In situ polarization modulation infrared reflection absorption spectroscopy (PM-IRRAS) at the air-water interface has been used to determine secondary structure of the pulmonary surfactant model peptide, Hel 13-5, in the absence and the presence of phospholipid monolayers. Herein, fully saturated phospholipids of DPPC and DPPG are utilized to understand the effect of specific interaction between anionic DPPG and cationic Hel 13-5 on the peptide secondary structure. The spectrum frequency in the amide region (1500-1700 cm− 1) obtained from PM-IRRAS has been confirmed by comparing with that from ATR-FTIR for the corresponding bulk films. The PM-IRRAS spectra of single Hel 13-5 monolayers indicate the α-helical contour in the amide region, which coincides with the result from CD measurements in aqueous solutions. In the presence of phospholipid monolayers, however, Hel 13-5 changes its conformation from the α-helix to the extended β-sheet as surface pressure increases upon compression at the interface, and this interconversion is found to be irreversible even during expansion process of monolayers. Furthermore, it is notable that the electrostatic interaction between DPPG and Hel 13-5 inhibits to some extent the interconversion to the β-sheet during compression. These features are completely different from the bulk behavior, which demonstrates different roles of native proteins in the bulk phase and at the interface for pulmonary functions. In addition, the conformational variation of Hel 13-5 does not indicate close correlation with surface activity, which is common characteristic even for reversible hysteresis curves in pulmonary surfactant systems. This suggests that the secondary structure of native proteins is not strongly related to the surface activity during respiration. This work contributes to secondary structure determination of Hel 13-5 in the phospholipid domains in situ at the air-water interface and will provide insight into the molecular and physiological mechanism for SP-B and SP-C actions across the interface.  相似文献

α-Chymotrypsin and lysozyme were solubilized in a water/O-[(2-tridecyl, 2-ethyl-1,3-dioxolan-4-yl)methoxy]–O′-methoxy poly(ethylene glycol) (CK-2,13 surfactant)/isooctane water-in-oil microemulsion solution at 1.5–2 and 10 g l−1 for 0.15 and 1.2 M CK-2,13, respectively. Upon contact with an equal volume of 0.1 M NaH2PO4/Na2HPO4 buffer, pH 5, a three-phase system (Winsor-III system) was formed, consisting of a surfactant-rich middle phase and aqueous and isooctane-rich “excess” phases. Both enzymes were rapidly released into the aqueous excess phase, with 70% recovery of each in 30 and 60 min for microemulsion solutions containing 0.15 and 1.2 M surfactant, respectively. The recovered enzymes retained >90% of their original specific activity.  相似文献

The influence of four non-ionic surfactants (isododecyl and cetyl polyoxyethylene ethers) on aqueous polysaccharide solutions (sodium alginate, guar gum, and sodium carboxymethyl guar), applicable for textile printing pastes, were studied via rheological measurements.

Rheology of polysaccharide–surfactant solutions in aqueous matrices is primarily governed by polymer content, which imparts marked shear-thinning and viscoelastic character to the system. Such properties are modulated in moderate but sensible way by changes in surfactant concentration or type. Above 3% surfactants addition to non-substituted guar gum solutions results in a significant impact leading to phase separation and a particular strongly associated phase is formed due to hydrogen bonds between ethylene oxy units from the surfactant and primary hydroxyl groups in guar.

A satisfactory fitting of viscosity data is obtained with both the Cross equation and the Roberts–Barnes–Carew model. The experimental results of mechanical spectra can be described quite satisfactory with both the Friedrich–Braun and the generalized Maxwell models.  相似文献

Surfactant protein A (SP-A), the most abundant protein in the lung alveolar surface, has multiple activities, including surfactant-related functions. SP-A is required for the formation of tubular myelin and the lung surface film. The human SP-A locus consists of two functional SP-A genes, SP-A1 and SP-A2, with a number of alleles characterized for each gene. We have found that the human in vitro expressed variants, SP-A1 (6A2) and SP-A2 (1A0), and the coexpressed SP-A1/SP-A2 (6A2/1A0) protein have a differential influence on the organization of phospholipid monolayers containing surfactant protein B (SP-B). Lipid films containing SP-B and SP-A2 (1A0) showed surface features similar to those observed in lipid films with SP-B and native human SP-A. Fluorescence images revealed the presence of characteristic fluorescent probe-excluding clusters coexisting with the traditional lipid liquid-expanded and liquid-condensed phase. Images of the films containing SP-B and SP-A1 (6A2) showed different distribution of the proteins. The morphology of lipid films containing SP-B and the coexpressed SP-A1/SP-A2 (6A2/1A0) combined features of the individual films containing the SP-A1 or SP-A2 variant. The results indicate that human SP-A1 and SP-A2 variants exhibit differential effects on characteristics of phospholipid monolayers containing SP-B. This may differentially impact surface film activity.  相似文献

The subject of this report was to investigate headgroup hydration and mobility of two types of mixed lipid vesicles, containing nonionic surfactants; straight chain Brij 98, and polysorbat Tween 80, with the same number of oxyethylene units as Brij, but attached via a sorbitan ring to oleic acid. We used the fluorescence solvent relaxation (SR) approach for the purpose and revealed differences between the two systems. Fluorescent solvent relaxation probes (Prodan, Laurdan, Patman) were found to be localized in mixed lipid vesicles similarly as in pure phospholipid bilayers. The SR parameters (i.e. dynamic Stokes shift, Δν, and the time course of the correlation function, C(t)) of such labels are in the same range in both kinds of systems. Each type of the tested surfactants has its own impact on water organization in the bilayer headgroup region probed by Patman. Brij 98 does not modify the solvation characteristics of the dye. In contrast, Tween 80 apparently dehydrates the headgroup and decreases its mobility. The SR data measured in lipid bilayers in presence of Interferon alfa-2b reveal that this protein, a candidate for non-invasive delivery, affects the bilayer in a different way than the peptide melittin. Interferon alfa-2b binds to mixed lipid bilayers peripherally, whereas melittin is deeply inserted into lipid membranes and affects their headgroup hydration and mobility measurably.  相似文献

An isolate of the fungus Myrothecium verrucaria was evaluated for its biocontrol potential against common purslane, horse purslane, spotted spurge, and prostrate spurge, all serious weed pests in commercial tomato fields in the southeastern US. In greenhouse and field tests, M. verrucaria was highly virulent against these weeds when applied as conidial sprays formulated in 0.2% Silwet L-77 surfactant, even in the absence of dew. In field test plots naturally infested with these weeds, seedlings in the two-to-three leaf growth stage treated with M. verrucaria at 2×107 conidia mL-1 in 0.2% Silwet, exhibited leaf and stem necrosis within 24 h following inoculation, with mortality occurring within 96 h. After 7 days, M. verrucaria had killed 90-95% of both purslane species and 85-95% of both spurge species. Tomatoes that were transplanted into plots treated with M. verrucaria remained healthy and vigorous throughout the growing season. Since M. verrucaria effectively controlled several common weeds under field conditions, this fungus appears to have potential as an effective bioherbicide for pre-plant weed control in production systems with transplanted tomato.  相似文献

A much studied oil-soluble surfactant, bis[2-ethylhexyl]sulfosuccinate, sodium salt, was ion exchanged into the silver ion form and dissolved into microemulsions of immiscible polyurethane step monomers. Coating and curing of these microemulsions produced polyurethane coatings that exhibit bactericidal activity against representative Gram negative bacteria. After 24 h exposure, 0.006–0.012% weight Ag relative to coating weight (0.0013–0.0025 μmol Ag/cm2) results in the three-log reduction in Escherichia coli. A slightly higher level of 0.031% weight Ag relative to coating weight (0.006 μmol Ag/cm2) killed all of the E. coli after 12 h exposure. Similar results were obtained for Pseudomonas aeruginosa. Since the double-tail surfactant anion promotes reverse micelle formation in many different kinds of oils and solvents, it appears an excellent vector for incorporating low and effective amounts of silver ion into many industrial, hospital, and household coating formulations.  相似文献

Polymerization of N-phenylmaleimide derivatives bearing a chiral oxazoline substituent at ortho-position on the phenyl group (namely, N-[o-(4,5-dihydro-1,3-oxazol-2-yl)phenyl]maleimides, OPMI) was carried out with diethylzinc (Et2Zn) as an initiator. The resulting polymers exhibited a quite high specific rotation and a unique split-type circular dichroism. Furthermore, an induced Cotton effect was observed in the pi-pi* transition region of the chromophores upon complexation of these polymers with copper(II) salt in tetrahydrofuran solution, indicating the formation of chiral supramolecular aggregates. The induced supramolecular chirality was found to be dependent on the absolute configuration of monomer units, that is, the polymers with (R)-configurational oxazolinyl chromophores tended to form chiral aggregates with levorotatory handedness, while polymers obtained from the (S)-monomer offered dextrogyrate one upon the addition of Cu(II) salt.  相似文献
