Evolutionary implications of hierarchical impacts of nonlethal injury on reproduction, including maternal effects 总被引:2,自引:0,他引:2

Nonlethal injury is pervasive in metazoans, but surprisingly little is known about its impact upon reproductive allocation. The impact of injury on reproduction has been explored in some detail in lizards and salamanders, which have tails that are adapted for fat storage but are also injured or lost during predatory and social encounters. We synthesize diverse insights from these studies and propose new hypotheses using graphical models which highlight three distinct, hierarchical effects of injury on reproduction: reproductive inhibition, reduction in propagule number and diminished per‐propagule investment (PPI), a maternal effect. Previous studies, which involved experimentally amputating the whole tail, have provided evidence of the first two effects, although there is little evidence of reduced PPI. We assayed these effects in free‐ranging Desmognathus salamanders exhibiting naturally occurring injury. Whereas earlier studies found that tail injury prohibits reproduction (precipitating functional conflict), we found that females missing 80% of their tails, including the smallest mature individuals, still reproduce. We also detected a negative correlation between magnitude of injury and PPI, a continuous maternal effect. Continuous (graded) effects of injury on PPI have not been reported previously; neither discrete nor continuous maternal effects due to injury have been previously demonstrated in free‐ranging vertebrates. The dearth of evidence for such effects may be due to the design of experimental manipulations that use all‐or‐nothing treatments. Future studies employing quantitative field data, or more realistic experimental treatments that mimic the continuous distribution of injury are likely to detect maternal effects arising from nonlethal injury. Although our comparative and empirical findings derive from studies of lizards and salamanders, we discuss how they apply in principle to all metazoans. BACKGROUND: The developmental toxicity, toxicokinetics, and hematological effects of the antimalarial drug, artesunate, were previously studied in rats and rabbits and have now been studied in cynomolgus monkeys. METHODS: Groups of up to 15 pregnant females were dosed on Gestation Days (GD) 20–50 or for 3–7‐day intervals. RESULTS: At 30 mg/kg/day, 6 embryos died between GD30 and GD40. Histologic examination of 3 live embryos (GD26–GD36) revealed a marked reduction in embryonic erythroblasts and cardiomyopathy. At 12 mg/kg/day, 6 embryos died between GD30 and GD45. Four surviving fetuses examined on GD100 had no malformations, but long bone lengths were slightly decreased. At the developmental no‐adverse‐effect‐level (4 mg/kg/day), maternal plasma AUC was 3.68 ng.h/mL for artesunate and 6.93 ng.h/ml for its active metabolite, dihydroartemisinin (DHA). No developmental toxicity occurred with administration of 12 mg/kg/day for 3 or 7 days, GD29–31 or GD27–33 (maternal plasma AUC of 9.84 ng.h/mL artesunate and 16.4 ng.h/mL DHA). Exposures at embryotoxic doses were substantially lower than human therapeutic exposures. However, differences in monkey and human Vss for artesunate (0.5 L/kg vs. 0.18 L/kg) confound relying solely on AUC for assessing human risk. Decreases in reticulocyte count occur at therapeutic doses in humans. Changes to reticulocyte counts at embryotoxic doses in monkeys (≥12 mg/kg/day) were variable and generally minor. CONCLUSIONS: Artesunate was embryolethal at ≥12 mg/kg/day when dosed for at least 12 days at the beginning of organogenesis, but not when dosed for 3 or 7 days, indicating that developmental toxicity of artesunate is dependent upon duration of dosing in cynomologus monkeys. Objective: This study determined whether obese women have an increased risk of cardiovascular defects in their offspring compared with average weight women. Research Methods and Procedures: In a case‐control study, prospectively collected information was obtained from Swedish medical health registers. The study included 6801 women who had infants with a cardiovascular defect and, as controls, all delivered women (N = 812, 457) during the study period (1992 to 2001). Infants with chromosomal anomalies or whose mothers had pre‐existing diabetes were excluded. Obesity was defined as BMI >29 kg/m2, and morbid obesity was defined as BMI >35 kg/m2. Comparisons were made with average weight women (BMI = 19.8 to 26 kg/m2). Results: In the group of obese mothers, there was an increased risk for cardiovascular defects compared with the average weight mothers [adjusted odds ratio (OR) = 1.18; 95% CI, 1.09 to 1.27], which was slightly more pronounced for the severe types of cardiovascular defects (adjusted OR = 1.23; 95% CI, 1.05 to 1.44). With morbid obesity, the OR for cardiovascular defects was 1.40 (95% CI, 1.22 to 1.64), and for severe cardiovascular defects, the OR was 1.69 (95% CI, 1.27 to 2.26). There was an increased risk for all specific defects studied among the obese women, but only ventricular septal defects and atrial septal defects reached statistical significance. Discussion: In this sample, a positive association was found between maternal obesity in early pregnancy and congenital heart defects in the offspring. Population genetics of the amphibian pathogen Batrachochytrium dendrobatidis ( Bd ) show that isolates are highly related and globally homogenous, data that are consistent with the recent epidemic spread of a previously endemic organism. Highly related isolates are predicted to be functionally similar due to low levels of heritable genetic diversity. To test this hypothesis, we took a global panel of Bd isolates and measured (i) the genetic relatedness among isolates, (ii) proteomic profiles of isolates, (iii) the susceptibility of isolates to the antifungal drug caspofungin, (iv) the variation among isolates in growth and phenotypic characteristics, and (v) the virulence of isolates against the European common toad Bufo bufo . Our results show (i) genotypic differentiation among isolates, (ii) proteomic differentiation among isolates, (iii) no significant differences in susceptibility to caspofungin, (iv) differentiation in growth and phenotypic/morphological characters, and (v) differential virulence in B. bufo . Specifically, our data show that Bd isolates can be profiled by their genotypic and proteomic characteristics, as well as by the size of their sporangia. Bd genotypic and phenotypic distance matrices are significantly correlated, showing that less-related isolates are more biologically unique. Mass spectrometry has identified a set of candidate genes associated with inter-isolate variation. Our data show that, despite its rapid global emergence, Bd isolates are not identical and differ in several important characters that are linked to virulence. We argue that future studies need to clarify the mechanism(s) and rate at which Bd is evolving, and the impact that such variation has on the host–pathogen dynamic. 相似文献

The external morphology of frog larvae is predicted to vary among habitats, for a variety of functional reasons. I performed a phylogenetic comparative study to test whether correlations between habitat and the shape of the tadpole and its oral disc are adaptive in 82 species from south‐eastern Australia in the families Hylidae and Myobatrachidae. Habitat distributions and phylogenetic relationships were compiled from the literature and shape was quantified using geometric morphometric analysis of published drawings. Results indicate that shape evolved towards different optima in different habitats while also showing appreciable levels of phylogenetic inertia. Within myobatrachids, evolution of terrestrial tadpoles was associated with a short and shallow head/body and a shallow tail. In aquatic species, the use of benthic microhabitats was correlated with a long shallow tail, dorsal eye position, shallow head/body and ventral mouth with robust jaw sheaths. Even traits with evidence for adaptation evolved slowly in response to habitat shifts, usually requiring ≥10 million years to evolve half‐way to a new optimum. Although these findings support adaptive evolution of tadpole body form, they also highlight a strong influence of ancestral character states and indicate that phenotypes in extant species are partly maladaptive. 相似文献
