel linear Bac analogs with cell selectivity and anti‐endotoxic activity, we designed and synthesized a series of linear Bac analogs with amino acid substitution in Cys3,11 and/or Val6,7 of Bac. Among Bac analogs, some analogs (Bac‐W, Bac‐KW, Bac‐L, Bac‐KL, Bac‐LW, and Bac‐KLW) with higher hydrophobicity showed the amalgamated property of cell selectivity and anti‐endotoxic activity. Furthermore, Bac‐W, Bac‐KW, Bac‐LW, and Bac‐KLW showed serum stability comparable with that of disulfide‐bonded Bac. Therefore, these Bac analogs (Bac‐W, Bac‐KW, Bac‐LW, and Bac‐KLW) can serve as promising antibiotics for the development of therapeutic agents for treatment against endotoxic shock and bacterial infection. In addition, our results suggest that a little increase in hydrophobicity may be responsible for the decreased cell selectivity of the multiple Arg‐containing peptides (Bac‐W, Bac‐L, and Bac‐LW) over the multiple Lys‐containing peptides (Bac‐KW, Bac‐KL, and Bac‐KLW). A new series of smaller peptides with tryptophan at C‐terminal and varying N‐protected amino acids/peptides were designed, synthesized and characterized by analytical and spectroscopic techniques. Analgesic and anti‐inflammatory properties of these peptides were carried out in vivo using tail‐flick method and carrageenan‐induced paw edema method, respectively, at different doses and different time intervals. Most of the peptides synthesized displayed enhanced activity, and particularly tetra and hexapeptides 29–31 were found to be even more potent than the reference standards used. Moreover, some peptides have exhibited promising activity even after 24 h of administration, whereas the reference standards were active only up to 3 h.   • 1 DISRUPT Micro‐Flake Verbenone Bark Beetle Anti‐Aggregant flakes (Hercon Environmental, Inc., Emigsville, Pennsylvania) were applied in two large‐scale tests to assess their efficacy for protecting whitebark pine Pinus albicaulis Engelm. from attack by mountain pine beetle Dendroctonus ponderosae Hopkins (Coleoptera: Scolytinae) (MPB). At two locations, five plots of equivalent size and stand structure served as untreated controls. All plots had early‐ to mid‐outbreak beetle populations (i.e. 7.1–29.2 attacked trees/ha). Verbenone was applied at 370 g/ha in both studies. Intercept traps baited with MPB aggregation pheromone were placed near the corners of each plot after the treatment in order to monitor beetle flight within the plots. Trap catches were collected at 7‐ to 14‐day intervals, and assessments were made at the end of the season of stand structure, stand composition and MPB attack rate for the current and previous years.
  • 2 Applications of verbenone flakes significantly reduced the numbers of beetles trapped in treated plots compared with controls at both sites by approximately 50% at the first collection date.
  • 3 The applications also significantly reduced the proportion of trees attacked in both Wyoming and Washington using the proportion of trees attacked the previous year as a covariate in the model for analysis of current year attack rates; in both sites, the reduction was ≥ 50%.
  • 4 The flake formulation of verbenone appears to have promise for area‐wide treatment by aerial application when aiming to control the mountain pine beetle in whitebark pine forests.

Predator mobbing is a widespread phenomenon in many taxa but the evolution of cooperative mobbing as an adaptive behavior is still subject to debate. Here, we report evidence for cooperative predator defense in a nocturnal solitarily foraging primate, the gray mouse lemur (Microcebus murinus). Several mouse lemurs mobbed a snake that held a non-related male conspecific until he could escape. Evolutionary hypotheses to explain cooperative mobbing include (1) by-product mutualism, when individuals defend others in the process of defending themselves; (2) reciprocity, where animals achieve a higher fitness when helping each other than when they do not cooperate; and (3) kin selection where animals help each other only if they share genes by common descent. Owing to the solitary activity of this species, reciprocity seems to be least likely to explain our observations. By-product mutualism cannot be ruled out entirely but, if costs of snake mobbing are relatively low, the available detailed socio-genetic information indicates that kin selection, rather than any of the other proposed mechanisms, is the primary evolutionary force behind the observed cooperative rescue.  相似文献
