Aloe vera dry rot caused by Alternaria alternata, is one of the most serious fungal diseases affecting the commercial cultivation of aloe vera in North India. Control of this disease through chemical is quite expensive and not ecofriendly. The plant extracts as biopesticdes act as a vital components for the management of this disease. Evaluation of some plant extracts was carried out against A. alternata in laboratory conditions. The extracts were prepared at 5% and 10% concentration and were evaluated through inhibition in radial growth (food poison technique) and spores (conidia) germination (hanging drop technique) against A. alternata. Neem leaf extract gave 58.6% inhibition in radial growth and 56.5% in spore germination at 10% concentration followed by Ocimum sanctum which was found effective and gave 54.7% inhibition in radial growth and 50.4% in spore germination over control. 相似文献

芦荟(Aloe)是美容和医疗保健工业的重要植物资源,然而基因工程途径改良芦荟鲜有报道。本实验研究了次氯酸钠、升汞不同浓度和时间对芦荟外植体的灭菌效果,比较了芦荟不同部位(叶片、叶鞘和茎段)的再生能力,利用GUS基因瞬间表达技术(X-Gluc染色)探讨了不同农杆菌对不同芦荟外植体的侵染效果,确定了G418筛选剂在芦荟转化后的最适筛选浓度,摸索了适宜于农杆菌—芦荟共培养用培养基组成和共培养条件,通过转化、筛选和移栽共获得了67棵抗性再生植株,进一步对抗性再生植株进行了PCR、Southern blotting和ELISA检测。结果表明,芦荟外植体灭菌方法为20.0%次氯酸钠溶液浸泡25min,效果优于利用0.1%升汞灭菌处理,茎部切段的再生能力高于叶片切段和叶鞘切段,适宜的共培养条件为芦荟外植体浸泡在含有农杆菌的液体共培养基中半小时后,在无菌滤纸上、24℃、10h光照共培养3天;EHA105农杆菌菌系对芦荟茎部细胞的侵染能力明显强于C58C1,EHA105侵染后GUS基因瞬时表达率达到了80.0%左右,而C58C1侵染后GUS基因瞬时表达率只有30.0%左右。G418用于筛选抗性再生芽和抗性植株的适宜浓度为10.0~25.0mg/L。PCR和Southern blotting检测证实外源基因已成功整合到芦荟基因组中,转化效率为0.9%,单拷贝整合占80.0%,2~3拷贝整合占20.0%,ELISA检测证明外源基因已在转基因芦荟中稳定表达。综上所述,初步建立了农杆菌介导转化芦荟的技术体系,为利用基因工程途径改良芦荟奠定了基础。 相似文献

用不同浓度NaCl和等渗聚乙二醇(PEG 6000)处理芦荟(Aloe vera L.)幼苗,10 d后测定叶片相对生长速率和厚度、叶片中主要有机溶质、无机离子含量及渗透调节能力.结果表明,-0.44、-0.88 MPa NaCl和PEG处理使芦荟叶片的相对生长速率和叶片厚度明显下降,且盐胁迫对幼苗生长的抑制和叶片含水量降低的效应明显高于等渗的水分胁迫,其叶片渗透调节能力随处理渗透势的降低而增加, -0.88 MPa PEG胁迫的芦荟幼苗的渗透调节能力高于等渗盐分胁迫.在主要渗透调节物质可溶性糖、有机酸、K 、Ca2 和Cl-中,-0.88 MPa PEG处理下含量比相同渗透势的NaCl处理下显著增加的是有机溶质,因此推断有机溶质含量高是PEG胁迫下渗透调节能力较强的主要因素. 相似文献

研究了磷对海水胁迫下库拉索芦荟(A loe vera)幼苗干物质积累、植株含水量、在器官和组织水平上离子分布的影响。增磷显著缓解海水胁迫对芦荟生长的抑制,明显提高海水胁迫下芦荟幼苗的干物质积累和含水量。器官离子含量和X射线微区分析结果都表明,30%浓度海水胁迫下,外源磷水平的提高能显著降低芦荟幼苗根系N a 、C l-的吸收,增强K 、C a2 向地上部的运输和分配,从而维持叶片较高的K /N a 、C a2 /N a 比率,而这很可能是增磷提高芦荟对海水胁迫抗性的主要原因之一。 相似文献

提出了一种反相液相色谱法分析芦荟制品中有机酸的含量。采用C18 RP为色谱分离柱,磷酸盐溶液与乙腈作流动相;芦荟制品经简单处理后直接进行分离定量,在10min内把其中的苹果酸、乳酸和富马酸等完全分离定量,各种酸的回收率均大于98%。经多次实验证明:该方法是测定芦荟制品中有机酸的快速、准确和有效的定量方法。 相似文献

目的:对传统上用于治疗作用的6个不同品种芦荟的蒽甙(蒽衍生物)进行了提取,并进行了抑菌作用的比较研究。方法:琼脂扩散法。结果:从叶皮中提取的有效成分表现出不同程度的抑菌活性,而叶肉提取物几乎没有抑菌效果。其中,龙山芦荟,皂质芦荟和木立芦荟的叶皮提取物对革兰阴性菌和革兰阳性菌均具有较强的抑菌能力,而且这种抑菌活性与蒽醌成分呈正相关性。 相似文献

Natural flavonoids are associated with anti-proliferation of cancer growth. However, the antioxidant and anti-proliferation effects of AE (aloe-emodin) have not been well studied. We have investigated how AE affects the proliferation of hepatic hepatocellular carcinoma cells and exerts an anti-cancer effect. The cytotoxic effect of AE was demonstrated using an MTT [3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl-2H-tetrazolium bromide] assay and Huh-7 cells were inhibited by AE treatment in both dose- and time-dependent manners. The IC(50) level of AE was ~75 μM. AE also has anti-proliferative effects via induction of DNA damage and apoptosis. 2-DE (two-dimensional electrophoresis) revealed that several proteins were related to the anti-cancer effects of AE. CAPN2 (calpain-2) and UBE3A (ubiquitin-protein ligase E3A), which are associated with the apoptosis signalling pathway, were verified by Western blotting. AE exhibited potent anti-proliferative effects on Huh-7 cells via down-regulation of CAPN2 and UBE3A. The findings support the possibility of AE being a chemopreventative agent. 相似文献

Introduction – Rhamnus alpinus L. (Rhamnaceae), a traditional plants in the flora of the Abruzzo region, is known to contain active anthraquinone secondary metabolites. However, the content of anthraquinones varies among R. alpinus samples depending on collection season and site. Thus, using simple, reliable and accurate analytical methods for the determination of anthraquinones in R. alpinus extracts allows comparative study of different methods of extraction. Objective – After a partial validation of an HPLC method for the simultaneous determination of five anthraquinones, aloe‐emodine, rheine, emodine, chrysophanol and physcione, in the bark of R. alpinus, we compared three different methods of extraction. Methodology – Anthraquinones were extracted from the bark of R. alpinus using different techniques (methanol maceration, ultrasonic and supercritical CO2 extraction). Separation and quantification of anthraquinones were accomplished using a reversed‐phase C18 column with the mobile phase of H2O–methanol (40 : 60, v/v, 1% formic acid) at a wavelength of 254 nm. The qualitative analyses were also achieved at wavelength of 435 nm. Results – All calibration curves were linear over the concentration range tested (10–200 mM) with the determination coefficients ≥0.991. The detection limits (S/N = 3) were 5 mM for each analytes. All five anthraquinones were found in the samples tested at concentrations reported in experimental data. Conclusion – The described HPLC method and optimised extraction procedure are simple, accurate and selective for separation and quantification of anthraquinones in the bark of R. alpinus and allow evaluation of the best extraction procedure between the tested assays. Using short hairpin RNA against p53, transient ectopic expression of wild-type p53 or mutant p53 (R248W or R175H), and a p53- and p21-dependent luciferase reporter assay, we demonstrated that growth arrest and apoptosis of FaDu (human pharyngeal squamous cell carcinoma), Hep3B (hepatoma), and MG-63 (osteosarcoma) cells induced by aloe-emodin (AE) are p53-independent. Co-immunoprecipitation and small interfering RNA (siRNA) studies demonstrated that AE caused S-phase cell cycle arrest by inducing the formation of cyclin A-Cdk2-p21 complexes through extracellular signal-regulated kinase (ERK) activation. Ectopic expression of Bcl-X(L) and siRNA-mediated Bax attenuation significantly inhibited apoptosis induced by AE. Cyclosporin A or the caspase-8 inhibitor Z-IETD-FMK blocked AE-induced loss of mitochondrial membrane potential and prevented increases in reactive oxygen species and Ca(++). Z-IETD-FMK inhibited AE-induced apoptosis, Bax expression, Bid cleavage, translocation of tBid to mitochondria, ERK phosphorylation, caspase-9 activation, and the release of cytochrome c, apoptosis-inducing factor (AIF), and endonuclease G from mitochondria. The stability of the mRNAs encoding caspase-8 and -10-associated RING proteins (CARPs) 1 and 2 was affected by AE, whereas CARP1 or 2 overexpression inhibited caspase-8 activation and apoptosis induced by AE. Collectively, our data indicate AE induces caspase-8-mediated activation of mitochondrial death pathways by decreasing the stability of CARP mRNAs in a p53-independent manner. 相似文献
