Two key issues in the application of plant-cell-culture technology to the production of valuable secondary metabolites are reviewed: the selection of cell lines with suitable genetic, biochemical and physiological characteristics; and the optimization of bioreactor environments. Although great progress has been made in recent years in the design, selection and optimization of bioreactor hardware, optimization of environmental factors such as medium components, light irradiation and O2 supply needs detailed investigations for each case. With a better understanding of plant cell metabolism and physiology, further developments in cultivation processes, such as process integration and on-line monitoring and control, can be expected in the near future.J.-J. The effects of variations in temperature, salinity and water movement on the laboratory growth and reproduction of a single clone of the bougainvilliid hydroid Clavopsella michaeli were examined. Unfavourable conditions resulted in reproducible oscillations of growth rate. They ranged from overall stimulatory effects (hormesis) to irreversible inhibition and shrinkage of colonies. Exposure to low concentrations (0.1–5 µg l–1) of copper and mercury ions produced similar responses, which were therefore regarded as non-specific. A control mechanism, dampening these growth rate oscillations, supported higher colonial tolerance, that is adaptation, to environmental stresses of low intensity. When the counteractive capacity of the control mechanism was exceeded, inhibition occurred. Higher frequencies of gonozooids were observed in stressed colonies and were interpreted as an adaptive response. 相似文献

This article summarises the way in which mycorrhizal infection of roots affects the mineral nutrition of plants and how the symbiosis may interact with the evaluation of efficiency of nutrient uptake and use by plants. A brief account of the processes of infection and the way they are affected by host genotype and environmental conditions is given and the relationships between this and mineral nutrition (especially phosphate nutrition) are outlined.The interactions between mycorrhizal infection and P efficiency are considered at two levels. Mycorrhizas may act as general modifiers of efficiency regardless of the extent to which the plants are infected and in some mycorrhiza-dependent plants infection may change the ranking of genotypes. The extent of infection is also under genetic control and shows considerable variability between genotypes in some species. This variation could be used in programs to select varieties in which infection is rapid and nutrient uptake from nutrient deficient or low input systems is, in consequence, increased. 相似文献

Records of crop damage caused by Bibionidae are reviewed together with evidence for their beneficial activities, natural enemies and control methods. Bibionids are sporadic and infrequent pests. Although grass and cereals are most vulnerable, a wide range of crops is attacked. Damage is most severe after grass or when plants are under stress. The presence of organic matter during oviposition increases the likelihood of damaging populations. Many natural enemies exist but their impact on adult or larval populations has not been quantified. Given the pattern of attacks and lack of approved insecticides, cultural control methods are more appropriate than chemical. The role of adult bibionids in pollination and of larvae involved in soil processes could be of greater importance than any damage caused by the group. 相似文献

Summary Genetic variability in the non-compound portion of the genomes of compound-chromosome (CC) strains intended for genetic control can be increased by the use of bridging strains which can be crossed to both CC and normal strains. Two bridging systems are described for chromosome-5 CC strains of Lucilia cuprina. The first system relies on the established viability and fertility of males trisomic for chromosome 5R. Males carrying the (5L.YL)23 half-translocation, a C(5R), and a normal chromosome 5 were crossed successfully to a CC strain and a normal strain. The second system uses a pair of reciprocal whole-arm 4;5 translocations to generate gametes disomic for 5R and nullosomic for 5L, which in combination with C(5L)-bearing gametes form viable near-euploid offspring with only small duplications and deficiencies. These offspring (C(5L); (4L.5R)357; (4R.5R)194; (4L.4R)) were crossed successfully with both CC and T(4;5)357/ + individuals. The latter were in turn crossed successfully with normal strains. The T(Y;5)23 system allows replacement of the non-CC genome with wild material more rapidly than the T(4;5)357/T(4;5)194 system, but unlike the latter does not allow replacement of the Y chromosome in the CC strain. Summary The 6-phosphogluconate dehydrogenase (6-PGD) zymogram phenotypes of wheat, rye and their aneuploid derivatives were determined. Two genes involved in the production of 6-PGD isozymes were located on chromosome arms CRL (4 RL) and FRL (6 RL) of Imperial rye. On the basis of differential interactions between wheat and rye chromosomes, evidence was obtained that genes located on chromosomes 6 A, 6 BL and 7 BL control 6-PGD isozyme activities in Chinese Spring wheat. The wheat and rye 6-PGD zymogram phenotypes were indicative of homoeologous relationships between rye chromosome 6 RL to wheat chromosomes of group 6, and rye chromosome 4 RL to wheat chromosomes of group 7. 相似文献

Phosphorus release rates from profundal sediments of Lake Constance (Obersee) have been determined in D.O., pH regulated sediment-water systems. Above 10% O2 saturation (> 1.2 ppm D. O.) and with pH as in situ, no net release could be found. Sedimentations of diatom sludge (Asterionella formosa) and carbonate-phosphate coprecipitate (CaCO3.CaHPO4) increased the release to 0.5 mg × m–2 × d–1 which, however, will not be relevant to the P balance in Obersee. The LDH isozymes were surveyed in bacterized cultures of syngens 1, 3, 12, and 13 of Paramecium caudatum by polyacrylamide gel electrophoresis. All the examined stocks of different syngens except one stock in syngen 3 had a single common LDH isozyme, and intra- and intersyngenic variation was not observed except for the one stock in syngen 3. Breeding data using the exceptional stock indicated that the LDH isozymes of P. caudatum are controlled by two codominant alleles at a single locus whose products aggregate randomly, forming a dimer. 相似文献

The increased iron content in livers from iron-loaded rats is almost exclusively confined to the mitochondria. The ten- to twenty-fold higher level of nonheme iron in such mitochondria decreases the respiratory control with pyruvate-malate, but not with 3-hydroxybutyrate or succinate as substrates, and has no effect on the capacity for phosphorylation and substrate oxidation. Iron-loaded mitochondria have a malondialdehyde level which is about three times higher than that of control mitochondria, even after repeated washings with bovine serum albumin and EDTA. This is suggestive of an on-going process of lipid oxidation presumably catalyzed by the accumulated iron. Differences between the present in vivo data and in vitro results obtained by others are discussed. 相似文献
