Cells subjected to the events occurring before, during, and after freezing and thawing are exposed to major changes in the
osmotic pressure of the surrounding medium; i.e., the osmolalities can exceed 30. An important question in understanding the
mechanisms of injury is whether cells respond as ideal osmometers to these strongly anisotonic solutions. Mouse and bovine
embryos from eight-cell to blastocyst stage were used to investigate the question. They were found to behave as ideal osmometers
at room temperature over a wide range of tonicities; i.e., from four times isotonic to almost 1/3 times isotonic, ideality
being defined by a Boyle-van't Hoff equation. Embryo volumes increased from 40 to 200% of isotonic over this range and survivals
of mouse embryos were unaffected. However, outside this range the membrane apparently becomes leaky and the survival of mouse
embryos drops sharply. Osmolalities rise to high values during freezing and the paper develops the thermodynamic equations
to show how computed cell volumes as a function of subzero temperature can be translated into the Boyle-van't Hoff format
of cell volume as a function of the reciprocal of osmolality. 相似文献

Establishment and maintenance of friable,embryogenic maize callus and the involvement of L-proline 总被引:30,自引:0,他引:30
Friable, embryogenic maize (Zea mays L.), inbred line A188, callus was established and maintained for more than one year without apparent loss of friability or embryogenic potential. Embryoid development was abundant in these cultures and plants were easily regenerated. Frequencies of friable-callus initiation and somatic-embryoid formation increased linearly with addition to N6 medium (C.C. Chu et al. 1975, Sci. Sin. [Peking] 18, 659–668) of up to 25 mM L-proline. Proline additions up to 9 mM to MS medium (inorganic elements of T. Murashige and F. Skoog 1962, Physiol. Plant. 15, 473–497, plus 0.5 mg 1-1 thiamine hydrochloride and 150 mg 1-1
L-asparagine monohydrate) did not stimulate embryoid formation. A major part of the difference between MS and N6 media could be attributed to their respective inorganic nitrogen components. L-Glutamine was not a satisfactory substitute for L-proline. Of 111 regenerated plants grown to maturity from three independent friable, embryogenic cell lines ranging in age from three to seven months, only four plants were abnormal based on morphology and pollen sterility. Summary The early appearance of catecholaminergic neurons, as revealed by fluorescence histochemistry, has been determined in the central nervous system of quail, pheasant, and pigeon embryos. The first neuronal assemblies displaying specific fluorescence are the locus coeruleus and the nucleus subcoeruleus ventralis. Embryos at the morula, blastocyst and hatched blastocyst stage were obtained from superovulated and naturally ovulated Japanese native goats. They were bisected into halves with a glass needle, and transferred immediately or after culture (for morula) to recipients. None of five does which received bisected morula became pregnant. Three of nine goats became pregnant after transfer of bisected hatched blastocysts, six of eleven recipients became pregnant. Four of them produced monozygotic twins and the remaining two produced singles. The present study demonstrated that the hatched blastocyst is suitable for bisection in the goat. 相似文献

Summary Previous studies have shown that extracts from mouse embryos at mid and late stages of development contain factors that exhibit
transforming growth factor activity. The work reported here demonstrates that cultured mouse embryos at significantly earlier
stages of development produce and release factors that exhibit the characteristic property of transforming growth factors.
Specifically, the data demonstrate that embryos cultured from the blastocyst stage in serum-containing medium or in serum-free
medium release factors that promote the anchorage-independent growth of normal rat kidney fibroblasts. It is shown that these
factors are produced and released by cells derived from the inner cell mass and by trophoblasts. The precise developmental
stage when production of these factors first begins has not been determined but our findings suggest that these factors are
produced by cell types associated with early postimplatation embryos.

Summary This paper reports the inhibitory effects of calmidazolium (CDZ), a calmodulin inhibitor, on electrical uncoupling by CO2. Membrane potential and coupling ratio (V
2/V1) are measured in two neighboring cells ofXenopus embryos (16 to 64 cell stage) for periods as long as 5.5 hr. Upon exposure to 100% CO2, control cells consistently uncouple even if the CO2 treatments are repeated every 15 min for 2.5 hr. CDZ (5×10–8–1×10–7
m) strongly inhibits uncoupling. The inhibition starts after 30, 50 and 60 min of treatment with 1×10–7, 7×10–8 and 5×10–8
m CDZ, respectively, is concentration-dependent and partially reversible. In the absence of CO2, CDZ also improves electrical coupling. CDZ has no significant effect on membrane potential and nonjunctional membrane resistance. These data suggest that calmodulin or a calmodulin-like protein participates in the uncoupling mechanism. 相似文献

As germination of axes of Pisum sativum L. seeds progressed, profound quantitative and qualitative changes occurred in the patterns of protein synthesis. This was shown by fluorography of gels following two-dimensional polyacrylamide gel electrophoresis separation of [35S]methioninelabelled proteins. The effects of desiccation during germination on these in-vivo protein-synthesis patterns were followed. Desiccation differentially affected the synthesis of proteins. Usually, however, upon rehydration following desiccation the types of proteins being synthesized were recognizable as those synthesized earlier during imbibition of control, once-imbibed axes: seeds imbibed for 8 h, and then dried, did not recommence synthesis of proteins typical of 8-h-imbibed control seeds, but rather of 4-h-imbibed control seeds. Seeds imbibed for 12 h, and then dried and rehydrated, synthesized proteins typical of 4-h-and 8-h-control seeds. Thus drying of germinating pea axes caused the proteinsynthesizing mechanism to revert to producing proteins typical of earlier stages of imbibition.
