In the gingival crevicular fluid (GCF) of control and chronic adult periodontitis (CAP) patients there is a spontaneous release of O2- radicals from polymorphonuclear leukocytes (PMN). The addition of the exogenous stimuli phorbol myrystate acetate (PMA) decreased the O2- formation in control GCF, while in CAP patients produced a marked enhancement of O2- generation.

The circulating PMN of control subjects did not show a spontaneous O2- formation, differently from CAP patients. On the contrary, a similar O2- production was measured when the circulating PMN were stimulated with PMA.

Moreover, the antioxidant activity measured in 10μl of cell free gingival supernatant (GS) of control and CAP patients had the same values by inhibiting 12.6% and 18.9% respectively of the O2- formation supported by a xanthine/xanthine oxidase system.

Probably, the protective or destructive effect of PMN in GCF of CAP patients depends on the variations of the rate of O2- formation in respect to the intrinsic antioxidant property of GS.  相似文献

The physico-chemical characteristics and possible formation mechanisms of negative air ions are considered. It was found that the products of oxygen and nitrogen negative ionization reduce ferricytochromec and nitroblue tetrazolium, and that these reactions were inhibited by superoxide dismutase. The interaction of negatively ionized oxygen with water led to hydrogen peroxide accumulation, which was inhibited by tetranitromethane or catalase. Nitrogen ionization under these conditions caused the formation of the hydrated electron e aq and the superoxide anion O 2 . The data obtained indicate that the biological activity of negative air ions may be dependent on superoxide. The generation of reactive oxygen ions in the gas phase and also at a gas/water interface is described. A scheme for superoxide production under oxygen and nitrogen ionization is proposed.  相似文献

Effects of deficient (20mmol m?3) and sufficient (1000 mmol m?3) magnesium (Mg) supply and of varied light intensity (100 μmol m?2 s?1 to 580 μmol m?2 s?1) on paraquat-dependent chlorophyll destruction in bean (Phaseolus vulgaris) plants grown in nutrient solution were studied over a 12-d period using leaf discs or intact primary leaves. Treatment of leaf discs with 10mmol m 3 paraquat for 15h caused severe chlorophyll loss, especially with increasing light intensity. This chlorophyll destruction by paraquat was very much higher in Mg-sufficient than Mg-deficient leaves. The occurrence of paraquat resistance in Mg deficient leaves was already apparent after 6d growth in nutrient solution, i.e. before any decrease in chlorophyll or growth by Mg deficiency was evident. Also, following foliar application of paraquat (10–140 mmol m?3) to intact plants, Mg-deficient plants were much more resistant to paraquat, even following longer exposure duration (72 h) and four to 14 times higher paraquat concentrations than those received by Mg sufficient plants. From experiments where exogenous scavengers of superoxide radical (O2.-), hydroxyl radical (OH·) and singlet oxygen (1O2) were applied to leaf discs, it appears that O2.-, and partly, OH· are the main O2 species which contribute to chlorophyll destruction by paraquat. The results demonstrate that Mg-deficient bean plants become highly resistant to O2.--mediated and light-induced paraquat injury. The mode of this paraquat resistance is attributed to well-known stimulative effects of Mg deficiency on O2.- and H2O2 scavenging enzymes and antioxidants.  相似文献

The effect of high CO2-concentration on photoacoustic signals from tobacco leaves is studied by means of a recently developed pulse modulation method which provides simultaneous information on photothermal and photobaric components in the millisecond time domain. High CO2-concentrations are found to induce large gas-uptake signals. Simultaneous measurements of chlorophyll fluorescence suggest that the uptake signals are correlated with energy-dependent fluorescence quenching. Very similar CO2-concentration dependencies are found in the absence and presence of methylviologen, which is known to catalyze O2-reduction, and in the presence of glyceraldehyde, which blocks Calvin cycle and photorespiration. It is suggested that the CO2-enhanced uptake signal is likely to reflect O2-uptake in the Mehler reaction. However, it is not ruled out that also rapid CO2-solubilisation or CO2-binding caused by light-induced stroma alkalisation are involved. Strong uptake is also induced when the CO2-concentration in the closed photoacoustic chamber increases due to dark-respiration. The consequences of these findings with respect to the interpretation of photoacoustic data (e.g., low-light effect) and to the regulatory role of O2-dependent electron flow are discussed.  相似文献

Large multinucleated osteoclasts are the major cells responsible for bone breakdown and have been reported to produce high levels of superoxides which may contribute to the process of bone resorption (Key et al.: J Bone and Mineral Res 4 [suppl. 1]:S206, 1989). Osteoclasts also possess high levels of superoxide dismutase, a protective enzyme capable of converting toxic superoxides to less dtoxic H2O2 (Fridovich: J Biol Chem 264:7761-7764, 1989). The amino acid sequence of manganese and/or iron superoxide dismutase has a conserved region which exhibits substantial homology with a fragment obtained from a high molecular weight osteoclast surface marker glycoprotein which is reactive with monoclonal antibody 121F. In this report, evidence is presented substantiating immunological, biochemical, and functional similarities between the osteoclast membrane antigen recognized by the 121F monoclonal antibody and superoxide dismutase. Western blot and immunoprecipitation studies show that a monospecific polyclonal antibody generated against immunoaffinity purified antigen is cross-reactive with superoxide dismutase. Both the antigen and a high molecular weight superoxide dismutase activity have been detected in osteoclast plasma membrane preparations. The levels of superoxide dismutase activity and the membrane antigen have been found to correlate in antigen depletion studies and in western blots probing osteoclasts and closely related marrow-derived giant cells. Moreover, regions of osteoclast superoxide dismutase activity identified by electrophoretic zymogram analysis have been shown by gel electrophoresis and western blots to contain the high molecular weight antigen, or complexes of the antigen with the 121F monoclonal antibody when these were premixed prior to nondenaturing electrophoresis. It is proposed that the osteoclast plasma membrane possesses a high molecular weight superoxide dismutase activity. Furthermore, it appears that this activity is associated with the osteoclast antigen recognized by the 121F monoclonal antibody.  相似文献

Six chronic uremic patients on regular hemodialysis treatment were given recombinant human erythropoietin (r-huEPO) in a dose of 50 U/kg of body weight intravenously thrice weekly for 14 weeks. Following r-huEPO therapy, unstimulated whole blood superoxide anion ( 2) generation did not change significantly, while opsonized zymosan-stimulated whole blood 2 generation increased. At the same time, erythrocyte superoxide dismutase and, in particular, glutathione peroxidase activities were found to be reducing with concomitant lowering of erythrocyte malonydialdehyde (MDA) concentrations and increase in plasma MDA concentrations.  相似文献

高浓度Ca~(2 )(0.1 mol/L)使叶绿体产生O_2~ 的能力下降,旋转相关时间(τ_c)增大34.3%,即膜的流动性降低,并抑制ACC形成乙烯;衰老时细胞内的Ca~(2 )作用却与此相反;O_2~ 的生成与乙烯的产量成正相关(r=0.941)。EGTA,吩噻嗪和W_7等加入到叶绿体反应体系中,可使O_2~ 的产量下降,ACC形成乙烯减少;相反,亚油酸作为Ca~(2 )载体,却使之明显升高,但亚油酸本身产生乙烯的量比ACC少得多。因此推测:高浓度Ca~(2 )可能影响叶绿体膜的状态,从而影响EFE的构象或者减少O_2~ 的生成,抑制ACC的转化,衰老时细胞内的Ca~(2 )可启动钙信使系统,使O_2~ 的产量升高,而其中膜脂过氧化是衰老的中心环节,因此O_2~ 的升高可能是诱发衰老启动的重要因素。  相似文献

The granule rich-fraction isolated from human resting polymorphonuclear leukocytes is capable of CN-insensitive NADH oxidation and O2-uptake, accompanied by production of superoxide anion, hydroxyl radicals and H2O2. We showed that H2O2 initiates and maintains NADH oxidation and O2-uptake but is also necessary for the formation of superoxide anion and hydroxyl radicals. It acts as a primary substrate for CN-insensitive protein-mediated formation of hydroxyl radicals, which in turn produce superoxide anions, probably through univalent oxidation of NADH as an intermediary.  相似文献

The results presented in this paper show that lysis of human erythrocytes by linoleic acid is not caused by peroxidation of the fatty acid. Peroxidase, superoxide dismutase and scavengers of O 2 and OH had no effect on the lysis while catalase showed only marginal inhibition suggesting that O 2 , OH, O 2 and H2O2 do not play any direct role in hemolysis by linoleic acid. Generators of H2O2 inhibited the lysis completely and methemoglobin cells were more resistant to hemolysis by linoleic acid. The fatty acid did neither bind to nor fomed complex with red cell ghosts. Membrane oxidation of sulphydryl groups was also not involved in the lysis. Β-Carotene, retinol and bile salts enhanced the lysis, while, cholesterol but not cholesterol acetate, inhibited it. Taurocholate-pretreated cells were more susceptible to linoleic acid lysis. These observations suggested-that lysis by linoleic acid may be due to its detergent property.  相似文献
