We isolated eleven strains of the harmful algal bloom (HAB)-forming dinoflagellate Karlodinium veneficum during a bloom event in the NW Mediterranean coastal waters and we studied the inter-strain variability in several of their physiological and biochemical traits. These included autotrophic growth parameters, feeding capabilities (mixotrophy), lipid composition, and, in some cases, their responses to biotic and abiotic factors. The strains were found to differ in their growth rates (0.27–0.53 d−1) and in the maximum cell concentrations achieved during stationary phase (6.1 × 104–8.6 × 104 cells mL−1). Their ingestion performance, when offered Rhodomonas salina as prey, was also diverse (0.22–1.3 cells per K. veneficum per day; 8–52% of their daily ration). At least two strains survived for several months under strict heterotrophic conditions (no light, low inorganic nutrients availability, and R. salina as food source). These strains also showed very distinct fatty acid compositions, with very low contents of monounsaturated and polyunsaturated fatty acids. According to a Bray Curtis similarity analysis, three or four strain groups able to perform different roles in bloom development were identified. We further analyzed one strain from each of the two most distinct groups with respect to prey concentration, light intensity, nutrient availability, and we determined the functional responses (growth and feeding rates) to food concentration. Taken together, the results served to highlight the role of mixotrophy and clone variability in the formation of HABs.  相似文献

Mt. St. Helens, a volcanic peak in the Cascade Range in southern Washington erupted violently on May 18, 1980, causing enormous damage to both terrestrial and aquatic ecosystems. The initial explosion evaporated, scoured or buried all springs and streams in the blast impact area. Ash fall and erosion from defoliated hillsides subsequently filled most of the lotic habitats with organic debris and volcanic ash. Recolonization of springs and streams by algae occurred quickly in areas where erosion through the ash progressed down to bedrock. Within 15 months or less of the eruption, algal communities were established throughout the blast impact area. However, as a result of the initial and continued disturbance these communities remained in an early successional stage. Floral assemblages were highly variable except that they were composed mostly of diatoms, with Achnanthes minutissima dominating most lotic sites. Springs showed the most rapid development toward stable floras.  相似文献

The species (cytospecies) structure and seasonal dynamics of a thermophilic bluegreen algal/bacterial community from Mimbres, New Mexico are described. Using stereological techniques, several common quantitative ecological parameters (biomass, density, distribution measurements, Species Diversity Indices, importance values) were described for the community growing ca. 1 m beneath the water surface on the inner walls of a concrete cistern. Temperature and pH were relatively constant while water flow rates, duration and intensity of light showed seasonal variation. The total volume of the mat occupied by living organisms did not exceed 2% during the entire year. The remaining volume was occupied by gas and water bubble chambers, gelatinous matrix, empty tubes, mineral deposits, and water. The total biomass remained constant except during a period of severe, seasonal light reduction (November). Species Diversity Indices, density and importance values for the bluegreen algal and bacterial populations showed seasonal changes. The three dominant bluegreen algal cytospecies which represented the greatest amount of biomass alternated in their abundance on a seasonal basis while the three dominant bacterial cytospecies remained the same throughout the period of study. Although considerable season change occurred in the species structure of this community, the total living volume remained relatively constant with the major changes in the bluegreen algal populations. Many of the seasonal changes observed appear to correlate with seasonal changes in light intensity and duration.  相似文献

The annual and lunar reproductive cycle of the widely distributed edible sea urchin Tripneustes gratilla (L) was examined through measurements of gonad index, histological examination of gametogenesis, and induction of spawning with KCl injections. The population density and morphological characteristics of urchins at Diani, Kanamai, and Vipingo reef lagoons were also studied as well as the effects of seawater temperature and light on reproduction. Gonad growth started early during the northeast monsoon and reached a peak in June at the beginning of the southeast monsoon followed by a sharp decrease in gonad size of 50% in July and August towards the end of the southeast monsoons. Histological examination of gonads, revealed many different stages of gametogenesis with gametes present throughout the year, indicating continuous reproduction. There was a significant relationship between gonad index and lunar day with spawning occurring between lunar day 7 and 21, but spawning was not in perfect synchrony in the population. The population density of urchins at each reef is variable from year to year and was highest on average at Vipingo. Urchins at Kanamai had the lowest gonad indices, the largest jaws and smallest individuals an indication of food limitation. The gonads (roe) of T. gratilla at all three sites, were perpetually ‘runny’ an attribute that is not suitable for urchin fisheries. Studies to develop techniques to improve roe quality are recommended.  相似文献

A total number of 40 potentially toxic microalgae and 5 taxa causing discolorations have been identified along the coasts of the Campania region (South Tyrrhenian Sea, Mediterranean Sea). This number is based on results of over 20 years of research at a coastal station in the Gulf of Naples, 4 years of monitoring activity along the Campania coasts and of a series of scattered and sporadic observations. Several species of the recently erected genera Karenia and Takayama are reported for the first time in the area. Information on the period of highest abundances or most probable period of occurrence indicates the late spring and summer as the periods of maximum risk of harmful events. Despite the variety of potentially toxic species, no human health problems nor fish kills have ever been recorded in Campania. The reasons for this apparent paradox are probably to be found in the ecological factors that regulate the abundance, toxicity and spatial distribution of the potentially harmful species and reflect as well the relatively low number of aquaculture plants in the area.  相似文献

The carbonic anhydrase (CA) activities were determined in three cyanobacterial species, namely Microcystis aeruginosa Kütz., Microcystis viridis (A.Br.) Lemm, and Microcystis wesenbergii (Kom.) Kom, which were dominant in a lake (Dianchi Lake) subject to major blooms. In more detailed experiments on M. aeruginosa, the effects of inorganic carbon, pH, temperature, nitrogen/phosphorus ratio, glucose, and light intensity on CA activity were also investigated. Because of the relatively alkaline pH value of the culture media for the optimum growth of algal cells, bicarbonate ions were the main form of exogenous inorganic carbon. The results showed that the CA activity of M. aeruginosa was influenced dramatically by the concentration of bicarbonate. Consequently, it was suggested that bicarbonate ions were the main form of exogenous inorganic carbon that M. aeruginosa could utilize. Cultures grown in the dark exhibited CA activity six times higher than that of cells cultured mixotrophically with the addition of glucose. Features of eutrophic water bodies promoted an increase in CA activity, and the resulting higher CA activity would accelerate the utilization of inorganic carbon and favor the growth and blooming of Microcystis spp. in eutrophic lakes. Although the experiments were carried out under controlled experimental conditions, they could provide some basic data that would prove useful for the control of cyanobacterial blooms in nature.  相似文献

Simple sequence repeats (SSRs) composed of extensive tandem iterations of a single nucleotide or a short oligonucleotide are rare in most bacterial genomes, but they are common among Mycoplasma. Some of these repeats act as contingency loci in association with families of surface antigens. By contraction or expansion during replication, these SSRs increase genetic variance of the population and facilitate avoidance of the immune response of the host. Occurrence and distribution of SSRs are analyzed in complete genomes of 11 Mycoplasma and 3 related Mollicutes in order to gain insights into functional and evolutionary diversity of the SSRs in Mycoplasma. The results revealed an unexpected variety of SSRs with respect to their distribution and composition and suggest that it is unlikely that all SSRs function as contingency loci or recombination hot spots. Various types of SSRs are most abundant in Mycoplasma hyopneumoniae, whereas Mycoplasma penetrans, Mycoplasma mobile, and Mycoplasma synoviae do not contain unusually long SSRs. Mycoplasma hyopneumoniae and Mycoplasma pulmonis feature abundant short adenine and thymine runs periodically spaced at 11 and 12 bp, respectively, which likely affect the supercoiling propensities of the DNA molecule. Physiological roles of long adenine and thymine runs in M. hyopneumoniae appear independent of location upstream or downstream of genes, unlike contingency loci that are typically located in protein-coding regions or upstream regulatory regions. Comparisons among 3 M. hyopneumoniae strains suggest that the adenine and thymine runs are rarely involved in genome rearrangements. The results indicate that the SSRs in the Mycoplasma genomes play diverse roles, including modulating gene expression as contingency loci, facilitating genome rearrangements via recombination, affecting protein structure and possibly protein-protein interactions, and contributing to the organization of the DNA molecule in the cell.  相似文献

Australian science has made rapid advances in the last decade in understanding eutrophication processes in inland waters and estuaries. The freshwater research on which these advances are based was triggered by well-publicised blooms of cyanobacteria during the 1980s and early 1990s, particularly a 1000 km long bloom on the Darling River. In estuaries the study which greatly enhanced our understanding but simultaneously served to stimulate further research into estuarine eutrophication, the Port Phillip Bay Study, was initially designed to address perceived problems of toxicants in the Bay but provided profound insights into drivers for, and ecosystem responses to, eutrophication. Subsequent estuarine research has largely been stimulated by management questions arising from Australia’s increasing coastal development for residential purposes. The research has shown that some of the beliefs extant at the time of the blooms were incorrect. For example, it is now clear that stratification and light penetration, not nutrient availability, are the triggers for blooms in the impounded rivers of southeastern Australia, although nutrient exhaustion limits the biomass of blooms. Again, nitrogen seems to play as important a role as phosphorus does in controlling the biomass of these freshwater blooms. The research has also shown that aspects of eutrophication, such as nutrient transport, are dominated by different processes in different parts of Australia. Many of the biophysical processes involved in eutrophication have now been quantified sufficiently for models to be developed of such processes as sediment-nutrient release, stratification, turbidity and algal growth in both freshwater and estuarine systems. In some cases the models are reliable enough for the knowledge gained in particular waterbodies to be applied elsewhere. Thus, there is now a firm scientific foundation for managers to rely upon when managing algal blooms. Whilst these findings have already been presented to managers and communities throughout Australia, there is still a considerable way to go before they are absorbed into their modus operandi.  相似文献

The performance of microflotation, dispersed air flotation with microbubble clouds with bubble size about 50 μm, for algae separation using fluidic oscillation for microbubble generation is investigated. This fluidic oscillator converts continuous air supply into oscillatory flow with a regular frequency to generate bubbles of the scale of the exit pore. Bubble characterization results showed that average bubble size generated under oscillatory air flow state was 86 μm, approximately twice the size of the diffuser pore size of 38 μm. In contrast, continuous air flow at the same rate through the same diffusers yielded an average bubble size of 1,059 μm, 28 times larger than the pore size. Following microbubble generation, the separation of algal cells under fluidic oscillator generated microbubbles was investigated by varying metallic coagulant types, concentration and pH. Best performances were recorded at the highest coagulant dose (150 mg/L) applied under acidic conditions (pH 5). Amongst the three metallic coagulants studied, ferric chloride yielded the overall best result of 99.2% under the optimum conditions followed closely by ferric sulfate (98.1%) and aluminum sulfate with 95.2%. This compares well with conventional dissolved air flotation (DAF) benchmarks, but has a highly turbulent flow, whereas microflotation is laminar with several orders of magnitude lower energy density.  相似文献
