The activity of spinach chloroplast phenolase which had been repressed by ammonium oxalate was restored by adding copper. Oxalate appears to bind to the enzyme at a single site, the binding paralleling the inhibition produced at neutral pH. The inhibition of oxalate is due to its binding with copper at the active centre to form an inactive complex, the oxalate moiety of which is releasable when more copper is added. Similar reactivation by copper was obtained with pure mushroom phenolase.  相似文献

Frog retinal rod outer segments appear to contain uncharacterized chemical components whose mass is roughly equivalent to 12--51% of the rhodopsin mass. Available data suggest that such components include soluble proteins and complex polysaccharides, and that hyaluronic acid accounts for a substantial fraction of this mass. Electron microscopic histochemical staining studies suggest that these polysaccharide components are located within the ROS disks. The oligosaccharide moieties of rhodopsin also appear localized within the disks. The interdisk cytoplasm may contain carbohydrates, but their quantity and identity are uncertain. Rhodopsin oligosaccharides as well as some fraction of the intradisk polysaccharide appear to have extended saccharide chains preferentially oriented perpendicular to the surface of the disk membrane. Possible roles for these polysaccharides in disk development and photoexcitation are discussed. The immediate need for complete rod outer segment chemical composition data is emphasized.  相似文献

Exogenously supplied ammonium ion is critical to alfalfa morphogenesis in vitro. In alfalfa, the ability to induce the formation of either roots or somatic embryos provided an opportunity to examine the effects of ammonium ion on each pattern of morphogenesis. Somatic embryo formation required a minimum of 12.5 mM NH 4 + in regeneration medium for optimal expression. Root formation was inhibited by NH 4 + levels of 50 mM and above, and occurred in the absence of exogenous NH4 +. At high levels of NH 4 + somatic embryos were formed from cells exposed to the root-inducing combination of hormones. This observation suggests that the growth regulators and exogenously supplied ammonium ion comprise an interactive system controlling the in vitro pattern of alfalfa morphogenesis.  相似文献

SUMMARY 1. The effects of increasing CO2 and nitrogen loading and of a change in water table and temperature on littoral CH4, N2O and CO2 fluxes were studied in a glasshouse experiment with intact sediment cores including vegetation (mainly sedges), taken from a boreal eutrophic lake in Finland. Sediments with the water table held at a level of 0 or at ?15 cm were incubated in an atmosphere of 360 or 720 p.p.m. CO2 for 18 weeks. The experiment included fertilisation with NO3 and NH4+ (to a total 3 g N m?2). 2. Changes in the water table and temperature strongly regulated sediment CH4 and cCO2 fluxes (community CO2 release), but did not affect N2O emissions. Increase in the water table increased CH4 emissions but reduced cCO2 release, while increase in temperature increased emissions of both CO2 and CH4. 3. The raised CO2 increased carbon turnover in the sediments, such that cCO2 release was increased by 16–26%. However, CH4 fluxes were not significantly affected by raised CO2, although CH4 production potential (at 22 °C) of the sediments incubated at high CO2 was increased. In the boreal region, littoral CH4 production is more likely to be limited by temperature than by the availability of carbon. Raised CO2 did not affect N2O production by denitrification, indicating that this process was not carbon limited. 4. A low availability of NO3 did severely limit N2O production. The NO3 addition caused up to a 100‐fold increase in the fluxes of N2O. The NH4+ addition did not increase N2O fluxes, indicating low nitrification capacity in the sediments.  相似文献

The sex pheromone blend of the butterbur borer, Ostrinia zaguliaevi (Lepidoptera: Pyralidae) was analyzed by means of gas chromatography-electroantennographic detection (GC-EAD), GC-mass spectrometry and a series of wind-tunnel bioassays. Four EAD-active compounds were detected in the female sex pheromone gland extract, and these were identified as tetradecyl acetate (14:OAc), (Z)-9-tetradecenyl acetate (Z9-14:OAc), (E)-11-tetradecenyl acetate (E11-14:OAc) and (Z)-11-tetradecenyl acetate (Z11-14:OAc). The average amounts ± s.d. of the four compounds in a single sex pheromone gland were 7.9±3.7 ng, 10.1±3.2 ng, 1.1±0.5 ng and 11.6±5.1 ng, respectively. In a wind-tunnel bioassay, the ternary blend of Z9-, E11- and Z11-14:OAc at a ratio found in the sex pheromone gland (45:5:50) elicited the same behavioral responses from the males as did virgin females and pheromone gland extract. Removal of any single compound from the ternary blend significantly diminished the pheromonal activity, whereas addition of 14:OAc to the ternary blend had no effect on the males' behavioral responses. Therefore, it was concluded that the sex pheromone blend of O. zaguliaevi is composed of Z9-14:OAc, E11-14:OAc and Z11-14:OAc at a ratio of 45:5:50.  相似文献

Nitrogen stable isotope (15N, 14N) natural abundance has been much less used than carbon isotopes (13C, 12C) in plant physiology and ecology. Analytical problems, the lower fractional abundance of 15N than of 13C in the biosphere, the greater complexity of the N cycle relative to the C cycle, and smaller expressed discriminations in nature, are contributing factors. The major N pools, globally, have different isotope signatures: atmospheric N2 is 15N-depleted relative to organic N (including sedimentary N), a situation resulting from a greater expressed discrimination in the organic N to N2 (via denitrification) reaction than of diazotrophy during accumulation of the reduced N. Essentially all of the enzymes except nitrogenase which transform N compounds show discrimination against 15N, although for glutamine synthetase, and the amination of 2-oxoglutarate and pyruvate, this is only seen in terms of NH4+ rather than the true substrate, NH3. Discrimination is expressed in various N interconversions within plants, leading to substantial differences in δ15N (up to 12‰) among N compounds and macroscopic plant parts. N isotope fractionation during assimilation of exogenous combined N is often much lower than that expected from studies of isolated enzymes due to processes which show very little discrimination, such as limitation by transport through aqueous solution and membranes. Application of 15N/14N discrimination studies to plant ecology have concentrated largely on distinguishing diazotrophy from N supplied from combined N, based on the lower 15N/14N in diazotrophs due to the higher 15N/14N of combined N sources not being offset by fractionation during uptake. While potentially very useful, a number of pitfalls are discussed in its ecological use in both terrestrial and aquatic systems. N isotope discrimination is also useful in tracking N through food webs, and hence, back to combined N sources for plants.  相似文献

Growth, chemical composition, and nitrate reductase activity (NRA) of hydroponically cultured Rumex crispus, R. palustris, R. acetosa, and R. maritimus were studied in relation to form (NH4 +, NO3 -, or both) and level of N supply (4 mM N, and zero-N following a period of 4mM N). A distinct preference for either NH4 + or NO3 - could not be established. All species were characterized by a very efficient uptake and utilization of N, irrespective of N source, as evident from high concentrations of organic N in the tissues and concurrent excessive accumulations of free NO3 - and free NH4 +. Especially the accumulation of free NH4 + was unusually large. Generally, relative growth rate (RGR) was highest with a combination of NH4 + and NO3 -. Compared to mixed N supply, RGR of NO3 -- and NH4 +-grown plants declined on average 3% and 9%, respectively. Lowest RGR with NH4 + supply probably resulted from direct or indirect toxicity effects associated with high NH4 + and/or low Ca2+ contents of tissues. NRA in NO3 - and NH4NO3 plants was very similar with maxima in the leaves of ca 40 μmol NO2 - g-1 DW h-1. ‘Basal’ NRA levels in shoot tissues of NH4 + plants appeared relatively high with maxima in the leaves of ca 20 μmol NO2 - g-1 DW h-1. Carboxylate to organic N ratios, (C-A)/Norg, on a whole plant basis varied from 0.2 in NH4 + plants to 0.9 in NO3 - plants. After withdrawal of N, all accumulated NO3 - and NH4 + was assimilated into organic N and the organic N redistributed on a large scale. NRA rapidly declined to similar low levels, irrespective of previous N source. Shoot/root ratios of -N plants were 50–80% lower than those from +N plants. In comparison with +N, RGR of -N plants did not decline to a large extent, decreasing by only 15% in -NH4 + plants due to very high initial organic-N contents. N-deprived plants all exhibited an excess cation over anion uptake (net proton efflux), and whole-plant (C-A)/Norg ratios increased to values around unity. Possible difficulties in interpreting the (C-A)/Norg ratio and NRA of plants in their natural habitats are briefly discussed.  相似文献

The higher levels of nitrogen in ammonium form stimulated the growth of mycelia and increased the accessible as well as the total acid phosphatase activity of Paxillus involutus (Batsch) Fr. isolates grown in pure culture. Rates of mycelia growth and acid phosphatase activities varied widely from one isolate to another. Scots pine (Pinus sylvestris L.) seedlings were inoculated with different P. involutus isolates in axenic conditions. Shoots of pine seedlings with mycorrhizae contained more phosphorus than shoots of non-mycorrhizal seedlings. The relations between growth and phosphatase activity of P. involutus isolates and their efficiency in supplying the host plant with phosphate are discussed.  相似文献

Growth of maize seedlings can be improved by enhanced ammonium nutrition, but placing fertilizer anhydrous ammonia close to seedlings introduces the risk of ammonia toxicity. In this study, growth and root elongation response to rates of closely placed NH4OH bands were investigated in two contrasting maize hybrids. Seven rates of NH4OH, ranging from 0 to 200 mg N kg-1 soil were injected into the center of each pot. A single rate of Ca(NO3)2-N was included to compare hybrids for N form preference at a moderate N rate. Three seedlings per pot were planted 5.7 cm from the injection point.Hybrid B73×LH51 produced a quadratic response in shoot growth to NH4OH rates, whereas LH74×LH123 exhibited a significant linear decline in response to NH4OH rate. Root length density sampled from the fertlized zone declined linearly in response to NH4OH rate while a slight increase in root length density in unfertilized zones was observed at intermediate NH4OH rates. Hybrids did not differ in root length density in either zone.The hybrid with greater tolerance of NH4OH rates (B73×LH51) also showed a preference in shoot growth for NH4-over NO3-N at 66.7 mg N kg-1 compared to LH74×LH123. On average across hybrids, nitrate concentrations of xylem exudate collected from detopped plants were 14.5 mmol g-1 for Ca(NO3)2 treatments and 1.5 mmol g-1 for NH4OH treatments, indicating that contrasting N-form nutrition resulted from fertilizer treatments. Malate concentrations were higher in the NH4OH treatment indicating that this organic acid anion may substitute for the negative charge of nitrate during enhanced ammonium nutrition in maize.The results suggest that potentially useful genetic variation exists in maize for N form preference and for tolerance to increasing ammonical-N rates.  相似文献
