Resistance to the alfalfa weevil (Hypera postica (Gyllenhal)) and the potato leafhopper (Empoasco fabae (Harris)) is lacking in cultivated alfalfa. However, a closely related annual Medicago, Medicago scutellata, possesses dense glandular stem and leaf hairs which provides a mechanism for resistance. Several attempts have been made at transfering the glandular haired traint from M. scutellata to perennial alfalfa with limited success. Earlier studies have shown that one reason for the lack of success is embryo abortion. Therefore, this study was initiated to observe zygotic embryo-genesis and to develop an embryo rescue technique for M. scutellata and M. sativa. Observations of zygotic embryogenesis showed that the two species are similar in morphology and can be described from youngest to oldest as globular, heart, torpedo, and hook shaped embryos. M. sativa embryos are smaller than M. scutellata embryos and develop three to four days later. Self pollinated M. scutellata (PI 307446) and sib mated M. sativa (Saranac AR) embryos were cultivated on Murashige and (2,4-D), indolacetic acid (IAA), 6-benzylaminopurine (BAP), and kinetic (KIN). Embryos from both species were also cultured on Schenk and Hildebrandt's (SH) basal medium with the addition of L-glutamine and L-proline. The experimental design was a completely randomized factorial for each experiment. Heart and torpedo shaped embryos from M. scutellata grew best (27.5% plantlet recovery) when cultured on MS medium with 0.05 mgl-1 of both IAA and BAP. After 15 to 30 days on this medium, the embryos had only developed shoots. Therefore, it was necessary to transfer the shoots to MS basal medium without phytohormones for rooting. Rooting occurred in 15 to 30 days and the plantlets could be acclimatized to soil within 2 to 4 weeks. M. sativa embryos grew best (31% plantlet recovery) on SH medium with 50 mM L-glutamine. M. sativa embryos developed both shoots and roots on this medium. This information may now be applied to the development of an embryo culture method for recovering insect resistant hybrids between M. scutellata and M. sativa. We propose a technique for separating the climatic signal which is contained in two tree-ring parameters widely used in dendroclimatology. The method is based on the removal of the relationship between tree-ring width (TRW) and maximum latewood density (MXD) observed for narrow tree rings from high latitudes. The new technique is tested on data from three larch stands located along the northern timberline in Eurasia. Correlations were calculated between the temperatures of pentads (five consecutive days), TRW chronologies and MXD chronologies calculated according to the standard and proposed methods. The analysis confirms the great importance of summer temperature for tree radial growth and tree-ring formation. TRW is positively correlated with the temperature of four to eight pentads (depending on the region) at the beginning of the growth season, but MXD as obtained by the standard technique is correlated with temperature over a much longer period. For maximum density series from which the relationship between MXD and TRW has been removed (MXD′), there is a clear correlation with temperatures in the second part of the growing season. These results are consistent with the known dynamics of tree-ring growth in high latitudes and mechanisms of tree-ring formation.  相似文献

The aim of this work was to analyse the ultrastructure of storage crypts and stored spermatozoa, and to describe changes during the annual reproductive cycle of the bluemouth Helicolenus dactylopterus dactylopterus , which has internal fertilization and a zygoparous mode of reproduction. Spermatozoa had elongated heads and long midpieces, two characteristics which are thought to be fairly advanced and correlated with internal fertilization, as is the case of the bluemouth. A remarkable spermatozoon feature was the retention of a significant quantity of cytoplasm around the head, a condition that appeared to be related to nourishment during the long storage period, up to 10 months in the intraovarian crystal structures of the female. Male sex cells' protection against the female immune system was ensured by junctional complexes between the crypt cells composed of tight junctions and desmosomes.  相似文献

Abstract Lesquerella stonensis (Brassicaceae) is an obligate winter annual endemic to a small portion of Rutherford County in the Central Basin of Tennessee, where it grows in disturbed habitats. This species forms a persistent seed bank, and seeds remain viable in the soil for at least 6 years. Seeds are dormant at maturity in May and are dispersed as soon as they ripen. Some of the seeds produced in the current year, as well as some of those in the persistent seed bank, afterripen during late spring and summer; others do not afterripen and thus remain dormant. Seeds require actual or simulated spring/summer temperatures to come out of dormancy. Germination occurs in September and October. Fully afterripened seeds germinate over a wide range of thermoperiods (15/6–35/20°C) and to a much higher percentage in light (14 h photoperiod) than in darkness. The optimum daily thermoperiod for germination was 30/15°C. Nondormant seeds that do not germinate in autumn are induced back into dormancy (secondary dormancy) by low temperatures (e.g., 5°C) during winter, and those that are dormant do not afterripen; thus seeds cannot germinate in spring. These seed dormancy/ germination characteristics of L. stonensis do not differ from those reported for some geographically widespread, weedy species of winter annuals and thus do not help account for the narrow endemism of this species.  相似文献

6-C-α-l-Arabinopyranosyl- and furanosylacacetins have been synthesized. They are isomerized by short acid treatment to give a mixture of the four anomer/ring size combinations without any Wessely-Moser isomerization. In the same conditions molludistin (8-C-α-l-arabinopyranosylgenkwanin) led only to a mixture of molludistin and 8-C-α-l-arabinofuranosylgenkwanin. This is the first demonstration of ring sugar isomerization in C-glycosylflavones. In usual solvent systems, α-anomers are easily separated from β-anomers, whereas corresponding pyranosyl and furanosyl anomers are not. However, they are easily separated after permethylation and characteristic features are found in the mass spectra of PM 6-C-arabinofuranosyl isomers.  相似文献

It has been established that organelles, such as mitochondria and plastids, contain organelle-specific DNA and arise from the division of pre-existing organelles (e.g., Possingham and Lawrence, 1983). We propose that organelle DNAs, such as mitochondrial DNA and plastid DNA are not naked in organellesin situ but are organized in each case to form an “organelle nucleus” with basic proteins (Kuroiwa, 1982). The concept of organelle nuclei has changed our ideas about the division of organelles. Thus, the process of organelle division must be composed of two main events: division of the organelle nucleus and organellekinesis (division of the other components of the mitochondrion or plastid). The latter term has been adopted as an appropriate analogue of cytokinesis. We were the first to identify the plastid-dividing ring (PD-ring), which is located in the cytoplasm close to the outer envelope membrane at the constricted isthmus of dividing chloroplasts in the red algaCyanidium caldirum. The PD-ring is about 60 nm in width and 25 nm in thickness, and is a circular bundle of actin-like, fine filaments, each about 4–5 nm in diameter. Since cytochalasin B, an inhibitor of polymerization of actin filaments, inhibits the formation of the PD-ring and, thus, prevents subsequent division of chloroplasts, the PD-ring is thought to be a structure that is essential for the division of plastids (plastidkinesis). The behavior of the PD-ring during a cycle of chloroplast division can be classified into the following four stages on the basis of morphological and temporal differences. The chloroplast growth stage: the small, spherical chloroplast increases in volume and becomes a football-like structure, while the PD-ring from the previous division disappears. Formation of the PD-ring: the somewhat electron-dense body (see below) is fragmented into many, somewhat electron-dense granules, which are aligned along the equatorial region of the chloroplast and fine filaments are formed from the somewhat electron-dense granules in the equatorial region. The fine filaments of the PD-ring align themselves according to the longest axis of their overall domain, i.e., circumferentially. Contraction stage: a bundle of fine filaments begins to contract and generates a deep furrow. Conversion stage: after chloroplast division, the remnants of the PD-ring are converted into somewhat electron-dense bodies. Similar events occur during the second cycle of chloroplast division. Since similar structures are observed extensively in the plastids of algae, moss and higher plants, the PD-ring appears to be an essential structure for the division of plastids in plants.  相似文献

  • 1 Seasonal population growth rates for the pea aphid, Acyrthosiphon pisum Harris, were determined in three different host plant habitats; alfalfa, Medicago sativa (L.), clover, Trifolium pratense (L.), and peas, Pisum sativum (L.); over four years and eight places. It was possible to estimate a common intrinsic rate of increase for each host plant habitat.
  • 2 An analysis of the relative influence of temporal, spatial and host plant habitat variation showed that the host plant habitat was most important in determining the growth rates of the populations, both in rate of build-up and decline.
  • 3 Patterns of alate production in the three different habitats differed substantially between the annual peas and the two perennial legumes. During the summer, alate production was large and rapid in peas and remained low and constant in clover and alfalfa
  • 4 Parasitism was highest in peas. The species composition of parasitoids differed between crops.
  • 5 Aphids in annual peas had a higher intrinsic rate of increase and a faster rate of decline than in the two perennial legumes. This explains the presence of both migratory and sedentary forms among pea aphids.
