Extracellular release of dissolved organic compounds by the bluegreen algal community of a brackish marsh was studied using 14C techniques. Mannitol and trehalose were identified as the most commonly released compounds. The proportions of these two extracellular compounds varied in response to light intensity and the water potential of the environment. The presence of mannitol, in particular, suggests that excretion of organic compounds in natural situations is a function of osmotic adjustment.  相似文献

Eutrophication of Dutch lakes has led to massive algal growth, disappearance of most of the macrophytes and disturbance of the food chain. The pike population has fallen sharply and bream developed very strongly, in the absence of this predator. Eutrophication problems are primarily being tackled by reducing nutrient loading. Restoration of water quality, however, seems to be impeded by the present structure of the food chain,i.e. the large bream stock. Biomanipulation, especially fish stock control with the aim of reducing the bream stock and increasing that of predatory fish, can accelerate the process of restoration. For the further development of biomanipulation it is very important that water authorities and managers of fish stocks agree on a common strategy.  相似文献

Metallothioneins, MT's, are low-molecular-weight, cysteine-rich, polypeptides that complex ‘soft’ metal ions in thiol clusters. They are structurally diverse. Some MT's are gene products, while others are secondary metabolites. Two of the three classes of MT have been identified in algae. Eukaryotic algae possess the secondary metabolites referred to as class III MT. There is no unequivocal evidence that MT genes occur in eukaryotic algae. However, the products of MT genes have been identified in cyanobacteria. These genes and their metal regulatory elements remain to be isolated and characterized. MT's have attracted interest from researchers involved in a wide range of disciplines including bioinorganic chemistry, biochemistry, molecular biology, physiology, toxicology, environmental science and medicine. Although, the precise physiological roles of these polypeptides remain undefined, a large number of functions have been speculated. These molecules chelate toxic trace metals, such as Cd, thereby reducing the concentration of cytotoxic, free-metal ions. Furthermore, some MT's are believed to be involved in zinc and copper homoeostasis. Future studies should reveal whether or not some of the diversity of MT structure reflects a diversity of function.  相似文献

当蓝藻的类囊体膜用超声波进行破碎,并在4℃下用聚丙烯酰胺凝胶电泳进行分离,有6条叶绿素带被分离出来,它们分别是 CPIa,CPIb,CP1,CPa1 CPa2,FC。CP1 在红区和蓝区的吸收峰分别位于674和435 nm 处。在液氮甲该组分在725和680 nm 处有两个荧光发射带。CPa1和 CPa2的吸收光谱相似,其红峰和蓝峰的位置分别位于667和431.5nm 处。它们在77 K 的荧光发射峰都位于684 nm 处。用超声破碎法分离的叶绿素蛋白复合物的光谱特性,除 CPa1和 CPa2在红峰和蓝峰的吸收位置蓝移了3—5 nm 之外,其余与用 SDS 增溶法分离的相应复合物相似。属于光系统Ⅰ的 CPIa-CPI 的叶绿素含量占总叶绿素的40.93%,而属于光系统Ⅱ的 CPa1和 CPa2的叶绿素则占总叶绿素的38.78%,二者之差仅有2.15%。  相似文献

Studies demonstrating differences in chloroplast structure and biochemistry have been used to formulate hypotheses concerning the origin of algal plastids. Genetic and biochemical experiments indicate that significant variation occurs in ribulose-1,5-bisphosphate carboxylase (Rubisco) when supertaxa of eukaryotic algae are compared. These differences include variations in the organelle location of the genes and their arrangement, mechanism of Rubisco synthesis, polypeptide immunological reactivity and sequence, as well as efficacy of substrate (ribulose bisphosphate and CO2) binding and inhibitor (6-phosphogluconate) action. The structure-function relationships observed among chromophytic, rhodophytic, chlorophytic and prokaryotic Rubisco demonstrate that: (a) similarities among chromophytic and rhodophytic Rubisco exist in substrate/inhibitor binding and polypeptide sequence, (b) characteristic differences in enzyme kinetics and subunit polypeptide structure occur among chlorophytes, prokaryotes and chromophytes/rhodophytes, and (c) there is structural variability among chlorophytic plant small subunit polypeptides, in contrast to the conservation of this polypeptide in chromophytes and rhodophytes. Taxa-specific differences among algal Rubisco enzymes most likely reflect the evolutionary history of the plastid, the functional requirements of each polypeptide, and the consequences of encoding the large and small subunit genes in the same or different organelles.  相似文献

The green alga Chlamydomonas reinhardtii is a facultative heterotroph and, when cultured in the presence of acetate, will synthesize chlorophyll (Chl) and photosystem (PS) components in the dark. Analysis of the thylakoid membrane composition and function in dark grown C. reinhardtii revealed that photochemically competent PS II complexes were synthesized and assembled in the thylakoid membrane. These PS II centers were impaired in the electron-transport reaction from the primary-quinone electron acceptor, QA, to the secondary-quinone electron acceptor, QB (QB-nonreducing centers). Both complements of the PS II Chl a–b light harvesting antenna (LHC II-inner and LHC II-peripheral) were synthesized and assembled in the thylakoid membrane of dark grown C. reinhardtii cells. However, the LHC II-peripheral was energetically uncoupled from the PS II reaction center. Thus, PS II units in dark grown cells had a -type Chl antenna size with only 130 Chl (a and b) molecules (by definition, PS II units lack LHC II-peripheral). Illumination of dark grown C. reinhardtii caused pronounced changes in the organization and function of PS II. With a half-time of about 30 min, PS II centers were converted froma QB-nonreducing form in the dark, to a QB-reducing form in the light. Concomitant with this change, PS II units were energetically coupled with the LHC II-peripheral complement in the thylakoid membrane and were converted to a PS II form. The functional antenna of the latter contained more than 250 Chl(a+b) molecules. When excited by ultraviolet radiation, leaves of a great number of species of higher plants exhibit emission of blue fluorescence, comparable in intensity to the red emission of chlorophyll. The fluorescence decay of the blue emission of spinach leaves recorded by single photon counting techniques is decomposed into exponential components and it is shown that at least three different components are present. The lifetime of the three components does not show significant variations with the excitation or emission wavelengths. The excitation and emission spectra of each component were determined. The nature of the chemical compounds which cause this emission is discussed in relation to these spectra.  相似文献

A Pikea species attributed to Pikea californica Harvey has been established in England since at least 1967. Previously, this species was believed to occur only in Japan and Pacific North America. Comparative morphological studies on field-collected material and cultured isolates from England, California, and Japan and analysis of organellar DNA restriction fragment length polymorphisms, detected using labeled organellar DNA as a non-radioactive probe, showed that English Pikea is conspecific with P. californica from California. Both populations consist of dioecious gametophytes with heteromorphic life histories involving crustose tetrasporophytes; 96% of organellar DNA bands were shared between interoceanic samples. A second dioecious species of Pikea, P. pinnata Setchell in Collins, Holden et Setchell, grows sympatrically with P. californica near San Francisco but can be distinguished by softer texture, more regular branching pattern, and elongate cystocarpic axes. Pikea pinnata and P. californica samples shared 49–50% of organellar DNA bands, consistent with their being distinct species. Herbarium specimens of P. robusta Abbott resemble P. pinnata in some morphological features but axes are much wider; P. robusta may represent a further, strictly sub-tidal species but fertile material is unknown. Pikea thalli from Japan, previously attributed to P. californica and described here as Pikea yoshizakii sp. nov., are monoecious and show a strikingly different type of life history. After fertilization, gonimoblast filaments grow outward through the cortex and form tetrasporangial nemathecia; released tetraspores develop directly into erect thalli. Tetrasporoblastic life histories are characteristic of certain members of the Phyllophoraceae but were previously unknown in the Dumontiaceae. Japanese P. yoshizakii shared 55 and 56% of organellar DNA bands with P. californica and P. pinnata, respectively; phylogenetic analysis indicated equally distant relationships to both species. Pikea yoshizakii or a closely similar species with the same life history occurs in southern California and Mexico.  相似文献

Epilithic bacterial and algal biomass were compared among a run, riffle, and pool along an open-canopy section of a third-order, temperate stream. Epilithic biofilms were sampled after 3, 7, 14, 21, 28, and 35 days colonization on unglazed ceramic tiles that were attached to plastic trays (n = 3) placed across each of the three habitats (i.e., run, riffle, pool). The diverse habitats and sampling regime were selected to provide a range in algal biomass so that potential covariation between epilithic bacterial and algal biomass could be assessed. There were significant differences among habitats and among trays within each habitat for both chlorophyll a and AFDM. Chlorophyll a and AFDM increased in the run and pool throughout the colonization period. In the riffle, chlorophyll a and AFDM increased rapidly early in colonization, then decreased. Epilithic bacterial biomass increased rapidly with no significant differences among the three habitats throughout colonization. Further, bacterial biomass did not correlate with either chlorophyll a or AFDM in any of the three habitats or on any of the sampling days. These results suggest that epilithic algal and bacterial biomass may be regulated by independent controls in some stream environments.  相似文献
