The potential habitat of the introduced estuary crab Rhithropanopeus harrisi (Gould, 1841) in Oregon and central California was surveyed to discover current sites of presence. In order to infer the direction of range expansion and attempt to trace the source population, a 523 bp sequence of the mitochondrial gene Cytochrome c oxidase I was compared between three Pacific populations (28–39 individuals/population), with one mid-Atlantic population included for assessment as a hypothetical source. Notably, this species persists at fairly low densities in the Pacific despite its apparent habitat tolerance, and range expansion and establishment over a broad geographic area. It was possible to deduce that the current Pacific populations descend from a single introduction event. Despite a genetic bottleneck, the historical record of range expansion in a northward direction is reinforced by the molecular data. In order to consider the plausibility of coastal advective transport versus ballast water or aquaculture transport, the new genetic information is evaluated together with the records of northward range expansion for this and other species introduced into San Francisco Bay, including the European green crab, which has had a similar population trajectory. The genetic and temporal patterns that should be expected under these contrasting mechanisms are discussed for typical NE Pacific oceanic conditions, and during years influenced by the El Niño-Southern oscillation.  相似文献

The hydrolysis of proanthocyanidins to anthocyanidins in n-BuOH-HCl (95:5) has been shown to be an autoxidation, the yield of anthocyanidin being critically dependent on trace metal-ion impurities. Reproducible yields of anthocyanidin may be achieved if iron (III) salts are added to the reaction medium, and a standard method of analysis of proanthocyanidins based on use of an n-BuOH-HCl-FeIII mixture is given. The ratio of absorbance maxima of the cyanidin (550 nm) produced to that near 280 nm for the original procyanidin polymer solution was ~ 3.5.  相似文献

Bioremediation of petroleum-contaminated sites is expected to be a cost-effective remediation technology. However, many potential users of the technology expect the reliability of this technology to be similar to other candidate technologies for widespread consideration. In particular, candidate technologies should possess the property of reliable experimental linkage — there should be reasonable confidence that experiments done at one scale can be reliably related to another. An important example is bench-scale treatability studies that should result in linkages with commercial-scale operations. In this respect comparison of bioremediation to other candidate technologies reveals that bioremediation is in an early stage of its evolution. It is being pursued at a variety of sites and scales with practitioners from a variety of disciplines. Integration of activities between disciplines and an ability to quantitatively compare results at different sites and scales is proceeding. This paper addresses a number of physical, chemical, biological, analytical, and statistical issues regarding the successful comparison of results between experiments.  相似文献

Renal cell carcinoma (RCC) is a disease with no specific diagnostic method or treatment. Thus, the evaluation of novel diagnostic tools or treatment possibilities is essential. In this study, a multiplatform untargeted metabolomics analysis of urine was applied to search for a metabolic pattern specific for RCC, which could enable comprehensive assessment of its biochemical background. Thirty patients with diagnosed RCC and 29 healthy volunteers were involved in the first stage of the study. Initially, the utility of the application of the selected approach was checked for RCC with no differentiation for cancer subtypes. In the second stage, this approach was used to study clear cell renal cell carcinoma (ccRCC) in 38 ccRCC patients and 38 healthy volunteers. Three complementary analytical platforms were used: reversed-phase liquid chromatography coupled with time-of-flight mass spectrometry (RP-HPLC-TOF/MS), capillary electrophoresis coupled with time-of-flight mass spectrometry (CE-TOF/MS), and gas chromatography triple quadrupole mass spectrometry (GC-QqQ/MS). As a result of urine sample analyses, two panels of metabolites specific for RCC and ccRCC were selected. Disruptions in amino acid, lipid, purine, and pyrimidine metabolism, the TCA cycle and energetic processes were observed. The most interesting differences were observed for modified nucleosides. This is the first time that the levels of these compounds were found to be changed in RCC and ccRCC patients, providing a framework for further studies. Moreover, the application of the CE-MS technique enabled the determination of statistically significant changes in symmetric dimethylarginine (SDMA) in RCC.  相似文献

Entamoeba histolytica is one of the least understood protists in terms of taxa, clone, and kin discrimination/recognition ability. However, the capacity to tell apart same or self (clone/kin) from different or nonself (nonclone/nonkin) has long been demonstrated in pathogenic eukaryotes like Trypanosoma and Plasmodium, free‐living social amebas (Dictyostelium, Polysphondylium), budding yeast (Saccharomyces), and in numerous bacteria and archaea (prokaryotes). Kin discrimination/recognition is explained under inclusive fitness theory; that is, the reproductive advantage that genetically closely related organisms (kin) can gain by cooperating preferably with one another (rather than with distantly related or unrelated individuals), minimizing antagonism and competition with kin, and excluding genetic strangers (or cheaters = noncooperators that benefit from others’ investments in altruistic cooperation). In this review, we rely on the outcomes of in vitro pairwise discrimination/recognition encounters between seven Entamoeba lineages to discuss the biological significance of taxa, clone, and kin discrimination/recognition in a range of generalist and specialist species (close or distantly related phylogenetically). We then focus our discussion on the importance of these laboratory observations for E. histolytica's life cycle, host infestation, and implications of these features of the amebas’ natural history for human health (including mitigation of amebiasis).  相似文献

The lesser kestrel (Falco naumanni) suffered a sharp population decline over much of its European distribution range in the middle of the twentieth century. Still declining in some areas, the species has recently experienced a notable population recovery in certain regions. We examined the genetic diversity variation in a growing population of lesser kestrels from Central Spain over a 6-year period (2000-2005). The population studied showed a rapid demographic expansion, increasing in the number of both breeding pairs and colonies. Annual average heterozygosity and allelic diversity increased and genetic similarity between potential mates decreased over the study period. Several immigrants regularly arrived in the study area and introduced new alleles into the local population, pointing to immigration as the main cause contributing to the observed genetic recovery.  相似文献

Domesticated rice (Oryza sativa) is one of the world’s most important food crops, culturally, nutritionally and economically ( Khush 1997 ). Thus, it is no surprise that there is intense curiosity about its genetic and geographical origins, its response to selection under domestication, and the genetic structure of its wild relative, Oryza rufipogon. Studies of Oryza attempting to answer these questions have accompanied each stage of the development of molecular markers, starting with allozymes and continuing to genome sequencing. While many of these studies have been restricted to small sample sizes, in terms of either the number of markers used or the number and distribution of the accessions, costs are now low enough that researchers are including large numbers of molecular markers and accessions. How will these studies relate to previous findings and long‐held assumptions about rice domestication and evolution? If the paper in this issue of Molecular Ecology ( Huang et al. 2012 ) is any indication, there will be some considerable surprises in store. In this study, a geographically and genomically thorough sampling of O. rufipogon and O. sativa revealed two genetically distinct groups of wild rice and also indicated that only one of these groups appears to be related to domesticated rice. While this fits well with previous studies indicating that there are genetic subdivisions within O. rufipogon, it stands in contrast to previous findings that the two major varieties of O. sativa (indica and japonica) were domesticated from two (or more) subpopulations of wild rice.  相似文献

The identification of population bottlenecks is critical in conservation because populations that have experienced significant reductions in abundance are subject to a variety of genetic and demographic processes that can hasten extinction. Genetic bottleneck tests constitute an appealing and popular approach for determining if a population decline has occurred because they only require sampling at a single point in time, yet reflect demographic history over multiple generations. However, a review of the published literature indicates that, as typically applied, microsatellite-based bottleneck tests often do not detect bottlenecks in vertebrate populations known to have experienced declines. This observation was supported by simulations that revealed that bottleneck tests can have limited statistical power to detect bottlenecks largely as a result of limited sample sizes typically used in published studies. Moreover, commonly assumed values for mutation model parameters do not appear to encompass variation in microsatellite evolution observed in vertebrates and, on average, the proportion of multi-step mutations is underestimated by a factor of approximately two. As a result, bottleneck tests can have a higher probability of 'detecting' bottlenecks in stable populations than expected based on the nominal significance level. We provide recommendations that could add rigor to inferences drawn from future bottleneck tests and highlight new directions for the characterization of demographic history.  相似文献

Mitochondria are descended from free-living bacteria that were engulfed by another cell between one and a half to two billion years ago. A redistribution of DNA led to most genetic information being lost or transferred to a large central genome in the nucleus, leaving a residual genome in each mitochondrion. Oxidative phosphorylation, the most critical function of mitochondria, depends on the functional compatibility of proteins encoded by both the nucleus and mitochondria. We investigate whether selection for adaptation between the nuclear and mitochondrial genomes (mitonuclear co-adaptation) could, in principle, have promoted uniparental inheritance of mitochondria and thereby the evolution of two mating types or sexes. Using a mathematical model, we explore the importance of the radical differences in ploidy levels, sexual and asexual modes of inheritance, and mutation rates of the nucleus and mitochondria. We show that the major features of mitochondrial inheritance, notably uniparental inheritance and bottlenecking, enhance the co-adaptation of mitochondrial and nuclear genes and therefore improve fitness. We conclude that, under a wide range of conditions, selection for mitonuclear co-adaptation favours the evolution of two distinct mating types or sexes in sexual species.  相似文献
