An intron-less phaseolin gene [15] was used to express phaseolin polypeptides in transgenic tobacco plants. The corresponding amounts of phaseolin immunoreactive polypeptides and mRNA were similar to those found in plants transformed with a bean genomic DNA sequence that encodes an identical -phaseolin subunit. These results justified the use of the intron-less gene for engineering of the phaseolin protein by oligonucleotide-directed mutagenesis. Each and both of the two Asn residues that serve as glycan acceptors in wild-type phaseolin were modified to prevent N-linked glycosylation. Wild-type (wti–) and mutant phaseolin glycoforms (dgly
1, dgly
2 and dgly
1,2) were localized to the protein body matrix by immunogold microscopy. Although quantitative slot-blot hybridization analysis showed similar levels of phaseolin mRNA in transgenic seed derived from all constructs, seed from the dgly
1 and dgly
2 mutations contained only 41% and 73% of that expressed from the wild-type control; even less (23%) was present in seed of plants transformed with the phaseolin dgly
1,2 gene. Additionally, the profile of 25–29 kDa processed peptides was different for each of the glycoforms, indicating that processing of the full-length phaseolin polypeptides was modified. Thus, although targeting of phaseolin to the protein body was not eliminated by removal of the glycan side-chains, decreased accumulation and stability of the full-length phaseolin protein in transgenic tobacco seed were evident.Abbreviations bp

Changed fire regimes and the introduction of rabbits, cats, foxes, and large exotic herbivores have driven widespread ecological catastrophe in Australian arid and semi‐arid zones, which encompass over two‐thirds of the continent. These threats have caused the highest global mammal extinction rates in the last 200 years, as well as significantly undermining social, economic, and cultural practices of Aboriginal peoples of this region. However, a new and potentially more serious threat is emerging. Buffel grass (Cenchrus ciliaris L.) is a globally significant invader now widespread across central Australia, but the threat this ecological transformer species poses to biodiversity, ecosystem function, and culture has received relatively little attention. Our analyses suggest threats from buffel grass in arid and semi‐arid areas of Australia are at least equivalent in magnitude to those posed by invasive animals and possibly higher, because unlike these more recognized threats, buffel has yet to occupy its potential distribution. Buffel infestation also increases the intensity and frequency of wildfires that affect biodiversity, cultural pursuits, and productivity. We compare the logistical and financial challenges of creating and maintaining areas free of buffel for the protection of biodiversity and cultural values, with the creation and maintenance of refuges from introduced mammals or from large‐scale fire in natural habitats. The scale and expense of projected buffel management costs highlight the urgent policy, research, and financing initiatives essential to safeguard threatened species, ecosystems, and cultural values of Aboriginal people in central Australia. 相似文献

Background and AimsPlants depend fundamentally on establishment from seed. However, protocols in trait-based ecology currently estimate seed size but not seed number. This can be rectified. For annuals, seed number should simply be a positive function of vegetative biomass and a negative function of seed size.MethodsUsing published values of comparative seed number as the ‘gold standard’ and a large functional database, comparative seed yield and number per plant and per m2 were predicted by multiple regression. Subsequently, ecological variation in each was explored for English and Spanish habitats, newly calculated C-S-R strategies and changed abundance in the British flora.Key ResultsAs predicted, comparative seed mass yield per plant was consistently a positive function of plant size and competitive ability, and largely independent of seed size. Regressions estimating comparative seed number included, additionally, seed size as a negative function. Relationships differed numerically between regions, habitats and C-S-R strategies. Moreover, some species differed in life history over their geographical range. Comparative seed yield per m2 was positively correlated with FAO crop yield, and increasing British annuals produced numerous seeds. Nevertheless, predicted values must be viewed as comparative rather than absolute: they varied according to the ‘gold standard’ predictor used. Moreover, regressions estimating comparative seed yield per m2 achieved low precision.ConclusionsFor the first time, estimates of comparative seed yield and number for >800 annuals and their predictor equations have been produced and the ecological importance of these regenerative traits has been illustrated. ‘Regenerative trait-based ecology’ remains in its infancy, with work needed on determinate vs. indeterminate flowering (‘bet-hedging’), C-S-R methodologies, phylogeny, comparative seed yield per m2 and changing life history. Aim Panzootic chytridiomycosis caused by the chytrid fungus Batrachochytrium dendrobatidis (Bd) is the proximate cause of rapid amphibian declines across diverse biomes. While the origin of Bd remains unclear, increasingly the global trade in amphibians is associated with the spread of the infection. Global samples of Bd genotypes from previously unsampled regions are essential to test this hypothesis. In this paper, we present a study of the prevalence and phylogeny of Bd in both invasive and native amphibian species in markets and in the wild in ten provinces of China. Location China. Method We used a nested PCR assay to amplify the ribosomal internal transcribed spacer region of Bd followed by sequencing. Result Our results showed 246 of 2734 amphibians testing positive for Bd, with 157 positive samples in the wild (7.6%) and 89 in markets (13.5%). 30 haplotypes of Bd were identified, including 20 first detections. Introduced Lithobates catesbeianus had the highest prevalence of infection and the largest number of Bd haplotypes in both the wild and markets. Phylogenetic analysis based on 73 haplotypes (57 from Asia and 16 from other continents) showed that a unique, well‐supported, basal haplotype is present in Asia. Phylogeographical analyses revealed that some geographical structure exists amongst a subset of global haplotypes. Main conclusions Strains of the basal haplotype infected Babina pleuraden, an amphibian that is endemic to China, and Andrias japonicus, endemic to Japan, showing that Southeast Asia harbours a novel endemic lineage of amphibian‐associated Bd. Our data suggest that Bd in Asia pre‐dates the expansion of a globalized lineage of Bd, a finding that is indicative of a broader association of amphibians and chytrids than has previously been recognized. More genetic data from Bd isolates are needed to reveal the phylogenetic relationship of Bd in China compared to that found elsewhere. 相似文献

Studies were undertaken on interactions between panicle size, insect density, host plant, and the environment for sorghum
head bug,Calocoris angustatus Lethiery on five sorghum genotypes in terms of bug population increase, grain damage and loss in grain mass across four panicle
sizes (5, 10 or 20 branches/panicle and whole panicle), and three infestation levels (5, 10 and 15 pairs of bugs/panicle).
Head bug numbers increased and grain damage decreased with an increase in panicle size in the head bug susceptible cultivars,
CSH 1 and CSH 5. However, the increase in bug numbers or decrease in grain damage was not significant in head bug resistant
genotypes, IS 17610 and IS 17645. Head bug numbers increased with an increase in infestation level in CSH 1 and CSH 5, however,
such an increase was not substantial in IS 17610 and IS 17645. Grain damage was significantly lower in IS 17610 and IS 17645
compared with CSH 1 and CSH 5 across infestation levels. Head bug population increased at a greater rate during the rainy
season compared with the dry season. Panicle size and infestation levels accounted for greater variation in grain damage and
percentage loss in grain mass during the rainy season than in the dry season. To identify reliable sources of resistance to
insects, it is important to study insect host plant-interactions across panicle sizes (levels of food availability), infestation

Insect oviposition on plants is widespread across many systems, but studies on the response of host plants to oviposition
damage are lacking. Although patterns of oviposition vary spatially and temporally, ovipositing insects that exhibit outbreak
characteristics may have strong effects on host plants during peak abundance. Periodical cicadas (Magicicada spp.), in particular, may reduce the performance of host plants when they synchronously emerge in massive numbers to mate
and oviposit on host plants. Here we provide the first experimental manipulation of host plant use by periodical cicadas to
evaluate the impact of cicada oviposition on plant performance across a diversity of host species within an ecologically relevant
setting. Using a randomized block design, we established a plantation of three native and three exotic host plant species
common to the successional forests in which cicadas occur. During the emergence of Brood X in 2004, we employed a highly effective
cicada exclusion treatment by netting half of the host plants within each block. We assessed multiple measures of host plant
performance, including overall plant growth and the growth and reproduction of individual branches, across three growing seasons.
Despite our thorough assessment of potential host plant responses to oviposition damage, cicada oviposition did not generally
inhibit host plant performance. Oviposition densities on unnetted host plants were comparable to levels documented in other
studies, reinforcing the ecological relevance of our results, which indicate that cicada oviposition damage did not generally
reduce the performance of native or exotic host plants. 相似文献
