Lack of synchronization between N released from prunings applied to the soil as green manures and crop uptake as well as optimization of protein digestibility for ruminants, remain major research objectives for the selection of multipurpose tree and shrub legumes (MPT) for mixed smallholder systems in the tropics. Prunings of the high tannin, low quality MPT Calliandra houstoniana CIAT 20400 (Calliandra) and the tannin free, high quality MPT Indigofera zollingeriana (Indigofera) were mixed in the proportions 100:0, 75:25, 50:50, 25:75, and 0:100 (w/w) in order to measure the aerobic rate and extent of N release in a leaching tube experiment, and the anaerobic extent of N degradation in an in vitro gas production experiment. Parameters measured in Calliandra:Indigofera mixtures were compared to theoretical values derived from single species plant material (i.e. 100:0 and 0:100). Aerobic N release and apparent anaerobic N degradation increased with increasing proportion of the high quality legume (Indigofera) in the mixture. While N release in the soil was lower than theoretical values in the mixture 50% Calliandra/50% Indigofera, this was not the case with apparent anaerobic N degradation with the same mixture. Aerobic N immobilization was more pronounced for the mixture 75% Calliandra/25% Indigofera than for 100% Calliandra and negative interaction was observed with apparent anaerobic N degradation in the mixture 75% Calliandra/25% Indigofera. Plant quality parameters that best correlated with aerobic N release and apparent anaerobic N degradation in the rumen were lignin + bound condensed tannins (r=−0.95 and −0.95 respectively, P<0.001). In addition, a positive correlation (r=0.89, P<0.001) was found between aerobic N release in the leaching tube experiment and apparent N degradation in the in vitro anaerobic gas production experiment. Results show that mixing prunings of MPT materials with contrasting quality is an effective way to modify aerobic N release pattern as well as apparent anaerobic N degradation and could possibly be applied to minimize N losses in the rumen and in the soil. In addition, apparent anaerobic N degradation was identified as good predictor of aerobic N release in the soil, which has resource saving implications when screening MTP to be used as green manures.  相似文献

The aim of this work was to demonstrate at pilot scale a high level of energy recovery from sewage utilising a primary Anaerobic Migrating Bed Reactor (AMBR) operating at ambient temperature to convert COD to methane. The focus is the reduction in non-renewable CO(2) emissions resulting from reduced energy requirements for sewage treatment. A pilot AMBR was operated on screened sewage over the period June 2003 to September 2004. The study was divided into two experimental phases. In Phase 1 the process operated at a feed rate of 10 L/h (HRT 50 h), SRT 63 days, average temperature 28 degrees C and mixing time fraction 0.05. In Phase 2 the operating parameters were 20 L/h, 26 days, 16 degrees C and 0.025. Methane production was 66% of total sewage COD in Phase 1 and 23% in Phase 2. Gas mixing of the reactor provided micro-aeration which suppressed sulphide production. Intermittent gas mixing at a useful power input of 6 W/m(3) provided satisfactory process performance in both phases. Energy consumption for mixing was about 1.5% of the energy conversion to methane in both operating phases. Comparative analysis with previously published data confirmed that methane supersaturation resulted in significant losses of methane in the effluent of anaerobic treatment systems. No cases have been reported where methane was considered to be supersaturated in the effluent. We have shown that methane supersaturation is likely to be significant and that methane losses in the effluent are likely to have been greater than previously predicted. Dissolved methane concentrations were measured at up to 2.2 times the saturation concentration relative to the mixing gas composition. However, this study has also demonstrated that despite methane supersaturation occurring, micro-aeration can result in significantly lower losses of methane in the effluent (<11% in this study), and has demonstrated that anaerobic sewage treatment can genuinely provide energy recovery. The goal of demonstrating a high level of energy recovery in an ambient anaerobic bioreactor was achieved. An AMBR operating at ambient temperature can achieve up to 70% conversion of sewage COD to methane, depending on SRT and temperature.  相似文献

Modeling product formation in anaerobic mixed culture fermentations   总被引:1,自引:0,他引:1  
The anaerobic conversion of organic matter to fermentation products is an important biotechnological process. The prediction of the fermentation products is until now a complicated issue for mixed cultures. A modeling approach is presented here as an effort to develop a methodology for modeling fermentative mixed culture systems. To illustrate this methodology, a steady-state metabolic model was developed for prediction of product formation in mixed culture fermentations as a function of the environmental conditions. The model predicts product formation from glucose as a function of the hydrogen partial pressure (P(H2)), reactor pH, and substrate concentration. The model treats the mixed culture as a single virtual microorganism catalyzing the most common fermentative pathways, producing ethanol, acetate, propionate, butyrate, lactate, hydrogen, carbon dioxide, and biomass. The product spectrum is obtained by maximizing the biomass growth yield which is limited by catabolic energy production. The optimization is constrained by mass balances and thermodynamics of the bioreactions involved. Energetic implications of concentration gradients across the cytoplasmic membrane are considered and transport processes are associated with metabolic energy exchange to model the pH effect. Preliminary results confirmed qualitatively the anticipated behavior of the system at variable pH and P(H2) values. A shift from acetate to butyrate as main product when either P(H2) increases and/or pH decreases is predicted as well as ethanol formation at lower pH values. Future work aims at extension of the model and structural validation with experimental data.  相似文献

NirD is part of the nitrite reductase complex NirBD that catalyses the reduction of nitrite to NH3 in nitrate assimilation and anaerobic respiration. The crystal structure analysis of NirD from Mycobacterium tuberculosis shows a double β‐sandwich fold. NirD is related in three‐dimensional structure and sequence to the Rieske proteins; however, it does not contain any Fe–S cluster or other cofactors that might be involved in electron transfer. A cysteine residue at the protein surface, conserved in NirD homologues lacking the iron–sulfur cluster might be important for the interaction with NirB and possibly stabilize one of the Fe–S centers in this subunit. A computational fluid dynamics (CFD) model that simulates mechanical mixing for high-solids anaerobic digestion was developed. Numerical simulations of mixing manure slurry which exhibits non-Newtonian pseudo-plastic fluid behavior were performed for six designs: (i) one helical ribbon impeller; (ii) one anchor impeller; (iii) one curtain-type impeller; (iv) three counterflow (CF-2) impellers; (v) two modified high solidity (MHS 3/39°) impellers; and (vi) two pitched blade turbine impellers. The CFD model was validated against measurements for mixing a Herschel-Bulkley fluid by ribbon and anchor impellers. Based on mixing time with respect to mixing energy level, three impeller types (ribbon, CF-2, and MHS 3/39°) stand out when agitating highly viscous fluids, of these mixing with two MHS 3/39° impellers requires the lowest power input to homogenize the manure slurry. A comparison of digestion material demonstrates that the mixing energy varies with manure type and total solids concentration to obtain a given mixing time. Moreover, an in-depth discussion about the CFD strategy, the influences of flow regime and impeller type on mixing characteristics, and the intrinsic relation between mixing and flow field is included.  相似文献

Biogas produced by anaerobic digestion, is mainly used in a gas motor for heat and electricity production. However, after removal of CO2, biogas can be upgraded to natural gas quality, giving more utilization possibilities, such as utilization as autogas, or distant utilization by using the existing natural gas grid. The current study presents a new biological method for biogas upgrading in a separate biogas reactor, containing enriched hydrogenotrophic methanogens and fed with biogas and hydrogen. Both mesophilic‐ and thermophilic anaerobic cultures were enriched to convert CO2 to CH4 by addition of H2. Enrichment at thermophilic temperature (55°C) resulted in CO2 and H2 bioconversion rate of 320 mL CH4/(gVSS h), which was more than 60% higher than that under mesophilic temperature (37°C). Different dominant species were found at mesophilic‐ and thermophilic‐enriched cultures, as revealed by PCR–DGGE. Nonetheless, they all belonged to the order Methanobacteriales, which can mediate hydrogenotrophic methanogenesis. Biogas upgrading was then tested in a thermophilic anaerobic reactor under various operation conditions. By continuous addition of hydrogen in the biogas reactor, high degree of biogas upgrading was achieved. The produced biogas had a CH4 content, around 95% at steady‐state, at gas (mixture of biogas and hydrogen) injection rate of 6 L/(L day). The increase of gas injection rate to 12 L/(L day) resulted in the decrease of CH4 content to around 90%. Further study showed that by decreasing the gas–liquid mass transfer by increasing the stirring speed of the mixture the CH4 content was increased to around 95%. A comprehensive study of anaerobic digestion requires an advanced turbulence model technique to accurately predict mixing flow patterns because the digestion process that involves mass transfer between anaerobes and their substrates is primarily dependent on detailed information about the fine structure of turbulence in the digesters. This study presents a large eddy simulation (LES) of mechanical agitation of non-Newtonian fluids in anaerobic digesters, in which the sliding mesh method is used to characterize the impeller rotation. The three subgrid scale (SGS) models investigated are: (i) Smagorinsky-Lilly model, (ii) wall-adapting local eddy-viscosity model, and (iii) kinetic energy transport (KET) model. The simulation results show that the three SGS models produce very similar flow fields. A comparison of the simulated and measured axial velocities indicates that the LES profile shapes are in general agreement with the experimental data but they differ markedly in velocity magnitudes. A check of impeller power and flow numbers demonstrates that all the SGS models give excellent predictions, with the KET model performing the best. Moreover, the performance of six Reynolds-averaged Navier-Stokes turbulence models are assessed and compared with the LES results.  相似文献

The aim of this study was to evaluate anthracene removal using activated soil reactors, previously inoculated, under both aerobic and anaerobic conditions. In the reactors, the soil was maintained at 60% moisture (weight basis), room temperature, in the dark, and under constant agitation at 100 rpm. Two experiments were run during and after acclimatization to evaluate anthracene removal under both aerobic and anaerobic conditions. The first one took place during inoculum acclimatization using three different concentrations of anthracene (50, 100, and 500 mg anthracene/L per day) during 90 days. The second experiment took place after acclimatization (during 132 days). The results of anthracene removal were compared with controls in which no additional inoculum was added. During the two experiments, the behavior of pH, chemical oxygen demand (COD), and biogas production was evaluated. Results indicate that the bacterial community adapted for removal of anthracene became enriched through the acclimatization process. Anthracene biodegradation occurred in the soil model with both types of reactors (aerobic and anaerobic), but the rates and extent of biodegradation in the aerobic reactor were higher (95%) than those in anaerobic conditions (74%). Microbial activity also contributed to enhancing bioremediation in the soil by reducing anthracene sorption.  相似文献

Thermodesulfatator indicus Moussard et al. 2004 is a member of the Thermodesulfobacteriaceae, a family in the phylum Thermodesulfobacteria that is currently poorly characterized at the genome level. Members of this phylum are of interest because they represent a distinct, deep-branching, Gram-negative lineage. T. indicus is an anaerobic, thermophilic, chemolithoautotrophic sulfate reducer isolated from a deep-sea hydrothermal vent. Here we describe the features of this organism, together with the complete genome sequence, and annotation. The 2,322,224 bp long chromosome with its 2,233 protein-coding and 58 RNA genes is a part of the Genomic Encyclopedia of Bacteria and Archaea project.  相似文献

Spirochaeta coccoides Dröge et al. 2006 is a member of the genus Spirochaeta Ehrenberg 1835, one of the oldest named genera within the Bacteria. S. coccoides is an obligately anaerobic, Gram-negative, non-motile, spherical bacterium that was isolated from the hindgut contents of the termite Neotermes castaneus. The species is of interest because it may play an important role in the digestion of breakdown products from cellulose and hemicellulose in the termite gut. Here we provide a taxonomic re-evaluation for strain SPN1T, and based on physiological and genomic characteristics, we propose its reclassification as a novel species in the genus Sphaerochaeta, a recently published sister group of the Spirochaeta. The 2,227,296 bp long genome of strain SPN1T with its 1,866 protein-coding and 58 RNA genes is a part of the Genomic Encyclopedia of Bacteria and Archaea project.  相似文献
